Objective To investigate the medical effect of the ethanol extract of Acorus gramineus Sol.on arthritis of mice induced by collagen-Ⅱ,and explore the potential pharmacological mechanisms.Methods Arthritis mouse model was established by injection of admixture containing type Ⅱ collagen and Freund's complete adjuvant (FCA) in male BALB/c mice.Mice were divided into five groups:the normal control group (0.9% of sodium chloride solution),the model control group (0.9% of sodium chloride solution),tripterygium group (15 μg·kg-1 of tripterygium tablets), the high-dose of extract of Acorus gramineus Sol.group (60 mg·kg-1 extract of Acorus gramineus Sol.) and the low-dose of extract of Acorus gramineus Sol.group (15 mg·kg-1 extract of Acorus gramineus Sol.).Each group was administered once a day,lasting 21 days.During the experiment,ankles of all mice were measured at predetermined time.At the end of the experiment,blood of the mice was exsanguinated and centrifuged to get serum for measuring the levels of IL-1β,RF and TNF-α.Spleens of mice were dissected and weighed to calculate the spleen index.All arthritis ankles were dissected to make tissue section,and observed under microscope.Results Compared with the model control group,the perimeter of ankle joints of the high-dose of extract of Acorus gramineus Sol.group significantly changed 6 days after administration (P<0.05);That of the low-dose of extract of Acorus gramineus Sol.group significantly changed 12 days after administration (P<0.05);That of tripterygium group significantly changed 9 days after administration (P<0.05).As compared with the normal control group, the spleen index of the model control group was significantly different (P<0.01).As compared with the model control group,the spleen index of tripterygium group,high-dose and low-dose of extract of Acorus gramineus Sol.groups were significantly different (P<0.05).As compared with the normal control group,levels of IL-1β,RF and TNF-α of the model control group were significantly different (all P<0.01).As compared with the model control group,levels of IL-1β,RF and TNF-α of tripterygium group,high-dose and low-dose of extract of Acorus gramineus Sol.groups were significantly decreased.Conclusion Ethanol extracts of Acorus gramineus Sol.have significant therapeutic effect on arthritis mice.The anti-arthritic mechanism is associated with its ability to regulate levels of IL-1β,RF and TNF-α.

Objective: To establish a GC method for determination of menthol, camphor and methylsalicylate in Cremor Mentholi Compositus. Methods: The determination was carried out on a PEG\|20M stainless steel column with temperature at 140℃, FID detector with temperature at 260℃. Results: The separating degree and the linearity were fine with the average recovery of menthol 100.17%, RSD =0.661%; camphor 99.89%, RSD =0.600%; methylsalicylate 100.17%, RSD =0.553%, respectively. Conclusions: The method is convenient, rapid and accurate. It can be used to control the quality of this preparation.

Objective To systematically compare the operative and non-operative treatments of acute injury to the lateral ankle ligaments in terms of efficacy and complications.Methods The databases of MEDLINE (From January 1966 to October 2011 ),EMBASE (From January 1988 to October 2011 ),CBMdisc (From January 1978 to October 2011) and CNKI (From January 1980 to October 2011 ) were researched for eligible randomized-controlled trials (RCTs),controlled clinical trials,quasi randomized controlled trials and cohort study which compared operative and non-operative treatments for acute injury to the lateral ankle ligaments.The methodological quality of the eligible studies collected was evaluated.The data of the studies included were extracted for a Meta analysis to compare the motional recovery,functional stability,re-injury,residual pain and post-treatment complications of the ankle between operative and non-operative treatments.RevMan 5.0 software was used for statistical analysis.Results In all the eligible 13 studies included,713 patients were treated operatively and 817 patients non-operatively.The functional stability of the ankle was significantly better in the operative treatment group than in the non-operative treatment group [ OR =0.72,95％ CI,(0.52,0.99),P ＜ 0.05].The incidence of ankle arthrocleisis in the operative treatment group was significantly higher than in the non-operative treatment group [ OR =3.41,95％ CI (1.56,7.44),P =0.002].There were no statistical differences in the motional recovery [ OR =1.14,95％ CI (0.58,2.21),P＞ 0.05],incidence ofre-injury [OR=0.68,95％ CI (0.35,1.31),P＞0.05],residual pain [ OR =0.81,95％ CI(0.56,1.16),P＞ 0.05],or ankle dyskinesia [ OR =2.38,95％ CI (0.91,6.25),P＞ 0.05]between the 2 groups.The incidences,scar tenderness [01R=7.46,95％CI(1.32,42.08),P ＜0.05]and sensory nerve loss [OR=12.16,95％ CI(2.24,66.02),P ＜0.05]were significantly higher in the operative treatment group than in the non-operative treatment group.The total rate of complications was significantly higher in the operative treatment group than in the non-operative treatment group [0R=6.20,95％ CI (2.67,14.41),P ＜0.05].Conclusions Compared with non-operative treatments,operative treatments for acute injury to the lateral ankle ligaments can significantly improve the functional stability of the ankle,but make no significant differences in the motional restoration,re-injury or the residual pain of the ankle.In addition,operative treatments may increase the risk of complications.

Objective To investigate the clinical effect of air compressor pump atomizing inhalation with pulmicort respules on idiopathic pulmonary fibrosis (IPF).Methods72 IPF patients were randomly divided into there groups with 24 cases in each group. The group A was treated with pulmicort respules suspension by air compressor pump atomizing inhalation, 1 mg each time, twice each day; the group B with pulmicort aerosol, 4 puffs each time, twice each day and the group C with prednisone, 1 mg/kg each day, reduce half of the dosage 4 week later. The clinical manifestation, pulmonary function testing, high-resolution CT of the chest, arterial blood gas analysis and side effects were observed before and 3 months after treatment.ResultsAfter treatment, the improvement rate of symptoms was 70.8% in group A, 25% in group B and 50% in group C, with a significant difference between group A and group B ( P<0.0125), but no difference between group A and group C ( P>0.0125). The improvement rate showed by lung CT was 66.7% in group A, 29.2% in group B and 45.8% in group C, with a significant difference between group A and group B ( P<0.0125), but no difference between group A and group C ( P>0.0125). PaO2 improved but no significant difference compared with that before treatment ( P>0.05). The incidence rate of side effect was 8.3% in group A, 8.3% in group B and 58.3% in group C with a significant difference between group A and group C ( P<0.0125).ConclusionAir compressor pump atomizing inhalation with pulmicort respules is effective in the treatment of IPF and has few side effects.

Objective To clone,express,and purify Tp 0319 outer membrane protein of Treponema pallidum and to develop an indirect ELISA for diagnosing syphilis.Methods The expression plasmid PQE32/Tp 0319 was conventionally constructed.The recombinant Tp 0319 protein was produced in E.coli M15 after induction by IPTG.The Tp 0319 protein was analyzed by SDS-PAGE and Western blotting,and then purified with Ni-NTA affinity chromatography.Indirect ELISA was developed to detect the syphilis antibody in human sera.Results The recombinant plasmid PQE32/Tp 0319 was constructed successfully and the fusion protein with relative molecular weight near 30 000 Dalton was revealed by SDS-PAGE.Western blotting proved that the recombinant protein specifically reacted with anti-Tp antibodies in sera from syphilis patients.The results of the indirect ELISA indicated the sensitivity and the specificity were both 100%.The concordance of 300 sera(150 from blood donors and 150 from syphilis patients)detected in parallel by the ELISA and the TPPA was 95.3%.Conclusions The data suggest that the prepared recombinant protein Tp 0319 of Treponema pallidum has high immunoreactivity.The recombinant protein can be used to develop ELISA kit for diagnosing syphilis.

Objective To study the diagnostic values of a distintegrin and metalloproteases-8 (ADAMS) in non-small cell lung cancer(NSCLC).Methods The serum protein level of ADAM8 was assayed by ELISA in 62 NSCLC patients,27 lung benign lesions and 32 healthy people.Results The serum levels of ADAM8 of NSCLC [(456.88±143.87)ng/L] were higher than those with lung benign lesions[ (271.63±74.20) ng/L] and normal controls(253.09±72.15 ng/L) (P<0.01), but there was no significant difference between lung benign lesions and normal controls (P0.05).No significant difference was found between adenocarcinoma and squamous cell carcinoma in serum ADAM8 level,the serum levels of ADAM8 in NSCLC with stages Ⅲ-Ⅳ [ (498.80 ± 151.80) ng/L] were higher than those with stages Ⅰ -Ⅱ[(385.80±95.85) ng/L] (P <0.01 ).The diagnostic sensitivity of detection of ADAM8 for NSCLC was 77.4% and the specificity was 90.6%.Conclusion The overexpression of ADAM8 in serum of NSCLC patients indicates that ADAM8 is related with occurrence and metastasis of NSCLC;De-tection of ADAM8 could assist the diagnosis for NSCLC.

Objective To investigate the correlation of microperimetric parameters,best-corrected visual acuity (BCVA) and central retinal thickness (CRT) in diabetic macular edema (DME) eyes.Methods It is a prospective,no controlled,open study.Twenty-four consecutive patients (40 eyes) with DME were included.There were 10 males (18 eyes),14 females (22 eyes);aged from 41 to 79 years,with the mean age of (56.84±8.96) years.All the patients were type 2 diabetes,the average duration of diabetes was 8 years.BCVA was evaluated using the international Snellen E vision test chart,and then recorded as logarithm of the minimum angle of resolution (logMAR).CRT was measured by Cirrus HD-OCT4000.MAIA microperimetric parameters were evaluated,including average threshold (AT) of retinal sensitivity,macular integrity index (MI),fixating points within a circle of 1° (P1) and 2° of radius (P2),bivariate contour ellipse area (BCEA) considering 63％ and 95％ of fixating points (A63,A95),and horizontal and vertical axes of that ellipse (H63,H95,V63,V95).Pearson correlation analysis was performed to evaluate the association between these variables.The independent factor influenced the type of fixation was analyzed by multiple linear regression analysis.Results Strong correlations of logMAR BCVA with CRT (r=0.58,P=0.000),V63 (r=0.44,P=0.004),V95 (r=0.41,P=0.008),MI (r=0.36,P=0.024),AT (r=-0.61,P=0.000),P1 (r=-0.41,P=0.009),P2 (r=-0.38,P=0.015) were found.AT was correlations with P1 (r=0.53,P=0.000),P2 (r=0.51,P=0.001),A63 (r=-0.39,P=0.012),A95 (r=-0.40,P=0.012),V63 (r=-0.53,P=0.000),V95 (r=-0.46,P=0.003),MI (r=-0.50,P=0.001).There was no correlation between AT and CRT (r=-0.21,P=0.190).Forty eyes were included in this study,8 eyes (20％) had stable fixation,14 eyes (35％) had relatively unstable fixation,18 eyes (45％)had unstable fixation.Multiple linear regression analysis showed that fixation classification was independently affected by P 1.Conclusions In DME eyes,logMAR BCVA was positively correlated with CRT,negatively correlated with AT,P1 and P2.There is no correlation between AT and CRT.The fixation classification was independently affected by P 1.

Objective To study the expression of distintegrin and metalloprotease (ADAM10) in serum of lung cancer patients and its clinical significance. Methods The serum protein level of ADAM10 was assayed by ELISA in 44 lung cancer patients,34 benign lung lesions and 31 healthy subjects. The serum protein level of CEA was assayed by radioimmunoassay. Results The serum concentration of ADAM10 and CEA was higher in patients with lung cancer than benign lung lesions and normal controls(P <0.01) ,theft was distinct statistical difference be-tween lung cancer and lung benign lesions,normal controls(P <0.01) ,but no statistical difference between lung be-nign lesions and normal controls(P >0.05) ; the serum concentration of ADAM10 of lung cancer with lymph node metastasis were significantly higher than those of lung cancer without lymph node metastasis, there was statistical difference (P < 0.01) ; the serum concentration of CEA of lung cancer with lymph node metastasis were slightly higher than those of lung cancer without lymph node metastasis, but there was no difference (P > 0.05) ; the serum concentration of ADAM10 and CEA in adenocarcinoma were higher than squamous cell carcinoma and small cell lung cancer(P <0.01 ,P <0.05). The single item detection of ADAM10 or CEA to the lung cancer diagnostic sensitivity is 51.7% ,56.9% respectively,the union detection reaches 84.5% . Conclusion ADAM10 is related with occur-rence and metastasis in lung cancer. The union detection of ADAM10 and CEA can raise the lung cancer diagnostic sensitivity.

Objective: To observe the safety and efifcacy of excimer laser coronary atherectomy (ELCA) for treating the patients with coronary chronic total occlusion (CTO). Methods: A total of 3 coronary CTO patients treated by ELCA in our hospital from 2015-01 to 2016-11 were analyzed. The patients received guide steel wire gone through occlusion segment under the guidance of offside coronary angiography; drug-eluting stent implantation was conducted after ELCA treatment. The operative success rate with complication was observed and the occurrence of MACE was followed-up. Results: The device performing and interventional therapy were succeed in all 3 patients. No coronary dissection, perforation, slow relfow and thrombosis were occurred during the operation; no angina, myocardial infarction, target vessel revascularization and death were observed by follow-up study. Conclusion: Application of ELCA combining drug-eluting stent implantation was safe and effective for treating the patients with coronary CTO.
