Objective Inhaled corticosteroids combined with long β2-adrenoceptor agonist are widely used in treatment of chronic obstructive pulmonary disease, but mechanisms remain unclear. Methods Macrophage cells differentiated from THP-1 cell line were stimulated with dexamethasone and/or formoterol with or without lipopolysaccharide (LPS).TLR4 mRNA and protein were examined by reverse transcriptase polymerase chain reaction (RT-PCR) and Western blotting. Results A dose-dependent up regulation in TLR4 mRNA and protein expression in the presence of LPS was found.Treatment with dexamethasone (10-10 ~ 10-6mol/L) resulted in a dose-dependent reduction in TLR4 mRNA and protein.Stimulation with formeterol (10-7 mol/L and 10-6 mol/L) caused up regulation of TLR4.Formoterol (10-6mol/L)partially reversed the inhibitory effect of dexamethasone (10-6 mol/L) on TLR4 expression in the presence of LPS.Conclusions Modulation of TLR4 in macrophages by dexamethasone and/or formoterol may be one of the mechanisms for combination and have important implications for the treatment of airway inflammation in response to gram-negative bacteria.

Objective To explore the factors affecting results of alanine aminotransferase(ALT)preliminary screening by using dry chemical method in blood donors,so as to establish standards of ALT preliminary screening and reduce waste of blood. Methods A total of 21 065 blood specimen of blood donors in Blood Center of Maoming City were collected from January to June 2013.The influences of specimen hemolysis, lipidemia, different sample amount and waiting time after adding sample on the detec-tion results of ALT by using dry chemical method were comparatively analysed, and standardized procedures and established appro-priate limits of ALT preliminary screening according to the results. Results The specimen hemolysis affected test results of ALT, which was not obvious when the hemolysis rate was less than or equal to 0.5%.There was no obvious interference of lipidemia on ALT test. When the sample amount was (32±3)μL or the waiting time after adding sample was 10 to 40 seconds, the detection re-sults of ALT were acceptable. The standard limit of ALT was less than or equal to 40 U/L of female and less than or equal to 45 U/L of male. Conclusion Hemolysis, sample amount and waiting time after adding sample can influence the results of ALT detec-ted by using dry chemical method, while lipidemia had no interference on ALT test.Standardizing operation and reasonably setting normal limits standard may ensure the accuracy and stability of determination, save blood resources, further ensure blood safety, which should be worthy of wide application.

Objective To investigate liver regeneration after transplantation of microencapsulated hepatocytes in rats with acute liver failure (ALF). Methods ALF rat model was established by intraperitoneal injection of D-galactosamine (D-GalN). After 18 h, rats were randomized into control group ( Ⅰ ), free hepatoeyte transplantation group ( Ⅱ ) and the microencapsulated hepatecyte transplantation group (Ⅲ). Six rats for each group were randomly selected and sacrificed at 6, 12, 24, 36, 48, 72, 120, 168 and 240 h after ALF induced and blood samples from inferior vena cava were collected. Liver functions were tested in blood samples, and the expression of proliferating cell nuclear antigen (PCNA) was detected by immunohistochemistry. Results Ten-day survival rates of 3 groups were 26.7% (4/15), 40.0% (6/15) and 73. 3% (11/15), respectively (x2 = 9. 349,P = 0. 009). Survival rate of group Ⅲ was significantly higher than that of group Ⅰ and Ⅱ. Levels of ALT and AST in each group increased significantly at 6 h after ALF induced, and peaked between 48 ~ 72 h. Levels of ALT and AST in group Ⅱ and Ⅲ declined from 36 h, which was more significant in group Ⅲ. Tbil levels in group Ⅰ gradually increased after ALF induced and peaked at 72 h. Tbil in group Ⅱ and Ⅲ declined from 48 h, which was more markedly in group Ⅲ. In normal rats, the expression of PCNA protein was almost negative, but it was strongly expressed in ALF rats and peaked at 48 h. The number of positive cells in group Ⅲ was higher than that in group Ⅰ and Ⅱ, and the differences were of statistical signifieance. Conclusion The transplantation of microencapsulated hepatocytes can promote the regeneration of liver, and it can improve the liver function and prognosis in rats with ALF.
