1.Analysis of 96 children in countryside with upper digestive disease by gastroscope

Objective To investigate the disease categories of the upper alimentary tract and the ratio of Hp infection in countryside children. Methods 96 children with complaint in upper digestive tracts underwent gastro-scope and in each patient,a piece of mueosal tissue in sinus ventrieuli was taken out to perform Hp urase test. Results All of the 96 children were detected abnormal including chronic superficial gastritis (n = 63), ulcer of duodenal bulb(n = 13) ,enteritis of duodenal bulb(n =7) ,gastritis with bile reflux(n =4) ,erosive gastritis(n =2) ,ascariasis (n = 6), aneylestomiasis(n = 3). The positive rate was 100%. 49/96 (51%) were positive in Hp test, including 36/ 63(57.1%) of chronic gastritis ,10/13(76.9%) of ulcer of duodenal bulb,2/7 (28.6%) of enteritis of duodenum. Hp were positive in 11 (35.5%) in the group with age range from 3 to 6 years old and positive in 38 (58.5%) in the group with age range from 7 to 12 years old. Conclusion Chronic superficial gastritis and ulcer of duodenal bulb were the main findings in gastrescopy in children with a upper digestive tract complaint. Hp infection ratio was highest in patients with ulcer of duodenal bulb in our study, and followed by the chronic gastritis. And the infection rate in-creased with the age increased.
2.Therapeutic Effect of Argon Laser Photocoagulation on Macular Choroidal Neovascularization.

Objective To observe the therapeutic effect of Argon laser photocoagulation on macular choroidal neovascularization. Methods 57 cases(58eyes)with macular choroidal neovascularization were treated with Argon laser photocoagulation,the visual acuity, the changing of macular choroidal neovascularization and macular hemorrhage before and after treatment were observed and compared with 34 cases(34eyes)with macular choroidal neovascularization who were treated with drugs.The average follow-up was 48(from2 to 84) months.Results The retinal hemorrhage decreased evidently in cases treated with laser coagulation.The hemorrhage absorb in therapeutic group is more evident than that in control group,There are statistically difference between the two groups(0.01<P<0.05).There are sig- nificantly difference between the two groups in increased visual acuity and choroidal neovascularization disappeared(P<0.01).Couclu- sions Argon laser photocoagulation is effective on macular cloroidal neovasculurition.
3.Relationship between the yield and Extraction Time of Volatile Oil

Objective: To establish the relationship equation of yield and extraction time of plant volatile oil. Methods: By means of the rate eqation of substance transfer, the relationship equation of yleld (v) and extraction time (t) of volatile oil was accomplished: ln(V s-V)=-K t+A,K and A were parameters. Results: The experiment of volatile oil of Rhizoma Nardostachyos indicated that A was 0.44, K was 2.7?10 -3 , and the correlation coefficient (r) was -0.99 whereas A was 1.33, K was 3.62?10 -3 and r was -0.99 in the experiment of volatile oil of Flos Syzygii Aromatici. Conclusion: This relationship equation is able to reflect the relationship between the yield and extraction time of volatile oil.
4.Study on adsorptive properties of spherical activated carbon on several kinds of active ingredient from Chinese materia medica

ObjectTo study the adsorptive properties of spherical activated carbon on some effective ingredients from Chinese materia medica (CMM) and to approach the application feasibility to CMM purification. Methods The spherical activated carbon with high strength has been successfully prepared by the carbonation and activation of polymeric resin. The apparent and theoretical adsorption capacities under the condition of static operation were investigated by Langmuir monolayer adsorption model. Elution by 75% alcohol was studied. Results The static adsorption capacities of the berberine hydrochloride and dioscin were 35.46 mg/g and 47.12 mg/g, and theoretical adsorption capacities were 96.16 mg/g and 102.04 mg/g, respectively. The apparent adsorption amount of rutin was 40.88% mg/g. The elution ratio were 83.71%, 91.45%, and 87.69%, respectively. Conclusion The spherical activated carbon adsorbent shows better comprehensive adsorption properties and can be used to purification.

Objective To study the clinical efficay of the therapy adefovir dipivoxil combined with reduced glutathione in the treatment of decompensated hepatitis B cirrhosis .Methods 76 cases of decompensated hepatitis B cirrhosis were divided into the treatment group and the control group ,38 cases in each group .Both groups take symptomatic and supportive treatment ,the obser-vation group plus adefovir dipivoxil combined with glutathione therapy .After 3 months treatment ,we observed liver function ,hepa-titis B virus DNA quantification and assessed the clinical efficacy .Results After treatment ,the treatment group liver function im-proved was significantly better than the control group (P<0 .05) ,the total effective rate was higher than the control group (P<0 .05) ,the rate of hepatitis B virus DNA turn to negative was significantly higher than the control group (P<0 .01) .Conclusion Adefovir dipivoxil combined with reduced glutathione therapy for decompensated hepatitis B cirrhosis can effectively improve liver function ,suppress hepatitis B virus DNA undetectable ,reduce the incidence of the complication ,and worthy of promote in our clini-cal practice .
7.Effect of endoplasmic reticulum stress on chondrocytes in a rat model of knee osteoarthritis

BACKGROUND: Endoplasmic reticulum (ER) stress has been proved to be related to the occurrence of diabetes, dilated cardiomyopathy and neurodegenerative diseases. Indeed, it is closely associated with osteoarthritis. OBJECTIVE: To explore the effect of ER stress on the chondrocyte viability as well as the occurrence and development of osteoarthritis in rats. METHODS: Rat chondrocytes were isolated and cultured, and the ER stress in the rat chondrocytes was by 10 mg/L tunicamycin. The expression levels of ER stress markers C/EBP-homologous protein and 78 kDa glucose-regulated protein were detected by western blot assay, and the proliferation and apoptosis of chondrocytes were detected by cell counting kit-8 assay and AnnexinV-FITC flow cytometry, respectively. In the in vivo experiment, 15 Sprague-Dawley rats were selected and subjected to anterior cruciate ligament transection and medial meniscectomy to establish an animal model of osteoarthritis. Tunicamycin, tauroursodeoxycholic acid and PBS (blank control group) were respectively injected into the articular cavity, and then the progression of osteoarthritis was assessed by hematoxylin-eosin staining at 4 weeks after treatment. RESULTS AND CONCLUSION: After addition of tunicamycin, the expression levels of C/EBP-homologous protein and 78 kDa glucose-regulated protein were significantly upregulated, the viability of chondrocytes was decreased gradually, while the apoptotic rate was increased significantly. Results from gross observation and hematoxylin-eosin staining suggested that tunicamycin promoted the progression of osteoarthritis and tauroursodeoxycholic acid delayed the deterioration of cartilage in the rats. These findings indicate that ER stress results in the decreased chondrocyte viability and increased apoptosis, which may be an important pathogenesis of osteoarthritis. Additionally, tauroursodeoxycholic acid can effectively alleviate osteoarthritis induced by ER stress.
8.Predictive value of combined detection tumor markers in the diagnosis of lung cancer

To evaluate the predictive value of combined five tumor markers (TMs) CEA, NSE, SCCA, CYFRA21-1 and ProGRP in the diagnosis of lung cancer(LC). Total of 305 hospitalized patients with LC were enrolled, 100 healthy subjects and 100 patients with benign lung diseases were selected as the healthy control (HC) group and BLD group, respectively. The levels of TMs in serum were detected by Flow fluorescence technique. Positivity rates were analyzed by using Chi-square test,The differences of tumor marker levels were compared using Mann-Whitney test and Kruskal-Wallis test. The Receiver Operating Characteristics (ROC) curve was performed to analyze the diagnosis efficacy of TMs. The combined detection had a higher positive rate in patients with LC, adenoadenocarcinoma, squamous cell carcinoma and SCLC (70.82%, 64.74%, 76.4% and 81.03%, respectively) than each TM considered individually. The serological levels of CEA, NSE, SCCA, CYFRA21-1 in LC group were higher than HC and BLD group. The differences of them among the three groups were statistically significant (χ2 =90.599, 32.802, 8.473, 40.397 respectively, all P values were<0.05), ProGRP level had no remarkable difference (χ2 =3.366, P>0.05), whereas ProGRP level in SCLC were significantly higher compared with adenocarcinoma ( Z=6.404, P<0.001) and squamous cell carcinoma ( Z=5.765, P<0.001) group. The combined detection difference of positive rate between the early stages(stageⅠ and stage Ⅱ)and the advanced stages (stage Ⅲ and stage Ⅳ) were statistically significant(χ2 =24.941, P<0.001).The positive rate of combined detection in the diagnosis of lung cancer lymph node metastasis(76.31%) or distant metastasis(78.18%) was significantly higher than that of single detection. Meanwhile, the positive rate of combined detection in patients with lymph node metastasis or distant metastasis was significantly higher than that in patients without metastasis(χ2 =24.60, 9.50 respectively, all P values were<0.05).The combined detection had a better sensitivity(70.82%), accuracy(69.10%)and negative predictive value (59.91%)in LC group than each TM considered individually.The ROC curve showed that the AUC of combined detection in the diagnosis of LC, lung adenocarcinoma, lung squamous cell carcinoma and SCLC were 0.769, 0.780, 0.766 and 0.831, respectively.The combined detection of five tumor markers of CEA, NSE, SCCA, CYFRA21-1 and ProGRP by flow fluorescence technique can improve the diagnostic efficiency of lung cancer.

T cells engineered with chimeric antigen receptor (CAR) have been successfully applied to treat advanced refractory B cell malignancy.However,many challenges remain in extending its application toward the treatment of solid tumors.The immunosuppressive nature of tumor microenvironment is considered one of the key factors limiting CAR-T efficacy.One negative regulator of T cell activity is lymphocyte activation gene-3 (LAG-3).We successfully generated LAG-3 knockout T and CAR-T cells with high efficiency using CRISPR-Cas9 mediated gene editing and found that the viability and immune phenotype were not dramatically changed during in vitro culture.LAG-3 knockout CAR-T cells displayed robust antigen-specific antitumor activity in cell culture and in murine xenograft model,which is comparable to standard CAR-T cells.Our study demonstrates an efficient approach to silence immune checkpoint in CAR-T cells via gene editing.
