1.Effects of aging on ventricular remodeling and cardiac rupture after acute myocardial infarction in mice

Objective To explore the effects of aging on ventricular remodeling and cardiac rupture after acute myocardial infarction in mice. Methods Male C57BL/6 mice of 3 months and 12 months old were randomly divided into sham operation group and myocardial infarction(MI)group.Following acute myocardial infarction(AMI)modeling induced by open-chest surgery,the events of cardiac rupture were monitored and the echocardiography and hemodynamics were performed on the 7th day after surgery.Zymography,immunohistochemical method and pathological staining were used to measure the activity of matrix metalloproteinases(MMPs),the content of collagen and the degree of inflammatory cell infiltration on the 3rd and 7th days after surgery,respectively. Results The incidence of cardiac rupture was higher in elderly group than that in young group(38.0% vs.16.0%,X2=6.139,P<0.05).Compared with young group,significant infarct expansion,left ventricular (LV)remodeling and hemodynamic deterioration were showed in elderly group on the 7th day after surgery(t=5.754,P<0.05).The degree of inflammatory cell infiltration and the expression of MMP-9 were significantly increased in elderly group on the 3rd day following AMI modeling(P<0.05),and the collagen content and the expression of type Ⅲ collagen were significantly increased (P<0.05)compared with young group. Conclusions Aging is a risk factor for post-infarct cardiac rupture in the mice model.The mechanisms which are responsible for this age-related difference of cardiac rupture are related to increasing degree of inflammatory cell infiltration, overexpression of MMP-9 and type Ⅲ collagen and aggravated early LV remodeling.
2.Recombinant adeno-associated virus serotyoe 9 with CaMEK gene transfected on myocardial cells leads to reduce I/R-induced apoptosis

Objective To determine whether recombinant adeno-associated virus serotyoe 9 with CaMEK gene transfected on myocardial cells leads to reduce I/R-induced apoptosis.Methods Establish an ischemic/reperfusion(I/R) model of myocardial cells in vitro and the cells were divided into four experimental groups:(1) control group;(2) I/R group;(3) I/R+rAAV9-CBA-CaMEK group;(4) I/R+rAAV9-CBACaMEK+PD98059 group,respectively.The P-ERK1/2 and the Caspase-3,Bax were quqntitated by Western blot.Results These data clearly demonstrate that AAV9-mediated CaMEK gene transfected lead to active ERK1/2 and reduce I/R-induced apoptosis notablely.Conclusion rAAV9-mediated CaMEK gene transfected on myocardial cells can reduce I/R-induced apoptosis.
3.Uncoupling protein and nonalcoholic fatty liver disease.

OBJECTIVETo review the current advances on the role of uncoupling protein (UCP) in the pathogenesis and progress of nonalcoholic fatty liver disease (NAFLD).
DATA SOURCESA comprehensive search of the PubMed literature without restriction on the publication date was carried out using keywords such as UCP and NAFLD.
STUDY SELECTIONArticles containing information related to NAFLD and UCP were selected and carefully analyzed.
RESULTSThe typical concepts, up-to-date findings, and existing controversies of UCP2 in NAFLD were summarized. Besides, the effect of a novel subtype of UCP (hepatocellular down regulated mitochondrial carrier protein, HDMCP) in NAFLD was also analyzed. Finally, the concept that any mitochondrial inner membrane carrier protein may have, more or less, the uncoupling ability was reinforced.
CONCLUSIONSConsidering the importance of NAFLD in clinics and UCP in energy metabolism, we believe that this review may raise research enthusiasm on the effect of UCP in NAFLD and provide a novel mechanism and therapeutic target for NAFLD.

4.Effects of over-expressing resistin on glucose and lipid metabolism in mice.

Resistin, a newly discovered peptide hormone mainly secreted by adipose tissues, is present at high levels in serum of obese mice and may be a potential link between obesity and insulin resistance in rodents. However, some studies of rat and mouse models have associated insulin resistance and obesity with decreased resistin expression. In humans, no relationship between resistin level and insulin resistance or adiposity was observed. This suggests that additional studies are necessary to determine the specific role of resistin in the regulation of energy metabolism and adipogenesis. In the present study, we investigated the effect of resistin in vivo on glucose and lipid metabolism by over-expressing resistin in mice by intramuscular injection of a recombinant eukaryotic expression vector pcDNA3.1-Retn encoding porcine resistin gene. After injection, serum resistin and serum glucose (GLU) levels were significantly increased in the pcDNA3.1-Retn-treated mice; there was an obvious difference in total cholesterol (TC) level between the experiment and the control groups on Day 30. In pcDNA3.1-Retn-treated mice, both free fatty acid (FFA) and high density lipoprotein (HDL) cholesterol levels were markedly lower than those of control, whereas HDL cholesterol and triglyceride (TG) levels did not differ between the two groups. Furthermore, lipase activity was expressly lower on Day 20. Our data suggest that resistin over-expressed in mice might be responsible for insulin resistance and parameters related to glucose and lipid metabolism were changed accordingly.

5.Clinical trial of naloxone injection combined with aminophylline injection in the treatment of acute respiratory failure in children

Objective To observe the clinical efficacy and safety of naloxone combined with aminophylline in the treatment of acute respiratory failure in children.Methods A total of 120 children with acute respiratory failure were randomly divided into the control group and the treatment group,with 60 cases in each group.The control group was given conventional treatment (3 mg nikethamide + 500 mL of normal saline or 5% glucose solution for intravenous infusion,50 mL · h-1,bid).The treatment group was additionally treated with intravenous injection of naloxone 0.01 ~ 0.05 mg kg-1 combined with aminophylline 4 ~8 mg,50 mL · h-1,bid,on the basis of the control group.Both groups were treated for 7 d.The clinical efficacy,blood gas parameters before and after treatment and the incidence of adverse drug reactions were compared between the two groups.Results After treatment,the total effective rates of the treatment group and the control group were 85.00% (51 cases/60 cases) and 66.67% (40 cases/60 cases),respectively (P < 0.05).After treatment,there were significant differences between the treatment group and the control group in the power of hydrogen (pH) [(7.01 ±0.06) vs (7.11 ±0.29)],arterial partial pressure of oxygen (PaO2) [(68.99 ± 11.44) vs (59.34 ± 10.30) mmHg],arterial partial pressure of carbon dioxide (PaCO2) [(63.24 ± 10.54) vs (70.69 ± 18.98) mmHg],arterial oxygen saturation (SaO2) [(85.57 ± 13.79) vs (69.01 ± 10.03) mmHg] and the ratio of forced expiratory volume in 1 second and forced vital capacity (FEV1/FVC) [(40.43 ± 12.47)% vs (35.07 ± 11.83)%] (P <0.05).The adverse drug reactions of the treatment group mainly included rash,vomiting and convulsions while adverse drug reactions of the control group mainly included rash,vomiting,convulsions and arrhythmia.The incidence rates of adverse drug reactions in the treatment group and the control group were 11.67% (7 cases/60 cases) and 26.67% (16 cases/60 cases),respectively (P <0.05).Conclusion Naloxone combined with aminophylline exerted considerable clinical efficacy with high safety profile in the treatment of acute respiratory failure in children.
6.A Photoelecteochemical Sensor Based on CdS Sensitized Fe:TiO2Nanosheets for Determination of Cu2+

Iron-doped titanium dioxide nanosheets was prepared by hydrothermal method using tetrabutyl titanate (C16H36O4Ti) and iron trifluoride (FeF3) and modified on ITO electrode.ITO/Fe : TiO2/CdS photoelectrochemical sensor was fabricated by successive ionic layer absorption and reaction(SILAR) method for determination of copper ion.The band gap of electode material was narrowed by iron doped TiO2 nanosheets,which made its absorption red-shifted and its response range of light was magnified.Based on the sensitization effect of CdS,the absorption and utilization of visible light of sensor was significantly enhanced and the photoelectric signal was amplified via reducing the recombination of electrons and holes.The sensor displayed excellent analytical performance for detection of copper ion with linear range of 0.2-4.0 μmol/L and 4.0-80.0 μmol/L and with the detection limit of 85 nmol/L.The sensor was used to detect copper ion in tap water,drinking water and Yongjiang river water with recoveries ranging from 94% to 111%.
7.Prevalence on overweight and obesity in Han, Uygur and Hazakh in adults from Xinjiang

Objective To investigate the prevalence and distributing feature of overweight and obesity in Han, Uygur and Hazakh population in adults from Xinjiang. Methods Four-stage selected random samples with maternal age at 35 or over were used to analyze the prevalence and distributing feature of self-reported congestive heart failure in different nationalities, age, sex. The sampled adult population were collected from 6 localities(Urumqi, Kelamayi, Fukang, the Turfan Basin locality, Hetian locality, Yili Hazakh autonomous prefecture), 23 municipalities and 7 locality and 5 autonomous counties in Xinjiang. Results 16 460 people were surveyed. The prevalence rates of overweight and obesity were 36.1% and 26.9% in Han, Uygur and Hazakh population in Xinjiang,respectively from February, 2007. The prevalence rates of overweight and obesity were 41.4% and18.4% in Han population, 34.9% and 28.9% in Uygur population, but 32.8% and 40.1% in Hazakh population. The prevalence rate of overweight and obesity was higher in males(x2= 135.00, P<0.05).The prevalence rates of overweight and obesity were different between different ethnic groups(x2=338.232, P<0.05). The prevalence of overweight was highest in Han population, with the highest seen in Hazakh population. The prevalence rates of overweight and obesity were increasing with age (x2=246.80,P<0.05). The overweight rate in 45-54 year olds and the obesity rate in 55-64 year olds reached their peak values. Results from logistic regression model analyses indicated that the prevalence of overweight and obesity in Xinjiang were statistically associated with age, educational level, jobs, smoking and alcohol consumption. Conclusion The prevalence rates of overweight and obesity were much higher in the population of Xinjiang but different among ethnicities. The prevalence of overweight was the highest in Han male population and the rate of obesity in Hazakh male population was the highest.
8.The expression of platelet-derived growth factor (PDGF) receptor-beta and its correlation with extracellular matrix in hepatic tissue in hepatic fibrosis rats.

OBJECTIVETo investigate the expression of PDGF receptor-beta and its correlation with extracellular matrix in hepatic tissue during hepatic fibrosis.
METHODSThe model of hepatic fibrosis in rats was induced by carbon tetrachloride. PDGF receptor-beta subunit, collagen I, collagen III and a-SMA in hepatic tissues of these rats were examined using immunohistochemistry. The correlation between PDGF receptor-beta subunit and collagen I, III was analyzed using SAS software after the results of immunohistochemistry were semi-quantified.
RESULTSPDGF receptor-beta subunit and a-SMA were not detected in normal controls. Collagen I and III were distributed in the portal tracts and beneath the endothelia of the central veins and of the Disse spaces. Two weeks after CCl4 injection, the PDGF receptor-beta and a-SMA were detected, and the expression of collagen I and III increased. At the end of 4 and 6 weeks, the above four proteins were further increased. Two weeks after CCl4 injection, PDGF receptor-beta had no apparent correlation with collagen I and III. However, PDGF receptor-beta had a significant correlation with collagen I and III 2 weeks later, and the correlation coefficient was 0.74 and 0.60 respectively at 4 weeks, and 0.83 and 0.67 respectively at 6 weeks. PDGF receptor-beta had a significant correlation with a-SMA during the whole process of hepatic fibrosis and the correlation coefficient was 0.62, 0.69 and 0.81, respectively at the time of 2, 4 and 6 weeks after CCl4 injection.
CONCLUSIONThe PDGF receptor-beta was overexpressed during the process of hepatic fibrosis development, and it significantly correlated with collagen I and collagen III.

9.Effect of ribozyme against platelet-derived growth factor receptor beta subunit mRNA on the biological characters of hepatic stellate cells.

OBJECTIVETo study the cleavage activity of hammerhead ribozyme targeting at platelet-derived growth factor receptor beta subunit (PDGFR- beta) mRNA in hepatic stellate cells (HSCs) and its effect on the biological characters of HSCs.
METHODSExpression vector of anti-PDGFR- beta ribozyme was constructed and transfected into rat-derived HSC-T6 cells with lipofectin. The positive cell clones were gained by G418 selection. The expression of PDGFR- beta, alpha-smooth muscle actin (alpha-SMA), and type I and type III collagen was detected by means of northern blot, Western blot and immunocytochemical staining respectively. The cell proliferation was determined with MTT colorimetric assay. The cell apoptosis was demonstrated with flow cytometry, acridine orange fluorescence vital staining and transmission electron microscopy.
RESULTSThe expression of PDGFR- beta at mRNA and protein level was markedly reduced in ribozyme-transfected HSCs only 43% to 51% of that in control cells (t > or = 3.957, P < 0.05), and alpha-SMA expression level, type I and type III collagen synthesis ability were also reduced (t > or = 6.790, P < 0.01). The proliferation of ribozyme-transfected HSCs was significantly decreased (t > or = 3.858, P < 0.05), and the proliferation response to PDGF BB was markedly inhibited. However the apoptotic rate was significantly increased in ribozyme-transfected HSCs (chi2 > or = 14.157, P < 0.01), and typical apoptotic cells could be found under transmission electron microscopy.
CONCLUSIONSThe anti-PDGFR- beta ribozyme can be expressed stably in HSCs, cleave the target RNA effectively, inhibit HSCs proliferation and collagen synthesis, and induce HSC apoptosis. The results suggest that inhibiting PDGFR- beta expression in HSCs may be a new therapy for liver fibrosis.

10.Effect of Xuezhikang Capsule on serum tumor necrosis factor-alpha and interleukin-6 in patients with nonalcoholic fatty liver disease and hyperlipidemia.

OBJECTIVETo evaluate the effect of Xuezhikang Capsule on the serum levels of inflammatory factors such as tumor necrosis factor-alpha (TNF-alpha) and interleukin-6 (IL-6) in patients with nonalcoholic fatty liver disease (NAFLD) and hyperlipidemia, and to explore whether it has anti-inflammatory effect.
METHODSA total of 84 patients were randomly assigned to two groups with stratified block randomization, the treatment group (42 cases) and the control group (42 cases). They were treated with Xuezhikang Capsule and polyene phosphatidylcholine capsule for twenty-four weeks, respectively. The changes in serum TNF-alpha and IL-6 were measured by enzyme linked immunosorbent assay before treatment and at the 12th and 24th week.
RESULTSCompared with those before treatment, the serum levels of TNF-alpha and IL-6 significantly decreased in both groups after treatment (P<0.01). There was no significant change between the two groups for the treatments at different time points (P>0.05) and between the two groups for treatments at the same time points (P>0.05).
CONCLUSIONXuezhikang Capsule can inhibit the serum inflammatory factor in patients with NAFLD and hyperlipidemia.
