Chinese Journal of Clinical and Experimental Pathology 2015;(10):1089-1094
Purpose To investigate the relationship between Helicobacter pylori L-form ( Hp-L) infection and expression of XPF and p53 in gastric carcinoma. Methods XPF, p53 and Hp-L were examined in 239 cases of gastric carcinoma and healthy individuals by means of Giemsa stain and immunohistochemical staining with ABC method. Results XPF and p53 expression in gastric carcinoma was significantly higher than that of the surrounding tissue (P<0. 01). There were positive correlation between the expression of XPF and p53 (r=0. 196, P<0. 01). The expression of XPF was related to the Laurén classification (P<0. 05). The expression of p53 was associated to the depth of invasion (P<0. 05). (2) 175 (73. 22%) cases were Hp-L positive by both Giemsa and immunohisto-chemical staining. Hp-L expression in gastric carcinoma were significantly higher than that of the surrounding tissue (P<0. 01). (3) In the Hp-L positive group, XPF and p53 were significantly higher than in that of the Hp-L negative group (P<0. 01). Positive corre-lation existed between positive Hp-L staining and the expression of XPF and p53 (r=0. 32, r=0. 296, P<0. 01). (4) Univariate a-nalysis showed that depth of invasion, distant metastasis, TNM stage, XPF and p53 were related to the prognosis of gastric carcinoma ( P<0. 05). Multivariate Cox model analysis showed that XPF was an independent prognostic factor for the patients (P<0. 05). The pa-tients with positive expression of XPF had a shorter survival time. When combined with the depth of invasion and distant metastasis, the correction risk increased by 3. 791 times. Conclusion XPF, p53 and Hp-L may play very important roles in the development of gastric carcinoma. The high expression of XPF implies poorer prognosis. Hp-L infection may be related to the up-regulated expression of XPF and p53.

Objective To analyze the antibody titer value of indirect haemagglutination test(IHA)in 135 confirmed acute schistosomiasis patients,so as to provide the evidence for improving the diagnosis and treatment of acute schistosomiasis. Meth-ods A total of 135 acute schistosomiasis inpatients were selected from 2001 to 2006. They all received the IHA antibody titer de-tection,and the correlation among the age, incubation period, and hospitalization days was calculated. Results The antibody titers of IHA were higher than 1:320 in all the cases. The percentages of 1:640,1:1280,1:2560,1:5120 and 1:10240 were 1.48%,28.15%,35.56%,20.00%,and 14.81%respectively. The mean age was(47.70 ± 14.58)years,average incuba-tion period was(38.03 ± 4.59)days and mean hospital stay time was(15.08 ± 3.79)days. The antibody titer value had no corre-lation with the age distribution(r=0.109,P>0.05). There was a negatively correlation between the antibody titer value and in-cubation period,(r=-0.558,P<0.01),there was a positive correlation between the antibody titer value and hospitalization time(r=0.791,P<0.01),and there were significant differences among different groups(F=17.07,64.53,both P<0.01). Conclusions The antibody titer of acute schistosomiasis cases detected by IHA is 1:640 and above. There is no correlation be-tween the antibody titer value and age,but the antibody titer value is higher, the incubation period is shorter and hospitalization time is longer.

Objective To study the protective effect of hydrogen-rich saline (HRS) on apoptosis in skin flap after ischemia/reperfusion injury. Methods Total 18 Sprague-Dawley rats were randomly divided into three groups:a HRS treated group and two physiological saline treated groups (controls 1,2).The rats were anesthetized and an extended abdominal skin flap (6 cm × 9 cm) was elevated in each animal.Ischemia was induced by clamping the left right pedicle for 3 h,then HRS was administered intraperitoneally 10 min before reperfusion, and physiological saline was injected in control groups 1 and 2.In the control group 2,the flaps were elevated without occluding the artery and vein.Five days postoperation,apoptosis,TNF-α level in flap were measured with ELISA,NF-κB in nucleus was determined by Westernblot. Results Apoptotic rate represented (39.72±8.09) %in HRS group and (69.43±13.27) % in control group 1,respectively.Treatment with HRS resulted in a marked reduction in apoptotic rate.TNF-α level was (516.408±38.674) pg/ml in the control group 1,a significant reduced TNF-α was measured in HRS group, accounting for (269.136 ±24.530) pg/ml.Moreover,NF-κB activation was significantly down-regulated by HRS.In control group 2,no significant apoptosis was observed because of non-blood occlusion, and there was no marked elevation of TNF-α and NF-κB.Conclusions HRS can protect skin from ischemia/reperfusion injury, attenuate apoptosis in flaps, which may be associated with the inhibition of TNF-α and NF-kb elevation.
