1.Effect of Grape Seed Proanthocyanidin on Growth Associated Protein-43 through Extracellular Signal Regulated Kinase 1/2 Pathway in Rats with Brain Radiation Injury

Objective To observe the effect of grape seed proanthocyanidin extract (GSPE) on growth associated protein-43 (GAP-43) through extracellular signal regulated kinase 1/2 (ERK1/2) pathway in rats with brain radiation injury. Methods 72 3-month male Sprague-Dawley rats were divided into control group (n=18), model group (n=18), high dose GSPE group (n=18) and low dose GSPE group (n=18). The brain radiation injury models were established by linear accelerator irradiation with 22 Gy. The learning ability was assessed with shuttle box. The morphological changes of neurons in hippocampus were observed with HE staining;the expression of GAP-43 was de-tected by RT-PCR; and the expression of phosphorylated ERK1/2 was detected by Western blotting. Results Compared with the model group, the active avoidance reaction rate in the shuttle box test increased and the passive avoidance latency shortened in GSPE groups (P<0.001); the nerve cell morphological injury reduced and the expression of GAP-43 mRNA and phosphorylated ERK1/2 increased in the GSPE groups (P<0.001), especially in the high dose GSPE group (P<0.001). The GAP-43 mRNA level positively correlated with phosphory-lated ERK1/2 level in the model group (r=0.764, P<0.001), the low dose GSPE group (r=0.814, P<0.001) and the high dose GSPE group (r=0.822, P<0.001). Conclusion GSPE could promote the expression of GAP-43 through ERK1/2 pathway in rats, and prevent the brain from radiation injury.
2.Postoperative radiotherapy in patient with early stage uterine sacomas

Purpose:To study the role of postoperative radiotherapy in early stage uterine sarcomas. Methods:Retrospective analysis of the impact of radiotherapy on progress-free survival rate (PRS )rate, local recurrence rate (LRR) and overall survival (OS)in stage Ⅰ,Ⅱ uterine sarcomas. 34 cases were treated with surgery alone ,42 cases were treated with postoperative radiotherapy. Results:The PRS at 2 and 5 years for surgery were 38.2% and 26.5% ,69.0% and 57.1%for postopeative radiotherapy. The LRR at 2 and 5 years treated with surgery were 52.9% and 64.7%,26.2% and 28.6% with adjuvant irradiation. The OS at 2 and 5 years treated with surgery were 83.4%and 47.0%,83.3% and 69.0% with adjuvant radiation. Conclusions:Postoperative radiotherapy showed a marked increase in the three parameters studied (PRS? LRR?OS ) for I,II stage uterine sarcomas.
3.Adjuvant therapy in patients with uterine sarcomas

Purpose:To study the adjuvant therapy in uterine sarcomas,to decrease local recurrence and metastases and improve survival.Methods:Analysis of the management of 113 cases of uterine sarccoma , retrospectively. 30 cases were treated weith surgery alone , 25 with adjuvance radiation , 34 with postoperative chemotherapy, 21 with surgery and chemo radiation.Results:The 2 year pelvic recurrence and metastases with surgery alone were 43.3 % and 30 0 %,32 0 % and 28 0 % with postoperative radiation,44.1 % and 20.6 % with postoperative chemotherapy, 23.8 % and 23.8 % with postoperative combination of chemo radiation.But there was no significant difference between surgery alone and postoperative adjuvant therapy.Conclusions:Postoperative radiotherapy can improve local control,adjuvant chemotherapy can decrease metastases,both can improve disease free survival,but can not improve overall survival for uterine sarcomas.
4.Protective effect of Grape seed proanthocyanidin on learning disabilities in a rat model with radiation brain injury

Objective To observe the protective effect of grape seed proanthocyanidin extract ( GSPE) on learning disabilities after radiation injury brain in rats.Methods 120 male Wistar rats were divided into control group,model group,high and low dose GSPE groups.The radiation injury brain models were established using a method of linear accelerator irradiation in 22 Gy.The morphological changes of neu-rons in hippocampus were observed with HE staining; the expression of phosphorylated extracellular signal regulated kinase 1/2 ( ERK1/2) and growth associated protein-43( GAP-43) were detected by immunohisto-chemistry;the contents of malonalde hyde(MDA) and superoxide dismutase(SOD) were measured respec-tively by thiobarbituric acid( TBA) and xanthine oxidation( XTO) methods;the learning ability was assessed with shuttle box.Results Compared with the model group,the GSPE groups showed a decreased degree of nerve cell morphological injury; and increased ERK1/2 activities((13.20±1.45)/view,(27.40±2.52)/view,(19.80±1.30)/view),GAP-43 expression level((52.93±2.07)/view,(66.50±0.77)/view,(73.05± 2.40)/view) and SOD activities((79.82±5.26) U/mg,(76.20±6.86) U/mg,(80.12±5.76) U/mg)(P<0.05);and decreased the contents of MDA((71.62±1.88) μmol/g,(76.41±1.94) μmol/g,(72.32±1.98)μmol/g)(P<0.05).Shuttle box testing showed that the active avoidance reaction rate was increased(7 d:(56.23±2.56)%;14 d:(44.66±2.79)%;28 d:(50.40±2.16)%)(P<0.05)and passive avoidance latency was shorted(7 d:(34.11±1.57)s;14 d:(45.52±1.88)s;28 d:( 39.52±1.79)s)(P<0.05) in GSPE groups. The above mentioned changes were more significant in high dose of GSPE(P<0.05) .Conclusion GSPE has protective effect on learning disabilities of radiation injuries brain,which is related to enhance ERK1/2 activ-ity and the expression of GAP-43.
5.Plasma fibrinogen in patients with facial paralysis

OBJECTIVE To study the level of plasma fibrinogen in patients with common facial paralysis, and investigate the relationship between the plasma fibrinogen and the pathogenesis of facial paralysis.METHODS A total of 157 inpatients with different kinds of facial paralysis were studied from December 2006 to December 2008.They were etiologically divided into five groups:severe Bell's palsy group(60 cases), moderate Bell's palsy group(19 cases), Ramsay-Hunt syndrome group(28 cases), temporal bone fracture with facial palsy(28 cases), and facial nerve tumor group(22 cases).They were tested for plasma fibrinogen level right after admitted to hospital.RESULTS Plasma fibrinogen level in severe Bell's palsy group was(4.08?0.90)g/L, which was significantly higher than that in other groups and the normal value(2.0~4.0)g/L(P
6.Determination of 31 Triazine Herbicides in Farmland Soils by High Performance Liquid Chromatography-Tandem Mass Spectrometry

A high performance liquid chromatography-tandem mass spectrometric method ( HPLC-MS/MS ) was developed for the simultaneous determination of 31 kinds of triazine herbicides in farmland soils. Samples were extracted by accelerated solvent extraction ( ASE ) and purified by Oasis MCX solid phase extraction ( SPE) cartridges, and then analyzed by HPLC-MS/MS in multiple reaction monitoring ( MRM) mode with positive electrospray ionization. The analytical column was Phenomenex Luna C18 ( 150 mm í 2. 0 mm í3 μm) and the mobile phases were acetonitrile and water containing 0. 1% (V/V) formic acid. The limits of detection (S/N≥3) were 0. 008-0. 440 μg/L. All of the triazine herbicides had good linear responses with r≥0 . 996 and the average recoveries in the spiked levels of 0 . 40-40 . 0 μg/kg ranged from 76 . 9%-102 . 0%with the RSDs of 3 . 4%-10 . 3%. The HPLC-MS/MS method had been applied for the detection of the triazine herbicide residues in farmland soils from Shenyang region and the results showed that atrazine, simazine, prometryne and atrazine-desethyl were the main triazine herbicides in the region.
7.Expression and clinical significance of SALL4 expression in gastric carcinoma tissues

Objective To study the expression and clinical significance of the SALL4 in human gastric carcinoma tissues. Methods The expression of SALL4 in 91 samples of gastric carcinoma and 37 samples of normal gastric tissues was detected by RT-PCR,Western blot and immunohistochemistry,and its relationship with the clinical data were analyzed statistically.Results The positive expression rate of SALL4 in gastric carcinoma(74.7%)was significantly higher than that(18.9%)in normal gastric mucosa tissues(P <0.05).Moreover,with the decreased with the differentiation of gastric carcinoma,the positive expression rate of SALL4 was increased.The expression of SALL4 mRNA and protein in gastric carcinoma tissues were significantly higher than that in normal gastric tissues(P <0.050).The expression levels of SALL4 were relevant to lymph node metastasis(P =0.001),infiltra-tion depth(P =0.029)and the differentiation degree of gastric carcinoma(P =0.050).Conclusion SALL4 was highly expressed in gastric cancer tissues and relevant to lymph node metastasis,infiltration depth and the differentiation degree,which may have play an important role in the development of gastric cancer.
8.PM2.5 induces VEGF expression via activation of NF-κB pathway in bronchial epithelial cells

Objective To investigate the molecular mechanism of vascular endothelial growth factor ( VEGF) expression in bronchial epithelial cells (Beas-2B)induced by particulate matter 2.5(PM2.5).Methods PM2.5 powder was dis-solved in DMEM medium and diluted into five concentrations , 0,12.5,25,50 and 100μg/ml, respectively.The double an-tibiotics ( streptomycin and penicillin ) and FBS were added into the solution to a 2% final concentration of serum system after being treated by ultrasound for 30 minutes.The cultured Beas-2B cells were then treated with different doses of PM2.5.Subsequently, nuclear factor-kappa B(NF-κB) transactivity and the transcriptional activation of vegf gene promot-er were tested by dual-luciferase reporter gene analysis system while phosphorylation of p 65 , expression levels of IκBαand VEGF were detected by Western blotting .Results PM2.5 induced up-regulation of VEGF expression in Beas-2B cells in a dose-dependent manner , accompanied by NF-κB transactivation at the highest level under 100 μg/ml of PM2.5 treatment. Moreover, PM2.5 induced degradation of the repressor protein IκBαand increase in the phosphorylation level of p 65 sub-unit in Beas-2B cells.Knockdown of NF-κB p65 expression significantly inhibited vegf gene promoter transcriptional activa-tion as well as VEGF protein expression in Beas-2B cells induced by PM2.5.Conclusion PM2.5 induces VEGF expres-sion via activation of NF-κB pathway in bronchial epithelial cells .
9.Expression and significances of Merlin and mTOR in spinal schwannoma

Objective To clarify the expression and clinicopathological significances of mTOR and Merlin proteins in spinal schwannoma.Methods Immunohistochemical SP method was used to detect the expression levels of mTOR and Merlin proteins in tumor tissues from 21 spinal schwannoma patients.The meaning of the two proteins expression changes on schwannoma was analyzed.Results In 21 cases of schwannoma patients,the mTOR was positive expression in 16 cases,negative expression in 5 cases,while in the normal neural tissue,mTOR was all negative expression.In 21 cases of schwannoma patients,the Merlin protein was negative expression in 18 cases,positive expression in 3 cases,but it was positive in all of normal neural tissue.Merlin protein expression was negatively correlated with mTOR protein expression (r =-0.785,P < 0.001).Conclusion The expression level of mTOR proteins in schwannoma is significantly higher than that in normal nerve tissue,while the expression level of Merlin protein in schwannoma tissue is significantly lower than that in normal nerve tissue.There is an internal relationship between mTOR and Merlin.
10.Effects of butylphthalide on brain edema, blood-brain barrier permeability and RhoA expression in cortex in focal cerebral infarction in rats

Objective To investigate the effects of butylphthalide on brain edema,blood-brain barrier permeability and RhoA expression in cortex in focal cerebral infarction in rats.Methods A total of 220 male Sprague Dawley rats were divided into a control,a model and a butylphthalile group (40 mg/kg,once a day,gavage) according to the random number table method.A model of rat focal cerebral infarction was induced by photochemical method.At 3,12,24,72,and 144 h after modeling,wet-dry weight method and Evans blue extravasation method were used to detect the brain water content and blood-brain barrier permeability.At 24 h after modeling,immunohistochemical staining and Western blot were used to detect the expression of RhoA protein in the periinfarction cortex.Results Compared to the control group,the brain water content (except at 6 h) and blood-brain barrier permeability in the model group and the butylphthalide group were increased significantly (all P < 0.05).The immunohistochemical staining and Western blot suggested that the RhoA expression in the periinfarction cortex was upregulated significantly (all P < 0.05).Compared to the model group,the brain water content and blood-brain barrier permeability at different time points in the butylphthalide group were decreased significantly (all P < 0.05).The expression levels of RhoA were also decreased significantly (P < 0.05).Conclusions Butylphthalide may reduce the brain edema of focal cerebral infarction in rats,inhibit disruption of the blood-brain barrier,and down-regulate the expression of RhoA.
