1.THE PROTECTIVE ACTION OF HYDROLYSATE OF ARCA SUB-CRENATA ON LIVER INJURIES IN MICE

Hydrolysate of Area subcrenata (HA) was found to be effective in lowering the elevated SGPT level induced by carbon tetrachloride, thioacetamide (TAA) and pred-nisolone. In addition, HA was shown to shorten sodium pentobarbital sleeping time in both normal and carbon tetrachloride intoxicated mice. The results indicated that HA possessesa significantl protective action on liver injuries in mice.
2.Effects of endotoxin on NF-κB mRNA expression and aldosteron secretion in rat hepatic stellate cells

Objecfive To investigate the effects of endotoxin on nuclear factor-κB p65(NF-κB p65)mRNA expression and ahtosteron secretion in rat hepatic stellate cells(HSCs).Methods Cultured rat HSCs(HSC-T6)were divided into endotoxin-treated group and control group.Cells in endotoxin-treated group were exposure to 1 mg/ml.endotoxin.Aldosteron secretions of HSCs were determined by radioimmunoassay,and NF-κB p65 mRNA expressions of HSCs were detected by one-step RT-PCR.Results At 6,12,24 and 48 h,aldosteron secretions in endotoxin-treated group were significantly hisher than those in the control group(t=3.063,4.577,6.847 and 9.317,P<0.05),and the expressions of NF-κB p65 mRNA in endotoxin-treated group were also higher than those in control group(t=5.155,6.095,7.875 and 9.313,P<0.01).Aldosteron secretions and NF-κB p65 mRNA expressions in HSCs displayed a positive correlation(r=0.886,P<0.01).Conclusion Endotoxin can up-regulate the aldosteron secretion and NF-κB p65 mRNA expression in rat HSCs,which may be one of the mechanisms of liver fibrosis induced by endotoxin.
3.Effects of ornithine aspartate combined with alprostadil in the treatment of severe chronic hepatitis B and its effect on serum tumor necrosis factor-α

Objective To investigate the effect of ornithine aspartate combined with alprostadil in the treatment of severe chronic hepatitis B (CHB),and its effect on serum tumor necrosis factor-α (TNF-α).Methods 80 patients with severe CHB were selected.The patients were divided into the observation group and the control group by digital random method,40 patients in each group.The control group was given conventional treatment of rest,isoglycyrrhizinate magnesium needle,glutathione needle,plasma,albumin,anti-hepatitis B virus.The observation group received ornithine aspartate combined with alprostadil treatment based on the treatment of the control group.After a continuous treatment of 4 weeks,the curative effect was observed,the changes of liver function before and after treatment was compared.Enzyme-linked immunosorbent assay (ELISA) was used to detect the plasma TNF-α level,and the adverse reactions during treatment were recorded.Results The total effective rate of the observation group was 92.50%,which was significantly higher than 80.00% of the control group (x2 =26.588,P < 0.05).The preoperative and postoperative alanine aminotransferase(ALT),prothrombin activity(PTA),total bilirubin (TBIL) of the two groups had statistically significant differences (all P < 0.05),and,there were statistically significant differences between the observation group and the control group after treatment[(86.56 ± 38.67)U/L vs.(121.23 ± 39.65)U/L,(62.45 ±5.46)% vs.(54.88 ±5.37) % and (175.67 ±42.34) μmol/L vs.(214.56 ±43.45) μmol/L,t =3.959,6.252,4.054,all P < 0.001].The plasma TNF-αt level had no statistically significant difference between the two groups before treatment (P > 0.05).After treatment,the TNF-α level of the two groups was significantly decreased,which of the observation group was significantly lower than that of the control group[(176.45 ± 20.41)ng/L vs.(221.43 ± 21.43)ng/L,t =9.613,P < 0.001].The incidence rate of adverse reactions of the observation group was 12.50%,which had no significant difference compared with 10.00% of the control group (x2 =0.313,P > 0.05).Conclusion Ornithine aspartate combined with alprostadil in the treatment of severe CHB on the basis of the conventional treatment can apparently enhance clinical effect,improve liver function,decrease plasma TNF-α level and adverse reactions.
4.Simultaneous Determination of Paeoniflorin and Paeonol in Dieda Pill by HPLC

OBJECTIVE:To establish a method for the simultaneous determination of paeoniflorin and paeonol in Dirda pill, and provide reference for improving its quality control standard. METHODS:HPLC was performed on the column of ZORBAX C18 with mobile phase of acetonitrile-0.1% phosphoric acid (gradient elutio) at a flow rate of 0.8 ml/min,detection wavelength was 230 nm(paeoniflorin),274 nm(paeonol),columne temperature was 35 ℃,injection volume was 20 μl. RESULTS:The linear range was 0.514 8-1.544 5μg for paeoniflorin(r=0.999 2)and 0.643 2-1.960 4μg for paeonol(r=0.999 8);the limits of quantifi-cation were 0.135 6,0.126 4 μg,limits of detection were 0.067 8,0.063 2 μg;RSDs of precision,stability and reproducibility tests were lower than 2%;recoveries were 95.78%-97.27%(RSD=0.62%,n=6) and 97.38%-98.38%(RSD=0.42%,n=6). CONCLUSIONS:The method is simple with good reprosucibility and high sensibility,and can be used for the simultaneous deter-mination of paeoniflorin and paeonol in Dirda pill.
