1.Effect of Annonaceous Acetogenin on Proliferation and Apoptosis of Raji Cell

Objective To investigate the effects of annonaceous acetogenin on proliferation and apoptosis in Raji cells and its mechanism.Methods Raji cells cultured in vitro were divided into control group,annonaceous acetogenin group and adriamycin group.Raji cells were effected by 6.25,12.5,25,50 ?g/mL annonaceous acetogenin.Proliferation of Raji cells were evaluated by MTT assays,apoptosis percentage was assessed by flow cytometry.Caspase-9 protein was detected by immunohistochemistry.Results The Raji cell proliferation rate of annonaceous acetogenin decreased compared with the control,that of 25,50 ?g/mL group were lower than adriamycin group,and it was related to the concentration,relying on the incubating time.The apoptosis rate was higher than control,that of 25,50 ?g/mL group were higher than adriamycin group,and it was related to the concentration and the incubating time.The expression of caspase-9 protein of annonaceous acetogenin group was higher than control,and it had a positively relationship with the concentration and incubating time.Conclusions Annonaceous acetogenin could inhibit cell proliferation in a dose-dependent and time-dependent manner in Raji cells,and it may induce Raji cells apoptosis by up-regulating caspase-9 expression.
2.Effect of traditional Chinese medicine in improving quality of life of patients with colorectal cancer

Objective To observe the effect of traditional chinese medicine(TCM)therapy on improving quality of life of patients with colorectal carcinoma(CRC).Methods Patients with CRC treated at the first affiliated hospital to Guangzhou university of traditional Chinese medicine and the second affiliated hospital of Guangzhou medical college from March 2005 to April 2006 were enrolled.90 valid cases were randomly divided into three groups:TCM group(30 eases),integrated traditional and western medicine group (30 cases)and western medicine group(30 cages).The FACT-C were used to evaluate the quality of life(QOL).Results The QOL wag best in integrated traditional and western medicine group.worst in western medicine group.When the trial finished,TCM group and integrated traditional and western medicine group gained a significant ameliorating effect of QOL compared with pre-treatment scores(P<0.05).Conclusion The results prompted that TCM have antagonistic effect on the adverse reactions of chemotherapy and Can improve the patients' quality of life in a certain degree.
3.Correlation between platelet-associated antibody and platelet infusion effects

Objective To investigate the correlation between platelet-associated antibody and platelet infusion effects.Methods SEPSA was used to detect the platelet-associated antibody of 79 patients with multiple transfusions of platelet,the platelet count before and after platelet infusion was monitored and the corrected count increment (CCI) in lh and 24h were calculated,for those judged to be ineffective platelet infusion,the platelet cross-matching was first done and then received matching infusion again and the infusion effects were analysed. Results 47 cases were detected with platelet antibody positive in 79 patients,accounting for 59.5% ,the CCI of lh and 24h after infusion in platelet antibody positive group were significantly lower than the negative group, the difference was statistically significant( t = 2. 462、2. 583, all P < 0. 05 ) ;40 cases in positive group had the invalid with the invalid rate of 85.1%which was significantly higher than the negative group, the difference was statistically significant ( x2 = 34. 46, P <0. 05 ) ;the CCI of 1h and 24h after infusion in matching success group were significantly higher than the unsuccessful group, the difference was statistically significant( t = 2. 152、2. 230, all P < 0. 05) ;27 cases in matching success group had the effective infusion with the effective rate of 87. 1% ,which was significantly higher than the unsuccessful group, the difference was statistically significant ( x2 = 4. 34, P < 0. 05). Conclusion The production of anti-platelet antibody was a major immune factor which lead to platelet transfusion refractoriness ,the blood platelet cooperative infusion could be a good solution for the platelet transfusion refractoriness.
4.Effect of Invigorating Spleen,Clearing Heat and Eliminating Stasis Drugs on EGF and EGFR Expression in Rat Model of Gastric Ulcer

Objective To study the effect and mechanism of invigorating spleen, clearing heat and eliminating stasis drugs on QOUH of gastric mucosa and expression of EGF and EGFR in rat model of gastric ulcer. Methods The rat model of gastric ulcer was constructed by injection of acetic acid. The rats were randomly divided into model group, ranitidine group, invigorating spleen group, clearing heat group, eliminating stasis group, complex prescription group and normal control group. The ulcer index (UI), regenerate mucosal thickness, lamina muscularis mucosal width were observed by HE, and the level of EGF was detected by ELISA. The expression of EGFR in the mucosa around the ulcer was detected by SABC kit immunohistochemical method. 5.Synthesis and preliminary identification of L-alanyl-L-glutamine dip eptide

Objective To explore the method of chemical synth esis of L-alanyl-L-glutamine dipeptide. Methods L-alanyl-L -glutamine dipeptide was prepared by condensation and ammoniation reaction of L -glutamine and 2-D-chloropropionyl chloride. The physico-chemical properties of this dipeptide were identified with melting point determination, amino acid analysis, elements analysis and MS and its purity was determined with HPLC. Results This dipeptide was confirmed good in quality by physico-chem ical determination. Conclusion Dipeptide synthetized by this me thod is simple, convenient and with good guality.
6.ASPS induces G_2/M arrest of H446 cells by activation of ERK signal pathway

Objective To investigate ASPS induced G_2/M arrest in lung cancer cell line H446 and its effect on ERK MAP kinase signal transduction pathways. Methods Cell cycle phases were inspected by flow cytometery (FCM) ; Western blot analysis was used to inspect the proteins of ERK, p-ERK. Results Compared with control group, G_2/M phase cells increased with concentration significantly, G_0/G_1 phase cells were not different, G_2/M phase cells and G_0/G_1 phase cells were not different when pre-incubated with PD98059 prior to exposure to ASPS of different concentrations, protein of p-ERK was significantly increased, expression of ERK was no different. Conclusion ASPS may induce G_2/M arrest of H446 cells possibly by activation ERK MAP kinase pathways.
7.Evaluation of cardiac function by echocardiography and Doppler tissue imaging in patients with myocardial infarction transplanted by autologus monouclear bone marrow cells

Objective To evaluate cardiac function after transplanting autologus monouclear bone marrow cells(MBMCs) in patients with myocardial infarction by echocardiography and Doppler tissue imaging. Methods Six male patients with myocardial infarction who need coronary artery bypass procedure were prospectively studied. Before cell transplantation and in the follow-up, echocardiography and Doppler tissue imaging was performed to observe the regional and global cardiac function of the patients, including infarction wall thickness(IWT), infarction wall movement velocity(IWMV),left ventricular end-diastolic diameter(LVDd), left ventricular end-systolic diameter(LVDs),left ventricular ejection fraction(LVEF), left ventricular shorting fraction(LVFS), ischemic mitral regurgitation. ~ 99m Tc-MIBI SPECT examination included immediate and prolonged image scors. 8.Cochlear implantation in patients with middle ear inflammation

OBJECTIVE Explore the waysof cochlear implantation(CI)surgery technique for bilateral profound sensorineural hearing loss patients with otitis media and mastoiditis. METHODS CI was performed in 29 patients with different of otitis media and mastioditis,26 of them underwent a single-stage operation,and 3 had a two-stage operation,according to the degree and extent of lesion. For patients with mild inflammation,cochlear implantation was performed after complete eradication of inflammation at the same protocol the patients with severe inflammation underwent an intact canal wall tympanomastoidectomy or reconstructed it initially,cochlear implantation was performed after the initial procedures. RESULTS One- stage or tow-stage operations of CI were carried out for 29 patients with otitis media and mastoiditis. All electrodes were implanted successfully,in which the CI went normally and electrode array were protected well. None of the cases showed recurrence of infection during an average follow-up period of 2-8 years. CONCLUSION The CI could be performed in otits media and mastoiditis patients after reversionary surgery . Individual management based on the degree and extent of the local lesion was emphasized.
10.The impulsivity and aggressivity, and its treatment outcomes of the inpatients with internet addiction disorder by intergrafion interview

Objective To explore the impulsivity and aggressivity,and its treatment outcomes of the inpatients with intemet addiction disorder (IAD) by intergration interview. Methods Barratt Impulsiveness Scale ( BIS), Buss Aggressive Scale (BAS) were used to assess the impulsiveness and aggressivity in 58 IAD patients, at the first week and after the point of intergration interview 3 months,54 normal subjects in the control group were conducted by BIS and BAS only when they were recruited. Results ①The intemet addiction group had significantly higher scores on the BIS and BAS total scores than the control group at the first week ( BIS score 99.3 ±15. 1vs 75.0 ± 15.4, t=8.47, P<0. 01; BAS score 78.3 ±24.4 vs 54.8 ± 17.7, t=5.87 , P<0. 01 ). IAD group had lower scores on the total scores of BIS-Ⅱ and BAS after intergration interview than before, but the subscales and total scores were higher than those in the control group ( BIS score 86.3 ± 16.6 vs 75.0 ± 15.4, t =4.98, P < 0.01 ;BAS score 66.2 ± 22.6 vs 54.8 ± 17.7, t = 3.65, P < 0. 01 ). ②Total score of SCL-90 in IAD group were significantly correlated with the BAS total score ( r=0. 376, P<0. 01 ) ,and the decreased SCL-90 total score was also related with the decreased BAS score ( r= 0.508, P< 0. 01 ). Conclusion This study suggests that IAD group exhibit more impulsivity and aggressivity than those in the control group. After interview, their impulsivity and aggressivity are significantly decreased, but are still higher than those in the normal control group.
