China Oncology 2016;26(3):230-237
Background and purpose:Osteosarcoma, a highly malignant bone tumor, develops rapidly. The current medicines for osteosarcoma present some limitations with serious side effects of long-term use. Isosteviol has the structure of tetracyclic diterpene which is the starting material of many anti-cancer drugs. However, its anti-tumor activity has been rarely reported. This study investigated the effect of isosteviol on proliferation of human osteosarcoma cell line U-2OS.Methods:The effect of isosteviol on U-2OS cell proliferation was assayed by MTT method. Cellular morphologic changes were observed under an inverted phase contrast microscope. The cell condition was observed with Hoechst 33342 and PI staining. Generation of reactive oxygen species and cell membrane potential were detected as well. The cell cycles were analyzed with lfow cytometry. The expressions of apoptosis-related proteins Bcl-2 and Bax were measured by Western blot assay. Results:The result indicates that isosteviol suppressed the growth of U-2OS cells in time- and concentration-dependent manner. Isosteviol could cause S phase cell cycle arrest at 24 h and apoptosis at 48 h. With the increased drug concentration, reactive oxygen species increased significantly, and the membrane potential gradually reduced. In addition, isosteviol treatment enhanced the expression of Bax but reduced that of Bcl-2.Conclusion:The inhibition of isosteviol on cell growth of U-2OS cells was possibly caused by promoting apoptosis through regulating the apoptosis-related protein expressions, such as the enhancement of Bax and reduction of Bcl-2 expression.

Shanghai Journal of Acupuncture and Moxibustion 2015;(3):246-248
Objective To observe the clinical efficacy of superficial needling in treating temporomandibular joint disorder. Method Sixty patients with temporomandibular joint disorder were randomized into a treatment group and a control group, 30 in each group. The treatment group was intervened by superficial needling, and the patients were asked to repeatedly open and close mouth after the treatment; the control group was intervened by regular acupuncture. The treatment was given once a day, 10 d as a treatment course. The clinical efficacies were compared after 2 treatment courses.Result After the first treatment course, the total effective rate was 89.7% in the treatment group versus 86.2% in the control group; after the second course, the total effective rate was 96.6% in the treatment group versus 93.1% in the control group. There was a significant difference in comparing the therapeutic efficacy between the two groups after the second course (P<0.05). The relapse rate was 5.0% in the treatment group versus 33.3% in the control group, and the difference was statistically significant (P<0.05).Conclusion Superficial needling is an effective method in treating temporomandibular joint disorder.

PubMed,Embase,Cochrane Library, and ISI web were searched without any limitations with regard to publication date or language,as well as the references of qualifying articles. All kinds of cohort studies comprising the risk of gastric cancer between diabetic patients and control subjects were included. We excluded studies that investigated only mortality but not incidence.25 studies met our criteria, and the qualities of these studies were evaluated using the Newcastle-Ottawa Quality Assessment Scale.Statistical analyses were carried out with STATA software version 12.0.A random-effects model meta-analysis showed an increased risk of gastric cancer in diabetic patients (RR =1.20,95 % CI 1.12-1.28).Subgroup analyses indicated that this result persisted in studies of both male(RR =1.15,95% CI 1.02-1.29) and female (RR =1.29,95% CI 1.09-1.53) subjects,in studies of European countries(RR =1.25,95% CI 1.07-1.46) and Asia countries (RR =1.18,95% CI 1.09-1.28).Compared with nondiabetics, the incidence of gastric cancer may be increased by approximately 20% in diabetics. Thus diabetes may be an independent risk factor for gastric cancer. This effect tends to be more significant in type 1 and female patients.

Objective To evaluate the early emergency management process for severe pelvic fractures combined with multiple trauma. Methods The study involved 113 patients with severe pelvic fractures combined with multiple trauma treated from January 2007 to May 2011.The patients were divided into two groups,ie,before trauma team establishment (26 patients treated from January 2007 to May 2008) and after trauma team establishment (87 patients treated from June 2008 to May 2011 ) in accordance with the establishment period of trauma team in June 2008.The mortality was compared before and after application of trauma-team-based algorithm with damage control resuscitation, external fixation and angiographic embolization.Variables including age and gender distribution,ISS score, injury type and mechanism, transportation time, associated injury number, initial systolic pressure hemoglobin and blood transfusion were analyzed for study of their relation with mortality. Results After trauma team establishment, the mortality (23%) was significantly lower than before trauma team establishment (46%).The mortality showed significant relation to ISSN score and over two regions of associated injury, but showed no correlation with age and gender distribution, injury type and mechanism, transportation time initial systolic pressure hemoglobin, and blood transfusion. Conclusion The application of trauma team is effective in treatment of severe pelvic fractures combined with multiple trauma and can decrease the mortality.

Clinical skill competition based on objective structured clinical examination could evalu-ate students' clinical abilities effectively and fairly in a comprehensive way. The competition results not only reflected the learning effectiveness of the competitors but all the medical students. Analysis and exploration of the inspiration from the competition could promot the all-round development of students and boost the continuous growth and progress of clinical teaching.

Objective To investigate the expressions and clinical significances of breast cancer stem cell markers such as aldehyde dehydrogenase 1 (ALDH1) and transforming growth factor-β2 (TGF-β2) in patients with triple negative breast cancer. Methods Samples of 60 triple negative breast cancer tissues were investigated for the expressions of ALDH1 and TGF-β2 proteins by immunohistochemical staining. The correlation analysis,disease-free survival analysis and overall survival analysis were performed. Results The positive expressions of ALDH1 protein and TGF-β2 protein in the 60 breast cancer primary lesions were 23 cases (38.33%) and 38 cases (63.33%) respectively. The expression of ALDH1 protein was not correlated with tumor size (x2 =0.307,P =0.580),histological grade (x2 =4.244,P =0.120),clinical stage (x2 =0.982,P =0.612) or lymph node metastasis (x2 =1.111,P =0.292).The expression of TGF-β2 protein was not correlated with histological grade (x2 =4.651,P =0.098),lymph node metastasis (x2 =3.513,P =0.061),clinical stage (x2 =1.310,P =0.519) or tumor size (x2 =0.629,P =0.428).The disease-free survival time [(38.43±3.86) months vs.(53.38 ±2.58) months] and the overall survival time [(42.00±3.11) months vs.(53.84 ± 2.19) months] of ALDH1-positive patients were significantly shorter than those of ALDH1-negative patients, and the differences were statistically significant (x2 =8.490,P =0.004;x2 =11.270,P =0.001).The disease-free survival time [(42.81 ±3.32) months vs.(54.72 ±2.50) months] and the overall survival time [(44.74 ± 2.68) months vs.(57.18 ± 1.55) months] of TGF-β2 positive patients were significantly shorter than those of TGF-β2-negative patients, and the differences were statistically significant (x2 =4.300,P =0.038;x2 =8.900,P =0.003).The expression of ALDH1 protein was positively correlated with the expression of TGF-32 protein (r =0.360,P =0.005).Conclusion The ALDH1 phenotype is an independent predictor of poor prognosis. The activation of TGF-32 signaling pathway may be involved in the regulation of triple-negative breast cancer stem cells.
