1.Research on Extraction and Purification Technology of Flavonoids in Dried Tangerine Peel

Objective To set up a method for extracting and purifying of flavonoids in Dried Tangerine peel.Methods Total flavonids being used as an index, UV spectrophotometer was used to optimize the extracting technology with the help of orthogonal experiment. Meanwhile macroporous resin was adopted for optimizing indexes of total flavonoids.Results The optimum conditions for purification were as followings: AB-8 macroporous resin, 50% of ethanol serving as eluting solvent, 0.4g/mL of sample concentration, 1:5 ration of diameter to height, 2BV/h of current velocity, and 5BV of flow column. Conclusion The content of total flavonoids was remarkably increased after purification.
2.Observations on the Efficacy of Acupuncture plus Jisheng Shenqi Pills in Treating Diabetic Neurogenic Bladder

Objective To investigate the clinical efficacy of acupuncture plus Jisheng Shenqi pills in treating patients with diabetic neurogenic bladder. Method Patients with diabetic neurogenic bladder were allocated, using a random number table, to treatment and control groups. The treatment group received acupuncture plus Jisheng Shenqi pills and the control group, oral administration of Western medicine. Both groups of patients were treated for four weeks as a course. Post-treatment clinical total efficacy rates, pre-treatment and post-treatment postvoid residual urine volumes, and adverse reactions during treatment were compared between the two groups. Result The total efficacy rate was higher in the treatment group (93.8%) than in the control group (68.8%) (P<0.05). The postvoid residual urine volume decreased significantly in both groups of patients after treatment compared with before (P<0.05). Post-treatment postvoid residual urine volume was lower in the treatment group than in the control group (P<0.05). The adverse reaction rate was lower in the treatment group than in the control group (P<0.05). Conclusion Acupuncture plus Jisheng Shenqi pills is remarkably effective in treating patients with diabetic neurogenic bladder. It can effectively decrease postvoid residual urine volume in the patients, has higher safety and is worthy of clinical popularization.
3.Effects of Recombinant Human Interferon α1b Combined with Ribavirin on Thyroid Function in Chronic Hepatitis C Patients

OBJECTIVE:To observe the effects of recombinant human interferon (IFN)α1b on thyroid function of patients with chronic hepatitis C(CHC). METHODS:Retrospective collection 90 CHC patients,according to whether to merge with hyper-thyroidism into hypothyroidism group,hypothyroidism group and CHC group,30 cases in each group. 3 groups were given IFN-α1b 40μg,qod,combined with ribavirin 0.1 g,tid,for antiviral therapy. Before and after therapy,RVR,EVR,ETVR and SVR were observed in 3 groups,and the levels of T3,T4,FT3,FT4 and TSH were observed in hypothyroidism group,hypothyroidism group before and after antiviral therapy. RESULTS:After treatment,RVR,EVR,ETVR and SVR levels of 3 groups increased sig-nificantly,with statistical significance,compared with before treatment(P<0.05).After treatment,there was no statistical signifi-cance in RVR,EVR,ETVR and SVR among 3 groups,there was no statistical significance(P>0.05);there was no statistical sig-nificance in the levels of T3,T4,FT3,FT4 and TSH before and after treatment(P>0.05). CONCLUSIONS:IFN-α1b combined with ribavirin can obtain good antiviral effect and not worsen existing thyroid disease in the treatment of CHC complicated with thy-roid disease.
4.The study of DNA content and cell proliferation during the course of development of golden hamster buccal pouch carcinoma

Purpose:To investigate the DNA content and cell proliferation during the course of development of golden hamster buccal pouch carcinoma.Methods:60 golden hamsters were divided into 6 groups,0.5% DMBA were application to the buccal pouch mucosas of 1~5 groups, and then animals were killed at 6W, 8W, 10W, 12W and 16W after application of DMBA, the animals of normal group were killed at 16W . Buccal pouch samples were taken and each sample was cut into two parts, one was processed histologically way, the other sample was detected and analysed with flow cytometry for DNA content and cell cycle.Results:Our results revealed that the DNA of normal mucosa and hyperplasia appeared diploid; there were 16.7% (2/12) of anuploid in the mild and moderate dysplasia; 33.3% (3/9) of DNA anuploid in severe dysplasia; 54.1(6/11)% of DNA anuploid in carcinoma in situ ; and 78.57% (11/14) of DNA anuploid in invasive carcinoma. DI value and S phase proliferation became higher as the mucosal lesion became heavier , the S phase of severe dysplasia was the highest (39.31%).Conclusions:DNA anuploid is the marker of malignant tumor; the cell rate of S phase can be considered a sign of malignant tumor proliferation , it is a valuable guide for clinical treatment and prognosis assessment in oral carcinoma .
5.Spectroscopic Property of AO-H_2SO_4-KBrO_3-Naphthol System and Its Application in Determination of Trace Naphthols in Urine

Objective To understand the spectroscopic property of AO-H2SO4-KBrO3-naphthol system and the enhanced mechanism of resonance light scattering(RLS),and to develop a method for the determination of trace naphthols in urine.Methods In the dilute H2SO4 medium,naphthols could react with KBrO3 and AO to form ion-association complexes,which produced a new RLS spectrum and resulted in the great enhancement of RLS.The characteristics of RLS spectrum,three-dimensions fluorescence spectrum,absorption spectrum and fluorescence spectrum and the optimum conditions of reaction were studied.Results The enhanced intensity of RLS was 468 nm.The linear range was at 1.41?10-7-2.80?10-5 mol/L for ?-naphthol,1.28?10-7-3.00?10-5 mol/L for ?-naphthol.The relative coefficient and the limits were 0.999 6 and 0.422?10-7 mol/L,0.999 3 and 0.385?10-7 mol/L for ?-naphthol and ?-naphthol,respectively.The urine samples analysis of the relative standard deviation was 4.8%-7.3% and the average recovery rate was 90.7%(n=6).Conclusion This method is sensitive,simple,rapid and applicable to the determination of trace naphthols in human urine.
6.Studies and prospects of the candidate virulence gene of familial medullary thyroid carcinoma

Familial non-medullary thyroid carcinoma (FNMTC) is de fi ned as the presence of two or more affected fi rst-degree relatives with non-medullary thyroid cancers without other known familial syndromes. FNMTC is one of the most inheritable forms of all cancers, with a high risk of a first-degree relative developing the disease. Compared with sporadic non-medullary thyroid carcinoma (NMTC), FNMTC presents at a younger age and is associated with a higher incidence of multifocal disease and metastasis. This in-creased aggressiveness has been hypothesized to translate into higher recurrence rates and decreased survival of patients with FNMTC. The genes involved in the pathogenesis of FNMTC are yet to be elucidated, although some recent studies identified several predisposi-tion loci with a high degree of genetic heterogeneity. Since 2005, next-generation sequencing (NGS) technologies have been developing as rapid, high-throughput, and cost-effective approaches to fulfill medical sciences and research demands. With the use of NGS, the un-derlying causative genes can be directly distinguished via systematic filtering, through which the identified gene variants are verified for novelty and functionality.
7.Apoptosis of bladder cancer cells induced by oligonucleotide telomerase inhibitor

Objective To study the potential effects of telomerase inhibitor on the apoptosis of bladder cancer cells. Methods A phosphorothioate oligonucleotide (PS ODN) with sequence 5′ d(TTAGGG) 3′, as the telomerase inhibitor, was incubated with a bladder cancer cell line EJ. Such treated cells were studied with many approaches for the telomerase activity, the growth status and the morphological changes. Results The PS ODN inhibited the telomerase activity in the cells, arrested the cell growth, and induced the apoptosis of the cells. Conclusions The apoptosis of bladder cancer cells could be induced by PS ODN with 5′ d(TTAGGG) 3′ sequence as a telomerase inhibitor.
8.Telomerase activity in the tissue adjacent to bladder cancer and its significance

Objective To study the telomerase activity in the tissue adjacent to bladder cancer and to investigate its clinical significance. Methods Telomerase activity was detected with telomeric repeat amplification PCR (TRAP) assay. The telomerase activity in the tissue adjacent to bladder cancer was evaluated. Results Telomerase activity was positive in the tissue samples adjacent to bladder cancer in 10 of the 24 cases(42%). Telomerase activity in the adjacent tissue has been related with the tumour grades and stages. The tumour recurrence was also related with the telomerase activity in the adjacent tissue. Conclusions The detection of telomerase activity in the tissue adjacent to bladder cancer could be a prognostic marker for bladder tumor recurrence.
9.Clinical observation of the effect of budesonide atomization on tracheal extubation in children

Objective To investigate the effect of budesonide atomization on tracheal extubation in children.Methods A total of 85 patients with tracheal intubation in Tianjin Children's Hospital from May 2013 to September 2016 were selected in this study.Patients were randomly divided into budesonide group (n=44) and dexamethasone group (n=41).The budesonide group was given 1 mg of budesonide for inhalation 30 min before extubation,and 1 mg of budesonide inhalation immediately after extubation.Then every 8 hours for 0.5-1.0 mg budesonide inhalation for 4 days.The dexamethasone group was given dexamethasone 0.2-0.3 mg/kg intravenously 30 min before extubation,and dexamethasone 2.5-5.0 mg inhalation immediately after extubation.Then dexamethasone 2.5-5.0 mg inhalation was given every 8 hours for 4 days.The incidence of laryngeal edema (stridor,hoarseness),the time of extinction,the expression of hypoxia,reintubation rate within 24 hours and secondary infection rate after extubation were compared between the two groups.Results There were no significant differences in the mission success rate,the incidence of laryngeal edema,the time of extinction,the incidence of hypoxia and re-intubation rate between the two groups (P > 0.05).Two patients were found secondary infection after extubation in dexamethasone group.Conclusion Both budesonide and dexamethasone show curative effects on the prevention and treatment of laryngeal edema after extubation.Budesonide atomization can replace systemic corticosteroids,thus reducing the adverse reactions of glucocorticoids.We recommend the use of budesonide in treating adverse events after extubation.
10.Research Progress in Prevention and Treatment of Diabetic Nephropathy with TCM Polysaccharides

Diabetic nephropathy (DN) is one of the most important microvascular complications of diabetes, which is one of the most important causes of death in diabetic patients. Because of its high efficiency and few adverse reactions, TCM has been widely studied and used in the treatment of DN, with unique advantages. The recent research showed that a variety of TCM polysaccharides have the effects of reduce blood sugar and protect kidney. This article reviewed the research on TCM polysaccharides on the prevention and treatment of DN, which can provide references for further study and application.
