1.The correlation of TCF7L2 genetic polymorphisms with intimal -medial thickness and stiffness in the hypertension and type 2 diabetic patients

Objective To explore the relationship between TCF7L2 genetic polymorphisms with intimal -medial thickness (IMT),carotid arterial stiffness in patients of type 2 diabetes mellitus(T2DM)with hypertension. Methods 872 patients of hypertension and T2DM were enrolled into this study,and they were divided into two groups according to IMT,carotid arterial stiffness and cPWV:the observation group(n =432,IMT >1.0mm,cPWV >12m/s),the control group(n =440,IMT≤1.0mm,cPWV≤12m/s).Meanwhile,400 healthy subjects(normal fasting gluose,blood pressure,without diabetes and hypertension family history)were selected as the healthy group.Typed gene polymorphisms of TCF7L2 by RT -PCR,the body mass index(BMI),waist -hip ratio,systolic blood pressure, diastolic blood pressure,total cholesterol(TC),triglyceride(TG),high density lipoprotein cholesterol(HDL -Ch), low -density lipoprotein cholesterol(LDL -Ch),insulin sensitivity index(ISI),glycated hemoglobin A1C(HbA1c) between groups were compared.Calculated each allele and genotype frequencies and implemented Hardy -Weinberg equivalent test.The distributional differences of alleles frequencies between groups were analyzed by χ2 test and geno-type frequencies through dominant model.Results Distributional differences of rs290487 genotype frequencies had statistically significant differences in the observation group vs the control group(χ2 =33.296,P <0.05 )and the T2DMgroup vs the normal group(χ2 =100.801,P <0.05).Statistically significant differences existed in C and T alleles frequencies as well:the observation group vs the control group(χ2 =16.280,P <0.05),the T2DM group vs normal group(χ2 =27.190,P <0.05).However,the gene polymorphisms of rs11196205 and rs11196218 showed no statistically significant difference (P >0.05 ).Conclusion rs290487 involved in the development of T2DM with hypertension,C and T alleles might be the risk factors of increasing the IMT and carotid arterial stiffness levels in patients of T2DMwith hypertension.Logistic regression analysis revealed that the increase of SBP,DBP,TC,TG,LDL-Ch and decrease of HDL -Ch may be the risk factors of hypertension and diabetic patients.Meanwhile,it dedicated that the increase of those biochemical indexes might be the important elements which would lead to the gene variations of C and T alleles.

Nerve growth factor (NGF) has been the focus in area of angiogenesis.In differential,survival,development of nerves,NGF plays a crucial role.In addition,NGF activates migration,differential of endothelial cells and vascular smooth muscle cells,and stimulate the activation of the endothelial progenitor cells in angiogenesis.Crosstalk between vascular endothelial growth factor and NGF is also critical in the development of vessels.The function of NGF in angiogenesis blazes a trail in therapy of ischemia diseases.

4.The effect of percutaneous coronary intervention on myocardial perfusion in coronary heart disease patients

Objective To evaluate the effect of percutaneous coronary intervention(PCI) on myocardial perfusion in coronary heart.Methods Fourty-two patients with coronary heart disease who were confirmed to have coronary artery steonsis by the coronary angiography were selected.They were checked by myocardial contrast echocardiography of intravenous before and after treatment by PCI.Then we determined quantitatively the perfusion of coronary microcirculation based on the extent of myocardial imaging and recorded it images.The myocardial contrast echocardiography result was compared before and after treatment by PCI for evaluating the situation of myocardial perfusion and effect of PCL So we can estimte the patients' prognosis.Results There were 39 eases of 42 patients with PCI that their lesion vascular were all opening and unblocked.Repeated radiography showed that their TIMI blood flow was the third level.The partial cross sectional area of all the capillaries [A =15.46 ± 3.27) min] blood flow velocity [β =(0.75 ±0.16)min/s],and myocardial blood flow volume[A - β =(12.00 ±4.51 ) min2/s] dependent on myocardial segments all increased more significantly than those before the PCI[A =(6.68 ± 1.76) min,β =(0.40 ± 0.12) min/s、A · β =(2.82 ± 1.38 ) min2/s] (t =37.2527.58、30.65,P < 0.05 ).After PCI three months the EF value of follow-up patients was increased mote than that of before PCI (t =13.77,P < 0.01 )but the scoring index of ventricular wall motion was decreased more than that of before PCI ( t =8.75,P < 0.01 ).Correlation analysis showed that after PCI A,β,A · β all had a strong correlation ( every P < 0.01 ).Conclusion PCI could effectively improve the situation of myocardial perfusion in myocardial infarction patients.It could save ischemia myocardial in infarction field and also improve the function of left ventricular if patients implemented PCI operation as soon as possible.

Objective To investigate the protective effect of adiponectin on angiotesin Ⅱ (Ang Ⅱ)-mediated cardiomyocyte apoptosis,and explore the related mechanism. Methods Cardiomyocytes isolated from neonatal rats were cultured in vitro. The identity of cardiomyocytes was confirmed by morphological examination and staining with anti-sarcomeric ?-actin antibody,and most (more than 95%) of the cells were identified as neonatal rat ventricular myocytes. After being placed in a serum-free medium for 24 hours,the ventricular myocytes were randomly grouped and received the following treatments respectively:placebo,or Ang Ⅱ (1?mol/L),or Ang Ⅱ (1?mmol/L) plus adiponectin (30mg/L). The protein levels of Bcl-2 and Bax were detected by Western blotting. The apoptotic rate and the levels of intracellular reactive oxygen species (ROS) in the ventricular myocytes were determined by flow cytometry. The survival rate of ventricular myocytes was evaluated by trypan blue staining. 6.Numerical assessment of impeller features of centrifugal blood pump based on fast hemolysis approximation model.

The impeller profile, which is one of the most important factors, determines the creation of shear stress which leads to blood hemolysis in the internal flow of centrifugal blood pump. The investigation of the internal flow field in centrifugal blood pump and the estimation of the hemolysis within different impeller profiles will provide information to improve the performance of centrifugal blood pump. The SST kappa-omega with low Reynolds correction was used in our laboratory to study the internal flow fields for four kinds of impellers of centrifugal blood pump. The flow fields included distributions of pressure field, velocity field and shear stress field. In addition, a fast numerical hemolysis approximation was adopted to calculate the normalized index of hemolysis (NIH). The results indicated that the pressure field distribution in all kinds of blood pump were reasonable, but for the log spiral impeller pump, the vortex and backflow were much lower than those of the other pumps, and the high shear stress zone was just about 0.004%, and the NIH was 0.0089.

7.Relationship between expression of PD-L1 and microsatellite instability of colorectal cancer and its significance

Objective:To investigate the expression and clinical significance of PD-L1 in colorectal cancer (CRC). Methods:A total of 210 CRC patients who accepted radical surgery in our hospital from January 2015 to January 2016 were divided into three groups, namely, high-frequency microsatellite instability (MSI-H), low-frequency microsatellite instability (MSI-L), and microsatellite stable (MSS). The expression of PD-L1 was detected by immunohistochemistry, and the expression characteristics of PD-L1 in different types of CRC were analyzed. Results:CRC cases with low differentiation had a higher expression of PD-L1 than CRC patients with high differ-entiation (P<0.05). PD-L1 had a positive rate of 75.8%in the MSI-H group and a rate of 9.3%in the MSI-L and MSS groups, wherein the difference between the two groups was statistically significant (P<0.05). Conclusion:PD-L1 was positively expressed in some CRC tu-mor tissues, and its positive rate was significantly higher in MSI-H than in MSI-L and MSS. The therapeutic effect of a PD-L1 blocker for patients with MSI-H CRC might be preferable.

9.Expression of the estrogen receptor α and β in mouse thymus

Objective: To detect estrogen receptor α and β(ER-α, ER-β)protein expression in different age of mouse thymus.Methods:Protein expression of ER-α and ER-β in thymus was analyzed via immunohistochemistry.Moreover,the relationship between ER-α and cytokeratin 18(epithelial cell marker)was further tested through fluorescence double-staining.Results: Immunohistochemical analysis revealed that both ER-α and β protein was found in nuclei of some thymocytes of 3 month mice.However,expression of ER-β was absence while ER-α was still positive in aged mice, such as 12 months and 16 months old.Double staining further confirmed that lots of ER-α/β positive cells were Foxp3 negative cells.Conclusion: Expression of ER-β is absent while ER-α is still positive in thymus of aged mice, which indicates ER-α is the critical estrogen receptor that involves in thymic involution.Moreover, ER-α/β do not participate in Treg development within thymus.
