2.Investigation and analysis of psychological consulting requirement for the hospital staff

Objective To learn the psychological needs of the hospital staff in terms of their life,medical care and psychological health.Methods 2910 hospital staff were interviewed with questionnaires and the outcomes analyzed with x2 test and descriptive analysis.Results 96.9%of the surveyed found themselves in need of psychological counseling; considerable consulting needs of the staff; most of them turn to friends to complain instead of their leaders.Conclusion The psychological counseling should be enhanced to build effective communication channels and ease stress of the staff.
3.Prediction of multifocal visual evoked potential for postoperative visual recovery in cataract patients

AIM: To determine multifocal visual evoked potential (mVEP) in the prediction of postoperative visual acuity in cataract. METHODS: We examined 30 normal eyes as control and 60 eyes of 52 cataract patients, senile cataract in 27 cases 30 eyes, cataract with glaucoma in 25 cases 30 eyes by mVEP examination. All patients underwent phacoemulsifi-cation (Phaco) and intraocular lens (IOL) implantation. After surgery,best corrected visual acuity (BCVA) was examined at 1 week, 1 month, and 3 months respectively.RESULTS: The mean amplitude and latency in mVEP responses between normal control group were 183±11nV, 95±8ms, and in senile cataract group were 177±10nV, 96± 8ms respectively, there were no significant difference between two groups (P>0.05). The mean amplitude and latency of cataract with glaucoma 138±7nV, 99±6ms were significantly different comparing to both control and senile cataract group (P<0.05). After surgery, the am-plitude and latency were 276±11nV and 93±8ms respec-tively, which did not change significantly comparing to the normal eyes (P<0.05), their visual function got no obvious damage and visual recovery were better (BCVA≥0.8). While those with central amplitude 221±6nV and latency 105±7ms that were significantly deviated from control group (P<0.05), and their visual function were seriously damaged and visual recovery were much poorer (BCVA<0.3).CONCLUSION: mVEP waveform might enable us to evaluate objective visual function detection before cataract surgery. A subject with visual function damage, their mVEP responses to stimulation were severely changed when it compared to controls.

Pain is one of the common clinical symptom, previous studies have implicated sodium channels as a key constituent in pain signaling. Sodium channel blockers with efficient sodium channel blockade effect play an important role in analgesic treatment. However, most drugs used in clinic have many drawbacks and can not meet the demand of the clinical use. Therefore, for the development of new generation of sodium channel blockers, it is of great significance to find small molecule sodium channel blocking lead compounds with novel chemical scaffolds and new structures, sodium channel blocking activity and structure-activity relationship are discussed in detail, and current problems and trends in future research are also emphasized.

5.Advanced glycation end products inhibit testosterone production in rat Leydig cells.

OBJECTIVETo study the expression of the receptor for advanced glycation end products (RAGE) and the inhibitory effect of advanced glycation end products (AGEs) on testosterone production in rat Leydig cells.
METHODSRat Leydig cells were primarily cultured and the expression of RAGE in the Leydig cells was detected by RT-PCR and immunofluorescence staining. The Leydig cells were treated with AGEs at the concentrations of 25, 50, 100 and 200 microg/ml, respectively, and the testosterone content was determined by ELISA.
RESULTSRT-PCR and immunofluorescence staining exhibited the expression of RAGE in the rat Leydig cells. AGEs remarkably suppressed hCG-induced testosterone production in the Leydig cells in a concentration-dependent manner in the 50, 100 and 200 microg/ml groups as compared with the control (P < 0.01).
CONCLUSIONRAGE exists in rat Leydig cells and AGEs can significantly inhibit the secretion of testosterone in primarily cultured rat Leydig cells.

7.Effect of Smoking on Clinical Prognosis in Male Patients With Acute Coronary Syndrome After Drug-eluting Stent Therapy

Objective: To investigate the compliance of smoking cessation and the effect of smoking status on long-term clinical prognosis in male patients with acute coronary syndrome (ACS) after drug-eluting stent (DES) therapy. Methods: A total of 656 ACS patients after DES therapy were studied, according to the post-operative smoking status, the patients were divided into 3 groups: Non-smoking group,n=226, Quit smoking group,n=283 and Persistent smoking group, n=147. The patients were followed-up for the average of 27 months, the major adverse cardio-/cerebral-vascular events (MACCE) were recorded in detail, and the effect of smoking status for MACCE occurrence were evaluated by multivariable Cox regression analysis. Results: The pre-operative smoking rate was 65.5% (430/656) of patients and post-operative smoking rate was 22.4% (147/656). Compared with Non-smoking group and Quit smoking group, the patients in Persistent smoking group had the younger age (P<0.001), more patients with abnormal blood lipids (P=0.005) and having lower level of education (P<0.001). The all cause death rates in Non-smoking group, Quit smoking group and Persistent smoking group were at 1.8%, 1.1% and 6.1% respectively,P=0.004; the MACCE occurrence rates were at 7.1%, 5.3% and 15.0% respectively,P=0.002. Multivariable Cox regression analysis showed that post-operative smoking was the independent risk factor for MACCE occurrence, HR =1.404, 95% CI (1.206-1.793),P=0.008. Conclusion: Smoking is the independent risk factor for MACCE occurrence in male ACS patients after DES therapy.
8.Inhibition of β1-integrin on apoptosis of rabbit corneal epithelial cells

· AIM: To investigate the effect of β1-integrin overexpression on the apoptosis of rabbit corneal epithelial cells and the related mechanism. · METHODS: The plasmid expressing β1-integrin-GFP fusion protein was constructed by polymerase chain reaction (PCR), and this plasmid (β1 group) or the empty vector (mock group) was transfected into rabbit corneal epithelial cells, respectively. The expression of β1-integrin-GFP fusion gene was confirmed by reverse transcription-polymerase chain reaction (RT-PCR) and Western blot. The adhesion of transfected cells to extracellular matrix (ECM) proteins was determined by adhesion assay. The apoptosis of rabbit corneal epithelial cells was assayed by Hoechst 33342 staining and DNA ladder. The phosphorylation of mitogen-activated protein (MAP) kinase was examined by Western blot. · RESULTS: Rabbit corneal epithelial cells overexpressing β1 -integrin-GFP fusion gene were successfully established. Compared with mock group, β1-integrin transfection significantly promoted the adhesive of rabbit corneal epithelial cells to ECM proteins such as laminin, fibronectin, collagen Ⅰ and collagen Ⅳ. Β1-integrin overexpression inhibited apoptosis and induced MAP kinase phosphorylation in rabbit corneal epithelial cells (P<0.05).· CONCLUSION: These data suggest that overexpression of β1-integrin confers resistance to apoptosis in rabbit corneal epithelial cells, and MAP kinase pathway may play an important role in this process.
9.Inhibition of Nuclear Factor-?B Activity by Mesenchymal Stem Cells in Rats with Myocardial Infarction

Objective To investigate the effect of mesenchymal stem cells (MSC) on the activity of nuclear factor (NF)-?B in rats with myocardial infarction.Methods MSC were isolated from SD rats (120—150 g in weight).SD rats (180—200 g in weight) were subjected to MI by left coronary artery occlusion,and were allo- cated into three groups randomly:1)sham group (without ligation of the artery,n=8);2)injection of PBS solu- tion (n=8);3)injection of MSC (n=8).MSC or PBS solution was injected into myocardium from epicardium instantly after MI models were established.Four weeks after transplantation,cardiac function was evaluated u- sing physiological recorder.Western blot were performed to investigate the nuclear factor-? activity.The ex- pressions of tumor necrosis factor (TNF)-? and interleukin (IL)-6 were evaluated by RT-PCR and Western blot. Results 1)Mortality was 20%(2/10) in sham group,33.3%(4/12) in PBS group and 20%(2/10) in MSC group with no statistic differences between them(P=0.646).2) Hemodynamic measurements showed that MSC trans- plantation caused significant improvement in cardiac function,comparing with MI+PBS group.3) MSC inhibi- ted the activities of NF-?B in myocardium and down-regulated the expression of TNF-? and IL-6 in mRNA and protein level.Conclusion Transplantation of MSC improved cardiac function in MI rats,which may partly at- tribute to their immuno-inflammatory regulation mechanism.
10.Studies on callus growth and phillyrin accumulation of Forsythia suspensa.

OBJECTIVETo investigate the effects of physical and chemical factors on callus growth and phillyrin contents of F. suspensa.
METHODThe cell growth index and phyllirin yield in different culture condition such as different plant hormones mixed, mediums, light and dark were compared. HPLC was used to examine phillyrin contents.
RESULT AND CONCLUSIONGrowth cycle of cells is twenty-eight days. During the course of callus growth, the processes of phillyrin biosynthesis were parallel with the cell growth. The optimum medium is MS. The optimum hormones concentrations are 1 mg.L-1 2,4-D, 0.5 mg.L-1 6-BA and 0.5 mg.L-1KT. The cell culture in light is more suitable than that in dark.
