5.Coping strategy and its effect on occupational stress among rail freight dispatchers.

OBJECTIVETo analyse the relationship between coping strategy and occupational stress in rail freight dispatchers.
METHODS115 rail freight dispatchers were investigated by using group sampling method, investigation contents included coping strategies, occupational stressors, strains and personalities.
RESULTSThe proportion of using coping strategy in rail freight dispatchers is lower. The scores of job future ambiguity, type A behavior and work locus of control in workers with insufficient coping strategy were higher than those in workers with sufficient strategy (P < 0.05 or P < 0.01), but the score of organization commitment score in workers with insufficient coping strategy was lower than those in workers with sufficient strategy (P < 0.05), the differences of scores of some occupational stressor, strain and personality variables between workers with insufficient and those with sufficient in social support, job-family balance, job involvement coping factors of coping strategy were remarkable significant (P < 0.05 or P < 0.01), the differences of scores of occupational stressor, strain and personality variables between workers with insufficient and those with sufficient in ask, logic and time management factors of coping strategy weren't significant (P > 0.05). Logistic regression analysis showed that risk of being job dissatisfaction and daily life stress in workers with insufficient social support coping was three or four times than those with sufficient coping (OR = 3.06 or 4.38, respectively), risk of being daily life stress in workers with insufficient job involvement coping was three times than those with sufficient coping (OR = 3.26).
CONCLUSIONThe proportion of using coping strategy in rail freight dispatchers is lower. Coping strategy has influence on the individual's perception of occuaptional stressors, strains and personalities.

6.The synergism and mechanism of action of rClone30-hDR5 in combination with TRAIL on HCC.

To investigate the cell-killing effect and its possible mechanism of rClone30-hDR5 in combination with TRAIL on human hepatic carcinoma (HCC) cell line, first of all, recombinant plasmid pee12.4-hDR5 was introduced into HepG2 cells by liposome transfection. After five rounds of screening by flow cytometry, HepG2 cells expressing high levels of DR5 on cell surface were isolated. The cytotoxicity of TRAIL to selected cells was higher than that of TRAIL to HepG2 cells by MTT method (P < 0.01). The result suggested that the cloned hDR5 gene had biological activity. MTT assay showed that, rClone30- hDR5 in combination with TRAIL more efficiently inhibited the tumor growth of HepG2 cells compared to rClone30-hDR5 or TRAIL in vitro. The results of Annexin V-FITC/PI staining and Quantitative Real-time PCR indicated that rClone30-hDR5 in combination with TRAIL significantly increased the mRNA levels of caspase 3 and caspase 8, and induced the apoptosis of tumor cells. HepG2 cells were infected with rClone30-hDR5 or rClone30 at MOI of 1. The expression of hDR5 on tumor surface increased significantly by rClone30-hDR5 compared to that by rClone30, which contributed to the sensitivity to TRAIL. In conclusion, rClone30-hDR5 in combination with TRAIL has potential application value in cancer treatment.

7.Preparation and release behaviour of mesoporous silica/ethylcellulose sustained-release mini-matrix.

Hot-melt extrusion was applied to prepare mesoporous silica/ethylcellulose mini-matrix for sustained release, and fenofibrate was used as a model drug, ethylcellulose and xanthan gum were chosen as sustained-release agent and releasing moderator, respectively. This novel matrix obtained the controlled release ability by combining mesoporous silica drug delivery system and hot-melt extrusion technology. And mesoporous silica particle (SBA-15) was chosen as drug carrier to increase the dissolution rate of fenofibrate in this martix. Scanning electron microscope, transmission electron microscope, small angle X-ray powder diffraction and N2 adsorption-desorption were introduced to determine the particle morphology, particle size and pore structure of the synthesized SBA-15. The results showed that SBA-15 had a very high Brunauer-Emmett-Teller specific surface area, a narrow pore size distribution, large pore volume and a ordered two-dimensional hexagonal structure of p6mm symmetry. Differential scanning calorimetry and X-ray powder diffraction results demonstrated that fenofibrate dispersed in an amorphous state inside the pores of the mesoporous silica which contributed to the improvement in the dissolution rate. The drug release of mini-matrices was influenced by ethylcellulose viscosity grades and xanthan gum concentration, which increased with the increasing of xanthan gum concentration and decreasing of ethylcellulose viscosity. Mini-matrix containing 22% xanthan gum exhibited a good sustained release performance, and the drug release behavior followed the first-order kinetics.

8.Job stress, gene polymorphism of beta2-AR, and prevalence of hypertension.

OBJECTIVETo study the interactive effect of job stress and genetic susceptibility (or gene polymorphism) on hypertension.
METHODSA cross-sectional epidemiological study was conducted in 452 workers from a thermal power plant in China. Extrinsic effort, occupational reward, and over-commitment were measured. Hypertensive patients were defined by three phases of screening, reexamination, and final diagnosis. beta2-AR genotypes and allele frequencies at amino acid positions 16 (beta2-AR-16: Arg-->Gly) and 27 (beta2-AR-27: Gln-->Glu) were identified by PCR-RFLP.
RESULTSJob stress was related with the prevalence of hypertension in males (P < 0.05), whereas no significant relationship was found in females (P > 0.05). Differences in genotypes and allele frequencies of the beta2-AR-16 were statistically significant between the hypertension and control groups (P < 0.05), whereas those of beta2-AR-27 were not (P > 0.05). The prevalence of hypertension was higher in individuals carrying Gly16 allele than in those carrying Arg16 allele of the high job stress group (P < 0.01 or 0.05).
CONCLUSIONHigh job stress and polymorphism of beta2-AR-16 have an interactive effect on the prevalence of hypertension in male workers.

9.Accelerated improvement of bacterial blight resistance of 'Shuhui527' using molecular marker-assisted selection.

'Shuhui527' is a promising restorer line bred by Rice Research Institute of Sichuan Agricultural University in recent years. However, this line is susceptible to Bacterial Blight (BB), which limits its use. The IRBB60, from the International Rice Research Institute (IRRI), contains dominant genes Xa21 and Xa4 conferring resistance to BB. The objective of this study is to improve the BB resistance of 'Shuhui527' by introgressing Xa21 and Xa4, the two broad-spectrum BB resistance genes, into 'Shuhui527' with IRBB60 as the donor, pTA248 and MP12, linking tightly with Xa21 and Xa4 respectively as DNA markers. BC1 F1 progenies of (Shuhui527 x IRBB60), containing Xa21 and Xa4 identified using PCR screening and with agronomic traits including plant type, grain type and days to heading etc similar to those of 'Shuhui527', were subsequently backcrossed to 'Shuhui527' and self-pollinated to generate BC2 F1 and BC1 F2 . The BC3 F1 and BC3 F2 were subsequently developed using the same approach. Among the 20 BC3 F2 plants, homozygous Xa21 and Xa4,10 plants were the most similar to 'Shuhui527' in the agronomic traits, and were screened using 120 pairs SSR and 100 pairs RAPD markers. Based on the results of the background screening and the performance of the agronomic traits, 5 plants were identified as improved-'Shuhui527' and designated as 527R-5, 527R-6, 527R-8, 527R-9 and 527R-10. The improved-' Shuhui527' lines expressed high resistance to Xanthomonas oryzae pv . oryzae (Xoo) stains C I - C VII, P1 and P6. The evaluation of the polymorphisms and selection accuracies of pTA248 and MP12 demonstrated that the polymorphisms of the two markers were obvious and co-dominant and the accuracies were more than 97% and 83% respectively, indicating the two markers are good for Xa21 and Xa4 in Molecular Marker-assisted Selection.

10.Variation and significance of serum and stool IL-18 and IFN-gamma levels in children with rotavirus enteritis.

OBJECTIVETo study the variation and significance of serum and stool IL-18 and IFN-gamma levels in children with rotavirus enteritis.
METHODSSerum and stool specimens from 50 children with acute rotavirus enteritis were collected before treatment. Serum and stool levels of IL-18 and IFN-gamma were measured using ELISA. Serum and stool specimens from 21 age and gender-matched healthy children were used as the Control group.
RESULTSSerum and stool levels of IL-18 and IFN-gamma in patients with rotavirus enteritis were significantly higher than those in the Control group. There was a negative correlation between the serum IFN-gamma level and the frequency of vomiting (r=-0.368, P < 0.05). The stool IL-18 level negatively correlated to the frequency of diarrhea (r=-0.414, P < 0.05). A positive correlation was found between the serum levels of IL-18 and IFN-gamma (r=0.416, P < 0.05).
CONCLUSIONSSerum and stool levels of IL-18 and IFN-gamma were increased and associated with the severity in children with rotavirus enteritis. IL-18 and IFN-gamma might have protective effects against acute rotavirus infection at the early stage.
