1.Curative efficacy of Xianlinggubao capsules combined with Lugua polypeptide injection in vertebral osteoporosis

Objective To explore curative efficacy of Xianlinggubao capsules combined with Lugua polypeptide injection in treatment of vertebral osteoporosis.Methods 90 patients of vertebral osteoporosis from March 2014 to March 2015 in our haspital were selected as research objects and randomly divided into the observation group and control group,with 45 cases in each group.The control group were treated with Lugua polypeptide injection, while the observation group were treated with Xianlinggubao capsules combined with Lugua polypeptide injection.Then bone mineral density, osteocalcin, visual analogue scale (VAS), Oswestry disability index (ODI) score, curative effect in two groups after treatment were compared.Results Ater treatment, bone mineral density, osteocalcin in observation group was than that in control group [(0.99 ±0.13)g/cm2 vs(0.89 ±0.14)g/cm2, (41.05 ±5.30)μg/L vs(35.80 ±5.23)μg/L],the difference was statistically significant(P<0.05); VAS、ODI score in observation group were lower than those in control group[(1.21 ±0.30)分vs(4.65 ±1.08)分、(18.24 ±4.97)% vs(31.26 ±5.73)%],the difference was statistically significant (P<0.05).The total effective rate of observation group was statistically higher than that in the control group 95.55%(43/45),the difference was statistically significant vs77.77%( 35/45 ) ( P<0.05 ) .Conclusion Xianlinggubao capsules combined with Lugua polypeptide injection is well for vertebral osteoporosis,which can improve the bone metabolism of patients, promote bone growth and improve treatment efficacy.
2.Quality Criteria of Guben Yangxue Oral Liquid

OBJECTIVE:To establish the quality criteria of Guben yangxue oral liquid. METHODS:TLC was employed to identify prepared Ligustrum lucidum,Cornus officinalis,Radix Astragali,Radix Rehmanniae Praeparata,and Angelica sinensis,and the content of polysaccharide was determined by ultraviolet spectrophotometry. RESULTS:The TLC spots were clear,well-separated and specific yet without interference from negative control. The linear range of polysaccharide was 0.020 6~0.072 1 mg?mL-1(r=0.999 2) with an average recovery of 98.64%(RSD=0.56%,n=9). CONCLUSION:The established quality criteria is suitable for the quality control of Guben yangxue oral liquid.
3.Study on Pharmacodynamics of Guben Yangxue Oral Liquid

OBJECTIVE:To observe the effect of Guben yangxue oral liquid on cyclophosphamide (CTX)-induced suppression of the immune and hematopoietic system in mice. METHODS:Immunosuppressive model mice were induced by intraperitoneal injection of CTX 50 mg?kg-1. Different dose groups were Guben yangxue oral liquid at the same time with Zhenqi fuzheng capsules as control. Several index such as peripheral blood cells, the weight of immune organ, serum hemolysin were determined. RESULTS:The levels of white blood cells (WBC,87.3、26.3、7.96 g?kg-1), lymphocytes (L,87.3 g?kg-1), neutrophils (N,87.3、26.3、7.97 g?kg-1), hemoglobin (Hb,87.3、26.3 g?kg-1), red blood cell (RBC,87.3、26.3 g?kg-1), platelets (PLT,87.3、26.3 g?kg-1), sp- leen index(87.3,26.3 g?kg-1), thymus index(87.3 g?kg-1), serum hemolysin (87.3、26.3、7.96 g?kg-1)in CTX-induced immunosuppressive mice were increased after administrated of Guben yangxue oral liquid. CONCLUSION:Guben yangxue oral liquid can enhance the non-specific immunity and humoral immune function of CTX-induced immunosuppressive mice, and it can improve CTX-induced hematopoietic dysfunction significantly.
4.Effect of Dihydroarteannuin on transplantation tumor growth in C57BL/6J mice with lewis lung cancer

AIM: To explore the anti-tumor effect and the mechanism of dihydroarteannuin in C57BL/6J mice with lewis lung cancer. METHODS: Fifty C57BL/6J mice subcutaneously planted with 3LL lung cancer cells (2?106) were randomly divided into 5 groups: blank control group, same volume of normal saline group, positive control DDP group, low, middle and high dose dihydroarteannuin groups. Changes of body weight and inhibitory rate of tumor in each group were observed. The cell cycle and apoptosis rates were analyzed by flow cytometry. RESULTS: The body weights were decreased in middle and high dose group compared with NS group and the inhibitory rate of tumor were 53.50% and 59.24% respectively. FCM assay indicated that Dihydroarteannuin could induce apoptosis of lung cancer cell. At the same time, the number of G0/G1 and G2/M phase cells was decreased. Most of the cells were arrested in S1 phase. CONCLUSION: Dihydroarteannuin has obviously effect on anti-tumor in C57BL/6J mice with lung cancer. Its possible mechanism might be involved in inducing cancer cell apoptosis.
5.Clinical comparative study of two fixation methods for thoracolumbar spine fracture

ObjectiveTo compare the clinical results of pedicle screw fixation via the injured vertebra versus across the injured vertebra for thoracolumbar spine fracture.MethodsThe study reviewed 56 patients (41 males and 15 females,at age range of 21-66 years,mean 41.5 years) with thoracolumbar spine fractures managed with the two fixation methods from June 2005 to December 2008.The fracture segment included T12 in 13 patients,L1 in 27 and L2 in 16.According to the AO classification,there were six patients with type A1.2,29 with type A3.1,nine with type A3.2 and 12 with type A3.3.McCormack load score was 5-8 points (average 6.3 points). The spinal cord injury was classified as grade A in three patients,grade B in four,grade C in eight,grade D in 17 and grade E in 24 according to Frankel scale.The patients were divided into two groups,ie,across vertebral fixation group (27 patients) and via the vertebral fixation group (29 patients).All patients were selectively treated with monosegment bone graft simultaneously.The Cobb' s angle,restoration of the anterior height of the injured vertebra,improvement of spinal canal stenosis rate and Denis scale in local pain and work status were compared between the two groups.The bone graft fusion and spinal cord recovery of both groups were observed during follow-up.ResultsAll patients were followed up for 12-48 months (average 25.8 months).Implantation loosening occurred in one patient 1.5 month after operation in across vertebral fixation group.There were no significant differences in aspects of correction of Cobb' s angle,restoration of the anteriorheight of injured vertebra and improvement of spinal canal stenosis rate postoperatively as well as in aspects of restoration of anterior height of injured vertebra and improvement of spinal canal stenosis rate at the latest follow-up between the two groups ( P > 0.05).The postoperative loss of correction rate of Cobb's angle of both groups existed,with significant difference (P <0.05). The differences of the Cobb's angle at the latest follow-up and after operation were significant in the across vertebral fixation group ( P < 0.05) but insignificant in the via vertebral fixation group (P > 0.05 ).Bone graft fusion occurred in 21 patients (78%) in the across vertebral fixation group and in 27 patients (93%) in the via vertebral fixation group ( P < 0.05 ).Denis scale indicated a better recovery in the local pain of via vertebral fixation group compared with the across vertebral fixation group ( P < 0.05 ),but showed no significant differenc e in work status between the two groups ( P > 0.05 ).ConclusionsCompared with across vertebral fixation,the pedicle screw fixation via the vertebra has the advantages of higher fusion rate and better correction rate of Cobb' s angle and is a better choice for thoracolumbar spine fracture with posterior approach.
6.The comparative study of three different fixation methods via muscle-splitting approach to treat single segmental lumbar vertebral diseases

Objective To compare the advantages and disadvantages of three different fixation methods via muscle-splitting approach for the treatment of single segmental lumbar vertebral diseases.Methods All of 90 patients with the single segmental lumbar vertebral diseases were treated from June 2012 to June 2013,including 34 males and 56 females,at the age of 27-76 years old,averagely (53.37± 15.41) years.Twenty-seven patients were treated with unilateral pedicle screws via unilateral muscle-splitting approach (unilateral fixation group).Thirty-three patients were treated with unilateral pedicle screws and contralateral translaminar facet screw via unilateral muscle-splitting approach (combined fixation group).Thirty patients were treated with unilateral pedicle screws via bilateral muscle-splitting approach (bilateral fixation group).Observe and compare the imaging and clinical results of these three groups.Results There were no significant difference in incision length and postoperative wound drainage between these three groups.The operation time was shortest in unilateral fixation group,while longest in bilateral fixation group.In the intraoperative blood loss,the unilateral fixation group was the same as the combined fixation group,while less than the bilateral fixation group.No infection was found after operations.Seven patients experienced epidermal necrosis of the incision,including 1 patient of unilateral fixation group,4 patients of combined fixation group and 2 patients of bilateral fixation group.Three patients suffered from nerve injury,including 2 patients of combined fixation group and 1 patients of bilateral fixation group.The follow-up time was from 12 to 36 months,averagely 25.5 months.The intervertebral height of lesion segments was apparently recovered after surgery,and maintained during the follow-up in these three groups.No significant changes of area and grade of multifidus muscle were detected at 1 year after surgery compared with preoperative.Seven patients experienced endplate cutting and cage partially embedded into vertebral body,including 3 patients of unilateral fixation group,3 patients of combined fixation group and 1 patient of bilateral fixation group.There was no loosening,displacement,breakage or cage shifting.Except 5 patients (2 patients of unilateral fixation group,2 patients of combined fixation group and 1 patient of bilateral fixation group),the remaining cases had well interbody fusion.At the final follow-up time,the JOA score significantly increased compared with preoperative.Besides,unilateral fixation group and combined fixation group were superior to bilateral fixation group.Conclusion Three different fixation methods via muscle-splitting approach for the treatment of single segmental lumbar vertebral diseases had well clinical results.The unilateral fixation and combined fixation via muscle-splitting approach had more advantages.The combined fixation via muscle-splitting approach is an innovation in approach,exposure and fixation aspects.
7.Incidence and risk factors of chronic post-surgical pain and its effect on quality of life: a large sample prospective study

Objective To investigate the incidence and risk factors of chronic post-surgical pain (CPSP) and its effects on the quality of life in a large sample prospective study.Methods A total of 1098 patients of either sex,aged 18-70 yr,of American Society of Anesthesiologists physical status Ⅰ or Ⅱ,undergoing elective surgery,were enrolled in the study.Data regarding patient age,gender,body mass index,educational level,marital status,living situations,occupation,pre-surgical pain in the site of surgery,complications,type of surgery,surgery time and anesthesia method were recorded.The highest numeric rating scale scores within 3 days after surgery were also recorded.The patients were followed up at 3,6 and 12 months after surgery to record the development of CPSP.The patients were divided into CPSP group and non-CPSP group according to whether or not CPSP developed.Multivariable logistic regression analysis was performed to identify the risk factors for CPSP,and the quality of life was scored.Results A total of 981 patients completed the 1 year follow-up after surgery,the incidence of CPSP was 35.7%,and the constituent ratio of the distribution of the course was as follows:3 months ≤ postsurgical course < 6 months was 33.1%;6 months ≤ postsurgical course < 12 months was 16.8%;postsurgical course ≥ 12 months was 50.1%.Female,no diabetes mellitus and pre-surgical pain in the site of surgery were the independent risk factors for CPSP (P< 0.05).Compared with non-CPSP group,the physiological function score,professional function score,body pain score,vitality score,social function score,mental health score and general health score were significantly decreased (P<0.01),and no significant change was found in the emotional function score in CPSP group (P>0.05).Conclusion The probability of development of CPSP is high and the course is long,and the quality of life score is decreased;female,no diabetes mellitus and pre-surgical pain in the site of surgery are the independent risk factors for CPSP.
8.Bone marrow mesenchymal stem cells combined with VEGF gene for the treatment of limb ischemia in rabbits

Objective To evaluate bone marrow mesenchymal stem cells combined with VEGF gene in the treatment of limb ischemia in rabbits.Methods The right hind limb ischemia model of New Zealand rabbit was established by superficial femoral artery excision and deep femoral artery ligation.Rabbits then were divided randomly into 4 groups: empty plasmid control group(EP group),bone marrow mesenchymal stem cells group(BMSC group),VEGF gene therapy group(VEGF group),combination bone mesenchymal stem cells and VEGF gene therapy group(BV group).There were 8 rabbits in each group.Angiogenesis was detected by arteriography on day 28 after treatment and expression of VEGF was detected by immunohistochemical staining on day 30 after treatment.Results There were no differences of collateral vessel count between the EP group,BMSC group and VEGF group.The collateral vessel count in BV group was higher than that of the other three groups.Immunohistochemistry of VEGF showed that the integrated optical density(IOD)in BMSC and VEGF groups increased significantly compared with the EP group; the IOD in BV group was the highest compared with the other three groups.Conclusions Combination bone marrow mesenchymal stem cells and VEGF gene in the treatment of limb ischemia in rabbits can obtain stable and effective expression of VEGF along with significant improvement of limb ischemia.
10.Inhibitory effect of Periplaneta Americana extract on 3LL lung cancer in mice.

BACKGROUNDIt has been known that Kangfuxin, a drug derived from Periplaneta Americana, can induce cell apoptosis of many cancer cell lines in vitro. The aim of this study is to investigate the inhibitory effect and mechanism of Periplaneta Americana extract (PAE) on 3LL lung cancer in mice.
METHODSThe C57BL/6J mice transplanted with 3LL lung cancer were divided into normal saline (NS), PAE high dose (PAE-H) and PAE low dose (PAE-L) groups. The body weight changes and inhibitory rate of tumor growth in each group were observed. In addition, the cell cycle, apoptosis index (AI) and the expression of apoptosis associated genes were analysed by flow cytometry (FCM).
RESULTSThe body weights were decreased in PAE-L and PAE-H treated group compared with NS group and the inhibitive rate of tumor growth was 41.24% and 81.08% respectively. FCM assay indicated that PAE could induce apoptosis of lung cancer cell, and the apoptosis rate was concentration-dependent. At the same time, the number of S and G2/M phase cells was decreased, most of the cells were arrested in G1/G1 phase. The result of TUNEL showed that there were apoptosis and necrosis associated with upregulated expression of Fas, FasR and p53 genes, and downregulated expression of Bcl-2.
CONCLUSIONSPAE may inhibit the growth of 3LL lung cancer in mice and induce apoptosis of 3LL lung cancer cells. It might be related to its effects on the regulation of apoptotic gene expression.
