1.Clinical Analysis on Coagulation Markers in Advanced NSCLC Patients

Objective To discuss the distribution of coagulation markers in patients with advanced non-small cell lung cancer (NSCLC) and the correlation of these markers with the prognosis of the patients, the TCM syndrome and the long-term thrombotic events. Method Hospitalized NSCLC patients were selected for retrospective investigation and the coagulation markers data were analyzed with SPSS13.0. Result There were 10 cases (38.5%) and eight cases (30.8%) with one or more abnormal coagulation markers in the non-advanced NSCLC group, while there were 95 cases (71.4%) and 77 cases (57.9%) in the advanced NSCLC group. Objective The study aimed to construct one severity of illness scoring system specially for children with complex congenital heart diseases and analyzed the applicability of this new score,which will be helpful to assist the medical workers in evaluating children's condition.Methods A total of 1284children patients with complex congenital heart diseases in Shanghai Children's Medical Center Affiliated to School of Medicine of Shanghai Jiaotong University.The general information and clinical data were collected.Logistic regression,method of percentiles and expert opinions were used to construct the new severity of illness scoring system.Area under ROC curve,Hosmer-Lemeshow,goodness of fit test were used to evaluate the performance and applicability of the new score.Results A total of 1284 children with complex congenital heart diseases split into construction group (1138 cases) and verification group (146 cases).The new score included seven indexes (systolic blood pressure,blood oxygen saturation,lactic acid,potassium,potential of hydrogen,glucose,positive inotropic drug score).The range of the score was 0-57 and the cut-off point was 10.The discriminatory power was sufficient (area under the curve > 0.75),and the calibration capability of the score was tolerable (P > 0.05).Conclusions The applicability of the new score is well and it is easy to use,which will not increase the workload for the medical workers while assisting them in the clinical environment.
3.Antigen-loaded dendritic cells and CD40L triggers the killing effects of cytotoxic T lymphocytes on K562 cells in vitro

AIM:To investigate the anti-tumor effects of special cytotoxic T lymphocytes(CTLs)activated by dendritic cells(DCs)loaded with antigens and CD40L in vitro.METHODS:Peripheral blood mononuclear cells were isolated by Ficoll density gradient centrifugation from normal human heparinized blood.The adherent cells were cultured with granulocyte-macrophage colony stimulating factor(GM-CSF),interleukin-4(IL-4),alpha tumor necrosis factor(TNF-?),DCs were co-cultured with frozen-thawed antigen of K562 cells and CD40L,then triggered T cells into specific CTLs.RESULTS:Most suspended cells exhibited distinctive morphological features of DCs which expressed CD40 96%,CD86 97%,CD80 77%,CD1a 69%,and gained the powerful capacity to stimulate proliferation of allogenic lymphocytes.Under the effector∶target ratio of 20∶1,CTLs derived from cultures with DCs and frozen-thawed antigen of K562 cells were showed 71.3% cytotoxicities against K562 cells.CTLs derived from cultures with DCs loaded with frozen-thawed antigen and CD40L were showed 86.9% cytotoxicities against K562 cells.Cytotoxicities by CTLs derived from cultures with unloaded DCs against K562 cells were 37.6% and cytotoxicities by monocytes were 21.1%.Cytotoxicities by CTLs derived from experiment groups were stronger than control groups(P
4.Clinical Distribution and Drug Resistance Analysis of 60 Stenotrophomonas maltophilia Isolates

OBJECTIVE To investigate the clinical distribution and drug resistance status of Stenotrophomonas maltophilia and provide clinical guidance for treatment.METHODS Sixty clinical isolates of S.maltophilia were identified with GNI+ cards of VITEK-32.Drug sensitivitiy were detected by Kirby-Bauer disc diffusion method.The data were analyzed by WHONET5.3 software.RESULTS Thirty-eight strains were isolated from sputum(63.3%).Infection caused by S.maltophilia mainly occurred at the departments of respiratory diseases,ICU,old cadre,et al.Sixty isolates of S.maltophilia were highly resistant to imipenem,aminoglycosides and most of ?-lactam antibiotic,but showed the lowest resistance rate(16.7%) to SMZ/TMP.Then resistance rate of S.maltophilia to minocycline,levofloxacin,cefoperazone/sulbactam,piperacillin/tazobactam,and ticarcillin/clavulanic acid was 18.3%,20.0%,21.7%,25.0% and 30.0%,respectively.CONCLUSIONS The drug resistance of S.maltophilia is extremely severe.Among different areas of china the drug resistance is obviously different.Treatment based on drug susceptibility test should be adapted as soon as possible.
5.Retinervus luffae fructus protects against the myocardial ischemic injury in mice

AIM: To observe the effect of Retinervus luffae fructus (RLF) on the myocardial ischemia in mice. METHODS: Mice were supplied with RLF (166.7,333.3, 666.7 g/L) twice daily (0.01 mL/g, ig). One week later, 30 U/kg pituitrin was injected intraperitoneally. The electrocardiogram was recorded, and the content of lactose dehydrogenase (LDH) in the serum and the activities of superoxide dismutase (SOD), maleicdialdehyde (MDA) in the myocardium were examined. RESULTS: RLF reduced the height of T wave in electrocardigram in myocardial ischemia mice and inhibited the decrease in heart rate. RLF also reduced the content of LDH in serum and the content of MDA in myocardium. The increase in the activity of SOD in myocardium was also observed. CONCLUSION: RLF may protect myocardium against ischemia injury. The mechanism may be related to the increase in activity of SOD and the suppression of the lipid peroxidation.
6.Effects of electrical stimulation of ventral septal area on discharge of pyrogen-treated thermosensitive neurons in preoptic anterior hypothalamus region of rabbits

AIM and METHODS: To investigate the effect of electrical stimulation of ventlal septal area (VSA) on discharge of pyrogen - treated thermosensitive neurons in preoptic anterior hypothalamus (POAH) region, the discharging rate of thermosensitive neurons in the POAH region of 32 New Zealand white rabbits were recorded by using extracellular microelectrode techinque. RESULTS: (1) Intraceretroventricularly (i c v) injection of interleukin - 1?(IL - 1?) caused decrease of discharging rate of warm - sensitive neurons and increased in discharging rate of cold - sensitive neurons in POAH regions. (2) These effects could be reversed by electrical stimulation of VSA. CONCLUSION: VSA may play a central role of negative regulation in thermoregulation of pyrogen - treated animal.

AIM: To investigate the effect of granulocyte colony stimulating factor (G-CSF) on arabinosyl cytosine (Ara-C)-induced apoptosis in HL-60 leukemia cells. METHODS: The proliferation of HL-60 leukemia cell was observed by hemopoietic cell culture. 8.Glycine protection against the myocardial ischemic injury in mice

AIM: To observe the effect of glycine on the myocardial ischemic injury in mice METHODS: Mice were supplied with 20% glycine twice daily (0 025 mL/g, ig), or 40% Injection salviae miltiorrhizae composita (ISMC, 0 025 mL/g, ig) One week later, 0 02 U/g pituitrin was injected intraperitoneally The electrocardiogram was recorded, and activities of nitric oxide synthase (NOS), superoxide dismutase (SOD) and the content of malondiadehyde (MDA) in the myocardium were examined RESULTS: Glycine reduced changes in J spot in electrocardiogram Both glycine and ISMC increased activities of SOD and NOS, inhibited increase in MDA content in the myocardium induced by pituitrin There was no difference in above parameters between glycine-treated and ISMC-treated mice. CONCLUSION: These results demonstrated that glycine can protect myocardium from ischemic injury, the mechanism may be related to increase in activities of NOS, SOD and supressing the lipid peroxidation.
9.Effect of food intake on functional activities and biochemical index in mice

AIM: To study the effect of food intake on weight, memory, staying balance and power, and some biochemical indexes in mice. METHODS: In accordance with the average food intake everyday, the animals were divided into five groups: group A took the average food intake fully, group B took 75% of the average food intake, group C took 50% of the average food intake, group D took 25% of the average food intake, and group E did not take any food at all. The weights, memory, staying balance and power were recorded every three days, the biochemical index was recorded on the 10th day. RESULTS: On the 3rd day, the weight and staying balance and power of mice in group E decreased. On the 6th and 9th day, the weight of mice in group ABC increased. Compared with group B and C, the memory group A and D decreased. On the 10th day, the blood glucose concentratoin in of group D decreased, total cholesterol and triglyceride also decreased with the reduction of food intake. CONCLUSION: A food intake of 75%-50% in all helpes to keep a good body status and memory, decreases total cholesterol and triglyceride in blood.
10.Determination of the Content of Entecavir and Its Related Substances by HPLC

OBJECTIVE: To establish a HPLC method for the determination of content of entecavir and its related substances. METHODS: The HPLC condition was consisted of a Luna C18 column with a column temperature at 25 ℃, using gradient eluate method, the mobile phase was acetonitrile,with a flow rate of 1.0 mL?min-1 and detection at 254 nm. RESULTS: The calibrated linear curve of entecavir was within 0.033 45~0.167 2 mg?mL-1(r=0.999 98). The average recovery was 100.09%,RSD=0.20%, The content of related substance was 0.54%~1.48%. CONCLUSION: This accurate and reliable HPLC method is applicable for the quality control of entecavir and its related substances.
