1.Application of epidermal growth factor receptor tyrosine-kinase inhibitors combined with chemotherapy in advanced non-small cell lung cancer

Currently, histological and molecular methods are considered for the treatment of advanced non-small cell lung cancer (NSCLC). Single-agent epidermal growth factor receptor tyrosine-kinase inhibitors (EGFR-TKIs) and anaplastic lymphoma kinase in-hibitors (ALK-TKIs) have been used as standard first-line therapies for patients with active EGFR mutation and ALK rearrangement, re-spectively. However, to date, the single-agent EGFR-TKIs as the first-line therapy for patients with known EGFR mutations has been demonstrated to provide a prolonged progression-free survival but does not affect overall survival (OS). Physicians these days focus on improving the OS of patients with advanced NSCLC. To patients with EGFR mutation, combining and maintaining EGFR-TKIs with chemotherapy could be a promising approach. In this article, various ways of combining EGFR-TKIs with chemotherapy were explored.

3.NATURALLY OCCURING ANTIBIOTIC RESISTANCE IN BACILLUS SPHAERICUS

The naturally occuring antibiotic resistance in twentyseven strains from 7 different serotypes of Bacillus sphaericus was investigated. The antibiotics used in this experiment are Streptomycin, Ampicilline, Tetracycline, Chloramphenicol, Kanamycin, Erythromycin, Qingdamycin, Midecamycin, Ribostamycin, Acriflavin, Rifampicin, Sulphanilamide and Azide. All strains are resist to streptomycin and azide at higher concentration and chloramphenicol at lower concentration. Most of strains are resist to tetracycline at lower concentration.
4.Pharmacological effects of anti-melanoma DC vaccine sensitized by fusion proteins of G3G6 and HST1

This study aimed to investigate the effects of fusion proteins GnRH-GRP(G3G6)and HSP65-STEAP1(HST1)on dendritic cells(DC)and the sensitization of DCs to B16F10 melanoma. The fusion proteins G3G6 and HST1 were obtained using the previous engineering strains in our laboratory. Group by unsensitized DC(US-DC), the G3G6 fusion protein sensitized DC, the HST1 fusion protein sensitized DC(HST1-DC)and the combined sensitized DC(GH-DC), the mouse bone marrow-derived DCs were sensitized with fusion protein to obtain the fusion protein sensitized DC vaccines. B16F10 melanoma cells were transplanted into C57BL/6J male mice to construct a melanoma model(1×106 cells per mouse), and DC vaccine was injected for treatment. The antitumor efficacy of DC vaccine was explored by in vitro and in vivo experiments. Flow cytometry analysis showed that the fusion protein can effectively stimulate DC into differentiation and maturation; in the animal experiment, the inhibition rate of melanoma treated with G3G6-DC was 35. 75%, that of HST1-DC group and combination group were 34. 03% and 55. 74%. It was initially proved that both G3G6-DC and HST1-DC can effectively inhibit the growth of transplanted tumors of melanoma B16F10 cells in mice, and the combination therapy is superior to the single therapy.

6.Three indicators combined detection of the application of ICU in early diagnosis of sepsis patients

Objective To investigate the application of combined detection of serum calcitonin(PCT),interleukin-6 (IL-6)and C reactive protein(CRP)in early diagnosis of sepsis in intensive care unit(ICU).Methods ICU ward diagnosed 89 cases of patients with early sepsis(sepsis group),fever and did not peak use of antibiotics,send blood to blood culture and detection of PCT,IL-6,CRP level,and at the same time to select 132 cases of healthy subjects(healthy control group),blood samples were collected to detect PCT,IL-6,CRP;PCT,IL-6 for the detection of electrochemical luminescence method.The detection of CRP by immunoturbidimetric method.Results ICU patients with sepsis in PCT,IL-6,CRP detection value is significantly higher than the control group,and the difference was statistically significant(P<0.01);combined detection sensitivity and specificity was significantly higher than that of single detection,PCT+ IL-6,PCT+ CRP,PCT+ IL-6-+-CRP sensitivity of three kinds of combined detection were 91.3 %,90.5% and 91.9%,the specificity was 89.5%,88.3% and 89.4%,and accuracy were 85.5%,85.3% and 85.6%.Conclusion Combined detection of PCT+ IL-6 + CRP or PCT+ IL-6 or PCT+CRP,is helpful to the clinical recognition of early sepsis.
7.Effects of propofol on learning and memorizing ability and hippocampus TLR4 in epileptic rats

Aim To explore the effects of propofol on learning and memorizing ability and the effects of com-bination of propofol and phenobarbital sodium on epi-leptic rats.Methods Thirty-six epileptic rats were di-vided into epileptic model group (EP),normal saline group (NS),lipid emulsion +epileptic group (LE), phenobarbital sodium +epileptic group (PB),propofol+epileptic pattern (Prof),and combination of propo-fol and phenobarbital sodium +epileptic group.Each group had 6 rats.Tests of Morris water maze were giv-en to the rats to evaluate their learning and memorizing ability.The protein expression of toll-like receptor 4 (TLR4 )was examined by ELISA.Results There were no effects of saline and lipid emulsion on learning and memorizing ability and the expression of TLR4 pro-tein in hippocampus in epileptic rats (P >0.05 ). Propofol could increase the incubation period in epilep-tic rats obviously,shorten the plateau period,and in-crease the expression of TLR4 protein in hippocampus (P <0.05 ).Phenobarbital sodium could shorten the plateau period in epileptic rats,and increase the ex-pression of TLR4 protein in hippocampus (P <0.05), but it had no effect on the incubation period (P >0.05).Compared with PB,combination of propofol and phenobarbital sodium +epileptic group had a lon-ger incubation period and a shorter plateau period with an increase of the expression of TLR4 protein in hippo-campus (P <0.05 ).Compared with propofol group, combination of propofol and phenobarbital sodium +ep-ileptic group had a shorter plateau period (P <0.05) with an obvious increase in the expression of TLR4 pro-tein in hippocampus (P <0.05),but it had no effect on incubation period (P >0.05 ).Conclusions Propofol damages the learning and memorizing ability of epileptic rats.Phenobarbital sodium had no obvious effect on the learning ability in epileptic rats,but harms the memorizing ability in epileptic rats.Combi-nation of propofol and phenobarbital sodium affects the learning and memorizing ability of epileptic rats.Hip-pocampus TLR4 may be involved in the effect of propo-fol and phenobarbital sodium on the learning and mem-orizing ability of epileptic rats.
8.Needs analysis of caregivers of patients with schizophrenia for mental health knowledge

Objective To investigate needs of caregivers of patients with schizophrenia for mental health knowledge and provide the scientific evidence for formulating the general intervention to improve the caregivers′mental health knowledge and skills . Methods Sixty-five caregivers of the patients with schizophrenia participated in the survey by a general state questionnaire and the requirement to mental health knowledge and skill questionnaire using the convenience sampling method. The survey included mental illness knowledge drug, knowledge rehabilitative therapy and self-adjustment. Results Among all the items of the questionnaire, the 3 items they needed most was mental illness knowledge, the preventing method and the symptoms of the recurrent diseases. The 3 items they needed most in respect of drug knowledge were what examinations are needed when taking special drugs, the adverse reactions of the antipsychotic drugs and the importance of persisting in taking drugs. The 3 they needed most in respect of rehabilitative therapy were the relationship between psychology and disease rehabilitation, the role of mental treatment and the role of work-entertainment therapy. The 3 items they needed most in respect of self-adjustment included sleep adjustment, self-adjustment of affects and self-management of daily life and instructions on dieting. Conclusions The caregivers′mental health knowledge and skills are not enough. Therefore, their mental health knowledge and skills should be strengthened so that they can take better care of the patients to realize comprehensive rehabilitation of patients.

Based on the optimized technology on 50L scale, B subtilis 754# was used to manufacture guanosine on pilot scale (12M 3) and plant scale (100M 3) to give the production of 29 4g/L and 21 4g/L successively Through scale down the process of plant scale to pilot scale, based on the theory of dynamic metabolic flux, DO was found to be another key factor that limited the scale up process After conquering the DO limitation, the optimized technology was successfully scaled up to plant scale and the production was further increased by 18% to 25 2 g/L
10.Relationship between plasma concentration of fluorouracil and the chemotherapy efficacy and prognosis in patients with advanced colorectal cancer

Objective To detect the plasma concentration of fluorouracil in patients with advanced colorectal cancer who uesd fluorouracil , and to explore the relationship between chemotherapy efficacy , tumor response, toxicity and survival of patients with advanced colorectal cancer after chemotherapy with fluorouracil.Methods 174 patients with advanced colorectal cancer were collected from the General Hospital of Ningxia Medical University from January 2014 to December 2016.Patients with advanced colorectal cancer were treated with FOLFOX scheme , venous blood was taken from patients 18-24 hours after intravenous infusion of fluorouracil .The concentration of 5-FU in blood was measured by nami-enhanced immune assay.The targeted AUC range of 5-FU was 20-30 mg· h/L.The patients were divied into three groups-targeted range group , lower than the targeted range group and higher than the targeted range group . The therapeutic efficacy, toxicity and survival of patients were recorded , in order to observe the difference of different target range groups.To analyze the correlation between the different target groups and the clinical characteristics,a logistic method was used.Chi-square test was used to compare the efficacy and toxicity of the three groups.The survival curves were plotted by kaplan-meier method. Results To detected the concentration of fluorouracil at the same time after infusion of fluorouracil .The AUC was 2.267-31.06 mg· h/L,it was very different, the median AUC was 15.368 mg· h/L, and the highest AUC was 31.06 mg· h/L,it was 13.7 times that of the lowest blood concentration (2.267 mg· h/L) .The relationship between the concentration of fluorouracil and the clinical indicators was found as follows :(1)Clinical characteristics : the blood concentration of 5-Fu after infusion was different , which was related to the preoperative CEA status (r=0.318,P<0.05), EGFR(r=0.558,P<0.05), Lymph node metastasis status(r=0.325,P<0.05), and the number of metastatic organs ( r=0.404, P<0.05 ) .( 2 ) Effect of chemotherapy : There were significant differences in the effect of chemotherapy between different target groups (χ2=6.78,P<0.05). The effect of chemotherapy in the targeted range group was lower than that in the target range group , and the difference has statistically significant(χ2=2.030,P<0.05).(3)toxicity: there have statistical difference among the three groups,the toxicity of higher targeted range group was higher than the other two groups (χ2=2.01,P<0.05).(4)Survival: The survival time of patients with targeted range group was lower than that of targeted range group, and the difference has statistically significant (χ2=7.263,P<0.05).Conclusion The method of testing the concentration of fluorouracil in patients with advanced colorectal cancer is valuable to predict the efficacy , toxicity and prognosis of patients with colorectal cancer.
