1.Effects of acupuncture on the ratio of CD4+ CD25+ regulatory T cells and expression of transcription factor Foxp3 in patients with septic shock

Objective To study the effect of acupuncture on the ratio of CD4+ CD25+ regulatory T cells and expression of transcription factor Foxp3 in patients with septic shock.Methods Sixty-four patients with septic shock were randomly divided into two groups by using the random number table method.Acupuncture group (34 cases) was treated with both western medicine and acupuncture,and control group(30 cases) was treated with western medicine.The period of treatment was 7 days.After treatment,the ratio of CD4+ CD25+ T cells in peripheral blood was determined by flow cytometry.And the expression of Foxp3 mRNA in peripheral blood was detected by quantitative real time PCR.Results After treating for 7 days,the ratios of CD4+ CD25+ Treg cells and CD4+ CD25+ Foxp3 + Treg cells were (20.23 ± 1.12) % and (78.70 ± 7.65) % respectively in peripheral blood of the control group,which in the acupuncture groupwere (17.32 ± 0.78) % and (68.53 ± 8.01) %,the differences were statistically significant between the two groups(t =2.587,2.749,all P < 0.05).The levels of Foxp3 mRNA in peripheral blood were (1.21 ±0.02) and (1.02 ± 0.04) in the control group and acupuncture group,the difference was statistically significant(t =2.119,P < 0.05).Conclusion Acupuncture can adjust immune status of patients with septic shock by reducing the ratio of CD4+ CD25+ Treg cells and down-regulating the expression of Foxp3 mRNA.
2.Study on fingerprint chromatograms of extract of cimicifugae rhizoma.

OBJECTIVETo establish the fingerprint chromatograms of the extract of Cimicifugae Rhizoma firstly.
METHODPhenolic acids and triterpenoid saponins were analyzed by HPLC. Hypersil BDS C18 (4.6 mm x 250 mm, 5 microm) column was used, the mobile phase was composed of acetonitrile -0.1% H3PO4 with gradient elution, flow rate was 1.0 mL x min(-1), column temprature was 30 degrees C, and the detection wavelength was set at 316 nm and 210 nm.
RESULTIn the fingerprint of phenolic acids, thirteen feature peaks were found and the RSD of relative retention time and relative peak area were all less than 3% in the precision and repeation experiments. The similarity of ten batches of samples were all more than 0.90. In the fingerprint of triterpenoid saponins, fourteen feature peaks were found and the RSD of relative retention time and relative peak area were all less than 4% in the precision and repeation experiments. The similarity of ten batches of samples were all more than 0.90.
CONCLUSIONThis method is comprehensive, stable, reliable and can be used to evaluate the quality of the extract of Cimicifugae Rhizoma. It has provided a reference to the analysis on pharmacodynamic deferences of Cimicifuga extracts and also laid the foundation for its further development.

3.Development of speed indexes in student physical fitness tests

Abstract
Speed is an important component of physical fitness testing for students around the world. On the whole, the different testing systems usually set up various indexes based on the moving speed, action speed and reaction speed; the indexes are diverse but uniform in number; there are differences in the age of application of the indexes and no gender differences. As far as the classification of indexes is concerned, there are two types of movement:sprinting and shuttle run, for the moving speed test indexes; there are differences in the need for action speed and reaction speed tests. Based on this, from the perspective of energy metabolism and health benefits, the properties and testing methods for measuring moving speed and the need for action speed and reaction speed testing are analyzed. The study shows that acceleration capability should be a measured attribute of moving speed,the 30 m running can be used as a general index of moving speed for all students; the shuttle run is not suitable as an index of moving speed; action speed is less correlated with physical health, so it should not be included in the testing system for the time being; and reaction speed which closely relates to physical health,should be included in the testing system.
4.Study on rabbit mesenchymal stem cells differentiation to the adipogenic or osteogenic lineage in vitro.

Rabbit bone marrow-derived mesenchymal stem cells(MSCs) are multipotent. We studied the adipogenic and osteogenic differentiation potent using adipogenic supplement (AS) or osteogenic supplement (OS) in vitro. Specific markers of this induced adipogenic and osteogenic lineage were identified. The findings showed that the rabbit MSCs are capable of differentiating into adipogenic and osteogenic lineages spontaneously. On the 21st day, approximately 75% rabbit MSCs were induced to adipogenic or osteogenic cells in medium containing AS or OS, respectively. These results demonstrated that the differentiation of MSCs could be regulated in vitro. The underlying molecular mechanisms of adipogenic or osteogenic differentiation await elucidation.
