Chinese Journal of Clinical and Experimental Pathology 2015;(6):636-639
Purpose To assess the potentia1 of DNA image cytometry and urinary cyto1ogy in screening for urothe1ia1 carcinomas, and to find the idea1 method of ear1y diagnosis of urothe1ia1 cancer. Methods Voided urine specimens from 121 patients with urothe1ia1 carcinoma were random1y co11ected as test group and 95 patients with symptomatic uro1ogic disease but not urothe1ia1 carcinoma as con-tro1 group,a11 of the specimens were ana1yzed by both urinary cyto1ogy and DNA image cytometry. Results Urinary cyto1ogy had a higher sensitivity but 1ower specificity in diagnosis of urothe1ia1 carcinoma than that of DNA cytometry. Urinary cyto1ogy yie1ded an o-vera11 sensitivity of 85. 10%,specificity of 76. 80%. DNA cytometry revea1ed a sensitivity of 81. 80%,specificity of 81. 10%. Among the 121 urothe1ia1 carcinoma, there were 103 b1adder cancers,18 patients with upper urothe1ia1 carcinoma. For different types of urothe1ia1 carcinoma, both of the two methods demonstrated good sensitivity in the high 1eve1 tumor. And DNA cytometry had exce11ent sensitivity in the diagnosis of urothe1ia1 carcinoma in invasive b1adder cancer and upper urinary tract cancer, and both were more than 94%. Conclusions In 1ight of its high1y good sensitivity and specificity in urothe1ia1 carcinoma,especia11y in invasive b1adder cancer and upper urinary tract cancer,DNA cytometry shou1d be used to eva1uate suspect urothe1ia1 ce11s in urinary cyto1ogy specimens. Com-bined with the conventiona1 urinary cyto1ogy,the detection for urothe1ia1 carcinoma wou1d be significant1y improved.
4.Current incidence and management of gastrointestinal stromal tumors.
Chinese Journal of Gastrointestinal Surgery 2007;10(1):8-10
Gastrointestinal stromal tumor (GISTs) are a new recognized tumor entity. The incidence increases with time for the greater awareness of GISTs. Latest reports of the epidemiology of GISTs have been collected and sorted. Besides, it is still a controversy on the management of GISTs. While surgical en-bloc resection is the treatment of choice, imatinib is still optimal for patients with recurrent or metastatic GISTs. Preliminary promising effects have been showed on neo-adjuvant and post-operational adjuvant therapies. Imatinib-resistant GISTs become common and more targeted medication have been investigated and tested.

8.Preparation of a three-dimentional acellular cartilage matrix scaffold and its characteristics
Orthopedic Journal of China 2006;0(09):-
[Objective]To prepare a cartilage acellular matrix scaffold and to explore its feasibility in cartilage tissue engineering. [Methods]Microparticles about 100 ?m~154 ?m were prepared after calf cartilage physically shattered and experienced gradient centrifugation, and then treated by a modified Courtman's four-step method which was improved to produce acellular cartilage matrix. After this treatment the microparticles were made into 3% suspension which was placed into moulds. With the freeze-drying method,3-D cartilage acellular matrix (CACM) was prepared. The scaffolds were cross-linked by a neotype crosslinking agent genepin for 48h,and then placed into glycine solution server times for removing redundant genepin. The freeze-drying method was used to prepare CACM.The scaffolds were investigated by gross observation, histological staining (haematoxylin-eosin,toluidine blue) ,scanning electron microscope (SEM) observation and porosity measurement, water absorption rate and degradation rate analysis. After being cultivated for ten days, bone marrow stranal cells (BMSCs) of rabbit were seeded into the scaffold.MTT test and SEM were done to assess the growth and proliferation of BMSCs.[Results]Gross observation showed the scaffolds had a loosely porous and dark blue appearance after being cross-linked by genepin. The histological staining (haematoxylin-eosin,toluidine blue staining) showed that there were no chondrocyte fragments in the scaffold. The CACM scaffold had 90% porosity,(1314?337) % water absorption rate, and (13.69?7.3)% or (25.99?8.9) % degradation rate at 2 or 4 weeks.MTT test showed that BMSCs grew well in the 3-D CACM scaffolds of logarithmic trend, supporting that the scaffolds had no cytotoxic effect on BMSCs.SEM micrographs indicated that the scaffolds were porous and the cells covered the scaffolds firmly with cell processes.[Conclusion]The improved Courtman's four-step method makes a more thoroughly acellular scaffold. The 3-D CACM scaffold retains most of extracelluar matrix. After being cross-linked by genepin, the 3-D CACM scaffold has good biocompatibility and degradation rate of the scaffolds is decreased, which makes it a suitable carrier for cartilage tissue engineering.
9.Inhibitory effect of captopril on retinal neovascularization in mice
International Eye Science 2009;9(8):1448-1450
AIM: To study the inhibitory effect of captopril on retinal neovascularization (RNV).METHODS: Sixty seven-day-old mice were randomly divided into treated group and control group with thirty mice in each group. These mice were exposed to 750 50mL/L oxygen for 5 days and then to room air.The treated group had been injected captopril (2.7mL/kg), while control group had been injected 9g/L sodium chloride (2.7mL/kg) by intravitreal for 5 days. The mice were sacrificed at the 17th day after birth and the eyes were enucleated. Adenosine diphosphate-ase(ADPase) stained retina flat-mounts was performed to assess the retinal vascular profiles, Hematoxylin Eosin (HE)staining method was applied to count the number of new vascular cell nuclei and the expression of matrix metalloproteinase-2(MMP-2)and pigment epithelium derived factor (PEDF)was detected by immunohistochemical method.RESULTS: Comparing with control group, regular distributions and good branch and reduced density of RNV were observed in the treated group. The number of nucleus of new vessels vascular endothelial cells breaking through the internal limiting membrane was less in the treated group than in the control group (P<0.05). Stain of retinal MMP-2 was weaker in the treated group than in the control group and stain of retinal PEDF was stronger in the treated group than in the control group.CONCLUSION: Intravitreal injection of captopril (2.7mL/kg) may block the RNV in the oxygen-induced mouse model and may provide an effective method for prevent-ing RNV.
