1.Validity and reliability of social avoidance and distress scale (in Chinese version) in Mongolian college students

Objective To evaluate the validity and reliability of social avoidance and distress scale (SAD) for conforming it' s applicability in Mongolian college students.Methods Social avoidance and distress scale (SAD)security scale (SQ),self-esteem scale (SES) and life satisfaction scale (AULC) was used to measure 399 undergraduates whose can understand Mongolian and Chinese language.Results The total scale reliability was 0.872,avoidance scale was 0.726,distress scale was 0.742,total scale heavy reliability (r) was 0.86,avoidance scale (r) was 0.789,distress scale (r) was 0.814.(2)the confirmatory factor analysis verified the effectiveness of the structure,the revised model fitting index x2/df=4.864,NFI =0.92,TLI =0.90,GFI =0.90,IFI =0.87,GFI =0.92,AGFI =0.93,RMSEA =0.06,model fitting was better.(3) There was a significant positive correlation between avoidance scale and distress scale (r=0.634,P<0.01),and there was a negative correlation (r=0.202-0.499,P<0.01) between avoidance scale and SES,SQ and AULC.There was a negative correlation between distress scale and SES,SQ and AULC (r=0.192-0.556,P<0.01).Conclusion SAD (Chinese version) has a good reliability and validity in Mongolian college students.
2.Effects of two methods for deep endotracheal sputum suction

Objective To explore the effects of two methods for deep endotracheal suction.Methods Forty patients who had ineffective airway clearance or lung infections or refused suction were randomly divided into observation group (n=21) and control group (n=19).Temporay airway was established by using oropharyngeal airway.Then,the control group received subglottic suction after pading the shoulder with ordinary pillow (≤10cm),while the observation group received subglottic suction after pading the shoulder with homemade pillow (=15cm).The two groups were compared in terms of sputum suction effect and mucosal injury.Results There was no incidence of mucosal injury in the observation group but three in the control group.The times of suctioning in the observation group were fewer than those of the control group and the volume for suctioning was larger as well (allP<0.05).Conclusion Deep endotracheal suction through oropharyngeal airway,shoulder elevation for 15cm,temporary indwelling of suction tubes can achieve better suction effect without corresponding complications.
3.Silence of BRAF gene in human melanoma cells by plasmid mediated shRNA

Objective To construct the short hairpin RNA (shRNA)-expressing plasmid vectors specific for BRAF gene, and to test their effects in BRAF knockdown in human melanoma cell lines. Methods Two pairs of specific BRAF shRNA oligoes and a pair of randomly synthesized non-specific shRNA oligo were synthesized and inserted into plasmid pGenesil-1. Their fidelity was confirmed by double endonuclease digestion and sequencing. The constructed plasmids were transfected into human melanoma cell lines A375 and M14. The expression of BRAF mRNA and BRAF protein were detected by RT-PCR and Western blotting, respectively. Results The designed shRNA oligoes were precisely cloned into the plasmid pGenesil-1. The expression of BRAF mRNA and protein were down-regulated by specific plasmid braf 1 and braf 2, except to non-specific plasmid neg. The plasmid braf 1 was more effective, reducing BRAF gene expression by 90 per cent. Conclusions Plasmid mediated shRNA could successfully knockdown BRAF expression in human melanoma cells, and the suppression of the gene expression could maintain for 1 month at least.

Articular cartilage defects are common, which is one of the important factors leading to joint degeneration. Due to lack of vascular supply, the ability to regenerate itself is limited. SO the surgeons try a variety of ways to repair these defects. What specific methods are adopted should be based on the pathological types of cartilage defect in order to develop optimal strategies.
5.Influence of bone morphogenetic protein on articular cartilage regeneration after periosteal grafting

AIM: Bone morphogenetic protein (BMP) as polyphenic morphogen can induce the formation of bone and cartilage. This study investigates the effect of BMP on articular cartilage regeneration after periosteal graft. METHODS: Experiments were performed at the Animal Laboratory (absl-3) of Weifang People's Hospital from September 2006 to January 2007. Sixteen New Zealand white rabbits (32 knees) (2.5-3.0 kg) were divided into experimental and control groups randomly, each 8 rabbits (16 knees). The 3.5 mm in diameter of full-thickness articular cartilage defect was made on femoral intercondylar fossa in all rabbits, and 3.5 mm in diameter of periosteum was cut out from the anteromedial part of the upper tibial bone. In the experimental group, the cartilage defect was covered with periosteum, into which 20 ?g BMP and 20% Pluronic were injected. In the control group, the cartilage defect was covered with periosteum, into which the same dosages of 9 g/L saline and 20% Pluronic were injected. All the rabbits were sacrificed in 4, 8 and 12 weeks postoperatively. Motion of joint, conjunction of repair tissue and perienchyma were examined macroscopically. Haematoxylin-eosin staining and toluidine blue staining were used to observe the characteristics of repair tissues. Histological scores on samples in each group were measured by Wakitani score standard at different time points with light microscope. Ultramicrostructure of transplanted tissues was observed with scanning electron microscopy (SEM). RESULTS: Sixteen rabbits were included in the final analysis. Macroscopic observation: 4 weeks after the surgery, the defect was covered with tissue like cartage in the experimental group, and with periosteum in the control group. 8 weeks after the surgery, the surface of the defect was smooth, with boundary unclear in the experimental group. In the control group, the outcome was the opposite. In 12 weeks, cartilage had formed in the experimental group, and tissue like cartilage began to happen in control one. Histological observation: 4 weeks after the surgery, the defect was filled with cells and matrix with abundant proliferation of periosteal cambium layer in the experimental group, and slight proliferation in the control group. 8 weeks after the surgery, the periosteum in the experimental group became fibrocartilage with little hyaline cartilage. Just little fibrocartilage with on hyaline one was detected in the control group. In 12 weeks, the repair tissue in the experimental group approached to normal cartilage. Objective To study the correlation between 8-Iosmerie Porastglnadin-2a(8-iso-PGF2α) 、hypersensitive C-reactive protein(hs-CRP) and coronary heart disease(CHD). Methods 153 CHD patients were divided into 3 groups,including 52 cases of acute myocardial infarction(AMI) ,50 cases of unstable angina(UAP) ,51 cases of stable angina(SAP) and control group consisted of 50 healthy people. The levels of hs-CRP and 8-iso-PGF2α were measured. Person correlation analysis was used to analyze the relationship between the level of hs-CRP and 8-isoPGF2α. Results The levels of hs-CRP and 8-iso-PGF2α were significantly higher in AMI, UAP and SAP group than those in control group(all P <0.05). Compared with SAP group,the levels of hs-CRP and 8-iso-PGF2α were increased in AMI and UAP groups (all P < 0. 05) . The level of hs-CRP was positively associated with the level of 8-iso-PGF2α. Conclusion hs-CRP and 8-iso-PGF2α should be the markers of coronary atherosclerosis and involved in the process of CHD. The levels of serum hs-CRP and 8-iso-PGF2α were correlated with the severity of CHD.
7.Application value of serum pancreatic isled autoantibodies and biochemical indicators in classification diagnosis of diabetes mellitus

Objective To investigate the application value of serum pancreatic isled autoantibodies and biochemical indicators in classification diagnosis of type 1 diabetes mellitus (T1DM ) and type 2 diabetes mellitus (T2DM ) .Methods The clinical data and laboratory detection results in 99 cases of T1DM and 577 cases of T2DM were retrospectively analyzed .The levels of pancreatic isled autoantibodies and biochemical indicators were compared between the two groups and their characteristics were analyzed .Re‐sults The positive rates of single detection and combine detection of glutamic acid decarboxylase autoantibodies (GADA) ,insulino‐ma‐associated antigen‐2 autoantibodies (IA‐2A ) ,islet cell autoantibodies (ICA ) and ZnT8 autoantibodies (ZnT8A ) in the T1DM group were higher than those in the T2DM group ,the differences were statistically significant (P<0 .05) .The onset age ,fasting and postprandial 2 h CP ,fasting and postprandial 2 h insulin(2 h INS) ,triglyceride(TG) and body mass index (BMI) in the T1DM group were lower than those in the T2DM group ,while the levels of fasting and postprandial 2h blood glucose ,glycosylated hemo‐globin(HbA1c) and high‐density lipoprotein cholesterol(HDL‐C) were higher than those in the T2DM group ,the differences were statistically significant (P<0 .05);however total cholesterol and low‐density lipoprotein cholesterol levels had no statistical differ‐ence between the T1DM group and T2DM group (P>0 .05) .Moreover ,the fasting and postprandial 2 h CP levels in the T1DM group showed decreasing trend as the T1DM course extending ,and the difference had statistical difference among different disease courses ;but the fasting and postprandial 2 h CP levels in the T2DM group had no obvious decreasing trend .The areas under the receiver operating characteristic(ROC) curve of fasting and postprandial 2 h CP for differential diagnosis of T1DM and T2DM in the patients with the disease course < 2 year were 0 .902(95% CI:0 .850-0 .954) and 0 .905(95% CI:0 .852-0 .958) respective‐ly .The suitable threshold value of fasting CP was 0 .283 nmol/L ,its sensitivity and specificity were 82 .6% and 89 .2% ,respective‐ly ,which of postprandial 2 h CP was 0 .421 nmol/L ,its sensitivity and specificity were 84 .8% and 89 .2% respectively . Conclusion T1DM and T2DM are different in onset age ,BMI value ,serum GADA ,IA‐2A ,ICA ,ZnT8A ,insulin ,CP ,glucose , HbA1c ,TG and HDL‐C levels ,which may assist clinic in their classification diagnosis .
8.Anterior cervical discectomy with fusion and posterior cervical expansive open-door laminoplasty for multilevel cervical spondylotic myelopathy: its postoperative stability

BACKGROUND: The treatment of multilevel cervical spondylotic myelopathy (MCSM) aims at sustaining the spinal cord compression and restoring the stability of the cervical vertebrae at most.OBJECTIVE: To analyze the clinical characters of anterior cervical discectomy with fusion and expansive open-door laminoplasty for MCSM.METHODS: Sixty-seven patients with MCSM were divided into two groups, and treated with anterior cervical discectomy with fusion (group A) or posterior cervical expansive open-door laminoplasty (group B). All the patients were followed up for 12 months, and the range of motion of cervical vertebrae, cervical curvature index were observed,as well as the Visual Analogue Scale and Japanese Orthopaedic Association scores were condueted. Moreover, the operation time, blood loss and adverse reactions were recorded.RESULTS AND CONCLUSION: (1) The loss of range of motion of the cervical vertebrae in the group B was significantly less than that in the group A (P < 0.05). (2) The cervical curvature index in the group A was significantly improved (P < 0.05), but the index had no significant change in the group B. (3) The axial systems were significantly improved in both groups, especially in the group A (P < 0.05). (4) The neurological function was significantly improved in both groups (P < 0.05), which showed no significant difference between two groups (P > 0.05). (5) The intraoperative blood loss in the group B was significantly more than that in the group A (P < 0.05). (6) The incidence of hoarseness and dysphagia in the group A was 19%. The incidence of wound infection, cerebrospinal fluid leakage and C5 nerve root palsy in the group B was 9%. (7) These results suggest that during choosing an appropriate method for MCSM,surgeons' skills and patients' situation should be considered.
9.Latent categories of adolescents' short video media use tendency and their relationships with personality traits

Objective:To explore the latent categories of short video media use tendency among adolescents and the influence of personality traits on different categories.Methods:Totally 1 362 adolescents were tested by the five factors of adolescent personality questionnaire and the problematic short video media use scale in March 2022.SPSS 26.0 software was used for data collation and descriptive statistics.Mplus 8.3 software was used for latent profile analysis (LPA) to explore the latent categories of adolescents’ short video media use tendency.Regression mixed model (R3STEP) was used to explore the relationship between different categories of short video media use tendency and personality traits.Results:The short video media use tendency among adolescents was divided into 4 latent categories (by proportion from low to high): " problematic short video media use tendency type" (C4, 11%), " safe short video media use tendency type" (C3, 21%), " social short video media use tendency type" (C1, 29%), and " transitional short video media use tendency type" (C2, 39%). Different personality traits had different effects on the 4 latent categories of short video media use tendency, and using C3 as a reference, openness ( OR=1.95) was a triggering predictor of C1, neuroticism ( OR=0.72) was a protective predictor of C1; cautiousness ( OR=0.46) was a protective predictor of C2, neuroticism ( OR=1.60) was a triggering predictor of C2; cautiousness ( OR=0.33) was a protective predictor of C4, neuroticism ( OR=3.24) was a triggering predictor of C4. Conclusion:There are four different latent categories of Chinese adolescents' short video media use tendency, respectively " safe" , " social" , " transitional" and " problematic" . Further research reveal that, personality traits have a significant impact on the different categories of adolescents' short video media use tendency.This study suggests that educators reasonably guide " transitional" , intervene " problematic" , and focus on helping adolescents with high neuroticism to effectively regulate their negative emotions and reduce the occurrence of problematic short video use.
10.Targeting microRNA-mediated suppression of vascular endothelial growth factor gene expression and proliferation in malignant melanoma cells in vitro.

OBJECTIVETo explore the inhibitory effect of targeting miRNA on the expression of vascular endothelial growth factor (VEGF) and cell proliferation in malignant melanoma (MM) SKmel-28 cells.
METHODSRecombination miRNA plasmid vectors targeting VEGF gene were transfected into SKmel-28 cells via Lipofectamine 2000. The integrity of the inserted fragments was detected using colony PCR and sequence analysis. The expression of VEGF mRNA and protein in SKmel-28 cells was detected by RT-PCR and Western blotting, respectively. MTS assay was used to determine the inhibitory effect of a selected targeting miRNA on SKmel-28 cell proliferation, and the apoptosis of SKmel-28 cells was detected using flow cytometry.
RESULTSTransfection with the targeting miRNAs significantly down-regulated the expressions of VEGF mRNA and protein in SKmel-28 cells (P<0.01), and the miRNA construct X-26-2n-1 showed the highest inhibitory effect. The miRNA X-26-2n-1 significantly suppressed SKmel-28 cell proliferation in a time-dependent manner (P<0.01) and increased the early, late and overall apoptosis rates of the cells (P<0.01).
CONCLUSIONThe targeting miRNA we constructed can effectively suppress the cell proliferation and induce apoptosis of SKmel-28 cells by down-regulating the expressions of VEGF gene.
