1.Effect of tripterygium wilfkrdii Hook,ginseng and arotinoid ethyl ester on the sun-induced aging cultured fibroblasts of the human dermis

BACKGROUND:The expression of matrix metalloproteinases (MMPs) is enhanced in fibroblasts of aging skin; tripterine and ginsenoside Rd possess immunosuppression and anti-aging effects. Arotinoid ethyl ester can used to treat sun-induced aging skin,which accompanied by many side effects. OBJECTIVE:To discuss the regulation role of ginsenoside Rd,celastrol,arotinoid ethyl ester on MMP-1 and MMP-3 of the cultured fibroblasts of the human dermis in vitro. METHODS:The human dermis circumcision by surgical excision on newborn was obtained from Department of Surgery,Luzhou Medical College,and the informed consent obtained from patients. Culture of fibroblasts were as follow:①No treatment in the normal control group. ②Radiated with 80 kJ/m2 ultraviolet and treated by 100 mg/L 8-methoxypsoralen in the positive control group. ③Treated by ultraviolet radiation+8-methoxypsoralen+arotinoid ethyl ester in the arotinoid ethyl ester group. ④Treated by ultraviolet radiation +8-methoxypsoralen+triptolide in the triptolide group. ⑤Treated by ultraviolet radiation+8-methoxypsoralen+ginsenoside Rd in the ginsenoside Rd group. The triptolide group was divided into 3 groups with doses of 10,20,and 40 mg/L. The ginsenoside Rd group was divided into 3 groups with doses of 20,50 and 100 mg/L. The immunohistochemistry staining of MMP-1 and MMP-3 of the fibroblasts in each group was observed. RESULTS and CONCLUSION:The expressions of MMP-1and MMP-3 were significantly increased in the positive control group,which had a significant difference to the normal control group (P 0.05). The results demonstrated that:①Ginsenoside Rd and tripterine exhibit comparably effect on prevent and cure sun-induced aging skin. ②The wide ranges of effective concentration lead to little side effect to the body. ③ The action mechanisms of ginsenoside Rd,celastrol and arotinoid ethyl ester in preventing and curing sun-induced aging skin is adjusting the expressions of MMP-1 and MMP-3.

Objective To explore the therapeutic effect and the mechanism of allicin on experimental Behcet′s disease (BD) . Methods BD models were established .Then ,model mice were randomly divided into experimental group ,positive control group and negative control group ,with respectively nasogastric infusion allicin ,colchicine and saline respectively .Before and after treatment , the changes of the skin lesions were visually observed .Cytokines of IFN‐γ,TNF‐αand IL‐4 in serum were detected by ELISA anal‐ysis ,while the markers of T‐AOC ,MDA ,SOD and GSH‐PX by chemical colorimetric methods .Results After modeling ,ulcers ap‐peared on the back ,abdomen and genital of model mice with IFN‐γ,TNF‐α,IL‐4 and MDA elevation ,while T‐AOC ,SOD and GSH‐PX decrease .Twenty days of treatment later ,ulcer on mice in the experimental group and positive control group healed .After 30 days ,only two mice in the experimental group recurred ,while 6 mice in the positive control group ,which showed a significant differ‐ence between two groups (P<0 .05) .After treatment ,IFN‐γ,TNF‐αand IL‐4 levels in the experimental group were markedly low‐er than pretreatment (P<0 .01) ,but no significant difference compared with the positive control group (P>0 .05) .Meanwhile ,T‐AOC ,SOD and GSH‐PX levels in the experimental group were significantly higher than those in the pretreatment ,but MDA was lower (P<0 .01) .In comparison with the positive control group ,levels of those markers in the experimental group changed remark‐ably (P<0 .05) .Conclusion Allicin is an effective drug in the treatment of experimental BD .The therapeutic mechanism of allicin may be the inhibition of inflammatory mediators and prevention of oxidative stress .

Objective To observe clinical effects and side reactions of Huayu pill combined with TC (Paclitaxel+Carboplatin) chemotherapeutic regimes on treating epithelial ovarian cancer. Methods All 60 patients with the epithelial ovarian cancer were randomly divided into the treated group (Huayu pill combined with TC chemotherapeutic, n=30) and the control group (TC chemotherapeutic regimes, n=30). OBJECTIVE To investigate the use of antibiotics in our Outpatient Department during 2006-2007,and to analyze and evaluate the trend and rationality.METHODS The data about consumption quantity and consumption sum of antibiotics in our Outpatient Department during 2006-2007 were retrieved by means of computer management software in the department of pharmacy for the analysis of the clinical use of antibiotics using the method of defined daily dose(DDD).RESULTS Fluoro quinolones,cephalosporins and plant antibiotics have dominated the first places among the antibiotics used in our hospital,and the consumption of cephalosporins kept increasing year by year.CONCLUSIONS The use of antibiotics in our hospital should be further standardized to ensure the drug using safe,efficient and economic in our hospital.

OBJECTIVE To promote clinical rational use of antibiotic prophylaxis through surveying it in Obstetrics and Gynecology Department (OG) by pharmacists. METHODS Data of 223 cases using of antibiotic prophylaxis in OG were retrospectively analyzed. According to unreasonable use of antibiotic prophylaxis,the clinical pharmacists put out reasonable measures and directed treatment in succedent 206 patients. RESULTS The rate of unreasonable use of antibiotic prophylaxis in 223 cases was 100% and rate of infection after operation was 9.87%. Objective: To investigate the effects of hyaluroni dase (HAase) and hyaluronan (HA) on proliferation of vascular endothelial cells and its mechanism. Methods: The cultured bovine aortic endothel ial cells (BAEC) were treated with HAase or HA. Cell proliferation rate was dete cted by MTT assay. The expression of CD44 and DNA content of the cells were meas ured by flow cytometry (FCM). Results: HAase (50 μg/ml) stimula ted cell proliferation [(50.10±1.23)% vs control, P<0.01], incre ased S phase cell rate and induced the expression of CD44, but HA (100 μg/ml) i nhibited cell proliferation and the expression of CD44. Conclusion: HAase may degrade antiangiogenic HA of extracellular matrix, which may stim ulate proliferation of endothelial cells and enhance the curative effect of grow th factors to myocardial ischemia.
8.Establishment of reference intervals of dry chemistry tests among healthy population in Chengdu

Objective To establish the reference intervals of 20 dry biochemical items in different age periods among healthy population in Chengdu to providing better support and service for clinical diagnosis and treatment .Methods The stratified random‐ized cluster method was used to collect healthy children and adults in 4 age periods(1 month-3 years old ,>3-7 years old ,>7-18 years old and >18 years old) .Totally 1 495 healthy people (740 males and 755 female) were screened out as the research sub‐jects by the questionnaire ,physical examination and laboratory screening .Fasting venous blood samples were collected from these cases ,then the VITROS 5600 dry biochemistry analyzer was used to detect 20 biochemical items .The obtained results were statisti‐cally analyzed .Results In different groups according to sex and age ,except the conjugated bilirubin (BC) was constant 0 μmol/L , the other items had statistical differences (P<0 .05) .After merging the different groups without statistically significant difference , the obtained reference intervals had significant differences compared with the reference intervals provided by the manufacturer .Con‐clusion Laboratory should establish the different reference intervals aiming at different age and gender populations according to the special character of hospital visiting populations in order to meet the clinical requirements .
9.Study on the Total Flavonoids Content and Antioxidant Activity in Different Parts of Eucommiae ulmoides

OBJECTIVE:To compare the total flavonoids content and antioxidant activity in the barks,leaves,male flowers and seeds of Eucommiae ulmoides. METHODS:UV spectrophotometry was used to determine the total flavonoids content in differ-ent parts;tests was conducted to clear 2,2′-nitrilobis(3-ethylbenzothiazoline-6-sulfonic acid)(ABTS+),1,1-diphenyl-2-trinitro-phenylhydrazine(DPPH)radicals and the reducing ability of Cu2+,using half clear/reduction concentration value(IC50)as evalua-tion indexes,and vitamin C was regarded as positive control. RESULTS:The total flavonoids content of the E. ulmoides from hight to low was as follows as leaves>male flowers>barks>seeds,except there was no significant difference in barks and seeds (P>0.05),the other parts had significant differences (P<0.05);the ability of different parts eliminating DPPH and ABTS+ free radical was as follows as leaves>male flowers>seeds>barks,except there was no significant difference in the indicators of leaves and male flowers(P>0.05),the other parts had significant differences(P<0.05);the ability of reducing Cu2+ free radical was as follows as leaves>male flowers>barks>seeds,there was significant difference in leaves and males flowers with barks and seeds (P<0.05). CONCLUSIONS:The content of total flavonoids in leaves and male flowers is high,and the antioxidant activity is strong,which has a great prospect of exploitation and utilization to make up for deficiencies in barks of E. ulmoides.
