1.The influence of T regulatory cells on experimental colitis of rate after treatment by probiotics

Objective To evaluate the effects of probiotics on experimental colitis of rats and to explore systemic and local changes of T regulatory cells(Tr) after the treatment. Methods The models of experimental colitis were established by enema with 2,4,6-trinitrobenzenesulphonic acid (TNBS). Live probiotics of combined bifidobacterium,lactobacillus and enterococcus (Bifico) were used. Four groups of rats were set up as negative group, TNBS alone group, probiotics group (high dosage and low dosage, with dose of 150 and 300 mg?kg-1?d-1, respectively), and combination group (probiotics plus prednisolone or sulfasalazine). Histologic scoring was used to evaluate the effect of medications; the proportions of CD4+CD25+ and CD8+CD28- Tr in peripheral blood, spleen mononuclear cells and intraepithelial lymphocytes of colon were detected by flow cytometry. 2.Changes of CD4~+CD25~+ and CD8~+CD28~- T regulatory cells in spontaneous tolerance of inbred rat orthotopic liver transplantation

Objective: To explore the roles of CD4+CD25+and CD8+CD28- T regulatory cells in spontaneous tolerance of inbred rats orthotopic liver Transplantation.Methods: The orthotopic liver transplantation models of spontaneous tolerance were established between BN (donor) and LEW (recipient) inbred rats whose survival time was more than 120 days, The expression of CD8+CD28- T and CD4+CD25+T cells in PBMC and spleen were determined by flow cytometry. CD8+CD28- T and CD4+CD25+T cells were sorted by immunomagnetic beads. The expressions of CD25, CD28, CD152 and CD45RO on CD8+CD28- T and CD4+CD25+T regulatory cells were also determined by flow cytometry. 3.Probiotics in the treatment of experimental colitis in rats and its influence on T cell subsets.

Objective To evaluate the effects of probiotics in experimental colitis of rats.Methods Models of experimental colitis were established.Several groups of rats were set up as probiotics group,negative group and positive group.Histologic scoring was used to estimate effects of drugs;the expression of CD~+_4 andCD~+_8 on T cell surface in peripheral blood,spleen mononuclear cells and intraepithelial lymphocytes of colon were detected by flow cytometry.Results Histopathology shows that probiotics of larger dosage was curative for experimental colitis in rats.Proportion of CD~+_4 and CD~+_8 T cells increased in colon of colitis,which was decreased by treatment of larger dose of probiotics.CD~+_4/CD~+_8 ratio and T cell subsets in systemic immune system were not influenced by probiotics.Conclusion Probiotics of large dosage are effective in the treatment of experimental colitis of rats incuced by TNBS,possibly associated with immune regulation and tends to be localized in mucosal immune system in colon.
4.The induction of CD8+CD28- regulatory T cells by donor specific transfusion and their effect on the acute rejection responses in rats liver transplantation

Objective To acquire antigen specific CD8+CD28-Tr by donor specific transfusion (DST) in vivo, and evaluate their suppressive effect on the acute rejection responses in rat liver transplantation through adoptive transfer experiment. Methods DST was used to induce CD8 + CD28 Tr to LEW antigens in naive Brown Norway(BN) rat. Then the induced CD8+CD28- Tr were adoptively transferred into the recipients of LEW→BN liver transplantation, which were acute rejection models. The survival time and histological changes were observed. Statistic analysis was performed with Log-Rank test by SPSS11.0 software, to compare the survival rate. Results Adoptive transfer of the DST-induced CD8+CD28-Tr attenuated the acute rejection responses in acute rejection models of rat liver transplantation. Conclusion DST can induce large numbers of CD8+CD28-Tr in naive BN rats in vivo. The adoptive transfer of the induced CD8+CD28Tr suppressed the acute rejection responses in rat liver transplantation. DST may become one of the methods that induce antigen specific CD8+CD28- Tr in vivo.
5.The suppressive effect of CDS+ CD28-regulatory T cells from spontaneous tolerance models on the acute rejection responses in rat liver transplantation

Objective To evaluate the suppressive effect of CDS+ CD28-regulatory T cells(Treg)in vivo from spontaneous tolerance models on the acute rejection responses in rat liver transplantation.Methods Spontaneous tolerance models of inbred rat liver transplantation were established.CDS+ CD28-Treg isolated from recipients of spontaneous tolerance models were adoptively transferred into the recipients with acute rejection responses one day before the operation.The survival time and histological changes were observed in the adoptive-transferred models.Results CDS+ CD28-Treg from spontaneous tolerance models(LEW→DA)prolonged the survival time of the recipients in acute rejection models(LEW→BN)(from 14.0±2.2 days to 24.0 ± 3.0 days,P=0.0049),and the severity of rejection was alleviated in liver pathology.Conclusion CDS+ CD28-Treg from spontaneous tolerance models can suppress the acute rejection responses in rat liver transplantation,with antigen-specific properties.
6.Relation of body mass index to vitamin D, PTH, and bone turnover markers levels among women in Shanghai area

Objective To examine the relationship of body mass index (BMI) with levels of vitamin D,parathyroid hormone (PTH),and bone turnover markers among women in Shanghai area.Methods Altogether 810 Chinese women aged 45 year and older were enrolled to study the associations by multiple linear regression analyses.Bone mineral density was measured by a dual energy X-ray absorptiometry (Lunar,Prodigy,GE),Serum 25-hydroxyvitamin D [25 (OH) D],PTH,C-telopeptide of type Ⅰ collagen (CTX),osteocalcin (BGP),and bonespecific alkaline phosphatase (BSALP) were measured.Results (1) 25 (OH) D,BGP,BSALP,and CTX levels were significantly lower in obesity group than in control group ; PTH level was significantly higher in obesity group than in control group.(2) In multiple linear regression analyses,BMI was significantly associated with lower 25 (OH) D (β =-0.017,P =0.006),BGP (β =-0.077,P =0.019),and higher PTH (β =0.025,P =0.000).Conclusions (1) There were siginificant associations between higher BMI and lower 25 (OH) D among women in Shanghai.(2) Serum osteocalcin was associated not only with bone metabolism but also with energy metabolism.
7.Association between the efficacy of alendronate and serum level of vitamin D in patients with osteoporosis

Objective To examine the association between vitamin D and alendronate response,and to investigate the proper vitamin D levels for the efficacy of alendronate treatment of osteoporosis.Methods In this retrospective study,559 post-menopausal women with osteoporosis were devided into two groups:non-responders and responders,based on the European Study of Forsteo (EUROFORS).Demographic and clinical data including mean 25-hydroxy vitamin D (25-OHD) levels between dual-energy X-ray absorptiometry (DEXA) scans were obtained.Mean 25-OHD levels were compared between responders and non-responders and multiple logistic regression analysis was performed to identify factors associated with non-response.Results There were 437 (75.5%) responsers to alendronate therapy and 142 (24.5%) non-responders.Responders with a mean serum 25-OHD level of (59.96 ±12.56) nmol/L,obtained a more favorable result than non-responders,whose serum 25-OHD level was (47.50 ±9.92) nmol/L (P<0.01).25-OHD level was significantly associated with response.Conclusion Patients with a mean 25-OHD ≥50 nmol/L had a substantially greater likelihood of maintaining bisphosphonate response.
9.Clinical characteristics of women with polycystic ovary syndrome without detection of polycystic ovary

876 women with polycystic ovary syndrome (PCOS) were collected from Sep 2004 to Oct 2006. Among them 800 patients with polycystic ovary ultrasound image belonged to group A; the other 76 women without polyeystic ovary ultrasound image belonged to group B. As compared with group A, group B showed significantly higher hirsutism scores, serum testosterone, cholesterol, and lower density lipoprotein. Significantly higher prevalences of diabetes mellitus and hypertension appeared in the first degree relatives in group B than in group A.
10.The clinical observation of disease outcome of early nasogastric nutrition on patients with acute decompensated chronic heart failure

Objective To investigate early nasogastric nutrition on patients with acute decompensated chronic heart failure,and observe its influence on patient's disease outcome.Methods From January 2012 to December 2013,100 cases with acute decompensated chronic heart failure were used as the research subjects,were randomly divided into experimental and control groups with 50 cases each group.Patients of two groups adopt expansion of blood vessels,strong heart,diuresis,and routine anti-infection treatment;Based on this scheme,experimental group adopt early nasogastric nutrition.The cardiac function index,the change of nutritional status and clinical curative effect and the result of follow-up within 1 year after discharge of two groups were compared.Results Left ventricular end-diastolic dimension (LVEDD) and left ventricular end-systolic dimension (LVESD) in two groups after treatment were decreased,and the descender of experimental group was superior to that of the control group with statistically significant difference (P < 0.05).After treatment,left ventricular ejection fractions (LVEF) of two groups were improved,and the rise of experimental group was superior to that of control group with statistically significant difference (P <0.05).In the two groups after treatment,the serum brain natriuretic peptide (BNP) levels were significantly decreased,and the descender of the experimental group was more than that of control group with statistically significant difference (P < 0.05).After treatment,body mass index (BMI),the level of serum prealbumin (PA),transferrin (TRF),and the total number of lymphocytes of two groups were increased significantly (P < 0.05);and BMI,the level of serum PA and TRF,and the total number of lymphocytes of the experimental group were significantly better than those of the control group (P < 0.05).The total effective rate of the experimental group was significantly higher than that of the control group (P < 0.05).The exacerbation rates,readmission and mortality rate of experimental group follow-up within 1 year after discharge were significantly lower than that of the control group (P < 0.05).Conclusions The early nasogastric nutrition on the patients with acute decompensated chronic heart failure was better able to improve patients' nutrition state,more conducive to patients with cardiac function recovery,improve the clinical curative effect,and reduce the exacerbation rates,and readmission and mortality rate.
