1.Security time limit of intrahepatic bile duct ischemia under common temperature: What is limiting value?

BACKGROUND: Biliary ischemia-reperfusion injury is one of the main reasons for the injury bile duct following liver transplantation, liver resection and hepatic artery thrombosis after chemotherapy. However, nothing has been decided yet concerning liver can tolerate long intrahepatic bile ducts ischemia under normal temperature. OBJECTIVE: To use the rabbit biliary ischemia-reperfusion injury, model, analyze the rabbit intrahepatic bile ducts ischemia security time. METHODS: Rabbits were randomly divided into sham operation, hepatic artery and common bile duct joint blocking 1.5, 2, 2.5, 3 h groups. Sham operation group only free common bile duct, hepatic artery and portal vein. Hepatic artery and common bile duct joint blocking 1.5, 2, 2.5, 3 h groups, left and right hepatic duct openings superior margin was clamped using artery clamps, and hepatic artery, common bile duct and loose connective tissue were occluded. Following 1.5, 2, 2.5 and 3 hours, artery clamps were removed to recover hepatic artery or biliary tract blood flow. After a week, animal survival was calculated and liver function was tested. RESULTS AND CONCLUSION: None animals in the sham operation and hepatic artery and common bile duct joint blocking 1.5 hour groups died;survival rate was 87.5% following 2 hours of blocking. Animal survival rate was decreased gradually with prolonged blocking time of blood flow. These indicated that the maximum safety time of blocking biliary duct blood flow was 2 hours. Pathological and histological changes were mild within 2 hours of blocking, mainly presenting cell edema and inflammatory cell infiltration, and necrotic focus was punctiform or fragmentis, reversible. While ischemia above 2 hours, bile duct epithelial necrotic shape was significant. The obvious, hepatic necrosis was multifocal, flake, irreversible damage. Histological change also confirmed that 2 hours may be maximum security limitation of rabbit tolerance intrahepatic bile duct flow blocking under normal temperature.
2.Air Disinfection with Chlorine Dioxide in Saps

Objective To observe the effect of air disinfection with chlorine dioxide(ClO2)in the saps and determine the concentration change in order to ensure the safety for people to enter the saps.Methods The disinfection was done at the concentration of 1 000 mg/L and the spray dose of 10 ml/m3 and then to count the total bacteria and fungi with the cascade impactors in the saps before and after air disinfection and to monitor the concentration of ClO2 with the air ClO2 monitor(iTX).Results The 94.6% of the total bacteria and 92.1% of the fungi died in 30 minutes after disinfection.The concentration of ClO2 in the air decreased from 10 mg/m3 to 0.8 mg/m3 81 minutes later and to 0.3 mg/m3 96 minutes later,after 120 minutes,it was 0 mg/m3.Conclusion The air disinfection effect of ClO2 used at the dose of 10 ml/m3 and the concentration of 1 000 mg/L is satisfactory.96 minutes after disinfection,the concentration of ClO2 is safe for human health.

Objective To understand the air pollution by microorganisms in the underground spaces. Methods 51 underground spaces were investigated, the impact and sedimentation sampling methods were employed and the total bacterial count, mycetes, streptococcus and anaerobes in air were determined. ELESA was used to detect aflatoxin B1 in aspergillus flavus isolated from air. Results The total bacterial count in the air in underground spaces accorded with the military health standard. 6.Investigation on ideal cardiovascular health status in 37664 health checkup subjects

Objective To investigate the distribution of ideal cardiovascular health status in Chinese health checkup population. Methods Subjects were enrolled from a health checkup population coming to the PLA health management center from Sept 2009 to Mar 2016. Modified with China's specifications, lifestyle and checkup data were collected and analyzed according to the American ideal cardiovascular health standard. Results A total of 37664 people were included in the study, of whom 72.88%were male and 27.12%were female. Comprehensive analysis showed that, among 7 health indicators including smoking, physical activity, diet, fasting blood glucose, blood pressure, total cholesterol and body mass index, there were only 43 subjects (0.11%) whose lifestyle reached the ideal cardiovascular health status, 11 subjects were in the poor cardiovascular health status, accounting for 0.03%. The rest of the subjects were in the intermediate levels of cardiovascular health status. There was a large gap between the daily diet intake and the dietary recommendation, and there was also a large gap between the actual level physical activity and the ideal level of physical activity recommended by related guidelines, indicating that unhealthy diet and inadequate physical activity are two bottleneck factors. Dairy product intake has the lowest satisfaction ratio, followed by vegetable and fruit intake. Most subjects (94.10%) showed insufficient physical activity. The percentage of three status of cardiovascular health among young, middle-aged and elderly subjects differed significantly (χ2=1200, P=0.000), and presented an increasing trend of ideal cardiovascular health and a declining trend of poor cardiovascular health status with age from youth to middle age, to the elderly, which reflected insufficient physical activity especially among the young people, and then the middle-aged. Meanwhile, the proportion of ideal cardiovascular health in men was higher than that in women. Conclusion The rate of ideal cardiovascular health is relatively low in the study population, unhealthy diet and inadequate physical activity are two bottleneck factors. Encouraging people to develop good eating and exercise habits might be the most effective method to improve population's ideal cardiovascular health status.
7.Effects of Vibrio vulnificus cytolysin on the numbers of mitochondria and the expression of CD62L in murine liver CD4+T cells

Objective To understand how Vibrio vulnificus hemolysin (VvhA) affects the viability of murine liver CD4+ T cells as well as its effects on the numbers of mitochondria and the expression of CD62L. Methods The primary murine liver monocytes (MNs) were isolated from C57BL/ 6 mice and then treated with recombinant VvhA (rVvhA) for 6 hours in vitro. The viability of murine liver CD4+T cells and the expression of CD62L were measured by staining with anti-mouse CD4, CD8, CD44, CD62L and cell via-bility fluorescent dye or fluorescent antibody. Moreover, the cells were simply incubated with MitoTracker or JC-1 probes to label mitochondria and mitochondrial membrane potential, which were further analyzed by using flow cytometry analysis. Results With the increase in the doses of rVvhA, the viability of murine liv-er CD4+T cells was decreased from 81. 5% to 15. 8% . The expression of CD62L on the surface of murine liver CD4+T cells was dramatically decreased. Both the murine liver na?ve and effector CD4+ T cells were sensitive to the cytotoxicity of rVvhA. Moreover, treating murine liver CD4+ T cells with rVvhA resulted in significantly decreased numbers of mitochondria and lower mitochondrial membrane potential. Conclusion The cytotoxicity of rVvhA to murine liver CD4+T cells might be achieved through inhibiting the expression of CD62L, decreasing the numbers of mitochondria and lowering mitochondrial membrane potential.
8.Identification of hemolysin BL from Bacillus cereus of patients with endophthalmitis and its biological activity detection in vitro

Objective To identify the hemolysin BL(HBL) gene from Bacillus cereus of patients with endophthalmitis comfirmed by API bacterial identification test strip,and detect its biological activity in vitro.Methods Three pairs of specific primers were designed according to the gene sequence of HBL(B,L1 and L2 components),then the PCR assay were established through condition optimization,and to further detect five Bacillus cereus strains isolated from clinical patients with endophthalmitis.HBL with biological activity was extracted by salt fractionation from a randomly selected strain,and a series of different concentrations of HBL were prepared and acted on sheep red blood cells(SRBC),Vero cells and Hela cells; virulence of HBL was also assessed through observating lethal effect of which on mice with intraperitoneal injection.Results Three genes of HBL were detected in all B.cereus strains from clinical patients; Strong hemolytic activity of HBL showed obvious time-and dose-dependent.In the study,we found the morphological changes of Vero and Hela cells caused by HBL were different,but cell death were the same result with contents released; Within 48 h after infection,lethality of HBL for mice showed 100% with the concentration of more than 2.0 HU/ml,and was also in a time-and dose-dependent manner.Conclusion HBL isolated from B.Cereus had high hemolysis activity and low concentration.The expression of all BL genes provided a strong basis for the clinical feature of B.Cereus infection,which was developed rapidly and with a poor prognosis.It also provided a new method for rapid diagnosis and molecular epidemiology investigation in clinical.
9.Diagnostic value of CT in adults with midgut malrotation

ObjectiveTo evaluate the diagnostic value of CT in adults with midgut malrotation.MethodsThe CT and clinical findings of 13 cases confirmed by surgery were analyzed retrospectively.ResultsCT found “whirl sign” in 9 cases,“transposition sign” in 5 cases,and the superior mesenteric vein located in right lower abdomen in 4 cases.At the same time,duodenum and upper jejunum located in right upper abdomen.Conclusion“Whirl sign” and “transposition sign” were the typical CT findings of midgut malrotation in adults.Ectopic superior mesenteric vein highly indicated slight midgut malrotation in adults.
10.Mesenchymal stem cells transplantation under the diagnostic ultrasound mediated microbubbles destruction in improving myocardial perfusion after rabbit myocardial ischemia

Objective To explore the value of diagnostic ultrasound mediated microbubble destruction in improving the myocardial perfusion and left ventricular systolic function when cooperated with the mecsenchymal stem cells(MSCs) transplantation in rabbit myocardial ischemia. Methods One week after myocardial ischemia (MI) modeling,36 rabbits were divided into 3 groups,the control group(group Ⅰ) ,intravenous injection of MSCs group(group Ⅱ) and ultrasound + microbubble + MSCs group (group Ⅲ). Myocardial contrast enhancement (MCE) was performed and quantification analysis of anterior wall was assessed with Photoshop. Left ventrieular systolic function was assessed with M-mode echocardiography and bi-plane Simpson's method. CD34 expression in heart was detected with immunohistochemisty(IHC). Western blotting was applied to detect the level of VEGF in three groups. Results The differences of gray scale analyzed with histogram of Photoshop in anterior wall of ischemia myocardium between the group Ⅰ and group Ⅱ or group Ⅲ were significant,and P value was 0. 032 and 0. 000 , respectively. There were significant differences of FS between group Ⅲ (30. 43±4.09)% and group Ⅱ (26.29±2.93)%, P<0.01, and similar to group Ⅰ (19.28 ± 2.84)%. The difference of EF(%) between group Ⅲ and group Ⅱ was significant [(61.5±5.8 vs 53.6±4. 71), P<0. 05] ,or markedly significant between group Ⅲ and group Ⅰ [(61.5±5.8 vs 42.6± 5.0), P <0.01]. EF(%) assessed with bi-plane Simpson's method was significantly increased from (34.64 ± 4.59) in group Ⅰ to (41.78 ± 4.21) in group Ⅱ and (48.6±3.96) in group Ⅲ. The expression of CD34 assessed with immunohistochemistry was the highest in group Ⅲ. The level of VEGF with western blotting in group Ⅲ was significantly higher than other two groups. Conclusions It is an efficacious transplantation means of MSCs infusion under the ultrasound mediated microbubles destruction in improving the myocardial perfusion and cardiac systolic function.
