Objective:To construct the expression vector of RNA interference(RNAi)system for CGI-100 gene and identify its inhibition efficiency.Methods:Three oligonucleotides targeting CGI-100 gene were devised and chemically synthesized,annealed to form double-strand DNA and inserted into plasmid PGenesil-1 whose segments were digested by BamHⅠ and HindⅢ.The recombinant vector were then transfected into K562 cells with lipofectamineTM 2000.CGI-100 expresstion in the transfected cells was assayed by RT-PCR and dot blot.Results:The recombinant plasmids of PGenesil-1-CGI-100 were successfully constructed by DNA sequencing analysis.The first of PGenesil-1-CGI-100 had the highest inhibition efficiency in mRNA levels,up to 54%.Conclusion:The RNA interference system targeting CGI-100 gene were successfully constructed and a interference vector with a higher inhibition efficiency was screened,which provides the foundation for further investigation.

Objective To discuss the relationship between antibiotic prophylaxis and surgical site infection(SSI) in colorectal cancer.Methods A survey of 880 patients with colorectal cancer from Jan.1998 to Dec.2008 were studied.Patients were divided into two groups by antibiotic duration: ≥24 h group(n=401) and 0.05).Conclusion Longer operation time,more blood loss and longer bleeding time are the risk factors of surgical site infection.Moreover prolonged antibiotic prophylaxis can not decrease the incidence of SSI.

Objective To evaluate the risk factors for cerebral infarction (CI) resulting from transient ischemic attack (TIA).Methods The clinical data of 133 patients with TIA were evaluated.The condition of CI happening was recorded.Seizure frequency,seizure duration,onset to treatment time,the number of seizure before treatment time,blood pressure,fasting blood glucose,total cholesterol,triglyceride and other factors were collected.The clinical data of TIA with CI and without CI were evaluated.Results Among the 133 patients,there were 48 patients suffering from CI (36.09％).Among 108 patients with TIA in the anterior circulation system,there were 39 patients (36.11％) suffering from CI.Among 20 patients with TIA in the posterior circulation system,there were 7 patients (35.00％) suffering from CI.There was no significant difference in the rate of CI between TIA in the anterior circulation system and TIA in the posterior circulation system (P ＞ 0.05).Among 5 patients with TIA in both the anterior and posterior circulation systems,there were 2 patients suffering from CI.In patients of TIA with seizure frequency ＞ 3 times/d,seizure duration ＞ 30 min,onset to treatment time ＞ 24 h or the number of seizure before treatment ＞5 times,the incidence of CI was significantly higher,and the difference was statistically significant (P ＜ 0.05 or ＜0.01).The onset to treatment time,systolic pressure,diastolic pressure,fasting blood glucose,total cholesterol and triglyceride levels in TIA with CI were significantly higher than those in TIA without CI (P＜0.05).Conclusion There is highly danger for patients to develop CI if patients with TIA has longer seizure duration,much more seizure frequency,longer onset to treatment time,especially those accompany with high blood pressure,hyperglycaemia and hyperlipoidemia.

Objectives To investigate the incidence of the heparin-induced thrombocytopenia (HIT) and positivity of anti-heparin/platelet factor 4 complex antibody (HIT-antibody) in inpatients treated with heparin preparations.Methods A total of 197 consecutive patients,including 120 men and 77 women,who were treated with unfractioned heparin (UFH) or low molecular weight heparin (LMWH),were enrolled in this study.HIT-antibody was detected by enzyme-linked immunosorbent assay (ELISA).All patients were classified into subgroups based on 4Ts (Pretest Clinical Scoring System) and types of underlying disorders.The incidence of HIT and positivity of HIT-antibody among different groups were analyzed.Results The overall incidence of HIT was 3.0％ (6/197),and the positivity of HIT antibody was 12.2％ (24/197).The positivity of HIT-antibody was 10.1％ (18/178),7.7％ (1/13) and 83.3％(5/6) in low,moderate and high HIT probability group respectively.There were significant differences of HIT positivity and mean level of HIT-antibody between the high HIT probability group and the low or moderate HIT probability group (P =0.000).Both the incidence of HIT and positivity of HIT-antibody were higher in surgical patients than in medical patients (5.8％ vs 0.9％,P =0.047 and 19.8％ vs 6.3％,P =0.004).Conclusions The incidence of HIT and positivity of HIT-antibody were 3.0％ and 12.2％ respectively,both of which were related with the types of disorders.Detection of HIT-antibody has a better applicable value in patients with high HIT probability.

Objective To investigate the result of colonoscopic positioning in laparoscopic treatment of colon tumors . Methods A retrospective analysis was made in 16 cases of colon tumors treated with laparoscopic colon resection from January 2014 to November 2015.During the operation, the lesion could not be locatized and the resection range could not be determinated because the serous layer was not involved or the lesion was located in the lateral abdominal wall .Transanal colonoscopic positioning of the lesion with light transmission method was carried out . Results The pathological changes were found under colonoscopy in all the 16 cases and the laparoscopic resection was successfully completed without conversion to open operation .No missed resection or incorrect resection occurred.The operation time was (112.5 ±31.0) min and the blood loss was (22.8 ±11.2) ml.Among 9 cases of preoperative pathological diagnosis of benign tumors , there were 6 cases of malignant tumors after surgery , including 4 cases of moderately differentiated adenocarcinoma , 1 case of moderate to severe atypical hyperplasia with cancerous lesions , and 1 case of moderately differentiated adenocarcinoma with mucous adenocarcinoma .A total of 15 patients were discharged from hospital at 12-24 days after the operation , whereas 1 patient was dismissed 1 month after the operation due to anastomotic fistula .The 16 patients were followed up for 3 months with no recurrence under colonoscopic examiantion . Conclusions During laparoscopic colon resection , if the tumor can not be locolized , colonoscopic positioning can help prevent missed resection or incorrect resection , which increases the indication and safety of laparoscopic and colonoscopic minimally invasive treatment .It has good clinical application value and deserves further promotion .

Objective To study the effects of nitric oxide (NO) synthesis on serum interleukin 1? (IL-1?) and tumor necrosis factor ? (TNF-?) levels in liver ischemic preconditioning (IP) in rats. Methods 169 rats were randomly divided into ischemia/reperfusion (I/R) group, IP group,L-arginine plus IP (L-Arg+IP) group and sham operation group (S group). The concentrations of plasma NO, serum IL-1? and TNF-? were measured at 2h, 24h and 1 week after models were set up. Results ⑴ Plasma NO concentrations: In group IP and group L-Arg+IP,the NO concentration at all the 3 time points were significantly higher than those in group S (P0 05). Conclusions Liver ischemic preconditioning could down-regulate the levels of blood IL-1? and TNF-?, which may relate to the increase of NO synthesis. The increase of NO synthesis could enlarge this down-regulation effect, and further enhance the protective effect of IP on the liver I/R injury.

It has unique optical properties and can be used for clinical research including cancer research, determination of metal content, microbe detection and developing of drug target. It will have the potential value in the study of disease mechanism, clinical diagnosis and treatment.

[Objective]To describe a new way of arthroscopic posterior cruciate ligament reconstruction and evaluate the early stage of follow up results.[Method]Twenty four consecutive cases of isolated and combined posterior cruciate ligament deficiency were studied.Quadriceps tendon patellar bone was used for preparation of "Y" shaped autograft.Double-tunnel of femur were drilled and tibia was inlayed at their anatomical footprint.[Result]All cases were followed up for 24 months to 30months,averaging 24.5 months.Lysholm rating scales were used to make clinical subjective evaluations.Lysholm scores raised from 49.20?1.44 before operation to 87.91?1.52 after operation.Finial Lysholm evaluation showed 7 patients were excellent,16 good,and 1 poor.The good to excellent rate was 95.8%.Anteroposterior translation as clinical objective evaluations was measured with KT-1000.KT1000 arthrometer measurement showed that the average posterior translation improved from 12.85?1.10mm preoperatively to 3.98?1.99mm postoperatively.One patient was graded as failure.[Conclusion]Arthrosopy assisted prodedure of double bundle posterior cruciate ligament reconsruction using quadriceps tendon patellar bone autograft by double-tunnel of femur and inlaying of tibia is effective and reproducible.The short follow up clinical results are encouraging.

[Objective]To observe changes of skin reinnervation during burn wound healing and determine remodeling of nerve fibers.[Method]Wound tissue and scars were harvested.Collagen was determined with modified Masson trichrome staining and nerve fibers were labeled with neuro filament protein by immunofluorescent technique.Three dimension reconstruction of nerve fibers regenerated was observed under laser scanning confocal microscope.[Result]The CVF(collagen volume fraction) increased during the process of wound healing.The regenerated nerve fibers were sparse,short and small,which was significant lower the normal control.The skin reinnervation improved during the wound healing process and came to peak NVF(nerve fiber volume fraction) in proliferative stage.Disintegration and fragmentation were observed frequently in samples from proliferative stage,which seldom occurred during mature stage.[Conclusion]The remodeling of skin nerve fibers comprises a sequential process of increasing number,shape distortion during wound healing and then disintegration,number decrease and normal shape.

Objective To study the HRCT appearances and diagnostic value of primary Sjgren’s syndrome ( pSS ) . Methods CT images of 12 cases of pSS were retrospectively analyzed . Results CT and HRCT scans showed pulmonary interstitial diseases in different degree mainly localized periphery of lung in all cases,the ground-glass opacity and interlobular septal thickening were preliminary features of pSS.Conclusion The ground-glass opacity of lung is active preliminary features of pSS,while the honeycombed lung is final feature of pSS.HRCT can help to determine the stage of pSS,and to estimate the prognosis in patients with pSS.
