1.Inhibitory Effect of N~G-Nitro-L-Arginine Methyl Ester on Growth of Colorectal Cancer Xenografts in Nude Mice

Objective To investigate the effect of N~G-nitro-L-arginine methyl ester (L-NAME), an inhibitor of nitric oxide synthase, on the growth of colorectal cancer xenografts in vivo and on tumor-associated neovascularization. Methods Twenty BALB/c nude mice were randomly divided into control group and study group equally. Human colorectal cancer cell line SL174T was inoculated subcutaneously into nude mice to form transplantation tumors. Saline 0.2 ml was intragastric-administrated to mice in control group and L-NAME (4 mg toties guoties) was administrated orally to mice in study group three times per week for four weeks. The changes of tumors in both groups were recorded and the microvessel density (MVD) was also measured by immunohistochemistry assay. Results L-NAME significantly inhibited the growth of colorectal cancer xenografts in nude mice. The weight of transplantation tumor reduced with the inhibitory rate of 41.36%, and the inhibitory rate of tumor volume was 43.48 % in study group. BACKGROUND:Nitrogen monoxide (NO) plays dual effects in the occurrence and development of tumor. High concentration of NO exerts anticancer effect through cellular cytotoxicity and inducing the apoptosis of tumor; under the changed microenvironment in tumor tissue, its effect on tumor tissue is mainly promoting the growth of tumor.OBJECTIVE: To study the effect of the nitric oxide synthase(NOS)inhibitor NG-nitro-L-arginine methyl ester(L-NAME) on invasion and metastasis of human colorectal cancer cell line LS174T.DESIGN: Single sample observationSETTING: Department of Surgery, Tumor Hospital affiliated to Harbin Medical University MATERIALS: The study was conducted in Cancer Research Institute of Harbin Medical University between January 2004 and October 2004. The human colorectal cancer cell line LS174T was purchased from Cancer Research Institute of Harbin Medical University. L-NAME was purchased from Sigma Company.METHODS: LS174T cells were treated with L-NAME of 0.2,0.4,0.8 and 1.0 mmol/L. Effect of drugs on invasion and migration of LS174T cells was observed in reconstructed basement membrane invading experiment.MAIN OUTCOME MEASRES: Inhibitory rate of L-NAME on LS174T line invading reconstructed basement membrane and the inhibitory rate of cellular migration.RESULTS: ①0.2, 0.4, 0.8, 1.0 mmoL/L L-NAME were used to treate LS174T cells for 72 hours, and the inhibitory rate in cell invading reconstructed basement membrane was 10.29%, 19.62%, 34.08%,42.23% respectively (P < 0.05 or 0.01 ). ② 0.2, 0.4, 0.8, 1.0 mmol/L L-NAME were used to treate LS174T cell for 72 hours , its inhibitory rate in inhibiting cells migration was 20.76%,24.95%,39.43%,46.85% respectively(P < 0.01 ).CONCLUSION: L-NAME has anti-invasive and anti-metastasic effects on LS174T cells.
3.Content Determination of the 2 Lignan Components in Mongolian Medicine Compound Nutmeg-5 by HPLC

OBJECTIVE:To establish a method for the determining contents of 2 lignan components[dehydrodiisoeugenol and 2,3-dihydro-7-methoxy-2-(3,4-methylened ioxyphenyl)-3-methyl-5-(E)-propenyl-benzofuran(referred to“lignanoid 2”)]. METH-ODS:HPLC method was adopted. The column was Elite C18 with the mobile phase of water-methanol(gradient elution)at the flow rate of 1.0 ml/min;the detection wavelength was 225 nm and the column temperature was 30 ℃. The sample size was 20 μl. RE-SULTS:There was a good linear relationship between sample quantity and the peak area in the range of 0.202-2.02 μg(r=0.999 9) and 0.204-2.04 μg(r=0.999 9)for 2 lignan components. The RSD of precision,stability and repeatability tests were less than 2%with the average recovery of 101.54%(RSD=0.60%,n=6)and 99.43%(RSD=1.09%,n=6). CONCLUSIONS:The method is simple,sensitive and accurate,and can be used for the quantization determination of dehydrodiisoeugenol and lignanoid 2 in nut-meg-5.
4.Expression of vascular cell adhesion molecule-1 in recurrent oral ulceration

Objective: To study the effects of soluble vascular endothelial cell adhesion molecule-1(sVCAM-1) and soluble intercellular adhesion molecule-1 (sICAM-1) in recurrent oral ulceration (ROU). Methods: sVCAM-1 and sICAM-1 in oral menbrane tissue sampls were measured with an enzyme linked immunosorbent assay using sandwich technices in 45 patients with ROU and 40 nomal controls. Results: The expression level of sVCAM-1 in the oral membrane tissue samples of ROU patients was higher than that in the controls (P0.05). Both sVCAM-1 and sICAM-1 expression were not significantly different with the stages (ROU-active and ROU-free) of the lesion. Conclusion: sVCAM-1 might play a role in ROU.
5.Cloning and characterization of Giardia intestinalis cyclophilin.

The cyclophilins (Cyps) are family members of proteins that exhibit peptidylprolyl cis-trans isomerase (PPIase, EC 5.2.1.8) activity and bind the immunosuppressive agent cyclosprin A (CsA) in varying degrees. During the process of random sequencing of a cDNA library made from Giardia intestinalis WB strain, the cyclophilin gene (gicyp 1) was isolated. An open reading frame of gicyp 1 gene was 576 nucleotides, which corresponded to a translation product of 176 amino acids (Gicyp 1). The identity with other Cyps was about 58-71%. The 13 residues that constituted the CsA binding site of human cyclophilin were also detected in the amino acid sequence of Gicyp 1, including tryptophan residue essential for the drug binding. The informatization of traditional Chinese medicine resources is the basis of modern medicine. With a spatial attribute traditional Chinese medicine resources could be carried out for in-depth spatial analysis, data mining and traditional Chinese medicine resources regional industrial layout. In this paper, we took the data of Glycyrrhiza uralensis in the third national Chinese medicine resources survey as the experimental data, described the principles and structure of traditional Chinese medicine resources spatial information database. We also described the establishment of analysis model with the help of this spatial database.

7.The effects of early rehabilitation therapy in mechanically ventilated patients

Objective To investigate the effects of early rehabilitation therapy in mechanically ventilated ICU in patients.Methods A randomized controlled trial was carried out.Sixty mechanically ventilated patients,with tracheal intubation or tracheostomy more than 48 h and less than 72 h,were admitted to the intensive care unit (ICU) of the Affiliated Hospital of Medical College Qingdao University from May 2010 to May 2012.These patients were randomly (random number) divided into two groups,the rehabilitation group and the control group,30 patients in each group.In rehabilitation group,rehabilitation therapy was taken twice daily,and the training time and intensity was adjusted according to the condition of the patients.Early rehabilitation therapy included head up actively,transfer from the supine to sit,out of bed,transfer to a chair,standing bedside bed and walking bedside bed.The patient' s body mass index,days to first out of bed,duration of mechanical ventilation,ICU stay,APACHE Ⅱ score,highest FiO2,lowest PaO2/FiO2 and hospital mortality of patients were compared between rehabilitation group and control group.The differences between the groups were compared using t test.Results There was no significant difference in body mass index,APACHE Ⅱ score,highest FiO2,lowest PaO2/FiO2 and hospital mortality between rehabilitation group and control group (P > 0.05).Patients in the rehabilitation group had shorter first out of bed time [(3.8 ± 1.2) d vs.(14.9 ±4.7) d; P =0.00],duration of mechanical ventilation [(5.6±2.1) dvs.(12.7±4.1) d; P=0.005] andICUstay [(7.3±2.8) dvs.(15.2±4.5) d;P =0.01] compared with control group.Conclusions Early rehabilitation therapy was safe and effective in improving the outcomes of mechanical ventilation patient.
8.Protective effect of ischemic preconditioning on diabetic rats with myocardial ischemia/reperfusion injury

Objective To investigate the protective effects of ischemic preconditioning on myocardial ischemia/reperfusion injury in diabetic rats.Methods Twenty-eight healthy male rats were injected streptozotocin at dose of 45 mg/kg by tail and be fed with normal diet for 4 weeks,then rats were randomly divided into iscbemia/reperfusion (I/R) group and ischemic preconditioning (IP) group.ST segment of electrocardiograph changes and arrhythmias of all rats were recorded before ischemia and 0,15,30 minuets after ischemia and 0.5,2 h after reperfusion.TTC staining was performed to determine myocardial infarct size.TUNEL assay was used to assesse cardiomyocyte apoptosis.The expression of antiapoptotic gene (Bcl-2) and proapoptotic (Bax) was determined by immunohistochemistry.Results Compared with I/R group,ST segment elevation of patients in IP group decrease from (0.675 ±0.150) mV to (0.489 ±0.161) mV at 30 min after ischemia(P <0.05).Meanwhile the onset of ventricular premature contraction(VPC) in IP group was (18.21 ± 5.36) min,later than that of control group ((6.47 ± 4.28) min,t =5.241,P =0.000).The duration of VPC was (6.07 ± 4.33) min,shorter than that of I/R group ((16.71 ± 5.48) min,t =4.924,P < 0.01)).The incidence of ventricular tachycardia (VT) and ventricular fibrillation (VF) of lP group remarkably decreased compared with I/R group (VT:57.14% (8/14) vs.14.29% (2/14),x2 =5.600,P =0.018 ; VF:50.00% (7/ 14) vs.14.29% (2/14),x2 =4.094,P=0.043).The myocardial infarct size in IP group was (12.50 ± 9.45) %,smaller than that of I/R group ((37.50 ± 11.40)%,t =3.211,P =0.006).Cardiomyocyte apoptotic index (AI) was attenuated in IP group than that of I/R group((24.31 ± 3.12)% vs.(19.01 ± 4.32)%,t =3.227,P =0.006),which was correlate with increased the ratio of Bcl-2/Bax((0.103 ±0.045) vs.(0.221 ±0.101),t =2.670,P =0.015).Conclusion IP treatment for diabetic rats shows a protect effect on myocardial I/R injury through attenuating myocardial apoptosis,and increasing the ratio of Bcl-2/Bax.
9.The effects of L-NAME on invasion and metastasis of human colorectal carcinoma cell line

Objective To study the effects of L-NAME on invasion and metastasis of human colorectal (carcinoma) cell line(LS-174T) and explore its possible mechanism.Methods The LS-174T cells were co-incubated with L-NAME at various concertrations.Griess techniqe was used to examine the effect of(L-NAME) on excretion of nitric oxide(NO) of the cells.The effects of L-NAME on invasion and migration of LS-174T were evaluated using Transwell chambers attached with polycarbonate filters and reconstituted(basement) membrane.Expression of MMP2 mRNA and TIMP2mRNA in LS-174T cells was measured by(RT-PCR).Results(1)L-NAME decreased the excretion of NO of the cells in a dose-dependent manner.(2)The ability of the L-NAME treated LS-174T cells to invade the reconstituted basement membrane(increased) significantly with the concentration and time,at the concentration of 0.2 mmol/L,0.4 mmol/L,0.8mmol/L and 1.0mmol/L,respectively,after 72 hour,the inhibition rates were 10.29%,19.62%,34.08% and 42.23%,respetively.(P

Wet age-related macular degeneration (AMD) is one of major causes of blindness in elder people.Intraocular injection of anti-vascular endothelial growth factor (VEGF) -A regimen has made big breakthrough for the treatment on choroidal neovascularization of wet AM D,while long-term follow-up and necessary retreatments are the key issues to remain obtained visual acuity.Multiple strategies of wet AMD have been used in following-up and retreating based on the visual acuity,optical coherence tomography (OCT),ophthalmoscope and fluorescein fundus angiography (FFA) in abroad.However,there also are major differences in the patient' s composition,treatment habits and distribution of medical sources in China from Western.So we suggest to standardize the follow-up and retreatment strategies about intravitreal injection of VEGF-A for wet AMD as to achieve a better effectiveness.OCT-guided individual follow-up and retreatment strategies should be very helpful for maintaining a long-term efficacy,minimizing the treatment time and reducing medical cost.
