1.Effect of Hypoxia on the Expression of Hypoxia Inducible Factor 1? and Vascular Endothelial Growth Factor in Lung of Mice

Objective To investigate the expression of hypoxia inducible factor-1?and VEGF in lung of mice during hypoxia and its relationship with angiogenesis.Methods The mice were divided into two group(hypoxia group and control group).The hypoxia group mice were placed in a hypoxia chamber with 10%,7%,or 5%O2 respectively for 3,6,9 days.Immuno- histochemical staining technique was used to examine HIF-1?,VEGF and MVD.The p rotein expression of HIF-1? and VEGF were observed by Western blot.Results HIF-1?proteins didn't express and VEGF proteins expressed weakly in control group mice;Compared with the controls,the expression of HIF-1?and VEGF increased dramadically.There were significant differences between the hypoxia groups and the control group;Positive relationships were found between the expression of HIF-1?,VEGF and MVD.Conclusion HIF-1?/VEGF pathway may play an important role in angiogenesis of hypoxia in the lung of mice.
2.Algorithm expression on the optimal reconstruction increment in coronal multi-planar reformation of temporal bone for multislice CT

Objective To study algorithm expression of the optimal reconstruction increment with which images could be reconstructed without loss of z-direction spatial resolution for multislice spiral CT.Methods Using Gauss function and signal sampling principle,an algorithm expression was deduced to calculate the optimal reconstruction increment with which images could be reconstructed without loss of zdirection spatial resolution.Spiral slice sensitivity profile (SSP) phantom was scanned using Somatom Sensation 64-slice spiral CT and temporal bone protocol as those used for clinic,axial images were reconstructed with slice thickness of 0.600 mm and increment of 0.100,0.300,0.400 and 0.500 mm respectively.Then SSPs and full width at half maximum (FWHM) were measured and modulation transfer functions were obtained by Fourier transfer from SSPs.Axial CT scan of 7 normal temporal bones in 6 patients were obtained by the same CT system and parameters as above.Coronal MPR images of temporal bone out of different reconstruction increment were obtained and the quality of reconstructed images were independently assessed by three senior radiologists using a four-point scale and blinded the information of reconstruction.Experimental data were processed and two-way ANOVA(in which Dunnett t test was selected for multiple comparisons) was performed with statistic software SPSS10.0.P < 0.05 was considered as significant difference.Results (1) The measured FWHM of reconstruction slice thickness of 0.600 mm was 0.665 mm,so the optimal reconstruction increment calculated with the algorithm expression in this article was 0.296 mm(≈0.300 mm).Objective evaluation showed that there was obviously aliasing in high spatial frequency range of MTF curve when reconstruction increment was more than 0.300 mm,i.e.both 0.400 and 0.500 mm.Clinical scores of coronal MPR images of temporal bone reconstructed with different increment had significant difference(F =505.374,P < 0.01).Treating reconstruction increment of 0.100 mm as a control group and comparing another groups against it,there was no significant difference for reconstruction increment of 0.300 mm (t =-0.222,P > 0.05),while there were significant difference for 0.400 and 0.500 mm (t =-1.333、-15.889,P < 0.05,P < 0.01).Conclusion It was proved that the angorithm expression deduced in this article was proper because of the consistence between the calculated value and the results from objective and clinical evaluation.The optimal reconstruction increment without loss of z-direction spatial resolution is 0.445 times as large as FWHM for multislice CT.
3.Research on Urine Metabolomics of HIV/AIDS Patients with Spleen-lung Qi-deifciency Based on H-NMR Technique

This study was aimed to explore the characteristics of urine metabolomics among HIV/AIDS patients with spleen-lung qi-deficiency. H-NMR technique was combined with principal component analysis and cluster analysis in the comparison of urine metabolic products among 24 HIV/AIDS cases with spleen-lung qi-deficiency and 20 healthy control cases. The results showed that urine metabolic products of HIV/AIDS patients with spleen-lung qi-deficiency and healthy people by H-NMR technique detection and PLS-DA analysis can classify the outline of urine metabolites. There were about 20 variables in the difference between two groups. The speculated substances contained glycyl-L-leucine, L-valine, α-aminobutyric acid, methyl succinic acid, glycine propionyl, and etc. It was concluded that H-NMR technique was able to classify the outline of urine metabolites between HIV/AIDS cases with spleen-lung qi-deficiency and healthy people. Part of the potential existed characteristic markers contributed to the clinical diagnosis of traditional Chinese medicine (TCM) syndrome differentiation of AIDS. It had certain effect in its quantitative analysis.
4.Nano-hydroxyapatite/silk fibroin composite materials loaded with recombinant human bone morphogenetic protein 2 used for spinal fusion

BACKGROUND:With the wide application of bone repair scaffold in the field of medicine, nano-hydroxyapatite (nHA) and silk fibroin (SF) both of which have good biological properties have become research hotspots in recent years.OBJECTIVE:To study the feasibility of nHA/SF composite materials loaded with recombinant human bone morphogenetic protein 2 (rhBMP-2) to restore the initial stability of the spinal segment in rabbits.METHODS: Thirty-six male health New Zealand rabbits were randomly divided into three groups, followed by preparation of spinal instability models. Autogenous iliac bone, nHA/SF composite, and rhBMP-2/nHA/SF composite were implanted into the L4/5 spinal segment in autologous bone group, nHA/SF group and rhBMP-2/nHA/SF group, respectively. X-ray exmaination was performed at 12 weeks postoperatively, and then the animals were killed for gross observation. The stability of the fusion segments was tested through a tensile machine. Histologically, bone graft fusion at the surgical site was observed.RESULTS AND CONCLUSION:(1) Findings from the gross specimen observation showed that the specimens at the fusion site presented with a hard texture. Obvious signs of fusion were visible in the autologous bone group, followed by the rhBMP-2/nHA/SF group, while no signs of fusion were detected in the nHA/SF group. (2) At 12 weeks postoperatively, a large number of trabecular bones grew into the boundary between the vertebral body and the iliac crest graft block in the autologous bone group. A little trabecular bone was found in the boundary in the nHA/SF group, while a lot of trabecular bone tissues were found in the boundary in the rhBMP-2/nHA/SF group. In accordance with the standard of fusion, there were 10, 3, and 9 rats in the autologous bone, nHA/SF and rhBMP-2/nHA/SF groups, respectively. (3) The range of motion of the spine in the rhBMP-2/nHA/SF showed no statistical difference from that in the autologous bone group, but was significantly higher than that in the nHA/SF group (P < 0.05). (4) Osseous connection was found around the bone graft in the autologous bone and nHA/SF groups, but no mature bone tissue was visible in the latter group. In the rhBMP-2/nHA/SF group, a large number of new capillaries was found and permeated into the spinal tissues. In summary, nHA/SF composite materials loaded with rhBMP-2 possess good biocompatibility, mechanical properties and bone induction ability, which can rebuild the initial stability of the spine in a short time.
5.Improvement effect of electromyographic biofeedback on wrist dorsiflexion function of patients with cerebral infarction at different Brunnstrom stages

Objective:To observe the effect of electromyographic biofeedback on the wrist dirsiflexion function of the patients with cerebral infarction at different Brunnstrom stages, and to clarify the treatment of electromyographic biofeedback,and to provide basis for its clinical application.Methods:A total of 100 cerebral infarction patients were selected.Among them 54 BrunnstromⅠ-Ⅱ patients were randomly divided into treatment group (n= 32)and control group (n = 22),and another 46 Brunnstrom Ⅲ patients were randomly divided into treatment group (n=23)and control group (n=23).The patients in four groups were treated with the same routine stroke rehabilitation therapy while the patients in treatment groups still received the electromyographic biofeedback therapy additionally.The maximum electromyographic contraction of muscle,active range of movement (AROM) and Fugl-Meyers Assessment (FMA)of the extension of wrist joint were evaluated before treatment and 4 and 8 weeks after treatment,respectively.Results:The maximum electromyographic contraction values of muscle of the patients in BrunnstromⅠ-Ⅱ treatment group and control group were significantly improved 8 weeks after treatment (P <0.05),and the value in treatment group was higher than that in control group (P <0.05).The maximum electromyographic contraction value of muscle in Brunnstrom Ⅲ treatment group began to improve 4 weeks after treatment compared with before treatment (P < 0.05) and it was significantly higher than that in control group (P <0.05).The maximum electromyographic contraction value of muscle in Brunnstrom Ⅲ control group began to improve 8 weeks after treatment (P <0.05).The AROM in Brunnstrom Ⅰ-Ⅱ treatment group began to improve 8 weeks after treatment (P <0.05)and it was significantly higher than that in control group (P <0.05)while the AROM in control group had no significant change (P >0.05).The AROM in Brunnstrom Ⅲ treatment group and control group were significantly improved 4 weeks after treatment (P < 0.05 or P < 0.01 ), and the value in treatment group was significantly higher than that in control group (P < 0.05).The FMA in BrunnstromⅠ-Ⅱtreatment group and control group were significantly improved 8 weeks after treatment (P <0.05),while the value in treatment group was higher than that in control group (P <0.05);the FMA in Brunnstrom Ⅲ treatment group began to improve 4 weeks after treatment (P < 0.05)and it was significantly higher than that in control group (P <0.05). The FMA in control group began to improve 8 weeks after treatment (P <0.05). Conclusion:Electromyographic biofeedback can increase the strength and improve the body function of the patients with cerebral infaction.
6.Effect of dexamethasone on expression of hypoxia inducible factor-1α and vascular endothelial growth factor in hypoxic mice.

BACKGROUNDIt has been proved that hypoxia is closely related to oncogenesis and development of tumor. The aim of this study is to observe the effect of dexamethasone on expression of hypoxia inducible factor 1α (HIF-1α) and vascular endothelial growth factor (VEGF) in lung tissues of hypoxic mice, and to investigate the relationship between hypoxia and angiogenesis and mechanism of dexamethasone.
METHODSThe Kunming mice were randomly divided into control group and three experimental groups (3-day hypoxia group, 6-day hypoxia group, and hypoxia+dexamethasone group). HIF-1α and VEGF protein expression was detected in lung tissues of mice by immunohistochemistry.
RESULTSExpression of HIF-1α and VEGF significantly increased in hypoxia group compared with control group (P < 0.05). Compared with hypoxia group, expression of HIF-1α and VEGF decreased dramatically in hypoxia+dexamethasone group (P < 0.05). A positive correlation was found between the expression of HIF-1α and VEGF (r=0.730, P=0.007).
CONCLUSIONSHypoxia can increase the expression of VEGF and HIF-1α in lung tissues of mice. Dexamethasone can inhibit the expression of VEGF and HIF-1α of hypoxic mice and it may have anti-angiogenetic effect.
7.Mechanism of Ferroptosis and Its Research Progress in Lung Cancer.

Ferroptosis is a recently recognized form of regulated cell death caused by an iron-dependent accumulation of lipid reactive species. However, little research on ferroptosis and lung cancer, one of the most common tumors, has been carried out. This paper tries to review the research progress of ferroptotic suppression and explain it from the different ways of ferroptosis occurrence. Furthermore, as inducing ferroptosis to treat cancer gets more attention, we introduce four kinds of ferroptosis-inducing compounds and new prospects for lung cancer therapy to provide new ideas for lung cancer treatment.
8.Investigation on anxiety and depression of employees in municipal and above administrative organs and public institutions in Qinghai region

BackgroundThe mental health problems of employees in administrative organs and public institutions are highly valued. However, there is a lack of understanding regarding the mental health status of employees in provincial and municipal administrative organs and public institutions in Qinghai region. ObjectiveTo analyze the anxiety and depression of employees in provincial and municipal administrative organs and public institutions in Qinghai region, so as to provide references for promoting their mental health and intervening in psychological problems. MethodsFrom October 23 to 28, 2022, a total of 3 096 employees in provincial and municipal administrative organs and public institutions in Qinghai region were enrolled using stratified cluster sampling technique, and were assessed using Self-rating Anxiety Scale (SAS) and Self-rating Depression Scale (SDS). ResultsA total of 1797 (58.04%) employees in municipal and above administrative organs and public institutions in Qinghai region completed a valid questionnaire survey. Depressive symptoms were noted in 579 (32.22%). SDS scores yielded statistical difference among the selected employees of different ethnicities, types of job positions and educational levels (F=9.074, 101.488, 4.477, P<0.01), and the severity of depressive symptoms also demonstrated statistical difference among the selected employees with different ethnicities, educational levels and types of job positions (H=49.196, 17.028, 160.848, P<0.01). Anxiety symptoms were detected in 711 (39.57%) employees. SAS scores exhibited statistical difference among the employees with different genders, ages, ethnicities, educational levels, marital status and types of job positions (t=-4.571, F=6.648, 3.950, 5.243, 12.008, 22.253, P<0.01). Statistical differences were also illustrated in the severity of anxiety symptoms among the selected employees with different ages, genders, ethnicities, educational levels, marital status and types of job positions (H=24.361, -3.788, 28.365, 24.268, 25.976, 56.204, P<0.01). ConclusionAbout one-third of employees in municipal and above administrative organs and public institutions in Qinghai region may have symptoms of depression and anxiety. The depression symptoms are more severe among Tibetan people, those with low education levels and ordinary civil servants.
