Objective To study the body temperature of athlete before and after exercise.Methods 291 cases of different layer athletes were chosed to be the object,51 from national racing boat brigade,60 from the provincial track and field brigade,60 from the amateur long-distance running brigade,60 from the middle-distance race brigade and 60 from the extra-long distance brigade.Before and after exercise,the body temperature was measured under the armpit with electronic thermometer.Results In 5 groups,the body temprature of athlete after exercise was obviously lower than before exercise,the difference showed significant.(P
2.Clinical observation and nursing of thirty-two children with severe hand-foot-mouth disease

Objective To explore the clinical characteristics of severe hand-foot-mouth disease in children and summarize nursing experience.Method The clinical data of 32 children with severe hand-foot-mouth disease were reviewed and analyzed retrospectively. Results After standard diagnosis and treatment,all cases were cured. The average stay in hospital was(8.2±1.6)d.Conclusion The close observation,prevention of infection,skin and oral nursing are of importance for promoting curative effectiveness and prognosis.
3.Inhibition of IKKα binding to IL-1β promoter by p50 in LPS tolerant THP-1 cells

Objective To study the effect of p50 on IKKα at IL-1β promoter in LPS tolerant cells and to reveal the mechanism of the inhibition of IL-1β mRNA by pS0. Methods THP-1 human promono-cyte model of endotoxin tolerance that simulates the sepsis leukocyte phenotype was used. Chromatin immu-noprecipitation assay(CHIP) and real-time PCR were applied to quantify the binding of p50 and IKKα to IL-1βpromoter. IL-1β mRNA transcription was studied after knocking-down of p50 and/or IKKα. Results With LPS stimulation, p50 binding did not reduce but somewhat increased at IL-1β promoter in tolerant THP-1 cells. Knocking-down of p50 increased the transcription of IL-1β mRNA, which revealed the inhibi-tory effect of p50 in tolerant cells. In contrast, the accumulation of IKKα to IL-1β promoter decreased with LPS stimulation in tolerant cells; However, IKKα binding increased after p50 gene knock-down. In the meantime, IL-1β mRNA transcription increased; At last, IL-1β mRNA decreased again after double-knoc-king down of p50 and IKKα. Conclusion p50 is an inhibitory protein at IL-1β promoter in tolerant THP-1 cells. The unresponsiveness of IL-1β mRNA transcription to LPS at least partly results from the inhibition of IKKα binding to IL-1β promoter by p50.

5.Observation and Nursing of thread-burying in acupoint in treating exercise-induced fatigue

Objective To study the observation and nursing for exercise-induced fatigue by thread-burying in acupoint. Methods 41 athletes-in-training were treated by thread-burying in acu-point for 2 courses of treatment, choosing main acupointa such as Guan yuan, Shen yu, Pi yu, Ming men,Zu sanli, San yinjiao. Then the changes of athletes' training and hemoglobin were observed. Results After two courses of treatment, athletes' quality of sleep, mental condition in the morning, diet, desire to train, physical fitness, training quality and concentration improved siguificantly.Level of hemoglobin in-creased distinctly. Conclusions The treatment of thread-burying in acupoint could improve sport fa-tigue. Combination with Chinese and western integrative medicine, dialectical treatment, emotion and diet nurse, western integral appraisal procedure to observe and asepsis operation played a positive part in the recovery from sport fatigue.
6.THE EXPRESSION OF SSEA-1 AND OCT-4 ON HUMAN EMBRYONIC GERM CELLS

Objective To culture human embryonic germ cells in vitro and identify them by observing the expression of the surface markers SSEA-1 and Oct-4. Methods The gonadal ridges and mesenteries of human embryos at 5~9 weeks post-fertilization were isolated and then cultured in vitro.Flow cytometry and immunocytochemistry were applied to detect whether the cells cultured express SSEA-1 and Oct-4.Immunohistochemistry was also carried out to observe the expression of SSEA-1 on the PGCs which locate in the gonadal ridge. Results Both the cells and the cellular colonies cultured in vitro expressed SSEA-1 and Oct-4.The immunohistology study of gonadal ridges also showed that there were many PGCs which were SSEA-1 positive in it.Conclusion The cells we obtained through tissue culturing are undifferentiated human embryonic germ cells,which originate from hPGCs.
7.Placental alpha-microglobulin-1 in vagina liquid to diagnose premature rupture of membranes

Objective To explore the value of placental alpha-microglobulin-1 in vagina liquid to diagnose premature rupture of membranes. Methods A prospective observational study to initial evaluation included both the standard clinical evaluation for rupture of membranes and placental alpha-microglobulin-1 immunoassay. Rupture of membranes was diagnosed if fluid was seen leaking from the cervical os or if two of the following three conditions were present: pooling of fluid, positive nitrazine test, or feming. Rupture of membranes was diagnosed definitively on review of the medical records after delivery. Results Placental alpha-microglobulin-1 immunoassay confirmed rupture of membranes at initial presentation with a sensitivity of 100% (89/89), specificity of 91% (10/11), positive predictive value of 99% (89/90), and negative predictive value of 100% (10/10),false positive rate of 9% (1/11). Placental alpha-nricroglobulin-1 immunoassay was better than the conventional clinical assessment in confirming the diagnosis of rupture ofmembranes (P<0.01). Conclusion Measurement of placental alpha-microglobulin-1 in cervicovaginal secretions is superior to conventional clinical assessment in the diagnosis of rupture of membranes.
8.Scoring methods for liver tissue fibrosis based on ultrasound radio frequency time series.

Trying to provide ultrasonic image-aid measures for quantitative diagnosis and dynamic monitoring of liver fibrosis, we propose two scoring methods for liver fibrosis tissue in vivo , based on ultrasound radio frequency (RF) time series in this paper. Firstly, RF echo signals of human liver were recorded in this study. Then one of the recorded frame RF data was demodulated to be B model image. After that, a region of interest (ROI) in the B model image was selected. For each point in the ROI, its all frame data were acquired so that RF time series were formed. An SMR (size measure relationship) fractal dimension and six spectral features were extracted from RF time series in the ROI. With relative deviation and Fisher's discriminant ratio, seven features were weighted and summed so that the liver tissues' scores were obtained, Score-rd and Score-fisher, respectively. Area under ROC curve (AUC) and a support vector machine (SVM) were used to evaluate whether these scoring methods would be useful in distinguishing normal and cirrhosis tissues. Experimental results are shown as follows: Score-rd's AUC was 0.843, while Score-fisher was 0.816, SVM classification accuracies were both up to 87.5%. This proved that our proposed scoring methods were effective in distinguishing normal and cirrhosis tissues. Score-rd and Score-fisher have potential for clinical applications. They can also provide quantitative references for liver fibrosis diagnosis.

9.Expression of vascular endothelial growth factor in bone marrow mesenchymal stem cells under hypoxic conditions

BACKGROUND:Whether transplanted bone marrow mesenchymal stem cells under hypoxic conditions can survive is crucial for the successful celltransplantation. Therefore, studies on the growth of bone marrow mesenchymal stem cells under hypoxic conditions in vitro can provide experimental evidence for in vivo celltransplantation. OBJECTIVE:To observe the expression of vascular endothelial growth factor in bone marrow mesenchymal stem cells under hypoxia. METHODS:Rat bone marrow mesenchymal stem cells were obtained and cultured, and observed under light microscopy. Passage 3 cells were cultured under normoxia (21%O2) and hypoxia (3%O2 hours. Then cellcounting kit-8 assay and flow cytometry were employed to detect cellproliferation in the two groups. Western blot assay was adopted to detect the expression of hypoxia-inducible factor-1αand vascular endothelial growth factor in the two groups. RESULTS AND CONCLUSION:(1)Rat bone marrow mesenchymal stem cells were obtained and cultured successful y, which were fusiform cells and had uniform shape under the light microscope. (2)The results of cellcounting kit-8 assay showed that the number of cells in the hypoxic group was higher than that in the normoxic group at each time point, and cellviability increased significantly at hours 36 and 48 (P<0.05). (3)The results of flow cytometry demonstrated that the proportion of cells in S phase and cellproliferation index in the hypoxic group were significantly increased, compared with the normoxic group (P<0.05). (4)Western blot results showed ), respectively, for 72 that there was a smal amount of the expression of hypoxia-inducible factor-1αand vascular endothelial growth factor in the normoxic group, but the expression of these two proteins in the hypoxic group was increased in a time-dependent manner (P<0.05). These findings suggest that hypoxia can induce proliferation of rat bone marrow mesenchymal stem cells cultured in vitro, and also raise hypoxia-inducible factor-1αand vascular endothelial growth factor expression in a time-dependent manner.
10.Evaluation of reliability and validity of application of the Chinese version of walking impairment questionnaire in patients with type 2 diabetic

Objective To evaluate the raliability and validity of walking impairment questionnaire applied to assess the walking ability of patients with type 2 diabetes. Methods One hundred and twenty-six patients with type 2 diabetes were selected. WIQ, SF-36 and 6-minute walk test were used to collect da-ta that was conducted for reliability analysis, correlation analysis and independent-samples t test to evaluate the reliability and validity. Results The internal consistency determined by Cronhach's α was 0.867 for the total WIQ score. Significnat correlations were found between WIQ and 6MWD, also between WIQ and physical domains of SF-36. compared with patients over seventy-one years old, the score of WIQ including the distance, speed, stair climbing and total score is significantly higher than that in patients aged seventy-one or less than seventy-one years old. Conclusions The Chinese version of WIQ is a simple, valid and reliable, clinically relevant tool to assess the walking ability of patients with type 2 diabetes.
