Clinical Medicine of China 2008;24(5):420-422
Objective To investigate human esophageal cancer Eca109 cells apoptosis induced by resveratrol. Methods Eca109 cells were treated with difierent concentrations of resveratrol and the change of morphology was observed by inverted microscope and transmission electron microscope in different time; The cell growth inhibitory rate of resveratrol on the proliferation of Eca109 cells were evaluated in vitro by MTT assay.Flow cytometry (FCM)analysis was used to determine the apoptosis of Eca-109 cells. Results Resveratrol was able to inhibit the growth of Eca109 cells, and the suppressive effect was increased with treating concentration and treating time.Resveratrol inhibited the growth of esophageal cancer Eca109 cells in a dose-and time-dependent manner and induced the apoptosis in esophageal cancer. Conclusion Resveratrol could inhibit the proliferation of Eca-109 cells and block the cell cycle at G0/G1.And in the end resveratrol could induce the apoptosis of Eca109 cells.
2.The comparison of the ratio of Th17/Treg cells in patients with ankylosing spondylitis and the normal controls
Chinese Journal of Rheumatology 2013;(7):481-484
Objective To analyze the variation of peripheral blood Th17 cells and Foxp3+CD4+CD25+regulatory T (Treg)cells in patients with ankylosing spondylitis (AS),and to interpret the role of the ratio imbalance of peripheral blood Th17/Treg cells in the pathogenesis of AS.Methods The percentage of peripheral blood Th17 cells and Foxp3 +CD4+CD25+ Treg cells in AS patients and healthy subjects were determined by flow cytometry (FCM).The concentrations of IL-2,IL-10,TGF-β,IL-6,IL-17 and IL-23 in the serum were measured by enzyme linked immunosorbent assay. The correlation between these cytokines and traditional parameters was analyzed.T test,ANOVA,SNK-q and Speannan's analysis were used for statistical analysis. Results Compared with healthy controls, the percentage of Th17 cells in AS patients increased significantly (P<0.05); Compared with the inactive AS patients, the percentage of Th17 cells in active AS patients increased significantly [(2.10±0.82)%,(2.89±1.77)%,P<0.05)]; the percentage of Foxp3+CD4+CD25+ Treg cells in active AS patients was markedly lower than healthy controls (P<0.05).Compared with healthy controls and inactive AS patients, the specific value of Th17/Treg in active AS patients increased significantly(P<0.05).The concentrations of IL-17 and IL-23 was found significantly higher in patients with AS than that in healthy controls (t=2.203,P=0.032 and t=2.088,P=0.041).The concentrations of IL-17 and IL-23 was found significantly higher in patients with active AS than that in inactive AS (t=2.808,P=0.008 and t=2.242,P=0.020).The correlation analysis revealed that these cytokines and traditional parameters was not relevant except IL-17/BASDAI (r=0.32,P=0.044).Conclusion The increased ratio of Th17 cells and decreased ratio of Foxp3+CD4+CD25+ Treg cells may play a critical role in AS progression.Manipulating the balance of Th17/Treg would provide a new approach for the treatment of AS.

Journal of Leukemia & Lymphoma 2011;20(7):389-391,394
Objective To investigate the possible mechanism of apoptosis effect induced by resveratrol in human acute T lymphoblast leukemia Molt-4 cells. Methods MTT method was used to analyze the inhibition rate. The cell cycle and apoptosis percentage of Molt-4 cells treated with resveratrol were detected by flow cytometry. The expression of WAVE1 mRNA was assessed by semi-quantitative RT-PCR. Results After treated with 12.5, 25.0, 50.0, 100.0, 200.0μmol/L resveratrol for 24, 48 and 72 hours, the inhibitory effects were at 29.32 %, 36.11 %, 53.92 %, 62.50 %, and 74.98 %, respectively. Resveratrol was able to inhibit the proliferation of Molt-4 cells in a time-dependent and dose-dependent manner (F =33.614, P <0.05).Compared with control group, after treated with 50.0, 100.0 μmol/L resveratrol for 48 h, the cell number in S phase of Molt-4 cells was 68.6 % and 78.1 %, respectively, and the cell cycle was arrested at S phase (F = 19.453, P < 0.01) after treated with 50.0, 100.0 μmol/L resveratrol for 48 h, the ratio of WAVE1/GAPDH was 0.356±0.03, 0.382±0.05. Compared with control group 0.586±0.06, the ratio of WAVE1/GAPDH was decreased (F =8.950, P <0.01). Conclusion Resveratrol could induce the cells to undergo apoptosis, which was probably related to downregulation the expression of WAVE1.
