This study examined the roles of the initial level of muscle glycogen content and available substrate on glycogen repletion in muscle. The rats were randomly assigned to normal, starvation and exercise groups. The glycogen content of muscle was lowered by starvation and exercise for the purpose of this experiment. The normal rats remained sedentary in their cage without any restriction of food and water. The exercise and starvation groups were divided each group into two subgroups depending on the degree of stress, i.e. 16 and 64 hours starvation, and 30 minutes and 2 hours exercise loading. All experimental aninals sacrificed 9~10 O'clock in the morning. The glycogen content of gastrocnemius and liver were 0.416+0.0433 and 1.70+0.410gm/100gm wet tissue in normal rats, respectively. The glycogen content of gastrocnemius in stravaton groups was reduced to 83.5 and 75.5% of the values of normal groups by starvation for 16 and 64 hours, respectively. In exercise group, the content of glycogen was reduced to 63.7 and 49.8% of the normal group by 30 minutes and 2 hours exercise loading, respectively, After above exercise loading and forced starvation, glucose, 2.0gm/100gm body weight was ingested, and 2 hours later the glycogen content was determined to evaluate the role of initial level of muscle glycogen content on the repletion in gastrocnemius, and the different amount of glucose, 1.0, 1.5 and 2.0mg/100gm body weight, was given orally, and 2 hours later the glycogen content of gastrocnemius was determined to evaluate the role of available substrate on the glycogen repleted in muscle of the lowest initial glycogen content, and the larger the amount of glucose ingestion, the larger amount of glycogen repletion in muscle. The experiment demonstrates that the reducing level of muscle glycogen and increased amount of available substrate are the important factors for the acceleration of muscle glycogen repletion, and in the aspect of repletion of glycogen, the repletion rate of liver glycogen is 2~5 times faster than that of muscle, whereas there is no difference of repletion rate of liver glycogen between starvation and exercise groups.

Epoxy resins,which are irritant and allergen, are now used for surface coatings, electrical insulations, adhesives, and construction industry throughout the world. However, epoxy resin contact dermatitis has not been reported in the Korean literature. We report two cases of epoxy resin contact dermatitis which were occupationally related. They have been in the same work, painting dye and resin mixtures on the glasses for decoration. They showed positive patch test reactions to epoxy resin 1, and their own materials containing resins and accelerators which were proved to be -countinue-

BACKGROUND: It is a popular notion that cutaneous ageing includes two distinct phenomenon; true ageing, a universal presumably inevitable change attributable to the passage of time alone, and photoageing, changes attributable to chronic habitual sun exposure that are neither universal nor inevitable. Numerous investigations with experimental animals, in vitro skin models have been conducted, although, few histological studies to date have attempted to announce fundamental morphological changes with innate ageing. OBJECTIVE: We compared skin derived from the breast of old and young persons using light microscopy to discern structural changes in epidermal and dermal morphology with advancing age. METHODS: The histological, immunohistochemical studies were performed with normal skin sections of thirty donors who were diagnosed with breast cancer. They were classified into three age cohort groups; nine into group I (22 to 38), twelve into group II(40 to 52), and nine into group III(54 to 87). We chose the breast as an area that might closely resemble intrinsically aged skin. This region is relatively shielded from photoageing by its anatomical location. Analysis of data was performed using the Kruskal-Wallis and ANOVA test for dermal parameters based on a 5-point rating scale, and a simple regression test for a positive rate of immunoreactants. Results : 1. Light microscopic appearance of aged skin revealed a more flattened epidermis than young skin. There was no trend for an increase in epidermal melanin content per unit area on Fontana-Masson staining. There was an age-associated decrease in the Ki-67 positive rate(p<0.001), the density of Ki-67 positive cells declined approximately 1.16% per decade in photoprotected skin(p<0.001). The number of S-100 positive cells declined approximately 4.4/mm width along the dermo-epidermal juction per decade in photoprotected skin(p<0.001). The expression of differentiation markers(keratin 1, involucrin, filaggrin, loricrin) were not different among the three age cohort groups. 2. With advancing age, there was an attenuation in the number and diameter of elastic fibers in the papillary dermis and an increase in the number and straightness of the same fibers in the reticular dermis. The collagen fibers are arranged in sparse bundles in disarray, and/or aggregates of loosely woven, straight fibers in the aged skin. There was an apparent, age-related decrease in the stainability of ground substances in the papillary dermis on colloidal iron staining. Conclusions : Our data documents semi-quantitative differences among three groups in intrinsically aged breast skin and provide the framework for future research to evaluate the ageing process.

BACKGROUND AND PURPOSE: One of the most common genetic causes of frontotemporal dementia (FTD) is mutation in the progranulin (PGRN) gene. The aim of this study is to assess the early effects of the PGRN mutations on brain volumes by longitudinal voxel based morphometric (VBM) evaluation in asymptomatic mutation carriers. METHODS: We recruited 17 asymptomatic members of families with FTD caused by PGRN mutations; 7 mutation carriers (51.0+/-11.6 yr) and 10 non-carriers (55.2+/-6.0 yr, p=0.404). The MRI follow-up intervals of carriers and non-carriers were 788.6+/-103.8 and 922.0+/-225.1 days (p=0.124) respectively. We performed cross-sectional and longitudinal VBM analysis on both groups. RESULTS: At baseline, the carriers had lower white matter (WM) volumes in left frontal regions (p<0.001, uncorrected), but had no gray matter (GM) volume reduction. The carrier's global GM (p=0.924) and WM volume (p=0.364) reduction rate were not different from the non-carrier's. However, statistical parametric mapping T-maps showed differentially increased GM volume reductions in the bilateral parietal areas of carriers (p<0.001, uncorrected). CONCLUSIONS: The findings from this study to examine WM and GM cross-sectional and longitudinal changes in PGRN mutation carriers suggest that WM atrophic changes could precede both GM changes and symptom onset in FTD. Asymptomatic PGRN mutation carriers have measurably higher rates of regional GM atrophy than non-carriers even in the pre-dementia stages.

BACKGROUND: Considerable evidences suggest that the beta-amyloid acts as a neurotoxin, and the epigallocatechin-3- gallate (EGCG) has the anti-inflammatory and anti-oxidant properties. The purpose of this study was to investigate whether the EGCG reduces the death of the cultured hippocampal tissues exposed to the beta-amyloid 1-42 fragments (A beta1-42). METHOD: We cultured the hippocampus of postnatal 7 days old Sprague-Dawley rat into slices of 450 micrometer. The tissue slices had been exposed with 100 micro M A beta1-42 at an interval of 3 days since 12 DIV (days in vitro). Following co-treatment of the tissue with 10 micro M EGCG and A beta1-42, we evaluated EGCG effect on A beta1-42 induced neurotoxicity by measuring the expression of Bcl-2 and NeuN protein and by morphological observation of the hippocampus slice cultures with propidium iodide (PI) and bromodeoxyuridine (BrdU) staining. RESULTS: The EGCG exerted a significant role in restoration of NeuN protein expression inhibited by A beta1-42, showed inhibitory effects fluorescence in PI stained tissues, and increased the anti-BrdU stained cell. CONCLUSIONS: 10 micro M EGCG reduced the A beta1-42 induced neurotoxicity of the hippocampus slice culture.

Thoracic radiculopathy (TR) is one of various neurologic complications of diabetes mellitus (DM), which can cause pain and motor weakness of chest or abdomen. Electromyographic study is well known to be useful for accurate diagnosis of TR. This study is conducted in order to answer following questions: To what extent does paraspinal EMG study show denervation potentials in neurologically asymptomatic DM patients? ; Is there any correlation between EMG abnormality and severity of diabetes mellitus? We performed thorough neurologic examination and review of medical records in 74 DM patients who were referred for neurologic evaluation and rountine nerve conduction studies. Among those patients, 16 were selected for EMG on lower thoracic paraspinal muscles, who had no clinical symptoms of thoracic radiculopathy and no evidence of peripheral neuropathy on routine nerve conduction studies. The EMG findings were evaluated with relation to serum fructosamine level and duration of DM. On EMG examination, ten of sixteen patients exhibited positive sharp wave in lower thoracic paraspinal muscle. Nine of those ten patients with denervation potential in thoracic paraspinal muscle showed high fructosamine level and eight had been exposed to DM for more than 5 years. It is suggested that subclinical diabetic thoracic radiculopathy is more frequent than expected arid that there is positive correlation between abnormal paraspinal EMG findings and the level of fructosamine and duration of DM. And it is also suggested that there could be selective involvement of posterior primary ramus in diabetic patients, although it needs further studies to be confirmed.

We observed three patients who showed widespread macular ashy-colored eruptions. They fit clinically and histopathologically with the typical erythema dyschromicum perstans (ashy dermatosis). Erythoma dyschromicum perstans is a peculiar pigmented condition of unknown etiology characterized by persistent ashy-gray colored macule, refractory to any modality. All of them have similar skin lesions which are multiple, variable sized oval-shaped, ash-colored macular eruption on the trunk and extremities. Histologic findings of 2 patients reveal the characteristic feature of focal hydropic degeneration of the basal cell layer of the epidermis, perivascular infiltrates of lymphocytes and histiocytes and melanin-laden macrophages in the corium, however, merely pigmentary incontinence into the corium in 1 patient. Literatures are briefly reviewed.
