1.Retrovirus mediated IL-23 gene expression in mouse high-metastasis mammary cancer cells MA-891

Objective:The mouse mammary cancer cell line IL-23/MA-891 expressing IL-23 protein was set up and the effect of IL-23 on growth and other biological character of the cells was studied.Methods:The IL-23 gene was sequencely transfected into two packing cell lines (ecotropic ?2 and amphotropic PA317) by a retrovirus vector (LXSN),and the positive cellular clones were screened by G418.After infection to MA-891 cells with the culture supernatant of IL-23/PA317 cells and selected with G418,the IL-23/MA-891 cells to express IL-23 mRNA and protein was detected with RT-PCR,ELISA and immunocytochemical stairing,respectively.The expression of H-2Kb(MHCⅠ),I-Ab(MHC Ⅱ),CD80,CD86 and FAS was examined with flow cytometry.The ability to secrete IFN-? by mouse splenocytes stimulated with the culture supernatant of IL-23/MA-891 cells was detected with ELISA.The proliferation of MA-891,LXSN/MA-891 and IL-23/MA-891 cells was detected by MTT colorimetry and flow cytometry in vitro.Results:IL-23/MA-891 cells express IL-23 stably in mRNA and protein level.The expressing of H-2Kb(MHCⅠ),I-Ab( MHCⅡ),CD80,CD86 and FAS protein by flow cytometry were showing no difference in three kinds of cells.The proliferation rate of IL-23/MA-891 cells were showing little decresed,but statistically showing no difference with parental cells.The culture supernatant of IL-23/MA-891 cells induced secretion of IFN-? by mouse splenocytes were increased compared to parental cells.Conclusion:The cellular clone IL-23/MA-891 with high level expression of IL-23 was produced.
2.Role of endogenous arginine vasopressin in soman-induced hypothermia in rats

AIM: This study was designed to examine whether endogenous arginine vasopressin (AVP) is involved in soman-induced hypothermic process. METHODS: Core temperature was measured at 60 min intervals with digital thermometer. Effect of AVP V 1 receptor antagonist (30 ?g/kg, ip) on soman-induced hypothermia was observed in rats, and plasma AVP concentration was measured at 2 h after administration of soman(60 ?g/kg, sc). RESULTS: Administration of soman led to a marked hypothermia. Core temperature recovered to basal levels at 7 h after soman treatment. Plasma AVP concentration increased markedly at 2 h after soman treatment, and administration of AVP V 1 receptor antagonist markedly blocked the hypothermic effect of soman. CONCLUSION: The data indicate that AVP is involved in soman-induced hypothermia in the rat.
3.Clinical aralysis of the increase in full three-dimensional conformal radiotherapy for esophageal cancer

Objective To evaluate the effct of full increase of three-dimensional conformal radiotherapy on esophageal carcinoma.Methods 86 Esophageal cancer patients which has been pathologically confirmed were randomly divided into study group and control group.The control group showed in tumor invasion scope simulator under barium meal positioning,with three fields outside irradiation;Total dose of DT with 63~70 Gy,35 times,seven weeks to complete.Study group showed that full use of additional 3D treatment planning system design individualized treatment programs,90% of the dose contains all planned target volume,future confonnal irradiation DY 44~50 Gy,after the wild-shrinkage increases to a total dose DT66~75 Gy,30 times,completed six weeks.Results The study group and the control group after radiotherapy in a local control rates were 86.4%and 64.3%(x2=4.5420,P<0.05).Overall response rate(CR+PR)in two group were 93.2%and 76.2%(x2=3.6014,P=0.0577).Two toxicity of the difference was not significant.Conclusion The increase in full three-dimensional conformal radiotherapy of esophageal emleer in the near future is better than conventional methods.
4.Synthetic rehabilitation treatment of congenital muscular torticollis

ObjectiveTo investigate the relationship between the prognosis and both of treatment way and curative age of congenital muscular torticollis.Methods138 children accepted conservative and/or operative treatment according to different ages.A prospective study had been made. ResultsConservative treatment was given to 82 children,71 of them were <1-year-old. 49 of them were follwed up and had satisfactory outcome.7 patients who was >1-year-old had operations later because of asymmetry of head.63 children received operations.56 of them were followed up over one year,47 cases had good-looking,the outcome of other 9 was not satisfied.Conclusions Operative intervention was necessary for >1-year-old patients. Manipulation was suitable way to cure <1-year-old patients.It was the best operating time at the age of 1-5 years old.
5.Mechanism of action of miRNA-146a in sepsis-induced acute lung injury

Object To study the expression of miRNA-146a in sepsis-induced acute lung injury (ALI) patients and its effect on the inflammation in mouse models.Methods miRNA-146a expression in peripheral blood was determined in sepsisinduced ALI patients and healthy volunteers by qRT-PCR.Sepsis-induced ALI mouse model were reproduced by LPS treatment and miRNA-146a mimic,miRNA-146a NC and miRNA-146a inhibitors were injected through trachea.The expressions of miRNA-146a,TNF-α,IL-1β and COX-2 mRNA were determined by qRT-PCR 24h after the intervention.Results The miRNA-146a expression in peripheral blood significantly increased in severe sepsis with ALI patients,compared with the control subjects.For mouse model,the expressions of TNF-o,IL-1β and COX-2 mRNA significantly decreased in miRNA-146a group,while increased in miRNA-146a inhibitor group compared with miRNA-146a NC group (P<0.05).The TNF-α and IL-1β expressions significantly decreased in miRNA-146a inhibitor+COX-2 inhibitor group compared with miRNA-146a inhibitor group (P<0.05).Conclusion MiR-146a treatment can effectively alleviate the lung inflammation in sepsis-induced ALI mice.
6.Diagnosis of Prenatal MRI in Placenta Implantation Abnormality

Purpose To discuss the value of MRI in the diagnosis of placenta implantation abnormality, and to explore preliminarily the relationship between MRI signs and types of placenta implantation abnormality. Materials and Methods The clinical preoperative data and postnatal pathological findings of 54 women at high risk of placenta accreta were collected. All the patients undertook the conventional pelvic MRI examination. The scanning sequences mainly included: sagittal, coronal and axial T2-weighted imaging-turbo spin echo, balance fast field echo. The MRI images were observed and the areas which showed low signal in all the three directions on T2WI were measured. Then the correlation between the areas of low signal on T2WI in placental and the types of placenta implantation abnormality was analyzed. Results The incidence of placenta implantation abnormality was 64.8% in our research (35/54). The main MRI signs were low signal on T2WI (68.5%, 37/54) and heterogeneous signal in placenta (57.4%, 31/54); the main sign of placenta percreta was tenting bladder (75.0%, 6/8). The types of placenta implantation abnormality were positively correlated with the areas of low signal on T2WI (r=0.454, P<0.05). Conclusion Pregnant women at risk of placenta accreta should be evaluated with imaging examinations, particularly with MRI scanning, to improve disease detection rate. The typical indirect signs of placenta implantation abnormality are low signal on T2WI and heterogeneous signal in placenta. The larger size of low signal area on T2WI in placenta, the deeper implantation of placenta.
7.Preparation of One-off Self-heating Material for Physical Therapy and Health Care

Objective To prepare a kind of one-off self-heating material for physical therapy and health care. Methods Polymer in salt solid electrolyte(SPE)was prepared with urea, thiourea and polyethelene glycol(PEG). The SPE was chosen to prepare a self-heating material through mixing with reduced iron powder and activated carbon powder. Results The self-heating material was in solid state and performed satisfactory. It released heat when contacted with air and was in dormancy when isolated from air. Conclusion SPE can be used for solid self-heating material through mixing with metal powder and activated carbon powder. The self-heating material can be used for mount of ointment and for other heat-wanted one-off medical use.
8.Preparation and identification of the polyclonal antibody against the 3D polymerase of foot and mouth disease virus

To prepare the polyclonal antibody against the 3D polymerase of foot and mouth disease virus (FMDV), the 3D polymerase gene of this virus was amplified by PCR and doubly digested with BamH I and Nde I. Then, it were cloned into expression vector pET-30a(+) to obtain the recombinant plasmid pET-3D and this plasmid was transformed to E.coli BL21(DE3) with induction by IPTG.The target protein was identified and purified with SDS+PAGE, and the inclusion bodies were extracted. The purified target protein was used as antigen to immunize New Zealand rabbits to prepare the polyclonal antibody against 3D polymerase of FMDV, which was then characterized by indirect ELISA and Western blotting. As demonstrated by SDS-PAGE, the target protein with a molecular weight at 46 ku was expressed. The polyclonal antibody showed high affinity and obvious specificity and its titer was above 1:8 000. This polyclonal antibody may lay a foundation for the further studies on the biological functions and epitopes of the 3D polymerase of FMDV.
9.The advantage by MRI of brain lesions after fetoscopic selective laser coagulation for twin-twin transfusion syndrome

Objective To demonstrate that MRI is more advantageous than ultrasound in the detection of ischemic cerebral lesions precisely in twin-twin transfusion syndrome(TTTS)fetus following fetoscopic selective laser coagulation(SLCPV).Methods The fetus with TTTS treated by SLCPV were collected.All fetuses underwent brain MRI within 1-5 days after the procedure and a follow-up MRI at 29-32 weeks'gestation.All fetuses also had frequent ultrasound evaluation until delivery.Results Twenty-five pregnancies with TTTS were included in the study.Six pregnancies resulted in survival of only one twin after the procedure(Group A),while the other 19 pregnancies resulted in the survival of both twins(Group B),as assessed at the time of initial MRI.Two fetus in Group A and three in Group B had evidence of bleeding in lateral ventricle.Two fetus in Group B had actue brain ischemia.One fetus in Group B had appeared brain atrophy and choosed to terminate pregnancy.Conclusion MRI can make a clearer diagnosis of cerebral damage of TTTS fetus in a short period after SLCPV.
10.The methods of reconstruction of pancreatic digestive continuity after pancreaticoduodenectomy:a meta-analysis of randomized controlled trials

Objective There have been many techniques proposed for the reconstruction of pancreatic digestive continuity to prevent fistula (PF) formation, but this is still highly debated. We carried out a systematic review and meta- analysis to determine the effectiveness of methods of anastomosis after pancreaticoduodenectomy (PD). Methods A full literature search was conducted in the Cochrane Controlled Trials Register Databases, Medline, et al.Randomized controlled trials(RCTs) were considered for inclusion. Analysis was carried out using Revman software. Results In all, 10 RCTs including a total of 1 408 patients were included, the pancreaticogastrostomy (PG) and pancreaticojejunostomy (PJ) groups, duct to mucosa PJ and PJ, binding PJ and PJ, pancreatic duct without anastomosis PJ and PJ. The meta-analysis showed that the PF, postoperative complications, biliary fistula, mortality, re-operation and hospital stay were not statistically different among four methods(P>0.05).Conclusions No ideal technique of pancreatic reconstruction after PD is found to be applicable to all kinds of pancreatic remnants in this systematic review and meta-analysis.
