Objective To observe the changes of plasma Von Willebrand factor(vWF) content in chronic obstructive pulmonary disease(COPD) and late lung cancer(LLC) patients with Qi deficiency.Methods The changes of vWF content in 26 COPD patients with Qi deficiency and in 15 LLC patients with Qi deficiency were observed before and after treatment,and were compared with those in 30 healthy volunteers.Results Plasma vWF content before treatment was increased in COPD and LLC patients with Qi deficiency(P0.05).Conclusion Abnormal platelet activation exists in COPD and LLC patients with Qi deficiency,the function of vascular endothelial cells decreases,and the blood is in hypercoagulable state,which accords with the traditional Chinese medical theory of Qi failing in promoting blood circulation.
2.Correlation Analysis of the Detection Rate of Carbapenems-resistant Gram-negative Bacillus with Drug Consumption in Our Hospital

OBJECTIVE:To provide reference for standardizing the clinical use of carbapenem antibiotics and controlling drug-resistant bacteria infection. METHODS:The detection of 3 kinds of carbapenems-resistant Gram-negative bacillus in our hospi-tal during 2011-2016 were analyzed retrospectively. The consumption,target cure rate and treatment course of carbapenem antibiot-ics were analyzed statistically. The correlation between detection rate of drug-resistant bacteria with the consumption of carbapenem antibiotics was investigated by Pearson test. RESULTS:During 2011-2016,1222 strains of carbapenems-resistant Acinetobacter bauman (CRAB),655 strains of carbapenems-resistant Pseudomonas aeruginosa (CRPA) and 53 strains of carbapenem-resistant Escherichia coli (CRE) were detected in our hospital. The detection rates increased from 23.88%,8.92%,0.09% in 2011 to 80.34%,35.74%,0.97% in 2016. The types of carbapenem antibiotics in our hospital were mainly imipenem and meropenem. The consumption of them increased from 4222,145 g in 2011 to 7218,4387 g in 2016. The both target cure rates were all lower than 60%,and the proportion of the patients with treatment course >14 d was more than 65%. The detection rates of CRAB,CR-PA and CRE were positively correlated with the consumption of carbapenem antibiotics (r>0.9,P<0.05). CONCLUSIONS:The detection rate of carbapenems-resistant Gram-negative bacillus and drug consumption increase year by year in our hospital,and they have certain correlation. The target cure rate of carbapenem antibiotics in our hospital is in low level,and there is a long treatment course. They are should be standardized in the clinic. The selection of carbapenem antibiotics should be strictly followed clinical in-dications so as to reduce the generation of drug-resistant strains.
3.Development and evaluation of a rapid test strip for gB antibody detection of pseudorabies virus

To evaluate the gB antibody level of pseudorabies virus (PRV) in swine,a rapid test strip was developed.In the strip,the expressed protein of gB was labeled with colloidal gold,the staphylococcal protein A (SPA) and swine anti PRV antibody were blotted on the nitrocellulose membrane for the test and control lines,respectively.The specificity and sensitivity of the strip were detected with standard positive,negative and immunized sera of PRV,the results indicated that the strip was high specificity and sensitivity.Field swine serum samples were tested by the new strip and commercial IDEXX PRV gB ELISA kit,simultaneously.The agreement rate of the two methods was 91.04%.Furthermore,the dipstick assay based on the strip is rapid (5 min),sensitive and easy to perform.This suggests that the new strip is an acceptable alternative for field diagnosis.
4.Surgical treatment of thyroglossal duct cyst in 36 adults

Objective To analyze clinical features and surgical treatment of thyroglossal duct cyst (TDC) in adult patients.Methods Clinical data of 36 adult patients with TDC were analyzed retrospectively.Results All the 36 patients received surgical resection of Sistrunk's procedure.One case suffered wound infection after the operation.Two cases recurred(recurrence rate:5.6%)but cured after the 2nd operation and showed no sign of recurrence during the follow up of 18 and 26 months respectively.No other complications were reported.Conclusions Etiological factors,pathological and anatomical features need to be analyzed.Surgical treatment should focus on hyoid bone and the upper area.The recurrence rate depends on total resection of cyst.
5.Differences of DNA copy number changes between hyperplastic scar and keloid

BACKGROUND: Clinical genetics and molecular biology studies have shown that the occurrence and development of the keloid is closely related to the inheritance. However, it remians unclear if the same is ture to the hypertrophic scar. OBJECTIVE: To investigate similadties and differences of genetic alteration between the hyperplastic scar and the keloid, DESIGN, TIME AND SETTING: A contrast observational experiment was performed in Guangdong Medical College between March 2007 and December 2008.MATERIALS: Scar samples were taken from 16 patients (in-patient and out-patient) in the Department of Plastic Surgery, the Affiliated Hospital of Guangdong Medical College, with10 patients with hypertrophic scars (3 males and 7 females, 20-50 years old) and 6 patients with keloids (1 males and 5 females, 19-46 years old). METHODS: The DNA of both hyperplastic scar and keloid tissues was extracted to investigate, using comparative genomic hybridization technique, the genomic imbalance (the lose or amplification of genetic material), so as to make a comparative study on differences of the DNA copy number changes between the two. RESULTS: Neither altofrequent loss nor amplification of DNA copy number was found in any specific DNA region of hyperplastic scar tissues; as for the keloid, special DNA altofrequent loss regions were also not found, but altofrequent DNA copy number loss regions presented in 1, 16, 20 and 22 chromosomes. Comparatively, the keloid presented much higher loss rate of the DNA copy number in 1,16,20 and 22 chromosomes than the hyperplastic scar (P < 0.05).CONCLUSION: The hyperplastic scar has no conspicuous DNA copy number lose or amplification compared with the keloid, which indicates that the occurrence and development of the hyperplastic scar may not have any direct relation with the inheritance.
6.Effects of nerve growth factor on N-methyl-D-asparate receptor 1 after spinal cord injury in rats.

OBJECTIVETo explore the effects of the nerve growth factor (NGF) on N-methyl-D-asparate receptor 1 (NMDAR 1) after spinal cord injury.
METHODSSpinal cord injury of Wistar rats was performed with Allen's method by a 10 gx2.5 cm impact on the posterior T8 spinal cord. NGF was given to the rats of the treatment group via subarachnoid space tube at once, 2, 4, 8, 12 and 24 hours after spinal cord injury, respectively. The expression of NMDAR1 mRNA in spinal cord was detected by in situ hybridization.
RESULTSRare expression sequence of NMDAR1 mRNA was found in rat spinal cord of the normal group. A strong expression sequence of NMDAR1 mRNA was found in rat spinal cord of the normal saline group. The expression of NMDAR1 mRNA in the NGF group was significantly decreased as compared with that in the normal saline group (P=0.01).
CONCLUSIONSNGF can relieve damage of injured spinal cord by prohibiting the expression of NMDAR1 mRNA.

7.The cranial MRI appearance of cerebral autosomal dominant arteriopathy with subcortical infarcts and leukoencephalopathy in a family

ObjectiveTo recognize the cranial MRI appearance in cerebral autosomal dominant arteriopathy with subcortical infarcts and leukoencephalopathy(CADASIL). MethodsFive patients with CADASIL from two generations in a family underwent routine MRI and MRA examinations.Three patients with CADASIL were confirmed by the Notch3 genetic testing and the vascular pathological Resultsand one was diagnosed on basis of MR and clinical manifestations. The imaging data from 4 patients with CADASIL were analyzed.ResultsFour cases achieved preliminary diagnosis of CADASIL and one was excluded by MRI.In 4 patients with CADASIL,bilateral symmetrical,confluent white matter lesions in the subcortical and Deriventricular regions were seen frequently in the temporal,frontal and parietal lobes,but the occipital lobes were less involved.These lesions appeared as long T1 and long T2 signal.O'Sul]ivan sign was shown in all cases and subcortical lacunar lesions was seen in 2 cases. In the centrum semiovale,well-defined,round or oval cystic infarcts(black holes)were demonstrated in 3 cases and multiple tiny round enlarged perivascular spaces(pepperpot appearance)in all cases.The corpus callosum was involved in all cases,and it was evidently atrophic in 2 cases.The anterior part of internal capsule and external capsule were were present in the basal ganglia and brainstem. Only one case revealed a small infarct in the right cerebellum. Four Datients shared mild or moderate atrophy of brainstem, cerebellum and cerebrum. No marked abnorillalitv of large vessels was seen in all cerebral MRA. ConclusionsThe cranial MRI appearance in CADASIL shows some characteristics.MRI may give some infotznation in the preliminary diagnosis or exclusion of CADASIL.
8.Effect of nerve growth factor on neuronal apoptosis after spinal cord injury in rats.

OBJECTIVETo explore the molecular mechanism of the protective effect of nerve growth factor (NGF) on injured spinal cord.
METHODSThe posterior T(8) (the 8th thoracic segment) spinal cords of 60 Wistar rats were injured by impacts caused by objects (weighing 10 g) falling from a height of 2.5 cm with Allen's way. Solution with nerve growth factors (NGF) was given to 30 rats (the NGF group) through a microtubule inserted into the subarachnoid cavity immediately, and at 2, 4, 8, 12 and 24 hours after spinal cord injury (SCI) respectively. Normal saline (NS) with same volume was given to the other 30 rats (the NS group) with the same method. And 5 normal rats were taken as the normal controls. The expression of bcl-2 and bax proteins in spinal cord was detected with immunohistochemistry. The apoptotic neurons in spinal cord were measured with terminal deoxynucleotidyl transferase-mediated dUTP-biotin nick end-labeling of DNA fragments (TUNEL) staining.
RESULTSThe positive expression of bcl-2 protein was strong in the normal controls, but decreased in the NS group, and increased significantly in the NGF group as compared with that of the NS group (P<0.01). The positive expression of bax protein was also strong in the normal controls, but increased in the NS group, and decreased significantly in the NGF group as compared with that of the NS group (P<0.01). Apoptotic neurons were found in the NS group, and they decreased significantly in the NGF group as compared with that of the NS group (P<0.01).
CONCLUSIONSNGF can protect the injured nerve tissues through stimulating the expression of bcl-2 protein, inhibiting the expression of bax protein and inhibiting the neuronal apoptosis after SCI.

9.Comparison of the predictive value of multiparametric MRI and prostate-specific membrane antigen PET/CT for pelvic lymph node metastasis in prostate cancer

Objective:To compare the diagnostic performance of multiparametric magnetic resonance imaging (mpMRI) and prostate-specific membrane antigen (PSMA) PET/CT in detecting pelvic lymph node metastasis in prostate cancer.Methods:This is a retrospective case series study. A retrospective analysis was conducted on the data of 115 prostate cancer patients who underwent both mpMRI and PSMA PET/CT before undergoing radical prostatectomy and extended pelvic lymph node dissection at the Department of Urology, Xiangya Hospital, Central South University, between March 2020 and September 2023. The age ( M(IQR)) was 67(10) years (range: 45 to 84 years), and the body mass index was 24(4) kg/m 2 (range: 18 to 30 kg/m 2). Pathological and imaging data were obtained from the patients. Lymph node pathology results were used as the gold standard to evaluate the diagnostic performance of mpMRI and PSMA PET/CT for detecting pelvic lymph node metastasis in PCa through diagnostic evaluation tests. Comparisons between groups were performed using independent samples t-test, Mann-Whitney U test, or χ2 test. Results:The positive rate for detecting pelvic lymph node metastasis was 18.3% (21/115) with mpMRI and 25.2% (29/115) with PSMA PET/CT. The pathological positive rate for lymph nodes was 28.7% (33/115). In patient-based analysis, the diagnostic sensitivity of PSMA PET/CT was significantly higher than that of mpMRI (63.6% vs. 30.3%, χ2=7.36, P=0.007). In lesion-based analysis, both the sensitivity and positive predictive value of PSMA PET/CT were significantly higher than those of mpMRI (sensitivity: 68.0% vs. 21.6%, χ2=42.20, P<0.01; positive predictive value: 50.0% vs. 23.1%, χ2=7.54, P=0.006). Conclusions:PSMA PET/CT and mpMRI both demonstrates good specificity in predicting pelvic lymph node metastasis in prostate cancer. However, PSMA PET/CT is significantly superior to mpMRI in terms of sensitivity and the detection rate of pathologically positive lymph nodes.
