1.Determination of Saccharide in Honeysuckle Extracts Produced by Three Classical Methods

Objective To determine the contents of fructose, glucose, sucrose and total saccharide in honeysuckle extracts which were produced by three classical methods, to provide scientific data for producing and controlling of those patent medicines containing honeysuckle extract. Methods Three classical extract methods were used for producing honeysuckle extracts. HPLC-ELSD method was used for determining the contents of fructose, glucose, sucrose, and a spectrophotometric determination method was used for determining the contents of total saccharide. Results The contents of fructose in three honeysuckle extracts were 5.44%, 3.47%, 4.74%, glucose were 4.35%, 2.73%, 4.21%, glucose were 17.41%, 8.47%, 14.47%, and total saccharide were 50.63%, 41.12%, 35.90%, respectively. Conclusion About 36%~51% of total components of honeysuckle extracts were determined except chlorogenic acids, and 19%~27% of components in the extracts were fructose, glucose, sucrose.
2.Comparative Study on Fingerprints of Root of Ophiopogon japonicus

Objective To establish the fingerprints for characterization of the chemical components of maidong(root of Ophiopogon japonicus) in two main cultivate regions of China,Sichuan(Chuanmaidong) and Zhejiang(Hangmaidong).Methods An HPLC-UV analytical methods was applied to detect 70% ethanol extracts of 20 samples from Sichuan and Zhejiang province,a "Fingerprint similarity evaluating system for TCM" issued by Pharmacopoeia Committee of P.R.China was used to evaluate the similarities all of the samples.Results The fingerprints revealed that the similarities were higher than 0.95 between samples from the same cultivate region,and were lower than 0.80 between samples from different regions of above two.Conclusion The fingerprints of Chuanmaidong and Hangmaidong were provided with high difference,and the difference can be taken as a most important proof for distinguishing the material medica of Chuanmaidong and Hangmaidong,but also in patent medicine of TCM.
3.Determination of Camphor, Borneol and Bornyl Acetate in Shenling Baizhu Powder by Gas Chromatography

Objective To establish a GC method for simultaneous determine of camphor, borneol and bornyl acetate in Shenling Baizhu Powder. Methods Capillary column was used with 100% methyl polysiloxane as stationary phase. The temperature-programmed was as follows:starting temperature of 60 ℃, 5 ℃/min, rose to 130 ℃, kept for 5 minutes, followed by 20 ℃/min up to 230 ℃, and kept for 5 minutes. Results The average recovery rate of camphor was 99.87%, RSD was 1.3%, and good linear relationship was showed in the range of 0.003 77-0.150 8 μg (r=0.999 9). The average recovery of borneol was 100.71%, RSD was 2.1%, and good linear relationship was showed in the range of 0.002 411-0.096 4 μg (r=0.999 9). The average recovery rate of bornyl acetate was 101.95%, RSD was 1.3%, and good linear relationship was showed in the range of 0.003 02-0.121 1 μg (r=0.999 9). Conclusion The method is simple, reliable, accurate, and can effectively control the quality of Amomi Fructus in Shenling Baizhu Powder.
4.Effects of hydrogen sulfide on mouse cortical neuronal injuries induced by oxygen gloucose deprivation/reoxygenation in vitro

Objective To explore the effects of H 2 S on neuronal injuries induced by oxygen glucose deprivation /reoxygenation ( OGD/R) in cortical neurons .Methods For OGD, the primary cultured cortical neurons were incubated with glucose-free EBSS media for 4h in N2/CO2/O2 (93%/5%/2%) atmosphere.Thereafter, the media were replaced by Neurobasal/B27 culture media and the neurons were incubated for 12 h in a 5%CO2 incubator at 37℃.NaHS was used as a H2S donor and cell survival rate was determined by cell counting kit 8(CCk-8).[Ca2+]i was determined using fura-2/AM and fluorescence microscopic imaging systems .The release rate of lactate dehydrogenase ( LDH) was determined by lactate dehydrogenase assay kit , and cell damage was analyzed by staining of propidium iodide ( PI ) .Results After pretreated with 200, 300 and 600μmol/L sodium hydrosulfide ( NaHS) for 30min before OGD/R, the cell survival rate of neurons significantly increased (n=4).[Ca2+]I(n=5), LDH release rate (n=4) and cell damage percentage (n=6) in the neuron pretreated with 300 μM NaHS were significantly lower than those in ODG/R cells.Treatment with 10μmol/L calcium chelator BAPTA also reduced the LDH release rate and cell damage percentage induced by ODG /R in neurons . Conclusion The results indicate that H 2 S may inhibit the OGD/R induced damage in cortical neurons via reducing calcium overload of neurons .
5.Clinical utility of tuberculosis protein chip in diagnosis of tuberculosis

Objective To explore the potential value of tuberculosis protein chip for clinical diagnosis of tuberculosis.Methods The antibody level of tuberculosis protein ESAT-6,CFP10,16 KD,38 KD and LAM was determined in 4 093 patients,inclu-ding 441 tuberculosis and 3 652 non-tuberculosis cases by protein chip.Results The tuberculosis antibody was positive in 297 of the 441 tuberculosis cases and 647 of the 3 652 non-tuberculosis cases.Tuberculosis protein chip provided a sensitivity of 67.35% and specificity of 82.28% in the diagnosis of tuberculosis.Conclusions Tuberculosis protein chip test is a quick,easy and effective method for identifying potential tuberculosis patients with good specificity.
6.Effect of NaCl stress on growth and antioxidant systems of Pogostemon cablin.

OBJECTIVETo study the effects of salt stress on the growth and resistant-oxidation enzyme activities of Pogostemon cablin.
METHODCutting-rooted seedlings were cultivated in the plastic flowerpot irrigated with various concentrations of NaCl (Hoagland solution) for different duration, and the height, fresh/dry weight, root vitality, SOD,POD and CAT activity, membrane permeability and MDA contents were determined.
RESULTThe growth and development of P. cablin was restrained after salt stress. The differences of salt stress of lower concentration and for a short time (less than 20 mmol x L(-1) and the 9th day) were not obvious. The fresh/ dry weight, growth rate and increasing rate of dry matter were obviously decreased with the increasing of NaCl concentration and prolonging of the time, but membrane permeability and contents of MDA were increased all along, and lipid peroxidation enhanced, while the root vitality and activities of SOD, POD and CAT were firstly increasing at lower concentration and then decreasing at higher concentration.
CONCLUSIONThe growth of P. cablin depends on NaCl concentration and the treated duration.

7.Residuals of organochlorine pesticides and heavy metals in soil and water of planting base and Liriope muscari.

OBJECTIVETo analyze the residuals of organochlorine pesticides and heavy metals in soil, water and Radix Liriopes from planting bases in Quanzhou city, and evaluate the quality of the herb.
METHODGC method was applied to determine residuals of organochlorine pesticides, the contents of Pb, Cd, Cu, Cr, and As were determined by IPC.
RESULTThe contents of residuals of organochlorine pesticides and heavy metals varied among the samples of soil, water and Radix liriopes, but all of the residuals met requirement of the national standard.
CONCLUSIONResiduals of organochlorine pesticides and heavy metals in soil, water and Radix liriopes from planting bases in Quanzhou city were conformed to GAP.
