1.Chemical constituents of Epimedium sutchuenense

Objective To study the chemical constituents of Epimedium sutchuenense.Methods The compounds were repeatedly separated and purified on column chromatography of silica gel,polyamide,Sephadex LH-20,and ODS.The structures were identified on the basis of spectral methods.Results Ten compounds were isolated from 70% ethanol extract of E.sutchuenense and were identified as sutchuenoside A(Ⅰ),sutchuenoside B(Ⅱ),daidzein(Ⅲ),baohuoside-Ⅰ(Ⅳ),kaempferide 3-rhamnoside(Ⅴ),icariin(Ⅵ),anhydroicaritin-3-O-?-L-rhamnopyranosyl(1→2)-?-L-rhamnopyranoside(Ⅶ),p-hydroxybenzoic acid(Ⅷ),daucosterol(Ⅸ),and ?-sitosterol(Ⅹ),respectively.Conclusion Compounds Ⅰ and Ⅱ are novel ones named as sutchuentins A and B respectively.Compounds Ⅲ,Ⅴ,Ⅶ,and Ⅷ-Ⅹ are isolated from this plant for the first time.
4.Non-iridoid constituents from roots of Gentiana straminea

Objective To study the non-iridoid constituents from the roots of Gentiana straminea.Methods The compounds were repeatedly separated and purified on column chromatography of silica gel,Sephadex LH-20,and ODS,and their structures were identified on the basis of spectral and chemical methods.Results Twelve compounds were isolated from 70% ethanol extract of G.straminea and were identified as(-)-syringaresinol-4,4′-bis-?-O-D-glucopyranoside(1),gentiaphyllide-D(2),erythrecentaurin(3),macrophyiioside D(4),gentiaphyllide-E(5),roburic acid(6),uvaol(7),?-sitosterol(8),daucosterol(9),gentianose(10),?-glucose(11),and ?-glucose(12),respectively.Conclusion Compounds 1-6 and 10 are isolated from this plant for the first time.

Antimicrobial peptides are a class of small peptides with anti-extrogenous pathogen activities.They are derived from organism and possess antibacterial,antifungus,antiviruses and anticancer cell actions.In recent years,it’s found that some microbial pathogens are able to resist antimicrobial peptides.The constitutive and inducible mechanism of a pathogen resists a given peptide were reviewed in this paper.
6. Characteristics of PLA2 activity and expression of PLA2 subtypes in rat pancreas after simple pancreatic trauma

Objective To study the characteristics of phospholipase A2 (PLA2) activity and expression of PLA2 subtypes in rat serum and pancreas tissue with an established simple pancreatic trauma model in rats. Methods One hundred and twenty Wistar rats were divided into two groups in a one-to-two ratio: control group and impact group. The two groups were subsequently divided into four subgroups at 6h, 24h, 72h, 7d time point. For the animals in the impact groups, the pancreas was injured by a BIM- III biotical impact machine, and the animals in control group were sham injured. The total catalytic activity of PLA2 in serum and pancreatic tissue, the expressions of mRNA and protein and immunohistochemical localization of secretory PLA2 (sPLA2), cytosolic PLA2 (cPLA2) and calcium independent PLA2 (iPLA2) in pancreatic tissue were determined after the corresponding pretreatments in each group. Results The total catalytic activity of PLA2 in serum and pancreatic tissue of impact groups increased at 6h after injury, and the PLA2 activity reached peak value at 24h in pancreatic tissue and at 72h in serum after trauma, and at 7d in serum and pancreatic tissue were still higher than that in control group at the same time (P<0.05). The results of RT-PCR, Western blotting and immunohistochemistry indicated that the expression of sPLA2 in pancreatic tissue was higher than that of cPLA2 and iPLA2. Conclusions PLA2 activity can be upregulated in serum and pancreas after pancreatic trauma. The expression of sPLA2 is increased most significantly in pancreatic tissue after pancreatic trauma. PLA2 activity may be detected in serum 6h-7d after pancreatic trauma, and is helpful for auxiliary diagnosis of simple pancreatic trauma.
7.Progressive Effects of Chronic Intermittent Hypoxia on Hippocampus Neuron and Growth Associated Protein-43 in Rats

Objective To explore the effect of chronic intermittent hypoxia (CIH) on hippocampus neuron and growth associated protein-43 (GAP-43) in rats. Methods 40 adult male Sprague-Dawley rats were randomly divided into control group, CIH 1 week group, CIH 3 weeks group and CIH 5 weeks group with 10 rats in each group. The CIH groups were exposed to intermittent hypoxia in designed cabin 8 hevery day for 1 week, 3 weeks and 5 weeks. The pathological changes of hippocampal neurons were observed by HE staining. The GAP-43 expression of hippocampus was detected by immunohistochemical staining. Results There was degeneration and necrosis of hippocampal neurons in the CIH rats, and it became worse gradually along with the hypoxia. Compared with the control group, the GAP-43 expression in hippocampus increased in CIH rats. The peak of GAP-43 expression appeared in the 1st week (P=0.038) and then it decreased in the 3rd week (P=0.382) and it was closed to the normal level in the 5th week (P=0.860). Conclusion Degeneration and necrosis of hippocampal neurons exacerbated gradually along with the hypoxia, while GAP-43 expression changed inconsistently.

9.Construction of the Expression Vector of Viruslike Particles Containing FMDV IRES RNA

The Coat protein and Maturase gene of E.coli bacteriophage MS2 was amplified by PCR,then the gene was cloned into pET32a to construct the intermediate vector pET32aCP.The conservative sequence of FMDV internal ribosome entry site(IRES) was cloned into the downstream of pET32aCP bacteriophage gene to construct the prokaryotic expression vector pCPES.The recombinant plasmid pCPES transformed into E.coli strain BL21(DE3) was induced to express with 1mmol/L IPTG.The expression products were purified by sucrose density gradient centrifugation.The expression products observed by TEM were circular viruslike particles,and the diameter of these particles was about 26nm.The stability of viruslike particles was detected,and the viruslike particles was identified by RTPCR.The results showed that the viruslike particles contain the FMDV IRES RNA and have good stability.The viruslike particles have great prospect as the standard and quality control in the area of RNA virus detection.
10.Effect of Point-of-care Hemoglobin/Hematocrit Devices and Autologous Blood Salvage on Reduction of Perioperative Allogeneic Blood Transfusion.

Objective To evaluate the effect of point-of-care hemoglobin/hematocrit (POC HGB/HCT) devices and intraoperative blood salvage on the amount of perioperative allogeneic blood transfusion and blood conservation in clinical practice.Methods A total of 46 378 medical records of 22 selected hospitals were reviewed. The volume of allogeneic red blood cell and plasma, number of patients transfused, number of intraoperative autologous blood salvage, total volume of autologous blood transfusion, and amount of surgery in the year of 2011 and 2013 were tracked. Paired t-test was used in intra-group comparison, while t-test of two isolated samples carried out in inter-group comparison. P<0.05 was defined as statistically significant difference.Results In the hospitals where POC HGB/HCT device was used (n=9), the average allogeneic blood transfusion volume per 100 surgical cases in 2013 was significantly lower than that in 2011 (39.86±20.20 vs. 30.49±17.50 Units, t=3.522, P=0.008). In the hospitals without POC HGB/HCT meter, the index was not significantly different between 2013 and 2011. The average allogeneic blood transfusion volume was significantly reduced in 2013 than in 2011 in the hospitals where intraoperative autologous blood salvage ratio [autologous transfusion volume/(autologous transfusion volume+allogeneic transfusion volume)] was increased (n=12, t=2.290, P=0.042). No significant difference of the above index was found in the hospitals whose autologous transfusion ratio did not grow.Conclusion Intraoperative usage of POC HGB/HCT devices and increasing autologous transfusion ratio could reduce perioperative allogeneic blood transfusion.
