PURPOSE To sutdy the effects of dibutyl pathalate (DBP) on the proliferation and apoptosis of leukemic cells from patients suffering from acute leukemia. METHODS The cell culture in vitro was used to study the effect of DBP on the proliferation of leukemic cells, while agrose gel electrophoresis of DNA fragments was used to study the effect of DBP on apoptosis of leukemic cells. RESULTS DBP could inhibit the proliferation of leukemic cells got from patients suffering from acute leukemia in vitro. The percentage of inhibition were 24.24% and 36.33% respectively when leukemic cells were exposed to 50?g/ml and 200?g/ml DBP for 24 hours. Leukemic cells from acute myelogenous leukemia were more susceptive to DBP than those from acute lymphocytic leukemia. DBP also induced the leukemic cells from patients to die via apoptosis and showed typical "ladder" of DNA fragments for apoptotic cells in agrose gel electrophoresis. CONCLUSION DBP could clear the leukemic cells in bone marrows of patients suffering from acute leukemia by suppressing the growth of leukemic cells and inducing them to die via apoptosis.

Objective To study the chemical constituents of Lignum Aquilariae Resinatum.MethodsThe compounds were isolated and purified by silica chromatography and their structures were identified on the basis of physicochemical constant and spectral analysis.Results The compounds were determined as 6-hydroxy-2-[2-(3'-methoxy-4'-hydroxy phenylethyl)] chromone(Ⅰ)and a triterpene,hederagenin(Ⅱ).Conclusion Compound I is a new compound and compound Ⅱ is found in this plant for the first time.

Objective To study the chemical constituents of Evodia lepta.Methods The chemical constituents were isolated by chromatographic methods and their structures were elucidated by physicochemical characteristics and spectral data.Results Two compounds were isolated and their structures were identified as erythro-3-(1',2',3'-trihydroxy) isopentyl-7-hydroxycoumarin(Ⅰ) and?-daucosterol (Ⅱ).Conclusion CompoundⅠis a new one named evodosin A while compoundⅡis isolated from E. lepta for the first time.

Objective To study the constituents in Melicope pteleifolia. Methods Plant material was isolated with 80% EtOH. Compounds were separated with chromatographic methods and their structures were elucidated on the basis of spectral analysis (EI-MS, 1H-NMR, and 13C-NMR) and chemical evidence. Results Five compounds were isolated from petrol ether or ethyl acetate soluble fraction. Their structures were identified as 3,5,3'-trihydroxy-4'-methoxy-7-(3-methylbut-2-enyloxy) flavone (pteleifolosin C, 1), 3,7-dimethoxyl kaempferol (kamatakenin, 2), vanillic acid (3), tricosanoic acid tetradecyl ester (4), and p-sitosterol (5), respectively. Conclusion Compound 1 is a new structure named pteleifolosin C. Compounds 2-4 are isolated from this plant for the first time.

Objective: To study the chemical components of Citrus medica L.var. sarcodactylis Swingle and to supply evidence for the quality control. Methods: Chromatography and recrystallization methods were applied to analyze the components. Results and Conclusion: Three compounds were extracted from Citrus medica L. var. sarcodactylis (Noot) Swingle. And their structures were identified as Limettin (I), stigmasta-5,22-dien-3-ol (II) and palmitic acid (III) by the methods of UV, MS, IR, 1HNMR, 13CNMR and DEPT. Stigmasta-5,22-dien-3-ol (II) was obtained by extraction.

Objective To determine the content of 5,7-dimethoxycoumarin in Fructus Citris Sarcodactylis. Methods HPLC was used. Mobile phase was methanol-water(65 ∶35),detection wavelength at 326 nm,flow rate 1mL/min and column temperature at 35 ℃.Results The linearity of 5,7-dimethoxycoumarin was in the range of 0.3528～1.7640 ?g. The regression equation was Y=2403.73 X-65.35,r=0.9998. The average recovery was 99.33 %,RSD=1.47 %(n=6). Conclusion The method is simple and accurate. It can be applied in the determination of 5,7-dimethoxycoumarin in Fructus Citris sarcodactylis.

Objective To investigate the function and characteristics of histamine receptors on the hemopoietic progenitor cells.Methods BDF1 mice (both male and female), inbred at our university, aged 8-12 weeks, weighing 20-24 g, were used in this study. Bone marrow cells were incubated for 1 hour at 37℃ with 2-AT (H1 receptor agonist) or impromidine (H2 receptor agonist) alone, or in combination with the antagonists pyrilamine or cimetidine respectively. Control experiment was performed in Dulbecco's modified Eagle's Medium (DMEM) alone. Cells treated with different drugs were performed by colony forming unit-granulocyte-macrophage (CFU-GM) and colony forming unit-megakaryocyte (CFU-Meg) assay.Results When bone marrow cells were treated with 10-8 mol/L to 10-5 mol/L of 2-thiazolylethylamine (2-AT) which had no influence on CFU-GM and CFU-Meg proliferation, 10-8 mol/L to 10-5 mol/L of impromidine could increase the number of CFU-GM and CFU-Meg colonies. The effects of H2 receptor agonists on CFU-GM, CFU-Meg could be antagonized by H1 receptor agonist.Conclusions Our findings suggest the existence of histamine H1 and H2 receptors on the hemopoietic progenitor cells and the antagonism between two different histamine receptor subtypes on the proliferation of CFU-GM and CFU-Meg.
