1.Determination of sugars in heteropolysaccharide by phenol-sulfuric acid method

AIM: To develop a new method based on phenol-sulfuric reaction for the determination of hetero-polysaccharide. METHODS: The proportion of sugars in the hetero-polysaccharide was determined by GC. According to standard solution obtained, Heteropolysaccharide can be determined through the standard curve. ( RESULTS:)The linear relationship of standard solution of mixed sugars was at the range of 8.56-38.52 mg/L~(-1). CONCLUSION: GC and phenol-sulfuric acid method can be used to determine the heteropolysaccharide accurately.
2.The clinical study on dinoprostone in premature rupture of the membranes at term cervical ripening

Objective To explore clinical effect of dinoprostone in premature rupture of the membranes at term cervical ripening.Methods 42 cases with premature rupture of the membranes at term cervical ripening were selected.All patients were randomly divided into two groups.The dinoprostone group had 21 cases,which applied the treatment of dinoprostone belt the posterior fornix of vagina medicine induced abortion.The oxytocin group had 21 cases,which applied the treatment of intravenous drip oxytocin.The time of induction labor,6h,12h cervical Bishop score after using the medication,drugs acting time,labor conditions and perinatal situation,pregnancy outcome and some other index were observed.Results It taken (6.18 ± 4.48 ) h until parturient and ( 12.62 ± 8.03 ) h until the cervical mouth full open in the dinoprostone group,which were shorter than the oxytocin group,that taken( 10.21 ±5.42) h,( 18.87 ± 9.14) h.( t =2.62,2.35,all P < 0.05 ).There was no significant difference about cervical Bishop score before using the medication.It's(7.52 ± 2.44 ) point 6h after using the medication and( 9.03 ± 1.96 ) point 12h after in the dinoprostone group.They were better than the oxytocin group,which were(5.97 ± 1.95 )point,(7.13 ±2.12 )point.It was significantly different( t =2.27,3.02,all P <0.05 ).The average incubation period of dinoprostone group wss (9.91 ± 1.73 )h.It's longger than the oxytocin group,which was( 8.72 ± 1.34)h.The active period of dinoprostone group was(4.36 ± 0.66 ) h.It's shortter than the oxytocin group,which was( 5.84 ± 1.02 ) h.The difference was significant( t =2.49,5.58,all P < 0.05 ).There was no significant difference in the second and third stage of laber between the two groups( t =0.56,0.33,all P > 0.05 ).There was also no significant difference about intrauterine distress,amniotic fluid contamination,cesarean section,intestinal bleeding,the Apgar score of newborns and some other index in the two groups ( x2 =0.00,0.00,0.52,t =0.84,1.04,all P > 0.05 ).Conclusion Dinoprostone had good promoting effect on cervical ripening for preterm premature rupture of membranes pregnant woman.Its effect was better than oxytocin and it was safe to use.
3.Kindness trait of personality in pupils abused

Objective To study the kindness trait of personality of pupils in grade 5 and 6,and to explore its relationship with childhood abuse. Methods 172 elementary school students were investigated by applying Female had higher scores of generosity, altruism, honesty and kindness trait than male ((41.1±4.4) vs (37.6±6.6),(19.2±3.9) vs (17.1±4.6),(17.3±3.0) vs (15.3±3.5),(77.6±8.8) vss (69.9±12.6), F=tions on kindness trait(P<0.05). Sexual abuse had not predictable functions on kindness trait (P>0.05). Con-clusion There are significant differences of kindness trait among subjects with different genders. Childhood abuse can severely impact the development of kindness trait of personality in puerile.
4.Effect of thyrotropin-releasing hormone (TRH) on endotoxic shock in rats

The effect of TRH on endotoxic shock in rats was studied, iv 0. 22-2 mg ?kg-1 TRH significantly reversed hypotension induced by iv coli E. endotoxin (40 mg ?kg-1) into rats and caused a 4. 2 kPa rise in mean arterial pressure (MAP). MAP after TRH administered could be stablized over a higher level than control for 30 min and maintained for 3 h during observation. Interestingly enough, the MAP rose gradually in TRH-treated rats as contrast with increasingly falling of that in control group during the late shock. TRH also improved 24 h sur-vival of shock rats. The dose-response relationship could be observed between 0. 22 ~0. 67 mg ?kg-1 TRH and disappeared over a highest dose (2 mg ?kg-1). It was shown that the best dose to reverse hypotension and to improve survival was 0. 67 mg ?kg-1 TRH. Naloxone 2 mg ?kg-1 showed the nearly same effect as 0. 22 mg ?kg-1 TRH in increasing MAP, but the former had higher 24 h survival of rats than the later.
5.THE EFFECTS OF CUMULUS CELL APOPTOSIS ON DEVELOPMENTAL POTENTIAL OF HUMAN OOCYTE IN VITRO

Objective To study the effect of cumulus cell apoptosis on the structure of human oocyte cultured in vitro and to probe into the reason why cumulus cell apoptosis rate can be used clinically to predict the developmental potential of oocyte. Methods Cumulus-enclosed oocyte complexes (COCs) at the GV stage were cultured in vitro. The apoptosis rate of cumulus cells for every COC was evaluated with HE staining, DAPI staining and TUNEL labeling methods. Oocytes were divided into two groups according to their cumulus cell apoptosis rate. Structure of oocytes was observed under light microscope and transmission electron microscope. Mature oocytes at MⅡ stage were fertilized and cultured in vitro for two days. Then the embryos were evaluated according to their morphology under light microscope.Results The oocytes with low cumulus cell apoptosis rate had a normal structure and a promising developmental potential, while the oocytes with high cumulus cell apoptosis rate had deformed structures and a poor developmental potential. Some deformed oocytes had uneven perivitelline space. For oocytes with a small perivitelline space, the organelle development was slow compared with those with a large perivitelline space. Some had unevenly piled organelles. Secondary lysosomes and large space probably caused by the decopmounding of lysosomes were found in the ooplasm of these oocytes. The deformed oocytes appeared to have lots of swollen mitochondria with blurred cristae and membrane. Some of these oocytes had fractured first polar bodies and abnormally thick or thin zona pellucida. The cell junctions and microvilli of cumulus cells reduced as well. Conclusion The effect of cumulus cell apoptosis on the structure of oocyte cultured in vitro was revealed. The reason why cumulus cell apoptosis rate prognosticated the developmental potential of oocyte was demonstrated in the aspect of oocyte structure.
6.Correlation between diabetes mellitus and atherosclerosis in the elderly

Objective To investigate the relationship between blood biochemical indicators levels (glycosylated hemoglobin and serum bilirubin) and atherosclerosis in elderly type 2 diabetic.Methods Totally 100 type 2 diabetic patients aged 62-85 years were divided into type 2 diabetes mellitus with carotid atherosclerosis group (T2DM-AS group,n=55) and type 2 diabetes mellitus group (T2DM group,n=45).Blood biochemical indicators levels (glycosylated hemoglobin and serum bilirubin) were determined and compared between the patients and the healthy controls (n=60)Results Serum levels of total cholesterol,triglyceride,low-density lipoprotein and Lp(a) were elevated in T2DM-AS group as compared with T2DM group [(5.86 ± 0.28) mmol/L vs.(4.63 ±0.32) mmol/L,(2.02±0.43) mmol/L) vs.(1.56±0.39) mmol/L,(3.74±0.76) mmol/L vs.(3.03±0.81) mmol/Land (297.3±102.2) mmol/Lvs.(203.5±110.4) mmol/L,t=20.91,5.54,4.51 and 4.40,all P<0.05].Glycosylated hemoglobin level was higher in T2DM-AS and T2DM groups than in control group [(9.58 ± 0.47) mmol/L,(7.21 ± 0.66) mmol/L vs.(5.31 ± 0.54)mmol/L,both P<0.05].There was a significant difference in glycosylated hemoglobin level between T2DM-AS group and T2DM group [(9.58 ± 0.47) mmol/L vs.(7.21 ± 0.66) mmol/L,t=20.9,P<0.05].Serum bilirubin level was lower in T2DM AS group than in T2DM and control groups [(8.02±1.23)μmol/L vs.(13.72±1.64)μmol/L,(19.66±1.41)μmol/L; t=19.8,47.0,both P <0.05].Conclusions Glycosylated hemoglobin and serum bilirubin may promote atherosclerosis in elderly type 2 diabetic.The control of glycosylated hemoglobin and blood lipids levels may help to reduce atherosclerosis in elderly type 2 diabetic patients,which has the important clinical significance for the prevention and treatment of type 2 diabetes mellitus.

10.Changes of Tolerance and Cardiovascular Responses to Head-up Tilte Combined with LBNP Following Preceding Exposure to Head-down Tilt

Objective To observe pre-syncopal limited tolerance and cardiovascular responses to head-up tilt combined with lower body negative pressure (HUT/LBNP) following exposure to head-down tilt (HDT, -1 Gz). Method Exposures to HUT/LBNP (-60 mmHg) in control session (without preceding 30 s -1 Gz treatment) and in simulated push-pull effect (PPE) session (with preceding 30 s -1 Gz treatment) were performed in 8 healthy adults. The changes of hemodynamic parameters were monitored by electrical impedance instrument during the experiments. Result The mean endurance time in presyncopal symptom limited HUT/LBNP in control session and in simulated PPE session were 8.4±2.1 min and 4.5±2.4 min, respectively, the two means were significantly different (P< 0.01). In simulated PPE session, as compared with baseline, heart rate (HR) during HDT was significantly lowered (P<0.01), while stroke volume (SV) and cardiac output (CO) were increased significantly (P<0.01). During HUT/LBNP, the increased percentage (relative to baseline) of HR in PPE session was lower than these in control session (P<0.05); the decreased percentages of SV and CO during HUT/LBNP in PPE session were both higher than those in control session (P<0.05). During HUT/LBNP, arterial pulse pressure (PP) of control session was significantly decreased than the value of baseline value (P<0.05); Total peripheral resistance (TPR) of PPE session was significantly increased than baseline value (P<0.05). Conclusion Tolerance time before the appearance of presyncopal symptoms during HUT/LBNP decreases and cardiovascular responses to HUT/LBNP are impaired, preceding exposure to HDT.
