1.Effects of specific interfering TACC3 gene expression on proliferation and apoptosis of CD133+CD44+ oral squamous cell carcinoma cells

BACKGROUND: Studies have indicated that the abnormal expression of TACC3 is closely related to the occurrence and development of many kinds of tumors, and the expression of TACC3 is up-regulated in these tumors. Therefore, in vitro specific inhibition of TACC3 expression may become an important target for the treatment or intervention of tumor growth.OBJECTIVE: To investigate the mechanism by which TACC3 gene expression regulates cell proliferation and apoptosis in oral squamous cell carcinoma.METHODS: CD133+CD44+ oral squamous cell carcinoma cells were sorted from human oral squamous cell carcinoma cell line Cal-27 by immunomagnetic beads. In experimental group, the shRNA sequence of TACC3 was designed and synthesized, which was then trasnfected into CD133+CD44+ oral cancer stem cells by LipofectamineTM 2000. Empty vector-trasnfected (negative control) and untransfected cells were used as callsed. Forty-eight hours after the transfection, effects of TACC3 gene silencing on proliferation and apoptosis in vitro in CD133+CD44+ oral squamous cell carcinoma were detected by MTT, clone formation test, and TUNEL assay. Western blot assay was used to detect the effect of TACC3 gene silencing on Ki67, Bax and Bcl-2 protein expression in CD133+CD44+ oral squamous cell carcinoma.RESULTS AND CONCLUSION: (1) Cell proliferation. The proliferation rate and expression level of Ki67 were significantly lower in the experimental group than the negative control and untransfected groups (P < 0.05). (2) Clone formation. The clone formation ability in the experimental group was significantly lower than that in the negative control and untransfected groups (P < 0.05). (3) Cell apoptosis. TACC3 gene silencing caused an obvious decrease in Bcl-2 protein expression and a significant increase in Bax protein expression. These findings further confirmed that specific interference of TACC3 gene expression could inhibit the proliferation of CD133+CD44+ cells and promote the apoptosis.
2.CT Scan Diagnosis in Gastrointestinal Tract Rupture after Blunt Abdominal Trauma

Objective To evaluate the role of CT in diagnosis of the gastrointestinal tract rupture after blunt abdominal trauma . Methods Twenty preoperative CT scans and clini cal data were obtained in 20 patients who subsequently had bowel ruptures verifi ed surgically. CT findings were analyzed retrospectively in these patients. Retr ospective interpretation was made by consensus of at least two radiologists. Results Twenty cases of CT scan showed intraper itoneal fluid (18 cases), pneumoperitoneum (18 cases), extravasations of gastro intestinal tract contents (2 cases), bowel wall findings (14 cases) and mesenter ic injury (15 cases). Conclusion CT is fast, sensitive and noninvasi ve in diagnosis of the gastrointestinal tract rupture after blunt abdominal tra uma.
3.Screening of lentiviral vectors carrying effective siRNA targeting S1P2 gene

AIM: To screen the lentiviral vector carrying siRNA with higher efficiency of suppressing the sphingosine-1-phosphate receptor 2 (S1P2) gene expression in the primarily cultured corpus cavernosum smooth muscle cells of spontaneously hypertensive rats (SHR).METHODS: SHR and SD rats (n=5 each) were used for primarily culturing corpus cavernosum smooth muscle cells.The cells were randomly divided into 6 groups: SHR siRNA-1, SHR siRNA-2, SHR siRNA-3, SHR GFP, SHR control (SHR non-transfection group), and SD control (SD rat control group).Each group had 5 samples with 1.0×105 cells of each sample.At 72 h after transfection (MOI=60) with lentiviral vectors carrying S1P2 siRNA into the SHR corpus cavernosum smooth muscle cells, the expression of GFP was observed under fluorescence microscope.The protein expression of S1P2, ROCK1, ROCK2 and eNOS in the corpus cavernosum smooth muscle cells, and the mRNA expression of S1P2, ROCK1 and ROCK2 were determined by by Western blot and RT-PCR.RESULTS: The transfection efficiency of the corpus cavernosum smooth muscle cells in SHR siRNA-1, SHR siRNA-2, SHR siRNA-3 and SHR GFP groups were>80%.Compared with SHR control group, the mRNA levels and the protein expression of S1P2, ROCK1 and ROCK2 in SHR GFP group showed no remarkable changes, while those in SHR siRNA-1, SHR siRNA-2, SHR siRNA-3 and SD control groups were significantly lower than those in SHR control group (P<0.05).The protein expression of eNOS in SHR siRNA-1, SHR siRNA-2, SHR siRNA-3 and SHR GFP groups were not significantly changed as compared with SHR control group, but that in SD control group was significantly higher than that in SHR control group.CONCLUSION: Three groups of siRNA lentiviral vectors targeting S1P2 inhibit the expression of S1P2 in the corpus cavernosum smooth muscle cells of SHR, and by silencing the S1P2 expression, the expression of ROCK1 and ROCK2 is inhibited.Among them, siRNA-1 has the highest inhibitory efficiency.
4.Follicular thyroid carcinoma: an analysis of 105 cases

Objective To investigate clinical biologic characteristics and factors that impact prognosis of patients with follicular thyroid carcinoma(FTC).Method Clinical data of 105 FTC patients treated surgically in the Department of Head& Neck,Tianjin Cancer Hospital from 1970 to 1990 was analyzed retrospectively.Results The overall 5-year,10-year and 15-year survival rates of these patients were 85.3%,76.7% and 72.9% respectively.The overall 15-year survival rates of patients ≥45 years and those < 45 years were 45.9%,89.8% respectively.The overall 15-year survival rates of patients with unilateral and bilateral carcinoma was 50.0%,76.2% respectively.The overall 15-year survival rates of patients with neck lymph node metastasis and without were 54.2%,79.2% respectively.The overall 15-year survival rates of patients at stage Ⅰ,stage Ⅱ,stageⅢ and stageⅣ were 89.3%,70.0%,45.5% and 35.3%respectively.During the follow-up period,11 patients were diagnosed with distant metastasis from 1 year to 33 years after surgical treatment and 9 died of cancer within 5 years after diagnosis.Seventeen patients had local recurrence from 3 months to 34 years after surgery and 10 of these patients died of local recurrence.Conclusions The factors influencing prognosis of patients with FTC were age,clinical stage,bilateral carcinoma and neck lymph node metastasis,therefore early treatment and close following-up are essential to improve the prognosis of patients with FTC.
5.Relationship between DNA methylation of glucocort icoid receptor gene promotor and glucocorticoid receptor gene mRNA expression in systemic lupus erythematosus patients

Objective To investigate the relatio nship between the DNA methylation status of gluco-corticoid receptor (GR) gene promoter and mRNA expression level of GRα gene of peripheral blood mononuclear cells (PBMCs) in patients with systemic lupus erythematosus (SLE). Methods Fifteen new onset SLE patients and fifteen healthy controls were enrolled in this study. The DNA methylation status of GR gene promoter 1 of PBMCs was detected through bisulfite sequencing polymerase chain reaction (PCR). The mRNA expression of GRα, DNA methyltransferases, growth arrest and DNA damage-induced 45α (GADD45α) of PBMCs was detected using the quantitative real-time polymerase chain reaction method. T-test and χ2-test were used to detect the differences between the two groups, Pearson's correlation coefficient was used to analyze the linear correlation between two variables. Results Compared with healthy controls, the mRNA expression of GRα was signi-ficantly declined in SLE patients (10±5, 17±7, t=2.69, P<0.05), and the mRNA expression of DNMT1 and GADD45α was significantly elevated in SLE patients (t=3.11, P<0.05 and t=2.98, P<0.05). The overall mean methylation status of the 142 CpG islands of the four promoters was significantly elevated in SLE patients [(16±8)%vs (11±6)%, t=2.75, P<0.05]. The global methylation status of PBMCs in SLE patients was obviously lower than healthy controls (t=4.39, P<0.05). Conclusion Hypermethylation of GRα promoter may result in GRαgene low expression in PBMCs of patients with SLE.
6.The investigation on impulsive and aggressive personality traits affecting college students self-injury sui-cide behavior

Objective To examine the different effects of impulsive and aggressive personality on college students' suicide attempt. Methods Four thousand nine hundred eighteen sophomores and juniors were selected according to the principle of cluster sampling from Beijing agricultural university and Beijing university of information science and tech?nology. Four thousand seven hundred ninety-seven of them completed the survey. Their impulsive and aggressive person?ality were evaluated by the Barratt Impulsiveness Scale-Chinese Versions (BIS-CV) and the Buss-Perry Aggression Questionnaire-Chinese Versions (AQ-CV). A self-made questionnaire were used to investigate suicidal idea and self-in?jury/suicidal behavior of college students. Results Among the sophomores and juniors, 18 participants had self-injury/suicidal behavior (3.75‰) and 1843 had suicidal idea (38.42%). Compared with the students who didn’t have self-inju?ry/suicidal behavior, those who once had self-injury/suicidal behavior had significantly higher BIS-CV score (P<0.01) and higher scores of non-planning and motor factors (P<0.05). They also had significantly higher score in AQ-CV and the factors of physical aggression, verbal aggression, anger, hostility and pointing to the self attacks (P<0.01). Conclu? sions The college students with self-injury/suicidal behavior have higher tendency in impulsive and aggressive personali?ty. Therefore, it is necessary to prevent suicide behavior, form high impulsive and aggressive intention and behavior.
7.Clinical value of 18 F-FDG PET-CT in aiding CT-guided percutaneous lung biopsy

Objective To investigate the clinical value of PET-CT in aiding CT guided percutaneous puncture biopsy for lung mas-ses.Methods The retrospective data were analyzed including 5 7 patients with lung masses who underwent CT guided percutaneous puncture biopsy combined with PET-CT (regarded as a research group)and other 75 similar patients with single CT-guided biopsy (control group).In two groups,it was divided with malignant,suspicious malignant,can not be diagnosed,inflammatory,benign by pathological findings,and was contrasted with the surgical pathology or final clinical results.To calculate sensitivity,specificity and accuracy of diagnosis for cancer through two puncture method.Recorded complication for pneumothorax and bleeding in two groups .Be compared analysis using the 2 test.Results In study group,it was higher for the diagnosis of malignancy sensitivity, specificity,accuracy.There were 96.3% (52/54),100.0%(3/3)and 96.5%(55/57)in research group,while those were 88.4%(61/69),83.3% (5/6)and 88.0% (66/75)in the control group,respectively.And no statistically significant.Between research and the control group,there was a significantly statistical difference in the rate of twice punctures in one mass (χ2= 10.122,P=0.001).Between the two groups,there was no statistically significant difference in the pneumothorax rate and the hemorrhage rate. Conclusion PET-CT in aiding CT-guided lung biopsy improves the sensitivity,specificity and accuracy of diagnosis.
8.The diagnosis, treatment and prevention of dysuria within 6 months after suprapubic transvesicle prostatectomy

Objective To summarize the diagnosis,treatment and prevention of dysuria within 6 months after the suprapubic transvesical prostatectomy. Methods Twenty-four cases were retrospectively reviewed,including the data on the diagnosis, treatment and prognosis. Results Twenty-one of the 24 cases had received surgery. There were 9 cases with bladder neck stricture,9 cases with posterior urethra stricture and 3 cases with of remnant glands. Nine cases received transurethral bladder neck incision, 9 received open surgery ( bladder neck,posterior urethral incision) and 3 received transurethral resection of the prostate (TURP) plus bladder neck incision. The other 3 diagnosed as detrusor weakness were conservatively treated by indwelling catheter and they were improved one months later. None of these patients was readmitted into hospital for dysuria within 6 months after treatment. Conclusion The recurrence of dysuria post prostatectomy mostly ( 75% ) occurred within 6 months after surgery. A majority of these patients need a second surgery. To avoid a second surgery for postoperative dysuria, much attention should be paid to the operating technique and postoperative management.
9.The effect of satir model for compulsory drug rehabilitation women

Objective To explore the feasibility and validity of Satir model on compulsory drug rehabilitation for female drug users in closed settings. Methods Satir Model-based group psychotherapy was adopted in the treatment on 33 female drug users. Results The findings are that the pre-treatment SCL-90 scores were all higher than norm,indicating very high significant differences(P<0.01) ; while the post-treatment SCL-90 scores,except the score of somatization, were approximate to norm, showing no statistical differences(P >0. 05). Conclusion Satir model played a positive role in the process of female drug users mental health recovery.
10.Reconstruction and transplantation of composite skin containing keratinocytes and fibroblasts.

OBJECTIVETo investigate the role of fibroblasts in reconstruction of composite skin, and evaluate the effect of composite skin on full-thickness skin defect.
METHODSKeratinocytes and fibroblasts were seeded on the surface of acellular dermal matrix and cultivated in vitro to reconstruct the composite skin. Adherence of keratinocytes to dermal matrix was observed. Then take rate and histological construction were investigated after the composite skin was used to cover full-thickness skin defect wound in nude mice (n = 16).
RESULTSKeratinocytes grew and proliferated to reach tho confluence on the surface of the acellular dermal matrix. Keratinocytes adhered more stablely and could not be torn down from dermal matrix in operation when few fibroblasts were seeded on the epidermal surface of the dermal matrix. After grafting, the composite skin closed the full-thickness wound in nude mouse. The total survival was achived in 10 mice (62.5%). The newly generated skin was with intact histological construction of base membrance containing laminin and type IV collagen.
CONCLUSIONComposite skin could close the full-thickness wound, and fibroblasts could improve adherence of keratinocytes to dermal matrix, which should benefit the survival of composite skin.
