Purpose To screen and identify T cell lymphoma invasion and metastasis 1 (Tiam1) related genes and expression profile during distant metastasis of colorectal cancer. Methods Colorectal cancer( CRC) in a mice model was established by intraperitoneal injection of dimethylhydrazine ( DMH) . Five panels of fresh primary tumor tissues from Tiam1 transgenic mice were collected and com-pared with that from wild type mice. Differentially expressed genes were detected by Affymetrix human genome-wide expression profile chip and verified by real-time quantitative PCR ( qRT-PCR) analysis. Results The genechip result showed that 794 genes were differ-entially expressed by at least 3 folds in Tiam1 transgenic mice with distant metastasis of colorectal cancer tissue, including 400 up-regu-lated and 394 down-regulated ones. Bioinformatics analysis and gene co-expression network building identified 3 genes (PIK3R5, FIGF and RPS6KA6) with specific expression in colorectal cancer tissue with distant metastasis, and this result was confirmed by qRT-PCR. Conclusion A specific Tiam1 gene expression profile related to colorectal cancer distant metastasis has been established through screening and identifying the genes related to Tiam1 by gene chip technique. These findings provide a basis for exploring the molecular markers of colorectal cancer with distant metastasis.

Journal of Zhejiang Chinese Medical University 2006;0(04):-
[Objective ] To evaluate the curative effect of urinary retention caused by hyperplasia of prostate gland after treatment of electric acupuncture to Ciliao and Zhongliao acupoints combined with ultrashort wave therapy.[Methods] 35 patients were treated by electric acupuncture to Ciliao and Zhongliao acupoints cooperating with ultrashort wave therapy.Residual urine volumn(RUV),frequency of urinary retention, the therapeutic effect and follow-up were recorded and statistically analyzed.[Results] 29 patients were able to urinate after the therapy, except for 6 patients who were still in urinary retention condition.Effective rate were up to 82.8%.[Conclusion] It proved that the less the frequency was,the better the clinical effect would be.However,no matter how frequent the retention was,our combination therapy still worked, and both effects of the near future and long-term were proved to be fine.Special therapeutic action from Shu acupoint, electric acupuncture's electric current effect, and physical therapy of ultrashort wave contributed to the outcome.
7.Effects of Candesartan on the Expression of Cerebral NAD(P) H Oxidase mRNA in Salt-loaded Stroke-Prone Spontaneously Hypertensive Rats
Chinese Journal of Hypertension 2007;0(03):-
Background Oxidative stress has been implicated in the pathogenesis of angiotensin Ⅱ(Ang Ⅱ)related hypertension.Superoxide anion, produced by NAD(P)H oxidase, plays important roles in hypertension and its complications.Ang Ⅱ infusion increases cerebral NAD(P)H oxidase activity, which is inhibited by angiotensin Ⅱ receptor blocker(ARB)candesartan and NAD(P)H oxidase inhibitor. However, the underlying mechanism of ARB in preventing hypertensive stroke is not elucidated clearly. Objective To investigate the effects of candesartan on the expression of cerebral NAD(P)H oxidase mRNA in salt-loaded stroke-prone spontaneously hypertensive rats(SHRsp). Methods Twelve-week-old salt-loaded SHRsp were treated with candesartan [1.0 mg/(kg?d)],trichlormethiazide [TCM,1.6 mg/(kg?d)] or vehicle(n=12 in each)for 2 weeks.Age-matched salt-loaded WKY rats were served as control(n=12).Blood pressure was measured every week. After two weeks, cerebrums were harvested and 24 h urine was collected.Urinary albumin was examined by ELISA.Cerebral cortex NAD(P)H oxidase subunits(p22phox,p47phox and gp91phox)mRNA expression were assayed by real-time PCR.Results The systolic blood pressure was increased significantly in salt-load SHRsp.Candesartan and TCM reduced SBP to the similar level.Urinary albumin excretion and cerebral cortex NAD(P)H oxidase subunits were markedly higher in salt-loaded SHRsp than those in salt-loaded WKY rats.Incidence of stroke was significantly reduced in candesartan treated group as compared with TCM treated SHRsp(P
8.Changes in the drug resistance of E.coli induced by fluoroquinolones in vitro
Journal of Medical Postgraduates 2003;0(06):-
Objective:The drug resistance of E.coli to fluoroquinolones is getting stronger in the recent years. This study aimed at the drug resistance of E.coli induced by fluoroquinolones in vitro,so as to provide a guidance for the rational use of antibiotics and effective control of nosocomial infections. Methods: Drug resistance was induced in vitro by a multi-step method in 10 strains of E.coli from clinical isolates with Ciprofloxacin,Levofloxacin and Gatifloxacin. The minimal inhibition concentration(MIC) of the E.coli was determined by the agar dilution method before and after the induction. Results: After in vitro induction, the E.coli acquired a high resistance(MIC ≥ 128 ?g/ml).And one drug could induce different degrees of resistance to the other two. Conclusion: The MIC of the E.coli induced by fluoroquinolones in vitro was increased by 8 to 8 205 folds compared with that before induction, which demonstrated a gradually developed resistance of E.coli strains to fluoroquinolones.

Chinese Journal of Anesthesiology 2010;30(9):1069-1073
Objective To investigate the effects of sufentanil postconditioning and sevoflurane postconditioning on myocardial ischemia/reperfusion (I/R) injury in isolated rat hearts. Methods Healthy male SD rats weighing 230-250 g were anesthetized with intraperitoneal 5% chloral hydrate 8 ml/kg. Their hearts were excised and perfused in a Langendorff apparatus with modified K-H solution saturated with 95% O2-5% CO2 at 37℃. Forty isolated rat hearts with I/R injury were randomly assigned into 4 groups ( n = 10 each): Ⅰ I/R group, Ⅱ sevoflurane postconditioning group, Ⅲ sulfentanil postconditioning group and Ⅳ postconditioning with sevoflurane and sulfentanil group. After 30 min of stablization, the hearts were subjected to 40 min of global ischemia followed by 120 min of reperfusion. Fifteen minutes of drug postconditioning were performed in group Ⅱ , Ⅲ and Ⅳ:3.0% sevoflurane was introduced into K-H solution in group Ⅱ , 100 nmol/L sulfentanil was added to K-H solution in group Ⅲ , sevoflurane postconditioning and sulfentanil postconditioning were performed simultaneously in group Ⅳ . Left ventricular systolic pressure (LVSP), left ventricular end-diastolic pressure ( LVEDP), left ventricular developed pressure (LVDP), maximum increase in rate of LVDP ( + dp/dtmax), maximum decrease in rate of LVDP ( - dp/dtmax), HR and coronary flow (CF) were measured at the end of 30 min stablization (baseline), and at 15, 30, 60, 90 and 120 min of reperfusion. Coronary effluent was collected at 5 min of reperfusion for determination of activities of creatine kinase (CK) and lactate dehydrogenase (LDH). Myocardial tissues were obtained at the end of reperfusion for determination of infarct size (by triphenyltetrazolium chloride staining) and expression of Bcl-2 and Bax (by Western blot). The ratio of Bcl-2/Bax was calculated. Results LVSP, LVDP, ± dp/dtmax,CF, Bcl-2 expression and ratio of Bcl-2/Bax were significantly higher, LVEDP, activities of CK and LDH and Bax expression were significantly lower, and the infarct size was smaller in group Ⅱ , Ⅲ and Ⅳ than in group Ⅰ (P < 0.05 or 0.01 ). There were no differences in the indices mentioned above between Ⅱ , Ⅲ and Ⅳ groups. Conclusion Sufentanil postconditioning can attenuate myocardial I/R injury, and there is no enhancement of myocardial protection when combined with sevoflurane postconditioning. The mechanism of myocardial protection is related to the inhibition of cell apoptosis via up-regulation of Bcl-2 expression and down-regulation Bax expression.
