1.Purification of rough cordyceps polysaccharides solution by inorganic ceramic micro-filtration membrane

Objective To apply the membrane technology for purifying rough cordyceps polysaccharides solution,the optimal operation condition and effective cleaning method were researched.MethodsTo mainly analyze the flux of membrane and its recovery.Results The optimal experiment operation condition obtained is 60℃,pH value 7-8,and pressure 0.3-0.4 MPa,by which the polysaccharides recovery was about 30%,more than by traditional ethanol extraction,and the most impurities as infusibility substance and protein colloid etc.were taken out.Moreover,the flux of membrane recovery can be above 90% after 80℃ water and 1% NaOH solution was alternately used to wash the fouling membrane under intermittent little back-pulsing. Conclusion The process of membrane technology purifying rough cordyceps polysaccharides is simpl and feasible.
3.Preliminary analysis of miRNA expression profile of gastric cancer tissues by microarray

Objective To analyze the expression profile of specific miRNA of gastric cancer tissues.Methods miRNA were isolated from five paired gastric cancer tissues and corresponding paracancerous normal tissues.To identify the profile of gastric cancer,miRNA hybridization and cluster analysis were processed by significance analysis of microarrays (SAM,version 2.1) and cluster software.The relative expression of hsa-miR-30c,hsa-miR-196a and hsa-miR-193b were validated by fluorescence Real-Time PCR using taqman probe.Further hsa-miR-30c taget gene was predictived by online software.Results Revealed by miRNA microarray detection,there were 16 miRNA with differential expression in gastric cancer tissues and para-cancerous normal tissues,among which HS_100,hsa-miR-27a,hsa-miR-214*,solexa-555-1991 and hsa-miR-196 were significantly up-regulated,and hsa-miR-502-3p,hsa-miR-29c*,hsa-miR-64,hsa-miR-193b,hsa-miR-551b,hsa-miR-30c,hsa-miR-582-5p,hsa-miR-625*,hsa-miR-660,hsa-miR-363 and hsa-miR-486-5p were down-regulation.The relative expression of hsa-miR-30c,hsa-miR-196a and hsa-miR-193b was in high concordance with microarray.Conclusion There is specific miRNA expression profile in gastric cancer tissues,among which differentially-expressed miRNAs may be involved in the regulation of gastric cancer progress and serve as new potential biomarkers.

County -level public hospitals reform is an important measure of deepening reform of the medical and health system.Medicare payments system reform is therefore an important part of public hospitals reform.By abolishing drugs addition,registration fee,treating fee and setting up medical service fee instead,our hospital carried on a diversified management style in the aspect of medical insurance total prepaid system.The implementation of the reform measures turns out a good result:reasonably keeping a control on the medicare spending and really embodying the transformed of interests patterns.On the premise of ensuring patients interests,medical personnel′s interests are improved to a new level and medical insurance fund are also brought under a reasonable control.The medicare payments system reform puts equity in access and people benefit as the starting point and footing of county -level public hospitals reform,thus lays a foundation of further development of public hospitals reform.
5.Study of MK promoting breast cancer cell MDA-MB-231 angiogenesis through EPCR/PAR1 pathway in vitro

Purpose To observe the effects of midkine ( MK) on human breast cancer cell line MDA-MB-231 angiogenesis in vitro, and to explore its mechanism. Method shRNA interference was performed to silence the expression of MK in MDA-MB-231 cells, and Western blot was used to identify the expression of MK and EPCR. After MK and EPCR knockdown, or treated with anti protease-activated receptor 1 (PAR1) antibody, the culture medium of MDA-MB-231 cells were collected and the conditioned medium were pre-pared. Human umbilical vein endothelial cells ( HUVECs) were cultured with conditioned medium, and the endothelial cells prolifera-tion was detected by CCK-8 assay, cell migration was detected by transwell method, vasculogenic activity was assessed by Matrigel-based tube formation assay. Results After knockdown of MK, the protein level of EPCR was decreased in MDA-MB-231 cells. Com-pared with control, knockdown of MK and EPCR decreased the proliferation, migration and angiogenesis ability of HUVECs significant-ly (P<0. 05), and the effect of EPCR knockdown group was stronger than MK knockdown group (P<0. 05). After treated with anti-PAR1 antibody, the proliferation, migration and angiogenesis ability of HUVECs were decreased compared with control and EPCR knockdown group (P<0. 05). Conclusion MK promotes human breast cancer cell line MDA-MB-231 angiogenesis through EPCR /PAR1 signaling pathway in vitro.
6.Relationship between the yield and Extraction Time of Volatile Oil

Objective: To establish the relationship equation of yield and extraction time of plant volatile oil. Methods: By means of the rate eqation of substance transfer, the relationship equation of yleld (v) and extraction time (t) of volatile oil was accomplished: ln(V s-V)=-K t+A,K and A were parameters. Results: The experiment of volatile oil of Rhizoma Nardostachyos indicated that A was 0.44, K was 2.7?10 -3 , and the correlation coefficient (r) was -0.99 whereas A was 1.33, K was 3.62?10 -3 and r was -0.99 in the experiment of volatile oil of Flos Syzygii Aromatici. Conclusion: This relationship equation is able to reflect the relationship between the yield and extraction time of volatile oil.
7.Study on extraction of coumarins in Angelica Dahurica by supercritical CO_2

AIM: To optimize the extraction process for coumarins in Angelica Dahurica by supercritical CO 2 as guided by the contents of oxyimperatorin, imperatorin and the total coumarins in the extract. METHODS: HPLC was applied to determine the contents of oxyimperatorin and imperatorin. orthogonal design was applied to optimize SFE process. Range and variance analysis as well as stepwise nonlinear regression analysis were applied to deal with experimental results. RESULTS: The optimum process was established as following: 21MPa as extracting pressure, 50℃ as extracting temperature, 3.5h as extracting time, 20 mesh as pulverized degree and 6.5MPa as separating pressure. The relative error between experimental data and calculated value was less than 5%. CONCLUSION: Changes in extracting pressure, temperature, time, pulverized degree would affect the extracting results remarkably. Some factors had interactions on the extraction of coumarins in Angelica Dahurica . Regression equation was reasonable and could forecast results precisely.
8.Study on adsorptive properties of spherical activated carbon on several kinds of active ingredient from Chinese materia medica

ObjectTo study the adsorptive properties of spherical activated carbon on some effective ingredients from Chinese materia medica (CMM) and to approach the application feasibility to CMM purification. Methods The spherical activated carbon with high strength has been successfully prepared by the carbonation and activation of polymeric resin. The apparent and theoretical adsorption capacities under the condition of static operation were investigated by Langmuir monolayer adsorption model. Elution by 75% alcohol was studied. Results The static adsorption capacities of the berberine hydrochloride and dioscin were 35.46 mg/g and 47.12 mg/g, and theoretical adsorption capacities were 96.16 mg/g and 102.04 mg/g, respectively. The apparent adsorption amount of rutin was 40.88% mg/g. The elution ratio were 83.71%, 91.45%, and 87.69%, respectively. Conclusion The spherical activated carbon adsorbent shows better comprehensive adsorption properties and can be used to purification.
9.Characterization of Rhodiola crenulata/?-cyclodextrin complex

AIM:To characterize the inclusion complex of volatile oil of Rhodiola crenulata/?-cyclodextrin. METHODS: Some analytical methods,such as DTA,IR,GC,TLC were apllied to the investigation before and after the inclusion. RESULTS: The difference between the inclusion and physical mixture in differential thermal(analysis,) infrared spectra,gas chromatogram and thin layer chromatogram. CONCLUSION: The inclusion of Rhodiola crenulata volatile oil has the characteristic of intramolecular inclusion.
10.Characteristic and Resistance of Fungi Isolated from Hemopathy Patients

OBJECTIVE To investigate the distribution and resistance of fungi isolated from hemopathy patients,and to provide the laboratorial data in order to prevent and treat infection caused by fungi more effectively.METHODS Strains were isolated from the upper respiratory tract,lower respiratory tract,genitourinary tract,alimentary tract;and perianal region,perineum,and the soft tissue and skin wounds.Antimicrobial susceptibility of clinical isolates was tested by broth microdilution susceptibility test.RESULTS Candida albicans,Aspergillus,tropicalis,C.krusei,and C.glabrata were the main fungi of total 3104 detected isolates.The resistance rates of C.albicans and C.tropicalis to fluconazole,itraconazole and 5-flucytosine were no more than 1.4%.The MIC50-MIC90 range of C.tropicalis for itraconazole was narrow(0.25-0.5 mg/L),but that was wide for fluconazole(0.25-4.0 mg/L).CONCLUSIONS C.albicans,Aspergillus,C.tropicalis,C.krusei,and C.glabrata are possibly the most common pathogens causing endogenous and exogenous fungi infection.The main infection sites of Aspergillus are the upper respiratory tract and lower respiratory tract,which may be associated possibly with ward's air quality.The MIC50to MIC90 range of C.tropicalis is wider for fluconazole than for itraconazole,which is possibly correlated with overexpression of CtMDR1 in C.tropicalis caused by the widely application of fluconazole in therapy.
