2.Effects of Astragalus Polysaccharides on Damage of Keratinocytes in UVB Radiation Skin

Objective To investigate the effects of Astragalus polysaccharide (APS) on damage of keratinocytes in ultraviolet B (UVB) radiation skin in HaCaT cells; To preliminarily explore its mechanism of action.Methods HaCaT cells were treated by UVB irradiated for modeling. Blank control group, model group, UVB irradiation group and low-, medium- and high-dose APS interventional groups were set up. Each medication group was given relevant medicine. MTT assay was used to determine the cell activity; GSH-Px, CAT, SOD activity and MDA content were detected by biochemical colorimetric method; contents of TNF-α and IL-6 were detected by ELISA.Results Compared with the control group, the contents of SOD, CAT and GSH-Px in the model group decreased significantly, while the contents of MDA, TNF-α and IL-6 increased significantly (P<0.05); compared with the mode group, the contents of SOD, CAT and GSH-Px in the medication groups increased significantly, while the contents of MDA, TNF-α and IL-6 were reduced significantly (P<0.01).Conclusion APS can reduce the oxidative stress injury of UVB to HaCaT cells to some extent.

Performance evaluation of hospitals has become a high priority of Beijing Municipal Administration of Hospitals nowadays,application of targeted governmental allocations being a key to such evaluation.As an oral specialist hospital,such allocations were mainly used to purchase major oral equipment as important revenue makers and social benefits creators.The finance department of the hospital has paid commendable efforts in assuring satisfactory use of such funds to score high in the performance evaluation.On the other hand,however,the hospital is also faced with problems in the implementation of the evaluation of the funds and there remain rooms of improvement.
4.The effects of different doses of propofol infusion on real-time monitoring of recurrent laryngeal nerve

Objective To evaluate the effects of different doses of propofol infusion on real-time monitoring of recurrent laryngeal nerve. Method 30 ASA Ⅰ-Ⅱ patients who underwent thyroid gland surgery were given propofol intravenously in different concentrations. BP, HR and real-time monitoring of recurrent laryngeal nerve were recorded with the propofol dosage varying from 3 to 6, 9, 12, 15, 18?g/(kg?min). Results The amplitude of recurrent laryngeal nerve reflex had no change with different doses of propofol infusion. The HR and BP were elevated with low doses of propofol infusion, but there were no changes when propofol concentration was 15 or 18?g/(kg?min). Conclusion Propofol infusion i.v. does not interfere with real-time intraoperative monitoring of recurrent laryngeal nerve, and it does not inhibit the action potential of recurrent laryngeal nerve, but can effectively decrease the cardiovescular reaction.
5.Effect of Pingchuanlingchongji on the Auricular Microcirculation in Mice

Objective To observe the effect of Pingchuanlingchongji (PCL) on the auricular microcirculation of mice and to investigate the effect mechanism of the treatment for asthma. Methods 50 mice were randomly divided into five groups, group 1 and group 2 were given 0.9% NS (25 mL/kg) as control group; group 3 was given aspirin (0.25 g/kg), 4 group was given low-dose (1.1 g/kg) PCL, group 5 was given high-dose (2.2 g/kg) PCL. Last for seven days. We injected Posterior Pituitary Injection (0.5 U/mL) on the rear vein of 2-5 group mices after the last time of given druy. The diameter of micro-artery in mice auricular, the diameter of micro-vein and blood flow speed were observed. Objective:To sutdy effects of Compound Anmei Oral Liquid on spontaneous activity, sleep, convulsion of experimental mice.Methods: The effects of oral liquid were observed in the spontaneous activity and the sleep time of mice when they were given Nembuta in a threshold dose or a subliminal dose and its anti convulsant effect to investigate the actions of oral liquid.Results: This oral liquid could inhibit the spontaneous activity and cooperate with pentobarbital sodium to effect the sleep time of mice. It also resist convulsion. Conclusions: Compound Anmei Oral Liquid has obvious sedation, hypnosis and anti convulsant effect.
7.Effects of inducible hypothermia on expressions of PGC-1 and SHP in rat model of hemorrhagic shock

Objective To investigate the effects of inducible hypothermia on expressions of peroxisome proliferator activated receptor gamma coactivator-1 (PGC-1) and small heterodimer partner (SHP) in rat model of hemorrhagic shock.Methods SD rats were randomly (random number) divided into three groups:control (C),normothermia (N) and hypothermia (H).Exsanguination was carried out in rats by continuously drawing out venous blood (25 mL/kg) over 15 minutes to establish hemorrhagic shock model.Then,rectal temperatures of rats were maintained by body surface cooling to 32℃ in H group and by body surface warming to 38℃ in N group,respectively.After a shock period of 60 minutes,rats received the infusion of whole blood of their own and lactated Ringer's solution (1 ∶ 2) treatment for 60 min.Rats were warmed to 38℃ by body surface warming and monitored for 3 h after resuscitation.Hematocrit (Hct),base excess (BE),lactate (Lac),and glucose (Glu) were recorded before modeling and after different lengths of hemorrhagic shock period (HSP).The expressions of PGC-1 mRNA and SHP mRNA and the levels of their protein in liver were tested by real-time reverse transcription polymerase chain reaction (qRT-PCR) and western blotting,respectively.Results The H group had lower lactate levels and higher BElevels than the N group [(6.3±2.1) vs.(10.4±1.5) and (-4.7±2.5) vs.(-9.0±3.2)] (P< 0.05).At 72 hours after modeling,there were four survivors in the N group and seven survivors in the H group (P < 0.05,Log Rank).The expressions of PGC-1 mRNA and SHP mRNA increased in N group.Hypothermia resuscitation down-regulated PGC-1 mRNA expression,meanwhile,increased expression of SHP mRNA.Both Hypothermia and Normothermia resuscitation increased SHP protein levels,but decreased PGC-1 protein levels.Conclusions Inducible hypothermia ameliorated acidosis and energy metabolism imbalance through adaptive regulation in PGC-1 and SHP.
8.Anatomical MRI study for rotational alignment of tibial component in total knee arthroplasty

Objective To compare the difference between a vertical line (AA) drawn to the line connecting the inner edge of the patellar tendon with the mid-point of the ending point in the posterior cruciate ligament, tibial posterior condylar line (PC), tibial plateau anterior line (AC), the maximal mediolateral distance (MMLD) and a vertical line (BB) drawn to aligning the mid-point of ending point in the posterior cruciate ligament with the medial 1 / 3 of the patellar tendon relative to the surigical transepicondylar axis (STEA) by MRI, and to explore a reliable reference to determine tibial component rotation in total knee arthroplasty , and whether it will change in knees with varus deformity. Methods Thirty healthy volunteers (Group1) and thirty osteoarthritis patients (Group2) were enrolled in this study. The angles were measured among the five tibial rotation axes and STEA after MRI. Results The angles were (-1.48 ± 2.38)°, (6.16 ± 4.53)°, (6.45 ± 5.24)° ,(5.08 ± 4.99)° and (3.24 ± 2.68)° respectively in group 1 and (-1.88 ± 2.21)°, (-3.13 ± 4.66)°, (11.13 ± 5.72)°, (4.11 ± 4.15)° and (5.12 ± 4.87)° respectively in group 2. The angle between AA and STEA was not affected by varus deformity (P > 0.05), but the others were (P < 0.05). Conclusion The angle between AA and STEA is the smallest which is used to determine tibial component rotation in knees with varus deformity is the most reliable one.
9.Induction of Astragalus Polysaccharides on Differentiation of Rat Bone Marrow ;Mesenchymal Stem Cells

Objective To investigate the effect of astragalus polysaccharides (APS) on the differentiation of rat bone marrow mesenchymal stem cells (BMSCs) to neurones, adipocytes, osteoblasts and chondrocytes, and provide basis for the development of effective and low toxic differentiation inducing agents. Methods BMSCs were isolated from SPF Wistar rats, purified, expanded and cultured to family 3. The appropriate concentration of APS was filtered out by MTT assay. The F3 cells were randomly divided into control group and induced group (neural induction, adipogenic induction, osteogenic induction, cartilage induction). The effects of APS and classical chemical drugs on differentiation were measured by toluidine blue, oil red o and alizarin red staining. The protein expression of NSE, LPL, collagen Ⅰ and collagen Ⅱ were examined by Western Blot. Results MTT assay showed that 1 g/L APS promoted the proliferation better than other concentrations especially in 48 hours. The morphologic change of the cell from BMSCs was uniformly positive to toluidine blue staining, and was negative to oil red o and alizarin red staining. Western blot showed that the protein expression of the cell from BMSCs was positive for NSE but negative for LPL, collagen Ⅰ and collagen Ⅱ. Conclusion BMSCs induced by APS can differentiate to neurone and fail to differentiate to adipocytes, osteoblasts and chondrocytes in vitro.
10.Effects of Volatile Oil of Fructus Evodia, Mustard Oil and Total Anthraquinone in Rheum on Percutaneous Absorption of Paeoniflorin in Vitro

Objective To investigate the effects of volatile oil of Fructus Evodia, mustar d oil and total anthraquinone in Rheum on the transcutaneous permeability of paeoniflorin. Methods Using intelligent transdermal diffusion cell and excised mice skin in vitro as transdermal barrier, the kinetics parameters such as cumulative permeation quantity, permeation rate and permeation lagged time of the three kinds of penetration enhancers on paeoniflorin were determined by HPLC in 12 hours. Results The penetration rate of volatile oil of Fructus Evodia, mustard oil and total anthraquinone in Rheum were 8.188 6, 3.411 7, 1.230 3 μg/(cm2?h), respectively, the enhancement ratios were 22.6, 9.40, 3.40, respectively, and the permeation lagged times were 0.93, 0.51, 0.83 h, respectively. Conclusion Three penetration enhancers all can enhance previously percutaneous absorption of paeoniflorin, which provides reference for the selection of the penetration enhancers of transdermal delivery.
