1.Significance of Marrow Histotomy in Diagnosing Malignant Hemology

Objective To promote the level of morphological diagnosis and estimate prognosis combining the result of bone marrow histotomy and the swear of 82 cases of malignant hemology disease. Methods We collected 82 cases of malignant hemology disease, extracted marrow by one step and routinly made bone marrow swear and dyeing, made histotomy by embedding plastic. Diagnosis of malignant hemology disease refered to the standard of WHO 2001. Results Bone marrow histotomy was superior to bone marrow swear in diagnosing MF and hypocellular MDS. Bone marrow histotomy was easier to find cell of lymphoma than bone marrow swear. Bone marrow histotomy was a more sensitive index of tumor load of MM.It was also helpful for judging acute change of CML and relapse of acute leukemia. Conclusion Level of diagnosing malignant hemology disease can be promoted efficiently by combining marrow histotomy with bone marrow swear
2.Cloning, Expression and Activity of Recombinant Human Angiostatin

Objective: To express recombinant human angiostatin for further application in clinic. Methods: The complete encoding eDNA of human angiostatin was isolated from human embryo liver with RT-PCR and expressed in secretory Pichia expression system. Recombinant human angiostatin was purified with heparin sepharose chromatography and its activity was determined in chick embryo chorioallantoic membrane (CAM) and wound healing assays. Results: Expressed in large quantity (yield=5 mg/L) and purified with heparin sepharose, recombinant angiostatin was showed to have a molecular weight of 43 kD in SDS-PAGE and potently inhibit angiogenesis and wound healing. Conclusion: Recombinant human angiostatin was expressed efficiently in a biologically active form.

Objective To preliminary investigate the cytoactive of HELA cell after PUMA gene promoting HELA cell apoptosis and whether or not the protein of cyt-c,AIF in the chondrosome have participated in the process of PUMA gene inducing HELA cell.Methods we Used AO/EB dyeing to detect the Morphologic change of cell induces by PUMA gene and Western Blot to detect whether or not the position of cyt-c,AIF in the chondrosome have transferred after PUMA gene induces HELA cell apoptosis.Result ①Fluorescence microscope and inverted phase contrast microscope display HELA cell have a series of apoptosis Morphologic change after transfecting PUMA into HELA cell,and this Morphologic change of cell is more obviously after transfecting 48h than after 24h.;②Western Blot display the protein cyt-c and AIF transferred toward to kytoplasm after HELE cell took place apoptosis,the mount of transfecting protein is more obviously after transfecting 48h than after 24h.Conclusion ① PUMA gene have the effect of promoting HELA cell apoptosis;②cyt-c and AIF take part in the process of promoting HELA cell apoptosis,but during this process if the membrane potential of chondrosome was changing need to do some more study to confirm it.

Objective:To study the clinical application potential of aFGF and try to produce aFGF as gene engineering medicine.Methods:The complete encoding cDNA of human aFGF was isolated from human lung fibroblast with RT PCR.Recombinant human aFGF was expressed in secretory pichia expression system and purified with heparin sepharose chromatography.The activity of aFGF was detected in NIH3T3 proliferation assay,chick embryo chorioallantoic membrane assay(CAM) and wound healing assay.Results:The recombinant aFGF was expressed in large quantity(yield=12 mg/L) and was capable to promote proliferation of NIH3T3,angiogenesis and wound healing.Furthermore,those activities of aFGF could be agonized by recombinant FGFR extracellular domain.Conclusion:Recombinant human aFGF was expressed efficiently and possessed natural biological activities.

Objective To prepare chitosan-polyaspartic acid-5 fluorouracil (CTSPasp-5FU) nanoparticles and to investigate its anti-neoplastic effect and toxicity.Methods CTS-Pasly5FU nanopartieles were synthesized by ion gelatifieation.BALB/C nude mice were injected with gastric carcinoma cell line SGC- 7901 mass subcutaneously near nape to establish human gastric carcinoma model.Then they were randomly al- located into chitosan-polyaspartie acid -5fluorouracil(CTS-Pasp-SFU,containing 5-FU 1.25mg/kg) group, 5-FU (1.25mg/kg) group and normal saline group.Tumor weight was measured and the colony forming unit- granulocyte and maerophage (CFU-GM) was investigated.Results The drug content of CTS-Pasp-5FU was 40.2% and the encapsulation efficiency was 34.9%.Compared with normal saline group,tumor volume of 5-FU group and CTS-pasp-5-FU group were significantly decreased 21 days after treatment (P

Objective The aim of the present study is to evaluate the reliability and validity of the Mandarin version of the PP (MPP) .Methods The first step in the establishment was to translate the original English version into mandarin version with the method of cross -culture translation .The reliability was performed with the internal consistency analysis and test -retest reliability .The validity was performed for the content validity and structure va-lidity .The samples were from 80 Chinese CI children ,and 43 parents answered this questionnaire again 1 month lat-er to evaluate the test -retest reliability .The average age at cochlear implantation were 26 ± 14 months ,ranging from 7 months to 68 months ,the average duration of CI use were 10 ± 7 months ,ranging from 0 month to 24 months .Results The reliability analysis indicates that the Cronbach'sαcoefficient was 0 .797 ,except for the well-being and happiness ,education ,whose coefficients are respectively 0 .303 ,and 0 .341 ,all of the other sundomainscoefficient were greater than 0 .5 ,indicating the internal consistency was good .Test -retest reliability of the scale Cronbach'sαwas satisfactory .All subdomains and total score of the scale coefficients were greater than 0 .70(P<0 .01) .The validity analysis indicated that the pearson correlation coefficients among the total scale and the 8 subdo-mains were 0 .395~0 .992 ,the correlation coefficients among each subdomains were 0 .09~0 .654 ,which confirmed with the psychological characteristics ,proving its good structure validity .Conclusion The Chinese version of the PP show s good reliability and validity and can be used to evaluation the quality of life in mandarin CI children.
7.Application of medical science popularization competition in nursing interns from the perspective of healthy China

Objective:To explore the application effect of medical science competition in nursing interns whocontribute to "healthy China" , and to improve their health education awareness, ability, method and self-confidence.Methods:A total of 205 nursing interns who worked in Union Hospital of Tongji Medical College of Huazhong University of Science and Technology from 2019 to 2020 were selected as the research objects. They were divided into the control group (105 cases) and the experimental group (100 cases) according to whether they participated in the medical science competition. The control group learned the form and method of health education in clinical rotation according to the traditional practice teaching plan. The experimental group volunteered to participate in the medical science competition, which required the dissemination of health knowledge through various forms. Before and after the competition, the health education ability assessment scale was used for comparison.Results:Before the medical science competition, there was no significant difference in the total score of assessment, planning, implementation, evaluation and health education between the control group and the experimental group ( t values were 0.765 - 1.749, all P>0.05). After the medical science competition, the total scores of assessment, planning, implementation, evaluation and health education ability of nursing interns in the experimental group were (24.38 ± 4.72), (17.98 ± 3.98), (25.16 ± 5.36), (12.57 ± 2.96) and (80.09 ± 15.65) respectively, while those in the control group were (22.45 ± 6.29), (16.61 ± 4.77), (23.04 ± 6.55), (11.31 ± 3.46) and (73.41 ± 19.69).The differences between the two groups were statistically significant ( t values were 2.226 - 2.795, all P<0.05). Conclusions:The medical science competition can improve the health education ability of assessment, planning, implementation, evaluation of nursing interns and contribute to "healthy China" .
8.Early changes of TSH and thyroid hormones after131I ablation treatment in patients with papillary thyroid cancer

Objective To investigate the patterns of change in thyroid functional parameters ( serum TSH,FT3, and FT4 ) in patients with papillary thyroid cancer (PTC) before and after the initial 131I treatment for thyroidal remnant ablation. Methods Seventy-four PTC patients, treated with 3.7 GBq 131 I therapy, were divided into two groups, group A with serum TSH<30 mIU/L and group B with serum TSH ≥30 mIU/L the day before 131I treatment. Five days after the treatment, the patients were re-examined for serum FT3, FT4, and TSH levels.Results In group A (22 cases), 5 days after the 131I ablation treatment, FT4significantly increased by 88% and FT3 by 87%, while TSH decreased by 87% (all P<0. 05 ), and 45% (10/22)cases manifested the signs of transient thyrotoxicosis. In group B (52 cases)after treatment, individual variance of FT3 and FT4 was obvious,with FT4 decreased by 13% and FT3 decreased by 14% ( both P<0. 05 ), while TSH slightly increased by an average of 6% ( P>0.05 ). Conclusion After the initial 131 I ablation therapy for thyroidal remnant, the thyroid hormone levels in some PTC patients significantly increase while in others may slightly decrease in the early stage. The supplementary and suppressive therapy after 131I ablation for PTC patients might be individualized depending on the thyroid hormone determination.
9.The Preventive Effects of N-Acetylcysteine on Contrast-Induced Nephropathy in Patients Undergoing Elective Percutaneous Coronary Intervention

Objective To investigate the preventive effect of N-acetylcysteine (NAC) on contrast-induced nephropa-thy (CIN) in patients undergoing elective percutaneous coronary intervention (PCI). Methods A total of 521 patients under-went PCI in Tianjin were randomly divided into conventional treatment group (n=261) and NAC treatment group (n=260). NAC treatment group was given oral NAC (600 mg twice daily) for 48 h and 72 h before PCI plug hydration therapy, and the conventional treatment group was given only hydration therapy. The serum levels of creatinine(Scr), urea nitrogen(BUN), creatinine clearance rate (Ccr), C-reactive protein (CRP),β2-microglobulin(β2-MG), tumor necrosis factor-α(TNF-α), inter-leukin-6 (IL-6), superoxide dismutase (SOD), glutathione peroxidase (GPX) and incidence of CIN were detected at admission and 72 h after the procedure. Results (1) There was no significant difference in the incidence of CIN between NAC treat-ment group (6.2%) and conventional treatment group (3.8%,χ2=1.48, P>0.05). (2) There were no significant differences in se-rum levels of Scr, BUN, Ccr, CRP,β2-MG, TNF-α, IL-6, SOD and GPX before PCI ( P>0.05). (3) The serum levels of CRP, SOD and GPX were significantly higher 72 h after the procedure in two groups ( P<0.05). There were significantly lower se-rum levels in CRP, SOD and GPX in NAC treatment group than those of conventional treatment group ( P<0.05). There were no significant differences in serum levels of Scr, BUN,β2-MG and Ccr between NAC treatment group and conventional treat-ment group ( P >0.05). Conclusion N-acetylcysteine may have no beneficial effect on the prevention of CIN after PCI.
10.Changes in myeloid-derived suppressor cells and cytokines in peripheral blood of patients with ankylosing spondylitis

Objective:To investigate the expression of myeloid-derived suppressor cells (MDSCs) and cytokines in peripheral blood of patients with ankylosing spondylitis (AS).Methods:From June 2018 to June 2019, 50 AS patients (AS group) and 50 healthy people (control group) in Lishui People's Hospital were selected in this study.The number of PMN-MDSC and monocyte MDSC(M-MDSC) and the expression of cytokines (IL-2, IL-4, IL-6, IL-10, TNF-α, γ-IFN and IL-17) in the peripheral blood were detected by flow cytometry.The differences of MDSCs expression frequency in each group and its relationship with cytokine expression were analyzed.Results:The PMN-MDSC, M-MDSC and IL-6 in peripheral blood of the AS group were (0.22±0.08)%, (1.48±0.32)% and (20.74±5.14)ng/L, respectively, which were significantly higher than those of the control group[(0.06±0.02)%, (0.43±0.11)% and (2.52±0.53)ng/L], and the differences were statistically significant ( t=7.933, 6.310, 10.553, all P<0.05). Conclusion:IL-6 may be one of the reasons to promote the infiltration and recruitment of MDSCs, and the increase of MDSCs may be related to the occurrence and development of immune tolerance after ankylitis infection.
