1.Association between c. 1311C > T and c. 1004C > A and the risk of G6PD deficiency in Guangxi population

Objective :
To investigate the association between c. 1311C > T and c. 1004C > A of glucose⁃6 ⁃phosphate dehydrogenase (G6PD) gene single nucleotide polymorphism ( SNP) with the risk of G6PD deficiency in
Guangxi population.
Methods :
417 patients with G6PD deficiency were randomly selected as case group , and 295 healthy patients were selected as control group. The c. 1311C > T and c. 1004C > A were genotyped using the SNPscanTM multiple SNP method , and the haplotype frequency of two sites were analyzed by SHEsis.
Results :
In the case and control group , there were statistically significant differences in the distribution frequency of genotype TT , CC + CT and allele T at c. 1311C > T locus [TT vs CC :(P = 0. 001 , OR = 0. 373 , 95% CI = 0. 204 - 0. 683) ; TT vs CC + CT :(P = 0. 001 , OR = 0. 371 , 95% CI = 0. 203 - 0. 678) ; T vs C :(P = 0. 002 , OR = 0. 601 , 95% CI = 0. 435 - 0. 829)] ;however, there was no significant difference in genotype and allele distribution frequency at c. 1004C > A locus (P > 0. 05) . The results of the rate method showed that compared with genotype CC , the genotype CT at c. 1311C > T increased the expression level of G6PD enzyme , while the genotype TT decreased the expression level of G6PD enzyme(P < 0. 05) , the haplotype analysis showed that C ⁃C and T ⁃C were associated with G6PD risk (P < 0. 05) .
Conclusion
In Guangxi population , c. 1311C > T locus genotypes TT , CC + CT and allele T were related to the decreased risk of G6PD deficiency.

3. Effect of abnormal oligonucleotide binding folding domain protein gene on the initiation of DNA replication in hepatocellular carcinoma

ObjectiveThere are few reports about abnormal oligonucleotide binding fold domain protein genes (OBGs) affecting the initiation of DNA replication in hepatocellular carcinoma through the microchromosome maintenance (MCM) complex. This study aims to explore the roles of reverse-transcription-related genes (RTGs) in Hepatocellular Carcinoma cells (HCC) and the correlation between gene polymorphisms and abnormal gene expression.Methods We created a mouse model by injecting hepatocellular carcinoma cell line H22 (logarithmic growth phase) and dissected the tumor bodies from tumor-forming mice. The control group was treated by isotonic saline without H22. The healthy liver tissue cells were taken from the control mice. The total RNA of the H22 group and control group were extracted, and differentially expressed genes were analyzed. Screening of differentially expressed reverse transcription-related DEGs (RDEGs), GO and KEGG analysis of RDEGs. The interaction analysis of RDEGs encoded proteins, and the correlation analysis of RDEGs polymorphism and gene expression.ResultsThere were 193 differentially expressed RTGs in HCCs, which were involved in two biological procedures, three cell components, one molecular function, three signal pathways, and three functional sites; Its function is mainly concentrated in DNA replication, especially the construction of MCM complex and telomere complex in which OBGs participate in the initiation of replication. Most related genes had OB fold domains. The results also showed that both AS and SNV caused gene polymorphism was positively correlated with gene expression, and most OBGs in HCC had SNV phenomenon, but not occurred in healthy liver tissue.Conclusion Collectively, AS and SNV may be important regulatory factors for gene expression. SNV may particularly affect the function of OBGs in the MCM complex to abnormally initiate DNA replication in HCC.
4.Application andevaluation of random quality control sampling in donor blood detetion by ELISA

【Objective】 To evaluate the effectiveness of random quality control sampling in blood sample detetion by ELISA. 【Methods】 Blood samples of 5 mL specification of blood donors from our blood station from May to July 2022 were selected for routine operation on a fully automated sampler. J standard substances(3 mL specification) as daily samples were added to A1 well, H12 well and random wells of HBsAg, anti-HCV, anti-HIV, and -TP, and then placed in a fully automated enzyme immunoassay analyzer for testing. With random well quality control as the internal quality control judgment standard, 20 consecutive tests were conducted and were divided into A1 (well) group, H12 (well) group and random (well) group according to different well positions. Quality control maps were drawn using Levey-Jennings quality control chart with random group as the framework, and were compared with the quality control map of A1 well and H12 well results in the same day. 【Results】 The mean quality control levels of infectious indicators of blood transfusion in blood donors by ELISA were: HBsAg 3.87±0.28, anti-HCV 3.79±0.38, anti-HIV 3.64±0.30 and anti-TP 4.53±0.51. 【Comparison】 of HBsAg, anti-HCV, anti-HIV and anti-TP, between random group, A1 group and H12 group were HBsAg 3.87± 0.28 vs 4.09±0.30 vs 3.64±0.26, anti-HCV 3.78±0.37 vs 3.96±0.38 vs 3.63±0.38, anti-HIV 3.63±0.31 vs 3.82±0.32 vs 3.48±0.28 and anti-TP 4.51±0.51 vs 4.71±0.52 vs 4.36±0.51, The S/CO value of each indicator were H12 group

Objective:To study the effect of Ganoderma triterpenoids combined with exogenous monosialoteterahexosyl ganglioside (GM1) on cognitive dysfunction in rats with epilepsy. Methods:A total of 75 Sprague-Dawley rats were divided randomly into blank control group, epileptic model group, Ganoderma triterpenoids group, GM1 group and GM1 combined with Ganoderma triterpenoids group (combination group), with 15 rats in each group. All the groups, except the blank control group, were intraperitoneally injected with pentylenetetrazol (PTZ) 35 mg/kg once a day for 28 days. Medication groups were given corresponding administration based on daily intraperitoneal injection of PTZ. They were tested with Morris Water Maze; and were observed with transmission electron microscopy and HE staining for hippocampal neurons. Real-time quantitative polymerase chain reaction was used to detect the expression of actin-binding protein (Cofilin), synaptophysin (SYN) and growth-associated protein 43 (GAP-43) mRNA in hippocampus of rats. 6.Experimental Research on the Effects of Zhuangtongyin Decoction on Arrhythmia after Myocardial Ischemia-re- perfusion

OBJECTIVE:To investigate the e ffects of Zhuangtongyin water extract (ZTYWE)on hemorheology and blood lipid level of myocardial ischemia-reperfusion model rats ,and preliminarily explore its molecular biology foundation of reducing arrhythmia after myocardial ischemia-reperfusion. METHODS :Totally 75 SD rats were divided into control group ,model group and ZTYWE low-dose ,middle-dose and high-dose groups. Myocardial ischemia-reperfusion model was established by ligating the left ventricle cyclotron endings for 30 min then reperfusing for 60 min. Since the first day after surgery ,control group and model group were intraperitoneally administrated with same volume normal saline ;ZTYWE low-dose ,middle-dose and high-dose groups were intraperitoneally administrated with medicine solution (6.8,13.6,27.2 g/kg),once a day ,for continuous 28 d. At the 7th, 14th,21st,28th day after surgery ,the incidence rate of arrhythmia of rats were detected. The hemorheology indicators and serum contents of TC ,TG,LDL-C,HDL-C were determined after the last administration. The morphology of myocardial tissue was observed by HE staining. The protein expressions of apoptosis-related factor FAS and FAS-L were detected by Western blotting assay. RESULTS :Compared with control group ,the incidence rates of arrhythmia of rats in model group were significantly increased;the whole blood contrast viscosity ,hematocrit(HCT),in vitro forming length and quality of thrombosis ,platelet aggregation rate ,erythrocyte filtration index (IF),as well as the serum contents of TC ,TG and LDL-C were significantly increased;in vivo thrombus formation time as well as the serum content of HDL-C were significantly decreased ;the protein expression of FAS and FAS-L were significantly increased (P<0.05 or P<0.01);obvious pathological changes were observed in myocardial tissue. Compared with model group , the incidence rates of arrhythmia of rats in ZTYWE groups were in obvious reducing trend ; except for the HCT in 163.com low-dose ZTYWE group without significance , the blood contrast viscosity ,HCT,in vitro forming length and quality of thrombosis ,platelet aggregation rate ,IF as well as the serum contents of TC ,TG and LDL-C were significantly decreased ;in vivo thrombus formation time as well as the serum contents of HDL-C were significantly increased ;the protein expressions of FAS and FAS-L were significantly decreased (P<0.05 or P< 0.01);the pathological changes of myocardial tissue were improved. CONCLUSIONS :Through the promoting blood circulation and removing blood stasis effect ,Zhuangtongyin decoction can improve indexes of hemorheology index of ischemia-reperfusion model rats ,reduce the contents of TC ,TG and LDL-C ,enhace the contents of HDL-C ,down-regulate the protein expression of FAS and FAS-L ,so as to inhibit myocardial cells apoptosis and reduce arrhythmia.
7.Effects of Zhuangtongyin on Blood Vessels Diastolic and Systolic Function and Cardiac Function in Rats with Coronary Heart Disease

OBJECTIVE: To observe the effects of Zhuangtongyin (ZTY) water extract on blood vessels diastolic and systolic function and cardiac function in rats with coronary heart disease (CHD) and its potential mechanism. METHODS: SD rats were randomly divided into control group, sham operation group, model group, positive group (Compound danshen dripping pills, 0.08 g/kg) and ZTY water extract low-dose, medium-dose and high-dose groups (6.8, 13.6, 27.2 g/kg, by weight of water extract), with 10 rats in each group. Except for control group (no operation) and sham operation group (only open chest without ligation), CHD model were set up in other groups by ligating the left ventricle cyclotron endings. One week after modeling, control group, sham operation group and model group were given constant volume of water intragastrically, and administration groups were given relevant medicine intragastrically; once a day, for consecutive 4 weeks. Two hours after last administration, cardiac function indexes (LVEDV, LVESV, SV, LVEF) of rats were detected in each group. The levels of inflammatory factors (CRP, IL-1β, IFN-γ and ET-1) in myocardial tissue were determined by ELISA. Histomorphological characteristics of myocardial tissue were determined by HE staining. RESULTS: The structure of myocardial fibers in control group and sham operation group was clear and orderly; there was no statistical significance in difference of each index between 2 groups (P>0.05). Compared with control group, myocardial fibers were disorderly arranged in model group, and the phenomena of rupture, dissolution and necrosis were observed, accompanied by infiltration of inflammatory cells. The levels of LVEDV and LVESV as well as the levels of CRP, IL-1β, ET-1 and IFN-γ were increased significantly, while the levels of SV and LVEF were decreased significantly (P<0.05 or P<0.01). Compared with model group, above symptoms of rats in each administration group were improved to different extents; the levels of LVEDV and LVESV as well as the levels of CRP, IL-1β, ET-1 and IFN-γ in positive group, the levels of LVEDV in ZTY water extract medium-dose and high-dose groups, the levels of LVESV and CRP, IL-1β, ET-1, IFN-γ in ZTY water extract groups were decreased significantly. The levels of SV and LVEF in positive group, the levels of SV in ZTY water extract groups as well as the levels of LVEF in ZTY water extract medium-dose and high-dose groups were increased significantly (P<0.05 or P<0.01). CONCLUSIONS: ZTY water extract could reduce the level of ET-1 and improve blood vessels diastolic and systolic function in CHD model rats, then restore blood supply of myocardial tissue and strengthen cardiac function, by acting on inflammation cycle.
8.Association of MMP14 gene polymorphisms and osteoporosis in Zhuang men from Baise region of Guangxi.

OBJECTIVETo assess the association between membrane type 1 matrix metalloproteinase gene (MT1-MMP, MMP14) polymorphisms and osteoporosis in Zhuang men from Baise region of Guangxi.
METHODSGenotypes of 5 loci (rs1003349, rs3751488, rs2269213, rs2236303 and rs743257) of MMP14 gene in 301 Zhuang men were determined with single base extension methods, and bone mineral density (BMD) at left calcaneus was evaluated with quantitative ultrasound with measured values of broadband ultrasonic attenuation (BUA). The subjects were divided according to BMD into osteoporosis group, osteopenia group and normal bone density group.
RESULTSAll selected loci were in Hardy-Weinberg equilibrium (P> 0.05). By multiple linear stepwise regression analysis, polymorphisms of the five loci were not associated with BUA. But a significant higher risk of osteoporosis was found in individuals with MMP14 rs1003349 GT genotype (vs. GG genotype; P<0.05) and rs2236303 CC and CT genotypes (vs. TT genotype; P<0.05). Genetic linkage between rs1003349 and rs2236303 was also discovered (D'= 0.839, r(2) = 0.458, P<0.01). Compared with the normal bone density group, the frequency of a G-T haplotype of rs1003349 and rs2236303 was significantly lower in the osteoporosis group (P<0.05). And the risk of osteoporosis for individuals with G-C and T-C haplotypes was 2.556 (95% CI: 1.029-6.349, P = 0.038) and 5.111 (95% CI: 1.341-19.485, P = 0.011) compared with G-T haplotype.
CONCLUSIONPolymorphisms of rs1003349 and rs2236303 loci of MMP14 gene are associated with the susceptibility of osteoporosis in Zhuang men in Guangxi. G-C and T-C haplotypes for loci rs1003349 and rs2236303 may increase the disease risk.

9.Effects of acute hypoxia on the blood pressure heart rate, microvessels response and free radical in rabbit.

AIMTo study the effects of different acute hypoxia on blood pressure, heart rate and microvessels and free radical in rabbits.
METHODSThe experiment model was carried out with acute hypoxia on two groups of rabbits, using artificial inspiration 12.5% O2 and 87.5% N2, 8.5% O2 and 91.5% N2 (equivalent to altitudes of some 4 000 m and 6 500 m) keeping hypoxia for 5, 10, 15, 20 min. During the course of it, the changes of blood pressure, heart rate and microvessels response, superoxide dismutase (SOD), malondialdehyde (MDA) were recorded accordingly.
RESULTS(1) systolic pressure was slightly up, then down in 5 mins. Diastolic pressure was significantly down (P < 0.05) in 20 min. (2) Heart rate showed reduced and prolonged, particularly in 8.5% hypoxia group (P < 0.05). (3) Vas bores of microvessle expanded (P < 0.05) and the blood stream became slow gradually (P < 0.05, P < 0.01) in following acute hypoxia time. (4) SOD was significantly down (P < 0.05), MDA was significantly increased (P < 0.05) in 20 mins.
CONCLUSIONAcute hypoxia could cause the blood pressure and heart rate to decrease, vas bore of microvessle to expand, the blood circulation to slow down and free radicals would increase.

10. Advances on experimental models of rheumatoid arthritis

Rheumatoid arthritis (RA) is a chronic immune-me- diated synovial disease with unknown aetiology, therefore, it is considered a chronic disease that cannot be cured. RA is caused by a complex interaction between genetic and environmental factors. As such long-standing efforts have been made for better understanding of disease pathogenesis and the development of new classes of therapeutics, the continuing elucidation of pathogenic events underlying RA mostly relies on animal model studies. Individually animal models allow molecular and spatiotemporal dissection of various pathological processes of RA development. We have herein comprehensively discussed different animal models in induction methods, pathogenesis, pathological events and disease characteristics, hoping to provide the basis and reference for the rational selection of experimental animal models for the basic research and drug screening of RA.
