1.Role of beta-adrenergic signal transduction pathway on myocardial ischemic preconditioning of rats.

To study the changes in every part of the beta-adrenergic signal transduction pathway and their effects on ischemic preconditioning of rat myocardium in vivo. SD rats were divided into three groups: IP group, I/R group and CON group. The IP group was further divided into PC1-, 2-, 3-, and PC1+, 2+, 3+ groups according to preconditioning procedure. The rats received surgical procedure and underwent left coronary artery occlusion and reperfusion. We analyzed the infarct size by TTC staining, measured serum myocardial enzymes, studied the beta-AR Bmax and Kd by radioligand binding assay of receptors, checked the activity of AC and PKA by the method of biochemistry and examined the content of cAMP by radioimmunoassay. The infarct area was much smaller in the IP group than in the I/R group (P < 0.001), while the enzymes were significantly higher in I/R (P < 0.001). The Bmax of beta-AR in IP was much higher than that in I/R (P < 0. 001), but no difference in Kd could be seen between IP and I/R groups. In IP, the activity of AC and PKA and the content of cAMP were higher than those in I/R (P < 0.05, 0.002 and 0.001, respectively). In the procedure of preconditioning, the content of cAMP and the activity of PKA showed the characteristic of cyclic fluctuation. Ischemic preconditioning can protect the heart from necrosis and reduce endo-enzyme leakage. The system of beta-adrenergic signal transduction pathway probably takes part in the protection effect of the IP, which might be elicited by the PKA.

2.The expression of survivin and P-gp proteins in epithelial ovarian carcinoma and its relationship with multidrug resistance in ovarian carcinoma

Objective To detect the expression of inhibitor of apoptosis protein survivin and mdr1 gene protein P-gp in ovarian epithelial carcinoma,and analyze their functions of drug resistance in chemotherapy and the relationship with pathological characteristics.Methods We detected the expression of survivin and mdr1 proteins in 8 cases of normal ovarian tissues,16 cases of benign epithelial ovarian tumor tissues and 46 cases of epithelial ovarian cancerous tissues(28 cases of serous cystadenocarcinoma,12 cases of mucinous cystadenocarcinoma,and 6 cases of endometrioid carcinoma) with immunohistochemistry.Results The positive rate of survivin and mdr1 proteins in ovarian cancerous tissues was 60.9% and 34.8%,respectively,which was significantly higher than that in benign epithelial ovarian tumor tissues(31.3% and 0%) and normal ovarian tissues(negative)(P
3.Clinical effect on baggy eyelid plasty: a report of 1 050 consecutive cases

Objective To investigate a myocutaneous flap method for eyelid baggy plasty conforming to the medical aesthetic requirements. Methods Before operation the amount of the lower eyelid skin excised should be determined. Asking the patient to look upward at supine position, the operator lifted properly the lower eyelid skin using fine forceps without teeth at the results of smooth superficialness at this region and slight ectropion. The incision lines were marked along the print pinched by forceps and the trace 2 to 3 mm below the ciliary margin. Intra-operatively severing soft issues to the inferior orbital fringe just on the sub-orbicularis level, the skin-muscle flap was made. Then the excessive muscle was removed in the light of the incision lines. The orbital fat teased out resected or filled up the concavity along the lower orbital edge. Results The tense and smooth appearance of lower eyelid was archived in 900 cases received in our hospital except 2 temporary ectropion cases and 2 temporary double vision cases. We repaired the 15 ectropions using adjacent skin flap, tarsal reduction, skin soft tissue expansion, or superficial temporary fascia sling suspension, eliminated 28 haematomaes and made the excision of scar and wound suture for a hypertrophic scar case in the 150 complications from other hospitals. The crow wrinkles were improved apparently. Conclusion The amount of skin excision determined pre-operatively, which not only successfully refrains interfering elements from blooding and swelling but also avoids giving rise to ectropion, is accurate than that performed during operation. Natural appearance after operation becomes better and lasting because of orbicularis raised which strengthens the anterior wall of orbital septum.
4.Combined ultrasound-assisted liposuction and dermolipectomy for abdominoplasty

Objective To study the clinical effects of abdominoplasty using ultrasound assisted liposuction and dermolipectomy. Methods With the patient standing, the relative areas of maximal adipose fullness in the abdomen and waist were marked topographically. With the patient in the supine position, the "pinch test" was performed by surgical operator advancing the tissue in the infraumbilical area toward the pubic region in order to estimate how much skin needed to be resected before operation. Inter-operatively tumescent technique with the transduction of ultrasound into fatty areas was applied in the first place, following classic abdominoplasty including excessive skin and fat resection, musculoaponeurotic plication and umbilical transposition. Results Sixteen patients followed up from six months to two years were satisfied with aesthetic effects because of smoothness of the operated areas, good sensory recovery and obvious shrinkage of skin with fewer complications. There were no infection, skin necrosis, and wound dehiscence. Conclusion The excellent effects of abdominal profile sculpture can be reproduced with the new method using UAL in vivo and dermolipectomy.
5.The effect of thermal ablation therapy on regulatory T cells in hepatocellular carcinoma

Objective To investigate the changes of regulatory T cells (Tregs) in hepatocellular carcinoma(HCC) patients with thermal ablation therapy(TAT),and to explore the mechanisms of immunity enhancement induced by TAT.Methods A total of 36 patients with HCC undergoing TAT (observation group) were enrolled in this study from December 2009 to December 2010.Another 20 healthy person were selected as control group.The percent of Tregs was detected by flow cytometry analysis.The level of serum IL-10 was measured with ELISA.Results Before treatment,the percent of Tregs in TAT group was significantly higher than that in control group [(11.44 ± 2.74)% vs.(2.45 ± 0.67)%] (P < 0.01).After treatment,the percent of Tregs in TAT group decreased significantly[(11.44 ± 2.74)% vs.(5.07 ± 0.82)%] (P < 0.01).The level of serum IL-10 in TAT group [(41.66 ± 9.24) mg/L] was significantly higher than that in control group [(14.32 ± 3.64) mg/L] (P < 0.01).After treatment,the level of serum IL-10 in TAT group decreased significantly[(41.66 ± 9.24) mg/L vs.(25.47 ± 5.43) mg/L] (P < 0.01).Conclusion TAT can enhance immunity in HCC patients,and which may accomplish by down regulation of Treg and IL-10.
6.Application of mini-phlebectomy treatment for varicosis of lower limbs

Objective:To investigate the clinical therapeutic effects on mini-phlebectomy treatment for varicosis of lower limbs.Methods:Of 46 cases(70 limbs) of varicosis of lower limbs,25(38 limbs) received flush ligation and stripping of the great saphenous vein (control group),and 21 (32 limbs) received mini-phlebectomy (mini-phlebectomy group).The data of operating time,bleeding quantity,incision number,incision length and hospital stay time was analysed retrospectively in both groups.Results:The operating time,bleeding quantity,incision number,incision length and hospital syay time of mini-phlebectomy group are much less than that of control group.The cure rate of control group and mini-phlebectomy is 71.9% and 91.3% respectively.The difference of cure rate is significant while they are compared.Conclusion:Mini-phlebectomy has the advantage of minimal invasion,less bleeding,efficacious and time saving,it will be an ideal procedure for varicosis of lower limbs
7.Safety and prognosis of ovarian preservationin young women with early-stage endometrial cancer

Objective The aim of this study is to evaluate the safety and prognosis of ovarian preservation in young women with early-stage endometrial cancer.Methods Women≤45 years of age with stageⅠendometrial cancer recorded from Jan 2005 to Dec 2011 in Peking Union Medical College Hospital were examined.They were further divided into two groups: ovarian preservation group and oophorectomy group.Clinical and pathological recording of these patients were compared.Results A total of 72 women, including 25 patients (34.7%) who had ovarian preservation, were identified.The ovarian preservation group was younger (P=0.007) and had a lower prevalence of lymphadenectomy (P<0.001).While there were no significant difference in stage, tumor grade, the invasion depth of myometrium and adjuvant treatment between two groups (all P>0.05).Of seventy-two cases, five patients relapsed and all survived after a median follow-up time of 89 months (rang: 7-131 months).The Kaplan-Meier curve and the log rank test showed no difference in recurrence-free survival (P=0.194).In Cox model analysis, ovarian preservation had no effect on recurrence-free survival(HR=3.08, 95% CI 0.54-18.44).Conclusions Ovarian preservation in young women with early-stage endometrial cancer is safe and has no negative effect on recurrence-free survival or overall survival.
8.A retrospective study of the application value of 18 F-FDG PET/CT in patients with fever of unknown origin

Objective To evaluate the application value of 18 F‐FDG PET/CT in patients with fever of unknown origin (FUO) to improve the etiological diagnosis accuracy of PET/CT in patients with FUO and provide better service for clinical prac‐tice .Methods Clinical data and imaging results of 144 cases of FUO patients diagnosed through 18 F‐FDG PET/CT examination in Wuhan union hospital from December 2009 to December 2011 were retrospectively analyzed .The final diagnoses were based on sur‐gical pathology ,aspiration cytology ,or discharge diagnosis combined with more than 12 months of follow‐up results .All the PET/CT images were analyzed and the clinical application value of PET/CT in FUO patients were evaluated through the diagnose rate , diagnose efficiency and clinical staging of malignant tumor .Results Among the 144 patients ,there were 130 cases ultimately had clear diagnosis ,which including 41 cases of malignant tumor ,89 cases of benign lesions and 14 cases the cause of unknown .PET/CT detected abnormal lesions for 97 .6% (40/41) of patients with malignant lesions and 88 .7% (79/89) of patients with benign le‐sions ,and the diagnostic sensitivity ,specificity ,accuracy ,positive predictive value ,and negative predictive value were :97 .6% (40/41) ,86 .5% (77/89) ,90% (117/130) ,76 .9% (40/52) and 98 .7% (77/78) .PET/CT detected the primary lesions for the first time in 46 .3% (19/41) patients with malignant tumor ,and detected more lesions than the traditional imaging methods in 92 .7% (38/41) of these patients .Conclusion 18 F‐FDG PET/CT imaging can provided important reference value for the detection of primary lesions ,the qualitative diagnosis of benign and malignant lesions and clinical staging of malignant lesions .
9.Biocompatibility of bone marrow mesenchymal stem cells on acellular collagen matrix double-layer material

Objective To study the biocompatibility of bone marrow mesenchymal stem cells (BMSCs) on acellular collagen matrix double-layer material.Methods BMSCs with acellular collagen matrix double-layer material were trained as experimental group,while separately cultured BMSCs as control group.The growth and proliferation of BMSCs on acellular collagen matrix double-layer material were investigated.Scanning electron microscope was used to observe the adherence of cells,and cell vitality was detected by CCK-8 method.Results The acellular collagen matrix double-layer material did not influence the growth and proliferation of BMSCs.The difference between the growth curves of experimental group and control group was not significant (P>0.05).Conclusions The acellular collagen matrix double-layer material has acceptable biocompatibility.This research provides a scientific basis for the application of this composite material.
10.Research progress of additives for improving therapeutic peptides and proteins stability in PLGA microspheres

Microspheres made of poly-lactic-co-glycolic acid (PLGA) have been frequently proposed as drug delivery systems.A very significant challenge in the development of controlled PLGA releasing systems is the instability of drugs especially therapeutic peptides and proteins.Additional approaches,particularly the use of additives,are needed to optimize PLGA delivery of drugs.This article reviews the effects of additives,especially the effects of stabilizing protein during the preparation of PLGA microsphere and the sustained drug releasing processes.
