Objective To understand the effects of drinking water and source water on DNA breakage of human peripheral blood lymphocytes (HPBL). Methods The organic compounds in drinking water and source water were adsorbed by GDX_102 resin for solid phase of gas choromatography. DNA damages of lymphocytes were detected by single cell gel electrophoresis. Results At the same exposure doses to organic extracts of water, the DNA damages of HPBL exposed to organic extracts of surface water were heavier than those exposed to organic extracts of deep underground water, and heavier DNA damages were also observed in HPBL exposed to organic extracts of finished water from water plant treating ground source water compared with those exposed to organic extracts of finished water from water plant treating deep underground source water. Significant dose_response relationships were observed between the exposure doses of organic extracts of water samples and the degrees of DNA damages of HPBL. Conclusion The organic extracts of source water samples collected from surface water and ground water and its tap water samples could cause DNA breakages of HPBL in different degrees in a certain city.

Objective To evaluate the effect of isoflurane or sevoflurane inhalation before and after gestation on the offspring brain development. Methods Thirty female adult Sprague Dawley rats were randomly assigned into 5 groups(n=6 each):control group(group C),group that ex?posed to isoflurane with the concentration of 1.6%for 6 hours before gestation(group BI),group that exposed to isoflurane with the concentration of 1.6%at the 6th,10th,14th and 18th day for 6 hours(group PI),group that exposed to sevoflurane with the concentration of 2.4%for 6 hours before gestation(group BS),and group exposed to sevoflurane with the concentration of 2.4%at the 6th,10th,14th and 18th day for 6 hours after gestation (group PS). Twelve offspring rats from pregnant rats in each group were chosen on the day of birth(T1),and 7th,14th and 28th days after birth(T2, T3 and T4)and sacrificed, and the hippocampi were then isolated. Hematoxylin and eosin stain were adopted to observe the tissue pathological change. Electron microscope was used to observe the neuron ultrastructure change of hippocampus. Immolunohistochemistry was used to detect cas?pase?3,the expression of growth associated protein?43(GAP?43)and neuronal nitric oxide synthase(nNOS). Results Compared with group C, no significant change was found in caspase?3,GAP?43 and nNOS expression in offspring rat hippocampus in groups BI and BS(P>0.05),and no damage change in hippocampal was seen by HE staining and electron microscopy. In group PI and PB,the expression of caspase?3 was significantly up?regulated, the expression of GAP?43 and nNOS was down?regulated at T1 to T3(P<0.01),and structural changes in cell were seen by electron microscopy. In group PI,significant pathological changes in hippocampal were seen by HE staining at T1 to T3. Compared with group PI,the expres?sion of GAP?43 and nNOS was significantly up?regulated(P<0.01),and the expression of caspase?3 was down?regulated at T1 to T3(P<0.01). Conclusion Isoflurane or sevoflurane inhalation before gestation does not affect the offspring brain development, while isoflurane or sevoflurane in?halation after gestation can induce transient abnormal change of offspring brain development, and isoflurane′s toxicity was greater than sevoflurane.

Objective Looking for the chemical components which have positive correlations with anti-inflammatiory activity of Flos Lonicerea to set up pharmacodynamic-fingerprint. Methods To gain the fingerprints of different parts by HPLC and to detect the pharmacological activities of anti-inflammation, then the correlationship between chemical components and pharmacological activities were detected by linear regression. Results Pharmacological activities of the methanol extracted part were best of all, so the fingerprint of the methanol part could represent the pharmacodynamic-fingerprint of Flos Lonicerea. Conclusion The fingerprint established by this way is more scientific and reasonable.

Objective To investigate the prevalence of excessive daytime sleepiness and explore the associated factors in university teachers and staffs in Kunming. Methods Epworth sleepiness scale was used as the tool of diagnosis. We consecutively collected the data of teachers and staffs on duty in Yunnan Nationalities University,Yunnan Agricultural University,Yunnan Normal University,Kunming Medical University and Yunnan University of Finance and Economics within 8 months. Results The prevalence of excessive daytime sleepiness in university teachers and staffs was 12.45%, the prevalence of male was 16.82% and female was 7.70%, respectively. The results of Logistic regression showed that the main associated factors of excessive daytime sleepiness were male (OR=1.898),age (30 years and less was as control group, the ORs of 31-40 year group, 41-50 years group and more than 51 years group were 1.480, 2.366 and 2.719, respectively), family history of snoring (OR=3.371), consumption sedative (OR=1.815), memory failing (OR=2.044), dizzy and feeble (OR=2. 076),choking (OR=2.143),report apnea (OR=2.684),etc. Conclusions The prevalence of excessive daytime sleepiness is high in the university teachers and staffs that can partially represent the residents of Kunming city. The associated factors of excessive daytime sleepiness hint that excessive daytime sleepiness may be caused by sleep apnea/hyperpnoea syndrome in the sample population, hence the people with excessive sleepiness need further sleep monitoring .

To rapidly select potent anti-VSTM1-v2 scFv (single-chain antibody fragment) by construction and screening of a humanized scFv library in which a murine VH-CDR3 library was grafted onto a human scFv framework. A murine VH-CDR3 library was amplified from anti-VSTM1-v2 murine cDNA and grafted on human scFv (VH3-VK1) framework. Anti-VSTM1-v2 scFv templates were selected and enriched through ribosome display, TA-cloned into expression vector, and transformed into BL21 (DE3) for soluble expression of target scFv. A total of 1000 clones were randomly picked. Positive ones were first identified using colony PCR, indirect ELISA, Western blotting and then verified with sequencing and dose response ELISA. At last an anti-VSTM1-v2 humanized scFv with good binding affinity (EC50 = 21.35 nmol x L(-1)) was selected from the humanized library of 10(12) members generated in this study. This scFv antibody might have potential applications. This study provides a new approach for rapid screening of humanized antibodies.
