1.Study of Fanggan Decoction,s death prevention on mouse and inhibition effects on Influenza A virus

Objective:To study the Fanggan Decoction,s death prevention on mouse and inhibition effects on Influenza A virus in vivo.Methods:After setting up the model of mouse infected with Influenza A virus(H1N1),we observed the death prevention with Fanggan Decoction,done hemagglutination test and detected the dynamic contents of virus with Real-Time Fluorescence Quantitative RT-PCR.Results:Fanggan Decoction can prevent the death of infected mouse and delay the survival time.The death rate was 66.67%,33.33% and 25% respectively in low,middle and high dose of Fanggan Decoction groups and the average survival time was respectively 8.75 days,11.41 days and 12.33 days.Virus contents reached peak on the 5th day,while compared with the model group,virus contents were lower in each Fanggan Decoction groups,especial in the middle and high dose groups.Conclusion:Fanggan Decoction had good effect in inhibiting Influenza A virus,and can prevent the death of infected mouse,delay the survival time,while get better antivirus dose-effect relationship at double dose.

Objective To study the novel methods of VMAT planning based dose volume histogram ( DVH) optimization, evaluated the dosimetry and planning efficiency in VMAT planning for Esophageal Carcinoma. Methods Twelve Esophageal carcinoma patients were enrolled in this study. The conventional VMAT planning as the reference, using multi?criterion optimization DVH ( MCO?DVH ) and overlapping volume histogram prediction DVH ( OVH?DVH ) two different methods to get ideal objectives function for informing VMAT plans, Then evaluate the dosimetric, planning efficiency for all new VMAT plans. The difference between the paired t?test groups. Results The two VMAT plans based DVH objective function can meet the clinical needs. Compared with the conventional VMAT plan, Conformity index ( CI ) and Homogeneity index ( 0. 77 vs. 0. 72, P=0. 017 and 0. 10 vs. 0. 12, P=0. 047 ) is better in DVH informed plans;lung V5 and spinal cord V50 are better in MCO?DVH informed plan (54. 66 vs.60. 23,P=0. 013 and 0. 98 vs.0. 49,P=0. 037).Furthermore,the DVH informed plans had higher planning efficiency (8. 2 vs. 19. 5,P=0. 023) . Conclusions DVH Objective informed VMAT Planning can achieve clinical needs with much uniform dose to target,lower OAR dose and higher planning efficiency.
4.DEVELOPMENT OF A DIPSTICK DYE IMMUOASSAY WITH SOLUBLE CERCARIA ANTIGEN FOR EARLY DIAGNOSIS OF SCHISTOSOMIASIS

Objective To develop a fast, simple immunodiagnosis assay for early diagnosis of schistosomiasis. Methods The soluble cercariae antigen(SCA) labeled colloidal dye was used as the detecting antigen for schistosomiasis. A dipstick dye immunoassay(SCA-DDIA) for early diagnosis of schistosomiasis was established. The antibodies in sera of infected rabbits in early stage of infection by SCA-DDIA were detected and compared with SEA-DDIA. The sera from people with acute and chronic schistosomiasis and other parasitic diseases and from healthy people were tested by SCA-DDIA and SEA-DDIA. Results In infected rabbits during early stage of infection, the average time of antibody detected by SCA-DDIA was 22 d , at day 30 post-infection all experimental rabbits were positive with SCA-DDIA, the detected time was earlier than that with SEA-DDIA. The sensitivity of SCA-DDIA for acute, chronic schistosomiasis japonica were 100.0% and 93.3% respectively. The specificity for healthy persons was 99.0%. The cross reaction rates with paragonimiasis westermani, clonorchiasis sinensis and fasciolopsiasis buski were 26.3%, 0 and 0 respectively. The results were similar to that by SEA-DDIA. Conclusion The SCA-DDIA is more useful for early diagnosis of schistosomiasis.
5.Analysis of early diagnostic fraction antigens of cercariae, adult worms and eggs of Schistosoma japonicum

Objective To find out the valuable early diagnostic antigen of Schistosoma japonicum. Methods The sera of rabbits were collected at different time after the rabbits were infected with cercariae of Schitosoma japonicum. The fractions of the soluble cercaria antigen (SCA), soluble adult worm antigen (AWA) and soluble egg antigen (SEA) were separated by SDS-PAGE and recognized by Western blotting with rabbits' sera of different time of post-infection. Results In Western blotting, the bands of 94, 48, 41, 40 kDa and 38 kDa of SCA appeared the earliest and were recognized by the rabbits sera of 2-week post-infection, the bands of 71 kDa and 23 kDa of SCA reacted with the rabbits sera of 3-week post-infection strongly. The bands of 71 kDa and 58 kDa of AWA appeared the earliest and were recognized by rabbits sera of 3-week post-infection. The bands of SEA reacted earliestly to the rabbits sera of 4-week post-infection were 270, 151, 73, 69, 50 kDa and 24 kDa. Conclusion The fraction antigens of 94, 71, 48, 41, 40, 38 kDa and 23 kDa of SCA, the fraction antigens of 71 kDa and 58 kDa of AWA and the fraction antigens of 270, 151, 73, 69, 50 kDa and 24 kDa of SEA could be recognized by sera of acute infected rabbits and might have potential early immuno-diagnosis value for schistosomiasis.
6.Relationship of postoperative systemic inflammatory response syndrome and preoperative midway through the urine and perioperative renal pelvis urine of percutaneous nephrostolithotomy

ObjectiveTo explore the relationship of postoperative systemic inflammatory response syndrome(SIRS) and preoperative midway through the urine and perioperative renal pelvis urine of percutaneous nephrostolithotomy(PCNL).Methods Participants included 450 patients with urinary calculus who underwent PCNL,preoperative midway through the urine and perioperative renal pelvis urine of PCNL was collected.ResultsOf 450 cases,preoperative midway through the urine germiculture positive 100 cases (22.2%,100/450 ),perioperative renal pelvis urine germiculture positive 85 cases (18.9%,85/450),46 cases ( 10.2%,46/450) occurred SIRS after PCNL.Decompression of perioperative renal pelvis urine germiculture positive 20 cases(23.5%,20/85) preoperative midway through the urine germiculture positive,perioperative renal pelvis urine the bacteria cultures negative 80 cases (21.9%,80/365 ) preoperative midway through the urine germiculture positive(P >0.05),preoperative midway through the urine germiculture positive 15 cases ( 15.0%,15/100) in SIRS,preoperative midway through the urine the bacteria cultures negative 31 cases ( 8.9%,31 /350) in SIRS (P > 0.05 ).Decompression of perioperative renal pelvis urine germiculture positive 18 cases (21.2%,18/85) in SIRS,perioperative renal pelvis urine the bacteria cultures negative 28 cases (7.7%,28/365) in SIRS (P < 0.05 ).ConclusionPreoperative midway through the urine has no correlation with the occurrence of SIRS,perioperative renal pelvis urine germiculture positive can predict the occurrence of SIRS,giving corresponding antibiotic treatment can improve the security of PCNL.
7.The effect of bone marrow mesenchymal stem cells transplantation on the expressions of retinal Fas/FasL and caspase-3 after ichemic reperfusion injury

Background Retinal ichemic-reperfusion injury (RIRI) affects vision and life quality and has no ideal treatment outcome up to now.The mechanism of RIRI is associated with apoptosis.Researches showed that subretinal transplantation of bone mesenchymal stem cells (BMSCs) can relieve RIRI,but its mechanism is unclear.Objective This study was to observe the influence of BMSCs subretinal transplantation on the expressions of apoptosis protein Fas/FasL,caspases-3 after RIRI,and to explore the neuroprotecitve mechanism of BMSCs transplantation to treat RIRI.Methods Bone marrow was isolated and extract form femur and tibia of SD rat by the density gradient centrifugation method and then the BMSCs were collected.BMSCs suspension was prepared using DMEM containing low-glucose with the cell density 1 x 106-2× 106/ml.RIRI models were established by ligation of optical nerve (Daniel M method).Sixty-four SD rats were randomly divided into the normal control group,model control group,BMSCs transplantation group and PBS control group.Twenty-four hours after RIRI,BMSCs suspension with the cells density 5× 104 cells was subretinally injected in the rats of the BMSCs transplantation group,and equal volume of PBS was injected in the same way in the rats of the PBS control group.The expressions of Fas/FasL and caspase-3 were dynamically detected using immunohistochemistry at 6,24,48 and 72 hours after BMSCs transplantation.Results The positive response cells for Hoechst33324 were seen in subretina 72 hours after BMSCs transplantation.A few of positive cells for Fas,FasL,caspase-3 proteins were expressed in the rats of the normal control group at 6,24,48 and 72 hours after BMSCs transplantation.However,the positive cells for Fas,FasL,caspase-3 protein were significantly increased in the model control group and the BMSCs transplantation group compared with the normal control group at various time points after injection (all at P<0.01),and the number of positive cells was significantly less in the BMSCs group than those of the RIRI group and the BMSCs transplantation group (P<0.05,P<0.01).No significant differences were found in the numbers of positive cells of Fas,FasL and caspase-3 proteins at various time points between the model control group and the PBS control group (all at P>0.05).Conclusions The subretinal transplantation of BMSCs down-regulate the expression of Fas/FasL and caspases-3 proteins in RIR1 rats,and thus alleviate the apoptosis of retinal neural cells.
8.Application of the double-center setup in the precise radiotherapy for middle esophageal carcinoma

Objective To compare the errors of double-center and single-center setup, and to study the role of both on reducing the rotational setup errors for the patients with esophageal carcinoma depend on rigid registration errors between online kV-cone-beam computed tomography (kV-CBCT) images and plans for CT images. Methods 20 patients with middle esophageal carcinoma received image scanning before treatment every week by using double-center setup and CBCT, and single-center setup images of 20 patients were taken from the X volume image (XVI) system. Then the images of both setup types, registration errors of CT image and rotational setup errors were compared respectively. Every patient received kV-CBCT scanning analysis before treatment every week, and 6 times in total. 240 group of kV-CBCT images from all of the patients were off-line matched with plans for CT images to calculate the errors of X-axis, Y-axis, Z-axis. Then the data of linear errors and rotational setup errors from patients were collected, aiming at putting the error data into the patients treatment program and analyzing the significances. Results The standard registration of double-center setup was as follows: T (X) (0.28 ±0.19) cm, T (Y) (0.27 ±0.19) cm, T (Z) (0.33 ±0.12) cm, R (X) (0.40 ±0.19)° , R (Y) (0.30 ±0.18)° , R (Z) (0.30 ±0.19)° . The standard registration of single-center setup was as follows:T(X) (0.32±0.20) cm, T(Y) (0.29±0.25)cm, T(Z) (0.31±0.16) cm, R(X) (2.2±0.68)°, R(Y) (0.5±0.32)°, R(Z) (2.10±0.60)°. There were statistical differences between linear errors in T(X) and rotational setup errors in R(X), R(Y) or R(Z) (P< 0.05). Conclusion Double-center position can reduce the rotational setup errors, especially in X-axis, Y-axis errors, and may provide more help for the radiation oncology departments without on-board CBCT.

KylinRay-IMRT is the advanced radiotherapy treatment planning module of accurate radiotherapy system (KylinRay) aiming to provide accurate and efficient plan design platform. In this paper the system design, main functions and key technologies of KylinRay-IMRT were introduced. KylinRay-IMRT supports three dimensional conformal radiotherapy (3D-CRT), intensity modulated radiotherapy (IMRT) and many other types of treatment plan design with function modules including patient data management, image registration and fusion, image contouring, image three dimensional reconstruction and visualization, three dimensional conformal radiotherapy planning, intensity modulated radiotherapy planning, plan evaluation and comparison, and report print. KylinRay-IMRT has been tested by the national standard YY/T 0889-2013, the results showed that the performance of KylinRay-IMRT can fully meet the standard requirements.
