1.Effects of Astragalus polysaccharides on 2,4,6-trinitrobenzenesuifonic acid-induced colitis in rats

Objective To investigate the effects of Astragalus polysaccarides (APS) on 2,4,6-trinitrobenzenesulfonic acid (TNBS)-induced colitis in rats. Methods Forty male Sprague-Dawley rats were randomly divided into five groups (n = 8 ): control group, TNBS group, low-dose APS group, high-dose APS group, and prednisone group. Experimental colitis was induced in rats by enema administration of TNBS. Rats in APS and sions and histological damages were determined, and the activity of myeloperoxidase (MPO) was measured in the excised colonic tissues. Cytokine levels including interleukin (IL)-4 and IL-10 were determined by enzyme-linked immunosorbent assay. Results Both macroscopic lesions and histological colonic damages induced by TNBS were reduced by low-dose APS treatment. These were accompanied by significantly attenuated colonic MPO activity (P = 0. 03) and the increase of IL-4 and IL-10 levels. The macroscopic lesions and MPO activities of high-dose APS group were higher than TNBS group, histological damage and level of IL-4 were lower, and level of IL-10 was higher, but all without statistical significance. Levels of IL-4 and IL-10 were lower than those of TNBS group, but there was no significant difference between prednisone group and TNBS group. Levels of IL-4 and IL-10 were significantly lower in prednisone group than in control group ( P = 0. 049, P = 0. 001 ). Conclusions Different doses of APS have different effects on TNBS-induced colitis. Lower dose of APS has the therapeutic potential inexperimental colitis, while higher dose of APS may aggravate the disease.
2.Modulation of dendritic cells phenotype by Astragalus polysaccharides in experimental colitis in rats

Objective To investigate the effect of Astragalus polysaccharides (APS) on 2,4,6-trinitrobenzenesulfonic acid (TNBS)-induced colitis in rats and on dendritic cells (DCs) in mesenteric lymph nodes.Methods Forty-four male Sprague-Dawley rats were randomly divided into four groups (n = 11) using simple random sampling: normal control group, TNBS group, APS group, and 5-aminosalicylic acid (5-ASA) group.Experimental colitis was induced in rats by TNBS enema in the last three groups.Rats in APS and 5-ASA groups were treated by gavage with APS (0.75 g ? kg-1 ? d-1) and 5-ASA (100 mg ? kg-1 ? d-1) on the 10 consecutive days following TNBS administration.The rats were then sacrificed and the colonic inflammatory scores of rats were measured, including the scores of disease activity index ( DAI) , macroscopic lesions, and histological damages,as well as the activity of myeloperoxidase (MPO).The expressions of major histocompatibility complex class Ⅱ(MHC Ⅱ ) and costimulatory molecule CD86 on DCs were determined by flow cytometry.Results Compared with the TNBS group, APS group had significantly decreased scores of DAI ( P = 0.007 ) , macroscopic lesions (P =0.017), and histological damages (P = 0.016).Moreover, its level of the activity of MPO dropped but without statistical significance (P =0.183).TNBS group had significantly higher expressions of MHC Ⅱand CD86 molecules on DCs than the normal control group (P = 0.005, P = 0.008), APS group (P = 0.023, P = 0.018), and 5-ASA group (P = 0.017, P=0.013).Conclusion APS may attenuate TNBS-induced colitis in rats and downregulate the activation of DCs in mesenteric lymph nodes.

Objective To explore the antigen presentation of CT26.WT via intra-peritoneal injection. Methods The intra-peritoneal injection model was made via injecting cell suspensions in mice. The spleen was isolated from BALB/c mice toco-culture with CT26.WT to detect tumor-killed ability. Phenotype identification methods and CCK8 massy were used to measure the ability of antigen presentation and stimulate T lymphocyte proliferation. IHC was used to detect the expression of B7H4 in normal and tumor tissues. Results Along with the extension of intra-peritoneal injection, the surviving number of cells was increased, contrary to the apoptosis. DC cells failed in maturation and impaired in stimulating T lymphocyte proliferation. B7H4 was higher in tumor tissues. Conclusions With the extension of intra-peritoneal injection, the mature DC cells were scared in number, resulting in the impairement of antigen-presentation. Moreover, the higher B7H4 expression in tumor tissues led to the lack of second signals which may stimulate T cells. Consequently, the ability of T cells in killing tumor cells was decreased so that they escape immunosurveillance.
4.Preparation and sintering of dental machinable zirconia/La-monazite diphase ceramic

BACKGROUND: The brittleness and low flexural strength of ceramic material hinder, to a certain extent, its application in prosthodontic dentistry. Zirconia is enhanced in its flexural strength and toughness by the transformation toughening mechanism, which makes up for the brittleness of the traditional all-ceramic material.OBJECTIVE: To preliminarily explore a new machinable zirconia ceramic material and investigate sintering properties of dental machinable zirconia/La-monazite diphase ceramics with nano-zirconia/La-monazite diphase ceramics. DESIGN: By adjusting the composition and ratio of raw materials, and by adopting different preparation and sintering method, this study was intended to measure the related parameters and to explore the best preparation and sintering method. SETTING: Department of Prosthodontics, College of Stomatology Shanghai Ninth People's Hospital Affiliated to Shanghai Jiao Tong University; Lab of Advanced Inorganic Material Technology, School of Material Science and Engineering, Shanghai University. MATERIALS: There were 3 mol yttria-containing tetragonal zirconia polycrystals (3Y-TZP) (size≤50 nm, purity 99.99%, Yixing Xinxing Zirconia-products Co., Ltd.) and La-monazite (purity 99.99%, Baotou Rare-earth Phosphate Institution). METHODS: Experiments were performed at the Department of Prosthodontics, College of Stomatology Shanghai Ninth People's Hospital Affiliated to Shanghai Jiao Tong University from January 2004 to December 2006. The pilot experiments found that the zirconia with less than 15% of lanthanum phosphate was high in strength but poor in machinability while more than 20% of lanthanum phosphate was decreased significantly in its strength. Therefore, 15%, 18% and 20% (volume percentage) of la-monazite was added to 3Y-TZP. The green bodies of the three groups were compacted by cold isostatic pressing (200 MPa) and were sintered in air atmosphere at different temperatures: 1 560 ℃, 1 580 ℃ and 1 600 ℃ to make the ceramic samples. MAIN OUTCOME MEASURES: The volume density, porous rate (Archimedes method) and three-point bending strength (EZ-100 universal testing machine) were tested of all the ceramic samples.RESULTS: ①With the increase of sintering temperature, zirconia/La-monazite ceramics with 15%, 18% and 20% lanthanum phosphate was increased in its bulk and density. The density was the highest for 1 600 ℃ and the respective density of the three groups were 5.77 g/cm3, 5.42 g/cm3 and 5.39 g/cm3. The porous rate decreased with the increasing temperature and was the lowest at 1 600 ℃ (0.88%, 1.21%, 1.49% respectively). There was no significant difference in volume and density at different temperatures (P > 0.05). ②The flexure strength of diphase ceramic with 18% and 20% lanthanum phosphate increased with the temperature increasing to 1 580 ℃. At 1 580 ℃, the flexural strength reached the highest level, respectively (772.22±43.43) MPa, (216.03±25.20) MPa and (157.21±9.79) MPa. When the temperature reaches 1 600 ℃, the strength was decreased. CONCLUSION: Zirconia/La-monazite diphase ceramics can be prepared by adopting cold isostatic pressing (200 MPa) and sintering at 1 580 ℃.
5.Screening of nutritional risk in 112 patients with inflammatory bowel disease

Objective To assess the nutritional risk of patients with inflammatory bowel disease (IBD).Methods The nutritional status of 112 IBD patients from PUMC Hospital were evaluated by Nutritional Risk Screening 2002 (NRS 2002).Using Chinese standard,a body mass index (BMI) that was lowered than 18.5 kg/m~2 according to clinical material was regarded as malnutrition,and the score was recorded as 3.The se-verity of ulcerative colitis (UC) was evaluated using True-Love criteria as mild,moderate,and severe.Crohn's disease (CD) was evaluated using Harvey-Bradshaw Index as in remission stage (≤4),moderately active stage (4-8),and severely actively stage (≥9).Results All these 112 patients,including 70 UC cases and 42 CD cases,were evaluated by NRS2002.Forty-five patients (40.2%) were judged as at the risk of malnutrition.The incidence of malnutrition was significantly higher in CD patients than in UC patients (52.4% vs.32.9%,P<0.05).It was also significantly correlated with the severity of disease.In patients with severe IBD,the rate of mal-nutrition evaluated by BMI according to clinical material was significantly lower than the rate of nutritional risk.Con-clusions The risk of malnutrition is high in IBD patients.NRS 2002 can be used for nutritional risk screening.
6.Analysis of Reaction Products of (Ethylenediamine) palladium(Ⅱ) Chloride and 5′-Deoxyguanylic Acid in Aqueous Solution by Liquid Chromatography-Inductively Coupled Plasma Mass Spectrometry and Electrospray Ionization Mass Spectrometry

A method based on HPLC-ICP-MS was established to separate the reaction products of ( ethylenediamine) palladium(Ⅱ) chloride([Pd ( en ) Cl2])and 5’-deoxyguanylic acid ( 5’-dGMP). Two reaction products were detected at pH 8. 0 with 25 mmol/L phosphate buffer solution as chromatography eluent. One was the main product with HPLC retention time of 2. 8 min, the other product’s retention time was 3.2 min. According to ESI-MS(MS/MS) study, m/z=510, 511, 512, 514, 516[M+1]+ parent ions ( abundances same to palladium isotopes) were detected. Further analysis showed that the main product was[Pd( en) ( N1-5’-dGMP) ]. However the other product was hardly to be detected by ESI-MS. By using HPLC-DAD and HPLC-ICP-MS, we found that the two reaction products had the same UV absorption spectra and palladium percentage content. Combined with other groups’research, the other reaction product was deduced as dimmer, trimer or tetramer form of[Pd( en) ( N1-5’-dGMP) ]. Further study revealed that[Pd( en) ( N1-5’-dGMP) ] was easily formed in acid solution while its polymer form was generated in alkaline solution. At pH 6. 0, [Pd(en)(N1-5’-dGMP)] was formed within 12 hours with good stability. Research also revealed that the total amount of two reaction products declined as reaction pH climbed.
7.Study on the Preparation Technology of Angelica sinensis Powder and Screening for the Optimal Particle Size

OBJECTIVE:To optimize the preparation technology of Angelica sinensis powder and conduct screening for the op-timal particle size thereof. METHODS:With the contents of volatile oil and ferulic acid as the indexes,screening was conducted for the optimal drying temperature,dry welding time and particle size of the crude drug A. sinensis,and the stabilities of the A. si-nensis powder with different particle sizes which was packed by simulating that sold in the market were investigated. The verifica-tion tests on the preparation technology were conducted. RESULTS:The optimal conditions for preparing the crude drug A. sinensis were as follows as drying temperature of 60-70℃,drying for 4.5-5 hours and being crushed into moderate-sized powder;the stabil-ity of the moderate-sized powder which was packed by simulating that sold in the market was the best. In the verification tests on the preparation technology,the average content of the volatile oil was 0.46 ml/g,with RSD of 0.034%(n=3);the average con-tent of the ferulic acid was 0.123 μg/ml,with RSD of 0.026%(n=3). CONCLUSIONS:The optimized technology is stable and feasible,and the stability of the moderate-sized A. sinensis powder is the best.
8.The short-term curative effect of anterolateral small incision total hip arthroplasty

Objective To explore the surgery way of anterolateral small incision total hip replacement and evaluate the curative effect after surgery.Methods Clinical data of 41 patients(48 hips)with anterolateral small incision total hip replacement were analyzed retrospectively.The incision length,operation time,intraoperative blood loss,postoperative volume of drainage,perioperative complications,hospitalization days,X -ray performance were recorded.Results The incision length was 7-8cm,mean (7.5 ±0.5)cm.The operation time was 60-70min,mean (65 ±5)min.The intraoperative blood loss was 165 -280mL,mean (235 ±44)mL and the postoperative volume of drainage was 85 -120mL,mean (95 ±15)mL.No perioperative complications occurred.The average follow-up time was (36 ±6)months.The preoperative hip joint Harris score was (30.3 ±28.2)points,and the last follow-up score was (98.0 ±4.0)points,the difference was statistically significant(t=15.665,P=0.000),and the excellent and good rate was 100%.Conclusion The anterolateral small incision total hip replacement has small trauma,less bleed-ing,less postoperative pain,quick recovery,better joint stability,and it is suitable for clinical promotion.
9.The facial nevi associated with hydrocephalus and intracranial anomalous venous return of 3 case reports

Objective To study the clinical manifestation of intracranial anomalous venous return associated with the facial nevi and hydrocephalus. Methods Along with reviewing of the literatures, the clinical records of 3 patients suffered from the facial nevi and hydrocephalus were analyzed. Results All of 3 patients also have sinovenous occlusion, which affect cerebral venous return. Conclusions Intracranial anomalous venous return, which is induced by extensive sinovenous occlusion, has been considered to be the main cause of hydrocephalus in the facial nevi. The formation of sinovenous occlusion and anomalous venous return may be due to developmental defect in the embryonic stage.
10.Effects of preoperative regional intra-arterial chemotherapy on lymphatic metastasis of pancreatic head carcinoma

Objective To evaluate the efficacy of preoperative regional intra-arterial chemotherapy (RIAC) in the treatment of resectable pancreatic head carcinoma. Methods The clinical data of 50 patients with resectable pancreatic head carcinoma who had been admitted to the Research Institute of Pancreatic Diseases of Fudan University from December 2006 to July 2007 were retrospectively analyzed. Patients were randomly divided into2 groups (n =25 in each group): patients in group A were treated with preoperative RIAC followed by regional pancreaticoduodenectomy, and patients in group B were treated with surgical procedure routinely. The lymphatic metastases in the 50 specimens of pancreatic head carcinoma were detected by histological examination with hematoxylin and eosin (HE) staining, and lymphatic micrometastases were detected by immunohistochemical method with staining of cytokeratin AE1/AE3 in 10 specimens with negative HE staining of the lymph nodes in each group. Results There was no significant difference in the incidence of complications, the length of hospital stay and the 1-, 2-year survival rates between the 2 groups (χ2 = 0.12, 2.88, P > 0.05). The incidence of positive lymph node metastasis in group A was 7.1% (52/734), which was significantly higher than 22.1% (118/532) in group B (χ2 = 60.01, P < 0.05). The incidence of lymphatic micrometastasis was 9.4% (30/319) in group A, and 9.1% (23/252) in group B, with no statistical difference between the 2 groups (χ2= 0.01, P > 0.05). Conclusions Preoperative RIAC is helpful in improving the prognosis of patients with resectable pancreatic head carcinoma by reducing the incidence of lymphatic metastasis and decreasing tumor stage.
