BACKGROUND:The biological extracellular matrix materials become the focus of pelvic floor repair materials research because of its excellent biocompatibility and mechanical compatibility. However, bacterial infection can damage the function of biological repair materials. <br> OBJECTIVE:To prepare the biologic antimicrobial materials for pelvic tissue function repair. <br> METHODS:Chitosan and tigecycline-loaded chitosan nanoparticles suspension was prepared by the electrostatic adsorption and self-aggregation of nanoparticles preparation techniques. Then the suspension was coated onto the surface of extracellular matrix materials. The morphological examination was performed by scanning electron microscope. The antibacterial property was detected by solid plate microbial culture method. <br> RESULTS AND CONCLUSION:Infrared spectra displayed that aromatic ring skeleton vibration peak of chitosan occurred on the biologic antimicrobial materials, and it was significantly widened at about 3 359 cm-1, indicating the composite coating was successful to modify the surface of extracellular matrix materials. As a very smal dose of tigecycline, there was no characteristic absorption peak on the infrared spectra. Scanning electron microscopy showed the porous structure of the material surface with some nanoparticles adhesion. The prepared materials had good antibacterial properties on Staphylococcus aureus and Escherichia coli, and the inhibition zone diameter was significantly increased with the increasing concentration of antibacterial agent. The biologic antimicrobial materials for pelvic tissue function repair were prepared successful y.

Background Researches demonstrated that CpG ODN,a immunostimulatory sequences, has preventing and treating effect on allergic conjunctivitis caused by protein allergen. However, its effect on allergic conjunctivitis caused by fungal allergen is unclear. Objective This study aimed to investigate into whether the Th1-Th2 switching immunostimulatory CpG ODN could reverse the response in the murine allergic conjunctivitis model caused by aspergillus fumigatus. Methods A mixture of spores and hyphae of aspergillus fumigatus strain was used to induce allergic conjunctivitis in male BALB/C mice aged 6-8 weeks. This experiment was designed into preventive or therapeutical treatment program.Under both settings, allergic conjunctivitis of the animals were treated with CpG ODN,nonstimulatory GpC ODN or PBS.After the last challenge with the allergen, the clinical symptoms of the animals were scored based on the criteria of Magone. The animals were sacrificed and the histopathological examination of conjunctiva was performed.Expression of TLR4 mRNA in conjunctiva was analyzed by real-time PCR assay. The responsiveness and populations of lymphocytes in spleen and draining lymph nodes were analyzed by flow cytometry. The use complied with the Standard of Association for Research in Vision and Ophthalmology. Results In the prevention mode.CpG ODN decreased subconjunctival infiltration compared with GpC ODN and PBS groups with the average neutrophil count index(21.25 ±11.59/section,30.75 ±11.44 section and 69.00±9.90/section,respectively).Expression of TLR4 mRNA was up-regulated significantly by CpG ODN.The clinical scores for CpG ODN group were insignificantly lower than those in GpC ODN group and PBS group(P>0.05).In the therapeutic mode, compared with GpC ODN and PBS groups, the allergic symptom score in CpG ODN group manifested significantly lower(t=4.000.t=2.750,P<0.01)and showed fewer cellular infiltration(t=4.870,t=3.829,P<0.01)and higher expression of TLR4 mRNA(P<0.01).In cultured splenic and draining lymph node cells, increased percentages of CD4+ CD25+ and CD4+ CD25+ CD69+ in CpG ODN group were observed compared with control groups(|P<0.05). Conclusion CpG ODN can relieve aspergillus fumigatus-induced allergic conjunctivitis via either subconjunctival injection or topical application by upregulating expression of TLR4 and activating Treg lymphocytes.

Objective To study early changes in biochemical markers of bone turnover for prodicting the bone mineral density(BMD) response to alendronate therapy in Chinese men with osteoporosis. Methods Seventy-eight men aged 60 to 82 years with osteoporosis [mean age (69.8 ±11.8) years]were treated with alendronate sodium 70 mg/week for 12 months. Serum bone gla-protein (BGP) and serum pyridinoline-eross-linked earboxyterminal telopeptide of type I collagen (ICTP) level were measured by chemiluminescence (equipment is Roche E170). Serum BGP, ICTP and BSALP levels were measured before and 3 months and 12 months after treatment. BMD in lumbar spine and femoral neck were measured with dual energy X-ray absorptiometry (GE PRODIGY Inc. ) at L1-4 and at the left hip(total hip, trochanter,Ward's area and femoral neck)before and 12 months after treatment. Results After 3-months and 12-months treatment, the percent reductions of serum ICTP levels were 45.8% and 51.6%, serum BGP level 32.0% and 37.5%, serum BSALP level 35. 3% and 39.9% ,respectively. After 12-months treatment, the percent increase of lumbar BMD was 11.8%, femoral neck BMD 11.4 %. The percent reductions of serum ICTP level at months 3 and 12 after treatment were positively correlated to the percent increase of lumbar BMD (r=0.28, 0.295,P <0.05 and P<0.01)and of fermoral neck BMD at 12 months after treatment(r=0.262, 0.333, P<0.05 and P<0.01)respectively. The percent reductions of serum BGP level at months 3 and 12 after treatment were positively correlated to the percent increase of lumbar BMD (r=0. 322, 0.401,all P<0.01) and of fermoral neck BMD at 12 months after treatment (r=0.277,0.284, all P<0.05)respectively. The percent reductions of serum BSALP level at months 3 and 12 after treatment were positively correlated to the percent increase of lumbar BMD (r=0.133,0.231,all P<0.05) and of fermoral neck BMD at 12 months after treatment(r=0.248, 0.317, all P<0.01)respectively.Conclusions Early changes in biochemical markers of bone turnover rates can predict BMD response to alendronate in Chinese men with osteoporosis.

Journal of Zhejiang University. Science. B 2009;10(1):46-51
BACKGROUNDCapsule endoscopy is a novel non-invasive method for visualization of the entire small bowel. The diagnostic yield of capsule endoscopy depends on the quality of visualization of the small bowel mucosa and its complete passage through the small bowel. To date, there is no standardized protocol for bowel preparation before capsule endoscopy. The addition of simethicone in the bowel preparation for the purpose of reducing air bubbles in the intestinal lumen had only been studied by a few investigators.
METHODSSixty-four participants were randomly divided into two groups to receive a bowel preparation of polyethylene glycol (PEG) solution (Group 1) and both PEG solution and simethicone (Group 2). The PEG solution and simethicone were taken the night before and 20 min prior to capsule endoscopy, respectively. Frames taken in the small intestine were examined and scored for luminal bubbles by two professional capsule endoscopists. Gastric emptying time and small bowel transit time were also recorded.
RESULTSSimethicone significantly reduced luminal bubbles both in the proximal and distal small intestines. The mean time proportions with slight bubbles in the proximal and distal intestines in Group 2 were 97.1% and 99.0%, respectively, compared with 67.2% (P<0.001) and 68.8% (P<0.001) in Group 1. Simethicone had no effect on mean gastric emptying time, 32.08 min in Group 2 compared with 30.88 min in Group 1 (P=0.868), but it did increase mean small intestinal transit time from 227.28 to 281.84 min (P=0.003).
CONCLUSIONBowel preparation with both PEG and simethicone significantly reduced bubbles in the intestinal lumen and improved the visualization of the small bowel by capsule endoscopy without any side effects observed.
