1.Effect of Annonaceous Acetogenin on Proliferation and Apoptosis of Raji Cell

Objective To investigate the effects of annonaceous acetogenin on proliferation and apoptosis in Raji cells and its mechanism.Methods Raji cells cultured in vitro were divided into control group,annonaceous acetogenin group and adriamycin group.Raji cells were effected by 6.25,12.5,25,50 ?g/mL annonaceous acetogenin.Proliferation of Raji cells were evaluated by MTT assays,apoptosis percentage was assessed by flow cytometry.Caspase-9 protein was detected by immunohistochemistry.Results The Raji cell proliferation rate of annonaceous acetogenin decreased compared with the control,that of 25,50 ?g/mL group were lower than adriamycin group,and it was related to the concentration,relying on the incubating time.The apoptosis rate was higher than control,that of 25,50 ?g/mL group were higher than adriamycin group,and it was related to the concentration and the incubating time.The expression of caspase-9 protein of annonaceous acetogenin group was higher than control,and it had a positively relationship with the concentration and incubating time.Conclusions Annonaceous acetogenin could inhibit cell proliferation in a dose-dependent and time-dependent manner in Raji cells,and it may induce Raji cells apoptosis by up-regulating caspase-9 expression.
2.Clinical Analysis of 102 Cases of ARDs Induced by Angiotension Converting Enzyme Inhibitors

OBJECTIVE: To analysis the adverse reactions of angiotension converting enzyme inhibitor( ACEI) , so as to deepen the understanding to the ARDs and improve the therapeutic effect of ACEI METHODS: The therapeutic effects and ARDs of ACEI were retrospectively reviewed and the incidence of ARDs induced by ACEI and the relationship between different ACEIs and their ARDs were analyzed RESULTS: ( 1) The incidence of ARDs of ACEI was 12% Stimulating cough, parageusia, gastrointestinal tract discomfort, skin rash, itch of skin, hypotension, headache, dizziness, proteinuria and angioedema were the common symptoms ( 2) Of five kinds of ACEIs, captopril induced the highest incidence of ARDs and stimulating cough, parageusia, gastrointestinal tract discomfort, skin rash and itch of skin were the most common manifestations CONCLUSION: Stimulating cough, parageusia, gastrointestinal tract discomfort and skin rash are the most common symptoms of ARDs, and the symptoms will disappear when drug administration is discontinued
3.Curative efficacy of Xianlinggubao capsules combined with Lugua polypeptide injection in vertebral osteoporosis

Objective To explore curative efficacy of Xianlinggubao capsules combined with Lugua polypeptide injection in treatment of vertebral osteoporosis.Methods 90 patients of vertebral osteoporosis from March 2014 to March 2015 in our haspital were selected as research objects and randomly divided into the observation group and control group,with 45 cases in each group.The control group were treated with Lugua polypeptide injection, while the observation group were treated with Xianlinggubao capsules combined with Lugua polypeptide injection.Then bone mineral density, osteocalcin, visual analogue scale (VAS), Oswestry disability index (ODI) score, curative effect in two groups after treatment were compared.Results Ater treatment, bone mineral density, osteocalcin in observation group was than that in control group [(0.99 ±0.13)g/cm2 vs(0.89 ±0.14)g/cm2, (41.05 ±5.30)μg/L vs(35.80 ±5.23)μg/L],the difference was statistically significant(P<0.05); VAS、ODI score in observation group were lower than those in control group[(1.21 ±0.30)分vs(4.65 ±1.08)分、(18.24 ±4.97)% vs(31.26 ±5.73)%],the difference was statistically significant (P<0.05).The total effective rate of observation group was statistically higher than that in the control group 95.55%(43/45),the difference was statistically significant vs77.77%( 35/45 ) ( P<0.05 ) .Conclusion Xianlinggubao capsules combined with Lugua polypeptide injection is well for vertebral osteoporosis,which can improve the bone metabolism of patients, promote bone growth and improve treatment efficacy.
4.Applying DNA Barcoding Technique to Identify Mori Cortex and Its Adulterants

To find a new method to identify Mori Cortex and its adulterants by analysis their ITS2 sequence of barcode,total genomic DNA was isolated from Mori Cortex and its adulterants. Nuclear DNA ITS2 sequences were amplified,and purified PCR products were sequenced. Sequence assembly and consensus sequence generation were performed using the CodonCode Aligner V3.0. The Kimura 2-Parameter (K2P) distances were calculated us-ing software MEGA 4.0. Identification analyses were performed using BLAST1, Nearest Distance and neighbor-joining (NJ) methods. The results showed the intra-specific genetic distance of Mori Cortex was 0, which were lower than inter-specific genetic distances between Mori Cortex and its closely related species (0.003-0.343). The ITS2 region is an efficient barcode for identification of Mori Cortex and its closely related species, which provides a scientific basis for fast and accurate identification and new method of Mori Cortex.
5.Nasal surgery and upper airway radiofrequency ablation in patients with obstructive sleep apnea hypopnea syndrome.

OBJECTIVE:
To investigate the efficacy and related factors of nasal surgery combined with upper air way radiofrequency ablation(RFA) for treatment of obstructive sleep apnea hypopnea syndrome (OSAHS) with chronic nasal blockage.
METHOD:
One hundred and three mild or moderate OSAHS patients with chronic nasal blockage were recruited, all cases had nasal surgery and upper airway RFA. All patients were evaluated by body mass index (BMI), Epworth sleep scale (ESS), snoring scale, and nocturnal polysomnography (PSG). Eighty-nine patients were reevaluated at least 6 months after surgery with the preoperative methods.
RESULT:
After operation, the apnea and hypopnea index (AHI) decreased from (18.67 +/- 9.48)/h to (9.22 +/- 7.18)/h; the lowest artery oxygen saturation (LSaO2) increased from (0.83 +/- 0.08) to (0.92 +/- 0.06); the Epworth sleep scale(ESS) decreased from (8.74 +/- 5.67) to (5.12 +/- 3.74); the snoring scale decreased from (7.16 +/- 2.85) to (3.56 +/- 2.26), the percentage of time with oxyhemoglobin saturation below 0.90 (CT90) decreased from (18.64 +/- 12.98) to (10.73 +/- 8.29). All of the differences were obvious (P<0.01). Success was defined as a postoperative apnea-hypopnea in dex < 10 events per hour and at least 50% less than the preoperative value. The surgical success rate was 75.3% (67/89). No major perioperative complications occurred.
CONCLUSION
Our findings suggest that nasal surgery combined with upper airway RFA can improve snoring and disease-specific quality of life in patients with anatomic na sal obstruction with mild or moderate OSAHS.

6.Effects of intratympanic injection of dexamethasone on endolymphatic hydrops and changes in guinea pigs inner ear CFTR expression.

OBJECTIVE:
To observe the effect of tympanic injection of dexamethasone in the guinea pig endolymphatic hydrops and the change CFTR expression, to explore the effect of glucocorticoid treatment endolymphatic and its possible mechanism.
METHOD:
Thirty guinea pigs were divided into three groups: hormone group, water group, control group. The animals(hormone group, water group) in study were injected DDAVP 4 μg/kg in the first 7 d, and will increase to 6 μg/kg in the second 3 d. The control group was given normal saline, continuous 10 d. After twelfth days, the hormone group transtympanic injection of dexamethasone (5 mg/ml, 0.5 ml), and water group, control group transtympanic given normal saline (0.5 ml), continuous injection 5 d. Using immuno- histochemistry and Western blot to detect the cystic fibrosis transmembrane conductance regulator cochlear factor (CFTR) expression.
RESULT:
The water group ABR thresholds was significantly higher than that before the experiment (P < 0.01), and the water group was significantly higher than the rest of the groups (P < 0.01); Hormone group compared with the control group increased threshold value (P < 0.05). The control group had no endolym- phatic hydrops, the water group showed varying degrees of endolymphatic hydrops, cochlear duct and vestibular plus cochlear duct area ratio compared with the control group, hormone group was significantly higher (P < 0.01). hormone group area ratio was higher than the control group (P < 0.05). CFTR was primarily expressed in the stria vascularis, Corti's, spiral ligament, basilar membrane, cochlear ganglion,etc . The expression of CFTR in the water group was increased than that in the control group, and the hormone group (P < 0.01); the expression of hormone group increased compared with the control group (P < 0.05).
CONCLUSION
Tympanic injection of dexa- methasone can alleviate the desmopressin acetatein guinea pigs caused by membranous labyrinth, and the improve- ment of the hearing; Tympanic injection of dexamethasone can make the endolymphatic hydrops cochlea of guinea pigs decreased CFTR expression, indicating that the expression and possible mechanisms of CFTR intratympanic steroids reduce endolymphatic hydrops changes.

7.Expression of ksdD Gene Encoding 3-ketosteroid-△ 1-dehydrogenase from Arthrobacter simplex in Bacillus subtilis and Analysis of Steroid Transformation

To improve 3-ketosteroid-△1-dehydrogenase (KSDH) activity and the transformation level for androst-4-ene-3,17-dione,3-ketosteroid-△1 -dehydrogenase gene (ksdD) from Arthrobacter simplex was cloned into plasmid pWB980 and expressed in B. subtilis WB600 under the control of promoter P43. The molecular weight of expressed enzyme was about 55kDa by SDS-PAGE analysis. The activitities assayed by spectrophotometrical method of intracellular and extracellular soluble enzyme were 110 ± 0.5mU and 15 ± 0.6mU per milligram of protein respectively. The transformation rate of androst-4-ene-3,17-dione by the B. subtilis recombinant cells was 45.3%. Compared with Arthrobacter simplex, the enzyme activity of KSDH expressed in B. subtilis was improved about 30 fold, and the transformation level of androst-4-ene-3,17-dione by the B.subtilis recombinant cells was improved about 10 fold. The recombinant B. subtilis cells used in biotransformation of steroids provided a new way for steroid medicines production.
8.Effects and mechanism of different doses of rosuvastatin on expression of liver X receptor and caveolin-1 in cultured human monocyte-macrophage cells induced by oxidized low density lipoprotein

Objective To investigate the effects of different doses of rosuvastatin on expression of liver X receptor(LXR) and caveolin-1 in cultured human monocyte-macrophage cells which induced by oxidized low density lipoprotein (ox-LDL).Methods The human monocyte-macrophage cells were divided into six groups:control group,ox-LDL group,different doses of rosuvastatin group (0.01 μmol/L,0.1 μmol/L,1 μmol/L,5 μmol/L).The expression of LXR mRNA and caveolin-1 mRNA were assayed by RT-PCR.Results LXR mRNA expression induced by ox-LDL in the control group and ox-LDL group were 1.00 ± 0.02,0.26 ± 0.02,and the difference was significant (t =56.39,P < 0.001).Meanwhile,caveolin-1 mRNA expression in ox-LDL is (0.27 ± 0.01) fold than that in control (t =31.27,P < 0.001).Meanwhile,There were significant differences among ox-LDL group and the different doses of rosuvastatin group in terms of LXR mRNA and caveolin-1 mRNA expressions (F =72.154,66.007,P < 0.001).Along with the increase the doses of rosuvastatin,there was an increased trends of LXR mRNA and caveolin-1 mRNA expressions (P < 0.05).Conclusion Rosuvastatin and upregulated the LXR mRNA and caveolin-1 mRNA expressions in a dose dependent manner.
9.Expression and purification of the soluble envelope protein of Japanese encephalitis virus

Objective To express and purify the envelope ( E) protein of Japanese encephalitis virus (JEV) in soluble form.Methods Various prokaryotic expression vectors , host strains and induction conditions including time and temperatures were screened to obtain an optimum prokaryotic expression system for JEV E protein in soluble form .The expressed protein was purified by using nickel column chromatography and gel filtration chromatography .Results The soluble JEV E protein , accounting for 23% of the totally bacteria soluble protein was effectively expressed by using the recombinant plasmid PBCX -E406 at low tem-perature.The purity of the expressed protein reached up to 85%after the purification by using nickel column and gel filtration chromatography .Conclusion Soluble JEV E protein was successfully expressed and puri-fied in a simple and efficient way .It would provide a useful tool for further investigation on JEV infection , attenuation mechanism of JE live vaccine strain SA 14-14-2 and the quality control of JE vaccine .It can also be used for the development of diagnosis assay for JEV .
