1.Canonical correlation of professional stress,social support and somatic symptoms of sub-health status in Ningbo normal senior school teachers

Objective To explore the relationship of professional stress,social support and somatic symptoms of sub-health status of Ningbo normal senior school teachers. Methods 128 normal senior school teachers were selected and investigated by professional stress scale,social support scale and somatic symptoms of sub-health status scale, and canonical correlation analysis was used. Results The canonical correlation coefficient were 0.563, 0.471( P <0.01) with the existence of social support variable. The somatic symptoms of sub-health status would be greater with the rise of their professional stress and the decline of their social support. The canonical correlation coefficient was 0.497( P <0.01) with the elimination of social support variable. The factor loading of "examination pressure","professional burden" and "career expectation" in canonical variable ξ add to 0.801,0.855,0.726 respectively. The factor loading of "feeling of fatigue","sleep disorders" in canonical variable η add to 0.774,0.859 respectively. Conclusion The social support have adjustment function between the professional stress and somatic symptoms of sub-health status.

To study the effect of high altitude on the expression of transforming growth factor beta 1 (TGF ?1) during the healing process of radius fracture, a rat radius fracture model was established at a high altitude. 3.Preparation and analgesic effect of α-cobrotoxin-loaded poly(lactide-co-glycolide) microspheres

Aim To study the preparation of α-cobro-toxin-loaded poly ( lactide-co-glycolide ) microspheres and their analgesic effect. Method The microspheres were prepared by modified water-in-oil-in-oil ( w/o/o ) emulsion solvent evaporation method. A tail flick assay was used to evaluate the analgesic activity of the micro-spheres after nasal administration. Result The micro-spheres with high entrapment efficiency ( >80%) and average diameter of about 25 μm could be prepared by modified water-in-oil-in-oil ( w/o/o ) emulsion solvent evaporation method. A tail flick assy showed the evi-dence for their analgesic effect. Conclusion When Chinese cobraneurotoxin is loaded into microsphere and then the microsphere is administered through nasal cav-ity, its analgesic effect is enhanced significantly.
4.Transwell contact co-culture promotes growth and differentiation of sin-gle-dissociated iPSCs

[ABSTRACT]AIM:ToinvestigatethepromotingroleofTranswellcontactco-culturesysteminthegrowthand differentiation of single-dissociated induced pluripotent stem cells (iPSCs).METHODS:Bovine corneal endothelial cells (CECs) at passage 1~2 (P1~2) were seeded on the underside of Transwell inserts placed into culture plates and were cultured in 37 ℃and 5%CO2 for 8 h.Accutase digestion and 40μm filter process disaggregated colony-aggregated iPSCs into single-dissociated iPSCs , and the cells were seeded on the inside of Transwell inserts with CECs in medium of mTeSR 1 for 3 d and then in low-glucose DMEM supplemented with 10% FBS for 2 weeks.The characteristics and differentiation markers were evaluated by real-time fluorescence quantitative polymerase chain reaction ( qPCR ) , immunofluorescence staining, live&dead cell staining and alkaline phosphatase (ALP) staining.The group of iPSCs cultured in conventional medium was used as control group 1.The group of single-dissociated iPSCs co-cultured with CECs was set as experimental group, while single-dissociated iPSCs without co-culture were as control group 2.RESULTS: The bovine CECs showed typical hexagonal cobblestone shape .iPSCs showed colony-like growth , while became single-dissociated cells after Tran-swell contact co-culture with bovine CECs for 3 d.The single-dissociated iPSCs positively expressed the undifferentiated markers, Nanog and Oct4.The mRNA expression levels of Nanog , Oct4 and Sox2 between experimental group and control group 1 were both positive and had no statistical significance difference (P>0.05).The dead cells in experimental group decreased significantly, and there was statistically significant difference compared to control group 2 (P<0.01).After 14 d of induced differentiation co-culture , the single-dissociated iPSCs showed rather uniform polygonal morphology , increased dimension and no obvious colony existence .Negative ALP staining, positive immunofluorescence staining for ZO-1, AQP1 and CD31, and negative for CD34 and CD133 were also observed.The results of qPCR showed that the mRNA expression of Oct4, Nanog and Sox2 significantly decreased , and had statistically significant difference compared with control group 1 (P<0.01).CONCLUSION: When co-cultured with bovine CECs, iPSCs morphologically changed to endothelial-like cells and expressed some markers of CECs .Transwell contact co-culture system not only enhances the growth of single-dis-sociated iPSCs , but also promotes their differentiation .
5.Influence of maxadilan on human adipose-derived stem cells

[ ABSTRACT] AIM:To investigate the effect of maxadilan, which specifically activates pituitary adenylate cycla-se-activating polypeptide type I receptor (PAC1 receptor), on the proliferation, apoptosis and differentiation potential of human adipose-derived stem cells ( ASCs) .METHODS:ASCs from human adipose tissue were isolated by enzymatic di-gestion and cultured.ASCs were confirmed by the analysis of the markers for cell phenotypes by flow cytometry ( FCM) and adipogenic/osteogenic induction.The effect of maxadilan on ASCs viability was analyzed by CCK-8 assay and FCM.ASCs were irradiated by ultraviolet C ( UVC) at 254 nm and the absorbance of apoptotic ASCs induced by various doses of UVC was measured by CCK-8 assay.ASCs were exposed to 702 J/m2 UVC for 24 h to induce apoptosis.The effect of maxadilan on ASC apoptosis was analyzed by FCM and the determination of caspase 3 and caspase 9 levels.RESULTS:Adipose-de-rived stem cells were confirmed by the detection of the positive expression of cell phenotypes including CD29, CD44, CD59 and CD105 by FCM.The data of CCK-8 assay revealed that ASCs treated with maxadilan (80 nmol/L) had the strongest ability of proliferation.The data of FCM also demonstrated that the addition of 80 nmol/L maxadilan to ASCs in experimen-tal group markedly improved the proliferation capacity of the cells compared with control group (P<0.05).The apoptosis of ASCs exposed to 702 J/m2UVC was dramatically inhibited by the treatment with maxadilan (80 nmol/L).Such process involved the caspase signaling pathway including caspase 3 and caspase 9.There was statistical significance (P<0.05) between experiment group ( ASCs irradiated by UVC and supplemented with maxadilan) and control group ( ASCs only irra-diated by UVC) .Meanwhile, adipogenic and osteogenic differentiation potentials were both positive in experiment group and control group.CONCLUSION:Maxadilan promotes proliferation and inhibits apoptosis of the ASCs.The differentia-tion potential of ASCs toward adipogenic and osteogenic lineages wouldn’ t be altered by maxadilan.Maxadilan would bene-fit to growth and expansion of ASCs in vitro.
6.Role of ROCK inhibitor in corneal reconstruction:promoting the proliferation, migration and adhesion of keratocytes

BACKGROUND:With the deepen understanding on the biological function of Rho/ROCK pathway, new ROCK inhibitors continue to be discovered, and ROCK inhibitors show good promoting effects on the survival, proliferation and migration of keratocytes. Research on ROCK inhibitors wil provide more donor materials or seed cels for regenerative medicine and clinical cel transplantation. OBJECTIVE:To summarize and explore the progress in the treatment and application of corneal disease using the ROCK inhibitors Y-27632 and Y-39983. METHODS:The PubMed database and CNKI database were retrieved by computer to search the relevant literature published between 2008 to 2015 using the key words of “corneal endothelial cel, corneal epithelial cel, ROCK inhibitor, Y-39983, Y-27632” in English and Chinese, respectively. Relevant articles in line with the theme were screened and analyzed. RESULTS AND CONCLUSION:Totaly 264 papers were initialy searched. At last, 45 papers were selected. Currently there are two main ROCK inhibitors: Y-27632 and Y-39983, but both of which are stil in basic research stage and clinical testing stage. Y-27632 promotes the proliferation and activity of corneal epithelial stem cel after resuscitation; Y-39983 as a novel ROCK inhibitor can be better to inhibit Rho kinases activity than Y-27632, thereby more effectively promoting the healing of the corneal endothelium. There are many studies on the application of ROCK inhibitors in corneal treatment, but not a stable method established to obtain seed cels. Each method has its own advantages and disadvantages, and how to overcome these disadvantages and to find fast and stable access to seed cels is the future direction of development.
7.Effect of morphine on pharmacokinetics and pharmacodynamics of P2Y12 receptor antagonists

P2Y12 receptor antagonists and morphine are often recommended in patients with acute myocardial infarction.P2Y12 receptor antagonists can rapidly and potently reduce the platelet activity and prevent future thrombotic events.Meanwhile,combined morphine is used to relieve symptoms of angina.A number of studies have confirmed that morphine can decrease plasma concentrations of clopidogrel and impair its anti-platelet activity,which may lead to poor response in clopidogrel-treated patients with acute coronary syndrome.The randomized trials in healthy volunteers and patients with acute myocardial infarction also confirmed the similar drug-drug interaction between morphine and ticagrelor or prasugrel.Although the P2Y12 receptor antagonists combined with morphine are still used for myocardial infarction patients,there are few report on the objective evaluation of this drug interaction.This review,based on the findings of experimental as well as observational and randomized clinical studies,summarizes completely the drug interactions between oral P2Y12 receptor antagonists and morphine.
8.Finite element analysis of biomechanical features of percutaneous C1-2 anterior and posterior transarticular screws

Objective To develop a three-dimensional finite element model of atlantoaxial instability and compare the biomechanical properties of percutaneous anterior transarticular screw (ATS) and posterior transarticular screw (PTS) fixations.Methods A pathologic three-dimensional finite element model of atlantoaxial instability was developed from CT images of the upper cervical spine of volunteers with the aid of softwares,such as Mimics,Freeform,and Ansys.Percutaneous C1-2 ATS and PTS fixation modes were simulated and implanted to the model.Under the preload of 40 N and force moment of 1.5 Nm in anterior flexion,posterior extension,lateral bending,and axial rotation,biomechanical properties of the two fixation modes were compared.Results Under the four loading modalities,both fixation techniques provided maximal inhibition on C1-2 movement.The maximal stress for ATS was larger than that for PTS and maximal displacement for ATS was smaller than that for PTS.Conclusions Both ATS and PTS provide similar outlook and are effective to stabilize the atlantoaxial joint.Biomechanical performance of percutaneous C1-2 ATS is better than that of percutaneous C1-2 PTS.
9.Contralateral radiculopathy after unilateral transforaminal lumbar interbody fusion

Objective To analyze the incidence and risk factors of contralateral radiculopathy in patients after unilateral transforaminal lumbar interbody fusion (TLIF) surgery.Methods A retrospective study was conducted within 587 patients (average age 57.1 years,range 19-71 years) who underwent unilateral TILF from January 2010 to January 2014 in our hospital,including 334 males and 253 females.Patients were divided into a symptomatic group and an asymptomatic group.The causes of contralateral neurological symptom were evaluated according to the radiological data.The difference of pre-and post-operative contralateral foramen area (CFA),segmental angle (SA) and the clinical treatment outcomes (VAS,JOA score) were compared between two groups.Results Patients were followed up for 9-21 months,average 15.1 months.Post-operative contralateral radiculopathy occurred in 28 (4.8%) of the patients who underwent unilateral TLIF,including contralateral foraminal stenosis in 16 (57.1%,16/28),screw malposition in 5 (17.9%,5/28),contralateral lateral recess stenosis and/or newly developed disc herniation in 3 (10.7%,3/28),hematoma in 1 (3.6%,1/28),cement compression in 1 (3.6%,1/28),and unknown origin in 2 patients (7.1%,2/28).Nineteen (3.2%,19/587) of the 28 patients received revision surgery because of ineffective conservative treatment.Compared with the asymptomatic group,the difference of pre-and post-operative CFA was significantly smaller (-13.8±13.2 mm2) in symptomatic group,while the SA was significantly greater (7.0°±9.8°) in symptomatic group.The JOA score at 3 months after the surgery was significantly improved in asymptomatic group (63.0%±18.1%,P<0.05).Conclusion The incidence rate of contralateral neurological symptom was 4.8% in the present study.The potential risk factors associated with contralateral radiculopathy were predominantly contralateral foraminal stenosis and screw malposition.The excessive restoration of SA might have an effect on contralateral nerve compression,which should arouse the attention of the surgeon.
10.The relation between the positive rate of platelet antibody and the frequency of platelet transfusion, the gender and the blood group of patients

Objective To study the relation between the positive rate of platelet antibody and the frequency of platelet transfusion,the total quantity of platelet transfusion,the gender of patients,the blood group of patients through statistic analysis of the data gathered from patients.Methods A total of 5 745 results of platelet antibodies detection assay throughout the year of 2016 were gathered,and statistical relation between the positive rate of platelet antibody and the gender of patients,the blood group of patients and the frequency of platelet transfusion were analyzed.Results The results of platelet antibodies detection assay show that 108 patients were positive among 5 745 patients,and the positive rate of platelet antibody was 1.87%.There were no relations between the positive rate of platelet antibody and the gender of patients or the blood group of patients,while the positive rate of platelet antibody and the frequency of platelet transfusion were positively correlated.Conclusion Platelet antibody production could impact the therapeutic effect of platelet transfusion.Patients with platelet antibodies should receive matching platelet to ensure the therapeutic effect.
