1.Exercise Performance and Exercise-related Factors of Korean and Korean-Chinese Nursing Students: A comparative Study.

PURPOSE: The purpose of this study was to compare exercise performance with related factors between Korean and Korean-Chinese nursing students. METHODS: This study used a cross-sectional comparative survey method. One hundred sixty five Korean nursing students in Seoul, South Korea and 161 Korean-Chinese nursing students in Yanbian, China were recruited through convenience sampling. They were at the age of 19-26. Exercise performance and exercise-related factors were measured using the exercise performance degree scale and the scales of self-efficacy, perceived barriers, perceived benefits, and affection related to exercise. RESULTS: No significant difference in exercise performance was seen between the two groups. Self-efficacy and perceived benefits related to exercise performance were significantly higher in the Korean nursing students than in the Korean-Chinese nursing students. However, there were no significant differences in affection and perceived barriers related to exercise. CONCLUSION: We suggest that the reasons for the differences in self-efficacy and in perceived benefits between these two groups need to be explored though future studies. We suggest more studies comparing different groups having the same ethnicity as Korean but who were born into different societies to give answers regarding the influence of immigrated transition on health.

2.Gardenia jasminoides extract and its constituent, genipin, inhibit activation of CD3/CD28 co-stimulated CD4+ T cells via ORAI1 channel

Gardenia jasminoides (GJ) is a widely used herbal medicine with antiinflammatory properties, but its effects on the ORAI1 channel, which is important in generating intracellular calcium signaling for T cell activation, remain unknown. In this study, we investigated whether 70% ethanolic GJ extract (GJEtOH) and its subsequent fractions inhibit ORAI1 and determined which constituents contributed to this effect. Whole-cell patch clamp analysis revealed that GJEtOH (64.7% ± 3.83% inhibition at 0.1 mg/ml) and all its fractions showed inhibitory effects on the ORAI1 channel. Among the GJ fractions, the hexane fraction (GJHEX, 66.8% ± 9.95% at 0.1 mg/ml) had the most potent inhibitory effects in hORAI1-hSTIM1 co-transfected HEK293T cells. Chemical constituent analysis revealed that the strong ORAI1 inhibitory effect of GJHEX was due to linoleic acid, and in other fractions, we found that genipin inhibited ORAI1. Genipin significantly inhibited IORAI1 and interleukin-2 production in CD3/ CD28-stimulated Jurkat T lymphocytes by 35.9% ± 3.02% and 54.7% ± 1.32% at 30 μM, respectively. Furthermore, the same genipin concentration inhibited the proliferation of human primary CD4+ T lymphocytes stimulated with CD3/CD28 antibodies by 54.9% ± 8.22%, as evaluated by carboxyfluorescein succinimidyl ester assay. Our findings suggest that genipin may be one of the active components of GJ responsible for T cell suppression, which is partially mediated by activation of the ORAI1 channel. This study helps us understand the mechanisms of GJ in the treatment of inflammatory diseases.
3.Luteolin reduces fluid hypersecretion by inhibiting TMEM16A in interleukin-4 treated Calu-3 airway epithelial cells

Rhinorrhea in allergic rhinitis (AR) is characterized by the secretion of electrolytes in the nasal discharge. The secretion of Cl– and HCO3 – is mainly regulated by cystic fibrosis transmembrane conductance regulator (CFTR) or via the calciumactivated Cl– channel anoctamin-1 (ANO1) in nasal gland serous cells. Interleukin-4 (IL-4), which is crucial in the development of allergic inflammation, increases the expression and activity of ANO1 by stimulating histamine receptors. In this study, we investigated ANO1 as a potential therapeutic target for rhinorrhea in AR using an ANO1 inhibitor derived from a natural herb. Ethanolic extracts (30%) of Spirodela polyrhiza (SPEtOH) and its five major flavonoids constituents were prepared. To elucidate whether the activity of human ANO1 (hANO1) was modulated by SPEtOH and its chemical constituents, a patch clamp experiment was performed in hANO1-HEK293T cells. Luteolin, one of the major chemical constituents in SPEtOH, significantly inhibited hANO1 activity in hANO1-HEK293T cells. Further, SPEtOH and luteolin specifically inhibited the calcium-activated chloride current, but not CFTR current in human airway epithelial Calu-3 cells. Calu-3 cells were cultured to confluency on transwell inserts in the presence of IL-4 to measure the electrolyte transport by Ussing chamber. Luteolin also significantly inhibited the ATP-induced increase in electrolyte transport, which was increased in IL-4 sensitized Calu-3 cells. Our findings indicate that SPEtOH and luteolin may be suitable candidates for the prevention and treatment of allergic rhinitis. SPEtOH- and luteolin-mediated ANO1 regulation provides a basis for the development of novel approaches for the treatment of allergic rhinitis-induced rhinorrhea.

6.Preoperative CT Assessement of Esophageal Carcinoma: Comparison between the Patients with and without Recurrence of Esophageal Carcinoma after Surgical Resection.

PURPOSE: To determine whether preoperative CT is helpful in predicting the development of recurrent tumor following surgical resection in patients with esophageal cancer. MATERIALS AND METHODS: Thirty patients with esophageal cancer in whom preoperative CT of the chest had been performed were included in the study. All had undergone esophagectomy, esophagogastrostomy and lymph node dissection at our institution between 1995 and 1997. They were divided into two groups according to the development of tumor recurrence during the follow-up period of three years. Sixteen patients (group I) suffered tumor recurrence, while the other 14 (group II) remained tumor-free after surgery. In each group, a review of the preoperative CT scans indicated the length, thickness, location and margin of the tumor, and the presence or absence of lymphadenopathy in the mediastinum and/or upper abdomen. Differences in preoperative CT findings between the two groups were assessed by statistical testing. RESULTS: Lymphadenopathy of the mediastinum and/or upper abdomen was seen in 11 (69%) of 16 patients in group I and three (21%) of 14 in group II (p<.05). The tumor margin was indistinct in seven patients (44%) in group I and in one (7%) in group II (p<.05). The average length and thickness of esophageal tumors were 5.2 and 1.4 cm, respectively, in group I, and 3.5 and 1.5 cm, respectively, in group II (p>.05). In group I, five esophageal tumors were located in the middle esophagus and eleven in the lower esophagus. In group II, such tumor was located one in the upper esophagus, six in the middle esophagus, and seven in the lower esophagus (p>.05). CONCLUSION: Patients with preoperative CT findings of lymphadenopathy and/or an indistinct primary tumor margin are more likely to develop tumor recurrence following surgical resection than those without these findings.

7.Quantitative Evaluation of Infectivity Change of Cryptosporidium parvum after Gamma Irradiation.

Cryptosporidium parvum is a well-known waterborne and opportunistic intracellular protozoan parasite that causes diarrheal illness. In this study, we quantitatively investigated reduction of the infectivity of C. parvum after gamma irradiation and repair of the infectivity during incubation time after irradiation. C. parvum oocysts were subjected to gamma irradiation at various doses (1, 5, 10, and 25 kGy), and the in vitro infectivity was measured by real-time PCR every day up to 7 days after irradiation. The in vitro infectivity of C. parvum on human ileocecal adenocarcinoma cells (HCT-8) was effectively reduced (> 2 log(10)) by irradiation at 10 kGy or more. However, in the experiment to find out repair of the infectivity, recovery was not noted until day 7 post-incubation.

8.Differential Diagnosis of Tuberculous Pleural Effusion and Malignant Pleural Effusion: CT Accuracy and Findings.

PURPOSE: To evaluate the diagnostic accuracy of CT in the differential diagnosis of tuberculous and malignant pleural effusion whether or not lung lesions are present, and to investigate the CT findings used for this differential diagnosis. MATERIALS AND METHODS: This study involved 30 patients with tuberculous pleural effusion (mean age, 44.6 years; M:F=19:11) and 20 with malignant pleural effusion (mean age, 57.2 years; M:F=10:10). All 50 patients underwent enhanced CT chest scans, and the respective conditions were pathologically confirmed. Two radiologists unaware of the pathologic results and distribution of patients reviewed these scans retrospectively and independently. They recorded the presence or absence of helpful lung lesions, CT findings of pleural effusions, their diagnoses, and the degree of confidence of their diagnoses. RESULTS: Among the total of 100 answeres, helpful long lesions were found in 57 cases. Fifty-three of 57 diagnoses (93%) were correct and 26 cases (46%) were diagnosed with a high degree of confidence. Thirty-two of 43 cases (74%) without helpful lung lesions were correct and 11(26%) were diagnosed with a high degree of confidence. All diagnoses made with a high degree of confidence were correct, even in cases without helpful lung lesions. Frequent CT findings in tuberculous pleural effusion included diffuse pleural thickening, enhancement of pleura, deposition of extrapleural fat, and pleural calcification; in malignant pleural effusion, nodular pleural thickening, pleural thickening over 1cm and associated lymphadenopathy were frequent. Mediastinal and circumferential pleural, as well as fissural involvement, were seen in both effusions; there were no statistical differences. CONCLUSION: In most cases, CT provided correct differential diagnosis between tuberculous and malignant pleural effusion. It can help determine the nature of associated lung and pleural lesions, and specific findings of the latter, and can accurately differentiate tuberculous and malignant pleural effusion.

9.Mitochondrial dysfunction reduces the activity of KIR2.1 K⁺ channel in myoblasts via impaired oxidative phosphorylation.

Myoblast fusion depends on mitochondrial integrity and intracellular Ca²⁺ signaling regulated by various ion channels. In this study, we investigated the ionic currents associated with [Ca²⁺]i regulation in normal and mitochondrial DNA-depleted (ρ0) L6 myoblasts. The ρ0 myoblasts showed impaired myotube formation. The inwardly rectifying K⁺ current (I(Kir)) was largely decreased with reduced expression of KIR2.1, whereas the voltage-operated Ca²⁺ channel and Ca²⁺-activated K⁺ channel currents were intact. Sustained inhibition of mitochondrial electron transport by antimycin A treatment (24 h) also decreased the I(Kir). The ρ0 myoblasts showed depolarized resting membrane potential and higher basal [Ca²⁺]ᵢ. Our results demonstrated the specific downregulation of I(Kir) by dysfunctional mitochondria. The resultant depolarization and altered Ca²⁺ signaling might be associated with impaired myoblast fusion in ρ0 myoblasts.

10.Serologic and Genetic Characterization of Hantavirus from Apodemus Mice Captured in Kyonggi - do and Gangwon - do , 2001.

Hantaan virus is widely distributed in Korea and has been known to cause hemorrhagic fever with renal syndrome (HFRS). Hantaviruses are carried by numerous rodent species throughout the world. Especially, the striped field mice, Apodemus agrarius, is natural host for Hantaan virus in Korea. In this study, a total 105 wild rodents of 3 species (101 of Apodemus agrarius, 2 of Eothenomys regulus, and 2 of Mus musculus) were trapped in Kyonggi and Gangwon provinces for April to June, 2001 to study serologic and genetic characterization. 8 Apodemus agrarius (7.9%) were immunofluorescent antibody (IFA) positive against Hantaan virus and Hantaan virus genome was found in 5 among 8 seropositive rodents. S gene of isolated Hantaan virus genome was amplified and directly sequenced. Based on 917 bases of S gene (411-1327 bases), 2001 Korean isolates showed 94.8% to 95.5% nucleotide homologies in comparison with prototype Hantaan virus 76-118 which was isolated from Apodemus agrarius in Korea, 1976. The partial M gene (1969-2240 bases) showed 94.1% to 100.0% nucleotide homologies in comparison with 76-118 strain. In phylogenetic analysis, 2001 Korean isolates made the distinct cluster. Therefore, Hantaan viruses isolated in 2001 were not significantly chinged in genetic level comparison with previous isolate from Korea.
