Objective To investigate the association of cytotoxic T lymphocyte associated antigen 4 (CTLA 4) gene polymorphism with type 1diabetesmellitusandautoimmunethyroiddiseases. Methods The A/G phenotype at position 49 of the CTLA 4 gene exon 1 was determined by PCR RFLP method in 33 classic type 1 diabetes patients, 57 latent autoimmune diabetes in adults (LADA) patients, 122 autoimmune thyroid disease patients and 84 healthy control subjects of Chinese Han. Results The frequency of the CTLA 4 G phenotype was significantly higherintype1diabetespatientsthanthatincontrol subjects (55.6% vs 36.9%, P=0.0005). Objective To investigate the association of gene polymorphism of cytotoxic T lymphocyte-associated antigen 4(CTLA-4) with autoimmune thyroid diseases. Methods The A/G transition polymorphism at position 49(exon 1,codon 17) of the CTLA-4 gene was determined by polymerase chain reaction restriction fragment length polymorphism (PCR-RFLP)method in 122 autoimmune thyroid diseases patients which included 87 Graves’ disease (GD) patients and 35 Hashimoto's thyroiditis(HT) patients, as well as 84 control subjects. We detected their thyroid function by ELISA technique, and the thyroid autoimmune antibodies (TGAb,TPOAb) by indirect immunofluorescent technique. Results The strong association of the CTLA-4/G49 allele with AITDs was seen in our study(66.4% vs 36.9% P<0.0001). The G allele in GD patients was significantly increased compared with control subjects(69.5% vs 36.9%, P<0.0001). In HT patients, the frequency of G allele was also higher than control subjects(58.6% vs 36.9%,P<0.01), and there was no significant difference between HT and GD groups. When GD and HT subjects were stratified with respect to sex, neither female nor male patients demonstrated evident association of G49 allele with gender.Conclusions The polymorphism of CTLA-4 gene (exon 1 condon 17 position 49)confers susceptibility to AITDs. This association is independent of sex.

5.The correlation between leadership and nurse physician cooperation among nursing managers

Objective To investigate the current state of nurse leaders′leadership and nurse physician cooperation, to explore the relationship between them, and to provide reference for improving nurse physician cooperation situation. Methods Totally 193 nurse leaders were recruited who were participated in Anhui nursing management training courses. A series of questionnaires were used to collect data, including general information, competency inventory for registered nurse, nurse physician cooperation scale. Results One hundred and eighty- two questionnaires were valid. The score of nurse managers′leadership was (27.85±6.27) points,while the score of nurse physician cooperation scale was (68.31±11.67) points. Hierarchical regression analysis showed that the control of general information, leadership for the interpretation of the nurse physician cooperation level increased by 27.3% (△R2 = 0.273, P = 0.000). Conclusions Hospital managers can adopt effective strategies to raise the level of nurses' leadership, which further increases the nurse physician cooperation level.

7.Near-infrared diffuse correlation spectroscopy for early diagnosis of chronic periodontitis

Objective:To study the feasibility of the early diagnosis of chronic periodontitis using near-infrared diffuse correlation spectroscopy(DCS).Methods:30 subjects were inclded.The relative gingival blood flow(GBF) at the labial,mesial and distal healthy gingiva(the control) or the gingiva with periodontitis of maxillary anterior teeth was respectively measured by DCS before periodontal treatment,a week and a month after treatment.Results:The GBF of healthy gingiva group,gingivitis group and periodontitis group was 129.73 ± 10.70,95.51 ± 11.83 and 67.84 ± 13.05 respectively.Periodontal indexes were decreased and the relative GBF increased significantly 1 week and 1 month after treatment.Gingival index and sulcus bleeding index were respectively correlated to relative GBF(r =-0.902,r =-0.893).Conclusion:Near-infrared diffuse correlation spectroscopy can non-invasively detect the relative gingival blood flow in different stages of chronic periodontitis,it may be a method of early diagnosis of chronic periodontitis.
8.Determination of dexamethasone in human plasma by solid phase extraction with ultra performance liquid chromatography-tandem mass spectrometer

Objective To develop the analytical method to determine the content of dexamethasone in human plasma by solid phase extraction with ultra performance liquid chromatography-tandem mass spectrometer. Methods The human plasma was extracted with a solid phase extraction(SPE) and determined by UPLC-MS/MS. LC-MS/MS was performed in ESI source with MRM mode for quantification. Results The lowest detectable limit was 0.05ng/mL, the linear range was 1~100ng/mL. The absolute recovery was more than 78.1%. The intra- and inter day precision was within 15% at three concentrations. Conclusion Since the procedure proved to be simple, quick and effective, it could be used for the determination of dexamethasone in human plasma.
9.Effect of flurbiprofen axetil pretreatment on level of central β-endorphin in a rat model of incisional pain

Objective To evaluate the effect of flurbiprofen axetil pretreatment on the level of central β-endorphin in a rat model of incisional pain.Methods Fifty-four SPF male healthy Sprague-Dawley rats,aged 6-7 weeks,weighing 180-230 g,were divided into 3 groups (n=18 each) using a random number table:control group (group C),incisional pain group (group Ⅰ) and flurbiprofen axetil pretreatnent group (group FA).At 30 min before the model of incisional pain was established,fat emulsion 1 ml was injected via the caudal vein in group Ⅰ,and flurbiprofen axetil 6 mg/kg (diluted to 1 ml in fat emulsion) was injected via the caudal vein in group FA.The mechanical paw withdrawal threshold (MWT) was measured at 1 day before establishment of the model and 1,6 and 12 h after establishment of the model (T1-3).The rats were sacrificed after measurement of pain threshold at T1-3,and the lumbar enlargement segment of the spinal cord and hypothalamic arcuate nucleus specimens were obtained for determination of β-endorphin content (by enzyme-linked immunosorbent assay) and β-endorphin expression (by immunohistochemistry).Results Compared with group C,the MWT was significantly decreased at T1-3 in I and FA groups,the content and expression of β-endorphin in the spinal cord were significantly decreased at T2,3,and the content and expression of β-endorphin in the hypothalamic arcuate nucleus were increased at T1 in group Ⅰ,and the content and expression of β-endorphin in the spinal cord and hypothalamic arcuate nucleus were significantly increased at T1-3 in group FA (P<0.05).Compared with group Ⅰ,the MWT was significantly increased,and the content and expression of β-endorphin in the spinal cord and hypothalamic arcuate nucleus were increased at T1-3 in group FA (P<0.05).Conclusion The mechanism by which flurbiprofen axetil pretreatment produces analgesic effect may be related to the increased level of central β-endorphine in a rat modal of incisional pain.
10.Bacteriological examination of digital X-ray imaging device before and after disinfection

The digital X-ray imaging device was disinfected with trichloro-isocyanuric acid solution.The samples were taken with cotton swab pouring method before and 1,2,3 and 6 h after disinfection and the bacteriological examination was performed.Before disinfection the colony count of samples was all above the standard (> 10 cfu/cm2) with bacilli and gram-positive cocci predominantly.The highest count was detected in the X-ray photographic bed (58 cfu/cm2) and the lowest was in the exposure button(19 cfu/cm2).After disinfection the colony count in radiation suits still exceeded the standard,the colony count in diagnosis bed 2 h after disinfection,that in all parts of device 3 h after disinfection was off levels.The results indicate that the chlorine disinfectants can only achieve short-term effect in disinfection of digital X-ray imaging device.
