1.The correlation between micrometastasis and hMAM in the peripheral blood of the patients with breast cancer

Objective To study the correlation between micrometastasis and human manmmaglobin mRNA(hMAM-mRNA)in tlle peripheral blood of the patients with breast cancer.Methods Peripheral blood samples were obtained from 92 patients with breast carcinoma,and 20 patients were with benign breast diseases,and 10 patients with other system malignant tumor,and 10 normal healthy volunteers.To identify breast carcinoma cells in periphersl blood,hMAM-mRNA Was amplified from total RNA extracted from whole blood by nested reverse transcription-polymerase chain reaction(Nested-RT-PCR).The expression of hMAM-mRNA wag contracted to MCF-7 breast cancer cell.Results The hMAM-mRNA wag only detectable in the blood from breast disease patients.Among 92 breast carcinoma patients,40 were nested-RT-PCR positive for hMAM-mRNA.The positive rate was 43.5%(40/92).The expression of hMAM-mRNA in pe-ripheral blood was not correlated with patient's estrogen or pregnant receptor;menstruation state;primarytumour size.The expression of hMAM-mRNA in peripheral blood wag correlated with patient's age:the state of c-erbB-2 and carcino embryonic antigen and carbohydrate antigen 15-3.Conclusion The hMAM-mRNA in peripheral blood may be indicator of the micmmetastasis of breast cancer.
2.Exploration of hapten-induced atopic dermatitis murine models for non-clinical pharmacodynamics study of drugs

Atopic dermatitis (AD) is a chronic, relapsing, inflammatory dermatosis with a variety of clinical manifestations and difficult to cure. Currently, many AD drug candidates have entered the research and development pipeline. In order to provide technical specifications for the clinical development of AD drugs, the Center for Drug Evaluation of National Medical Products Administration released the "Technical Guidelines for Clinical Trials of Drugs for AD Treatment" (Draft for Comments) in November 2022. Non-clinical pharmacodynamics evaluation is an important research before the drug enters clinical trials. Oxazolone (OXA)- and 2,4-dinitro-fluorobenzene (DNFB)-induced models are the most popular classical hapten-induced AD murine models, but variations of modeling are existing in the methods from different studies, including sensitization sites, haptens' dosages, the period of challenges, and the skin lesions severity evaluation as well. In this study, the investigation of OXA- and DNFB-induced AD murine models with various conditions of modeling was performed to compare the characteristics of hapten-induced AD murine models in the pathological process and severity according to the appearance of AD patients, and the guidance of pharmacodynamics evaluation of AD-therapeutic drugs in clinical trials as well, which may provide a proposal for AD treatment drug candidates in the non-clinical pharmacodynamics evaluation. All animal experiments were approved by the Animal Care & Welfare Committee of Institute of Materia Medica, Chinese Academy of Medical Sciences and Peking Union Medical College (approval No.: 00007782 and 00007784).
3.Effect of nourishing Yin, strengthening Qi and activating blood decoction on Fas/FasL in salivary glands of NOD mice with Sjogren's syndrome and their mRNA expression.

OBJECTIVETo observe the effect of nourishing Yin, strengthening Qi and activating blood decoction on Fas/FasL in salivary glands of NOD mice with Sjogren's syndrome and their mRNA expression.
METHODThirty-two NOD mice were randomly divided into the model group, the traditional Chinese medicine group (TCM group, orally given 0.4 mL nourishing Yin, strengthening Qi and activating blood decoction as per 100 g x kg(-1) everyday), the hydroxychloroquine group (given 0.4 mL hydroxychloroquine as per 60 mg x kg(-1) everyday), the traditional Chinese medicine and western medicine group (TCM WM group, given nourishing Yin, Strengthening Qi and activating blood decoction 50 g x kg(-1) and hydroxychloroquine 60 mg x kg(-1), 0.4 mL everyday), with eight mice in each group. Eight Balb/C mice were selected as the normal control group (normal group). All of mice were killed after eight weeks, and their submaxillary glands were dissected. The expression levels of Fas/FasL were examined by immunohistochemical method, and the FasL mRNA was detected by RT-PCR.
RESULTThe expression levels of Fas/FasL in salivary glands of the model group were higher than that of other groups (P < 0.05). The expression level of FasL of the normal group was much lower than that in the hydroxychloroquine group (P < 0.05). The relative expression level of Fas mRNA in salivary glands of the model group was higher than that in other groups, but the control group was notably lower than other groups (P < 0.05). The expression level of FasL mRNA in salivary glands of the model group was higher than that in TCM and TCM WM groups (P < 0.05). But the expression level in TCM WM group was notably lower than the hydroxychloroquine group (P < 0.05).
CONCLUSIONThe nourishing Yin, strengthening Qi and activating blood decoction could down-regulate the expression level of Fas/FasL in salivary glands of NOD mice with Sjogren's syndrome and their mRNA expression, and had a better efficacy after being combined with hydroxychloroquine. The nourishing Yin, strengthening Qi and activating blood decoction might treat the Sjogren's Syndrome by reducing apoptosis which is regulated by Fas/FasL

5.Manipulative reduction and percutaneous K-wires fixation for treatment of supracondylar fractures of the humerus in children.

OBJECTIVETo explore clinical effect of manipulative reduction and percutaneous K-wires fixation in treating supracondylar fractures of the humerus in children.
METHODSFrom July 2010 to December 2012, clinical data of 52 children with supracondylar fractures of the humerus, which treated with manipulative reduction and percutaneous K-wires fixation, were retrospectively analyzed. Among them, there were 35 males and 17 females with an average age of 6.7 (ranged from 2.5 to 12) years old. All fractures were type Garland II - III fractures, and 51 cases were extension type and 1 case were flexion type. Flynn evaluation standard of elbow performance score were applied to evaluate clinical effects.
RESULTSAll patients were followed up from 12 to 18 months with average of 16 months. According to Flynn evaluation standard of elbow performance score, 41 cases obtained excellent result, 8 good and 3 moderate.
CONCLUSIONManipulative reduction and percutaneous K-wires fixation for the treatment of supracondylar fractures of the humerus in children has many advantages, such as minimally invasive, rapid recovery, stable fixation. It could prevent osteofascial compartment syndrome, Volkmann Contracture and cubitus varus.

6.Construction of fast propagation system of Houttuynia cordata new line.

OBJECTIVETo construct the fast propagation system of Houttuynia cordata Thunb. new line by selecting the proper type and concentration of plant growth regulators and proper explants.
METHODThe different explants of Houttuynia cordata new line were cultured on MS media with different concentration of different plant grow regulators.
RESULTStem node over-ground and shoot tip were optimum explants for fast propagation. The stem which node grew on MS medium with KT at 0.5 mg x L(-1) could induce more leaves and grow higher. The shoot tip cultured on MS medium with 1.0 mg x L(-1) 6-BA could induce better callus. Adventitious shoots were better achieved from over-ground nodal explants cultured on MS medium supplemented with 6-BA at 1.0 mg x L(-1) and NAA at 0.2 mg x L(-1), or 6-BA at 0.5 mg x L(-1) and NAA at 0.2 mg x L(-1). MS medium with IAA at 0.2 mg x L(-1) was the best one on inducing roots.
CONCLUSIONThe propagat coefficient can be highly improved by inducing adventitious shoots through stem node over-ground, and thus plentiful seeds for production can be provided.

The feasibility of the case-based learning (CBL) of Acupuncture-Moxibustion Techniques is discussed in the aspects of the connotation application, implementation process and effect evaluation of CBL in teaching this course. The result shows that compared with the traditional teaching model, CBL achieves the same effects on the mastery of theoretic knowledge on the techniques of acupuncture, moxibustion and cupping for the undergraduates. Moreover, CBL presentes the obvious advantages on the improvements of the ability of the manipulation selection based on the clinical symptoms and the practical manipulative skills of the students. Hence, CBL is deserved to be promoted as an experimental unit in the professional technique courses of acupuncture-moxibustion.

8.Effect of Chinese herbal medicine for nourishing yin, supplementing qi, and activating blood on the Th1/Th2 immune balance in peripheral blood in patients with primary Sjogren's syndrome.

OBJECTIVETo observe the effect of Chinese herbal medicine for nourishing yin, supplementing qi, and activating blood on the expression of interferon-γ (IFN-γ)/interleukin-4 (IL-4) in peripheral blood and disease activity in primary Sjogren's syndrome (pSS) patients, and to study the relationship between the immune balance of Th1/Th2 and the disease activity.
METHODSA total of 66 pSS patients were randomized with tossing coins method into two groups: the integrative therapy group (34 cases) and the control group (32 cases); and 28 healthy subjects were taken as the normal group. The integrative therapy group was treated by Chinese herbal medicines for nourishing yin, supplementing qi, and activating blood combined with hydroxychloroquine sulfate tablets and the control group was treated with hydroxychloroquine sulfate tablets. The treatment course was 3 months for both groups. The levels of serum immunoglobulin G (IgG), erythrocyte sedimentation rate (ESR), IFN-γ and IL-4 in peripheral blood were measured before and after treatment.
RESULTSCompared with the normal group, the levels of IgG, ESR, IFN-γ and IL-4 were significantly increased in pSS patients (P<0.05). Remarkably, after 3 months of treatment, these levels were dramatically decreased in both the integrative therapy group and the control group, although still higher than the normal group. The levels of IgG, ESR, IFN-γ and IL-4 in the integrative therapy group were lower than the control group and the same group before treatment (P<0.05). The ratio of IFN-γ/IL-4 also significantly decreased after treatment. Moreover, the level of IFN-γ and the ratio of IFN-γ/IL-4 in the integrative therapy group were significantly lower than the control group (P<0.05). For all patients the ratio of IFN-γ/IL-4 before and after treatment was positive correlated with the levels of IgG and ESR.
CONCLUSIONChinese herbal medicine for nourishing yin, supplementing qi, and activating blood can alleviate the disease activity of pSS by regulating the immune balance of Th1/Th2.

9.Segmentectomy for hepatoma originated from segment IX.

OBJECTIVETo report the procedure of segmentectomy for hepatoma located in segment IX.
METHODS11 cases of hepatoma originated from segment IX were treated by segmentectomy without interruption of blood flow of the liver. Among total 11 cases, 10 cases were primary liver cancer, the other one was secondary liver cancer.
RESULTSTumor diameters from 6 to 14 cm (median 9.2 cm), no perioperative death occurred in this group. Intraoperative blood losses were 200-600 ml (median 350 ml) without severe postoperative complications. Postoperative hospitalization time were 9-14 days (median 11 days). Transhepatic artery chemoembolization (TACE) was given at 4-6 weeks after operation and repeated at intervals of 2 to 4 months for 1 year. During the follow up time of 5-29 months (media 17 months), 10 patients were tumor-free and 1 patient developed an intrahepatic metastasis.
CONCLUSIONSSegmentectomy without interruption of blood flow of the liver is safe and practical for hepatoma located in segment IX.

10.Therapeutic effect of Chinese herbal medicine for strengthening qi, nourishing yin, and removing stasis on serum osteopontin and quality of life of patients with primary Sjogren's syndrome.

OBJECTIVETo observe the therapeutic effect of Chinese herbal medicine for strengthening qi, nourishing yin, and removing stasis on serum osteopontin (OPN) and quality of life (QOL) in patients with primary Sjogren's syndrome (pSS) and to study the correlation between OPN level and the disease.
METHODSSixty-eight pSS patients were randomly assigned to two groups: the treatment group (35 cases) treated by Chinese herbal medicine for strengthening qi, nourishing yin, and removing stasis combined with hydroxychloroquine sulfate tablet (HCQ) and the control group (33 cases) treated by HCQ only. Both were treated for 3 months. Before and after treatment, immunoglobulin G (IgG) level, erythrocyte sedimentation rate (ESR), and OPN level were measured. The QOL in patients was evaluated using the Short Form-36 Health Survey (SF-36) before and after treatment. Thirty healthy females were taken as the normal control.
RESULTSBefore treatment, levels of IgG, ESR, and OPN in patients were higher than those in the normal control. After 3 months of treatment, those in both treatment groups decreased but were lower in the treatment group than those in the control group (P<0.05). The scores of 8 dimensions of SF-36 were lower in the pSS patients than those in the normal control (P<0.05) and higher in the treatment group than in the control group after treatment (P<0.05). Furthermore, there was a positive correlation between the levels of OPN and the levels of IgG and ESR, and a negative correlation between OPN levels and the overall score of SF-36 (P<0.05).
CONCLUSIONSChinese herbal medicine for strengthening qi, nourishing yin, and removing stasis could alleviate pSS disease and improve the QOL. In addition, the OPN level might be used as an evaluating index for pSS disease.
